  • Biosynthesis and processing of cytoplasmic mRNA from heterogenous nuclear RNA (hn-RNA) in Aspergillus phoenicis were studied by $^{3}H$-uridine labeling and synchronous culture techniques during their life cycle. Incorporations of $^{3}H$-uridine into hn-RNA and mRNA were most rapid in vesicle-phialide fromation stage and diminished in hyphal growth stage. The processing of cytoplasmic mRNA from hn-RNA was proceeded more rapidly in hyphal growth and conidiophore formation stages than in conidia and vesicle-phialide formation stages. The specific radioactivities of hn-RNA and mRNA were very high in vesicle-phialide formation stage.

  • A survey of fungal diversity in Gyeongsang Province, Korea, revealed two previously unreported Aspergillus isolates, named KMG411 and KMG412. The phylogeny of the isolates was analyzed based on β-tubulin (BenA) and calmodulin (CaM) sequencing. Morphological analyses further identified the KMG411 and KMG412 as A. insuetus and A. nomius, respectively. Here we provide detailed morphological descriptions of the previously unrecorded Aspergillus species.

  • The qualities of Deodeok Doenjang including physicochemical characteristics and sensory properties were investigated over the following range of Deodeok levels: 5, 10, 15 and 20% (all w/w). The strains used in the manufacturing of Doenjang were Bacillus sp. B-3 and Aspergillus sp. M-9 with the highest enzyme activities like amylase and protease. In case of Deodeok Doenjangs prepared with Bacillus sp. B-3, the amino-type nitrogen of Doenjang containing 10% (w/w) Deodeok was produced more than others during fermentation for 40 days. Amino-type nitrogen produced in Deodeok Doenjang prepared with Aspergillus sp. M-9 was more than one of Deodeok Doenjangs with Bacillus sp. B-3 and a commercial Doenjang. When Deodeok content exceeded 15% (w/w), higher content resulted in lesser amino-type nitrogen production. The results showed that Deodeok had influenced growth of Bacillus sp. and Aspergillus sp. Sensory evaluation showed that Deodeok Doenjangs containing 10% (w/w) Deodeok and 1% (w/w) Bacillus sp. B-3 and containing 20% (w/w) Deodeok and 1% (w/w) Aspergillus sp. M-9 were superior to the other Doenjangs tested.

  • A total of 1,192 patients, who complained a continued chronic cough, suptum or occasional hemoptysis, in spite of successful completion of antituberculous chemotherapy or had some suspected fungal infection, were included. Serum specimens were collected from all the patients studied and sputum or other specimens collected and cultured from the most of the patients. 405(34.0%) cases of the total patients studied showed a positive precipitin reaction to the one or more of the fungal antigens on immunodiffusion tests and 303 cases of them were found to have been infected with Aspergilli, of which Aspergillus fumigatus was involved in 287 cases, followed by Aspergillus flavus(1.7%), Aspergillus nidulans(0.3%), Aspergillus niger(0.3%) and Aspergillus nidulans var. latus(0.1%). pricipitin antibodies were produced to Candida albicans(8.1%) and Pseudallerscheria boydii(0.8%). In the chest radiographs of 186 precipitin positive patients, distinct fungus ball shadows were seen in 47 cases and 45 cases of them were formed by A. fumigatus. The isolates from sputum specimens of 724 patients were aspergilli which were consisted of the 46.4% of the total fungal isolates. Identification of 137 yeast like fungi from the sputum specimens of 413 patients revealed that C. albicans(64.2%) was a commonest yeast flora.

  • Current study was conducted to identify the effects of dietary supplementation of Aspergillus oryzae ferments (AOF) cultured under normal (NAOF) or nitrogen-deficient (NMAOF) environment on feed efficiency, nutrient digestibility for broiler chicks. Fecal microbes and ammonia gas production were also determined. A total of 168 male Avian chicks, 2-wk-old, were randomly assigned into 56 cages, three chicks per cage. There were seven treatments (Control, NAOF 0.05, 0.1, 0.5%, NMAOF 0.05, 0.1, 0.5%), with 8 replicates (cages) per treatment. There was no significant difference in nutrient digestibility between two AOF groups, but the digestibility was greatly(p<0.05) improved by AOF supplementation. Total microbial account significantly (p<0.05) differed between the treatment groups with the highest number for NNAOF, followed by NAOF and control. In the case of Escherichia coli and Salmonella, the AOF supplementation significantly (p<0.05) reduced their numbers in feces, with a particular reduction in NNAOF group. Levels of ammonia gas generation were in order of control>NAOP>NNAOP. The current data implied that AOF supplementation, particularly grown under nitrogen-deficient environment, would be a feasible way to improve feed efficiency for broiler production, as well as to reduce environmental cost. However, further studies remain for industrial application.

  • The synthesis of phospholipids and the composition of fatty acids were analyzed in the fungal cells of Aspergillus phoenicis, Rhizopus acidus and Candida albicans treated with carbon sources (glucose, sucrose, raffinose) during the culture. Growth ratios were predominantly in Aspergillus and Candida treated with sucrose and those in Rhizopus treated with glucose. The synthesis of total lipids were accelerated in Aspergillus and Rhizopus with glucose and the contents of total lipid was increased in Candida with raffinose. The glucose treatment increased Phosphatidylinositol(PI) content by 73.7% for Aspergillus and Phosphatidylcholine(PC) content by 292% for Rhizopus. In sucrose treatment the synthesis of PI was accelerated 112% for Aspergillus and that of PC was increased 77.7% for Rhizopus and 71.8% for Candida. In raffinose treatment, the content of PC was increased 79.5% for Aspergillus, the biosynthesis of Phosphatidylethanolamine(PE) was inhanced 50.9% for Rhizopus and 49.1% for Candida. In Aspergillus and Candida, oleic acid and linoleic acid were major fatty acids for biosynthesis of PC and PE when the three carbon sources were treated. The major fatty acids for biosynthesis of phospholipid was palmitic acid and linoleic acid in Aspergillus, palmitic acid in Rhizopus and oleic acid in Candida. Palmitic acid was one of major fatty acids utilized for the biosynthesis of phospholipid(PC, PE, PI) in the fungal cells treated with carbohydrates.
