Analyses on Environment-friendliness of Root Barrier Materials based on Fish Toxicity Test (어독성 실험에 따른 방근재 친환경 특성 분석)

  • The purpose of this study is to analyze the characteristics of environment-friendliness of Root Barrier materials based on comprehensive experiments on harmfulness of Root Barrier materials and fish toxicity of Oryzias latipes mortality to verify eco-toxicity of each method of construction and Root Barrier material, which are to be applied by taking eco-toxicity into account when building ecological flows in upper areas on natural and artificial grounds. As a result, the following conclusions have been reached in this study: 1. In regard of the harmfulness analyzed, each material showed a different result of analytical value in each lab tank. Compared to lab tank, pH (la), DO (lb), T-N (VI) and T-coli (la) were in the same grade, but COD, SS, T-P and F-coli were less than that of control, respectively increased or decreased by material were analyzed. 2. In the experiment of fish toxicity, Barrier sheet was found to have 66.7% of fish mortality, indicating strong fish toxicity. Synthetic rubber system root barrier sheet (20.0%) was analyzed to have medium fish toxicity, while Synthetic resin system root barrier-waterproof sheet (3.3%), Synthetic rubber system membrane root barrier sheet (3.3%) and Synthetic resin system root barrier sheet (0.0%) showed relatively lower mortality and fish toxicity. To sum up such results as found in the experiment mentioned so far, the values of harmfulness and root penetration resistance analyzed were different in each lab tank, but there was absolutely little correlation with the mortality gained from the experiment on fish toxicity. In the experiment of fish toxicity, environment-friendly root barrier materials were analyzed, and it was found that Synthetic resin system root barrier sheet, Synthetic resin system root barrier waterproof sheet and Synthetic rubber system membrane root barrier sheet are highly environment-friendly. In contrast, Synthetic rubber system root barrier sheet was found to have medium-level environment-friendliness. Also, Barrier sheet was analyzed to have low environment-friendliness.

Characteristics of Mycelial Growth and Enzyme Activities of Mattirolomyces terfezioides Collected from Robinia pseudoacacia Forest in Korea (국내 아까시나무 숲에서 수집한 감자덩이버섯(Mattirolomyces terfezioides)의 균사 생장 특성과 효소 활성)

  • Mattirolomyces terfezioides is a type of sweet truffles that predominantly form ectomycorrhizae with Robinia pseudoacacia. It is also worthy of artificial cultivation. This is the first report on characteristics of mycelial growth and enzyme activities of M. terfezioides collected from R. pseudoacacia forest in Korea. M. terfezioides showed the highest mycelial growth when cultured on potato dextrose agar (PDA) at $30^{\circ}C$ or in modified Melin-Norkran's liquid medium (pH 8.0). The biomass of M. terfezioides was higher in liquid medium containing nitrate-nitrogen than ammonium-nitrogen by 1.8 fold. The mycelia of M. terfezioides showed both carboxymethylcellulase and laccase activities on solid media for enzyme screening.

A comparative study on the relationship of investing medium to vertical occusal change and surface smoothing during denture processing (Resin processing시(時) 매몰재(埋沒材)에 따른 교합고경(咬合高涇) 및 의치표면(義齒表面) 활택도(滑澤度)의 변화(變化)에 관(關)한 연구(硏究))

  • A comparative study was conducted to evaluate the relationship of investing medium to the amount of vertical occlusal changes and to the differences of surface smoothness during denture construction. Three groups of 20 dentures, 30 sets of upper and lower were fabricated of conventional heatcuring acrylic denture base resin, using silicone-gypsum molding techniques, with or without covering the occlusal surfaces of the teeth by artificial stone and all-gypsum molding techniques. The distance between the two reference points indented by 1/2 round bur on the upper and lower frontal surfaces of each articulator were measured and recorded before processing and again after processing and remounting of each denture on the articulator. The differences between the two recordings indicated the amount of vertical opening during denture processing. The difference of surface smoothness were investigated and determined by 3 observers continual comparing of the two randomly selected dentures with each other, which were seperately selected as pairs from the different two groups of 20. The results obtained were as follows: 1. During resin processing no statistically significant differences of the amount of vertical occlusal changes were detected between any of the two groups of two silicone-gypsum and one allgypsum molding techniques, although the amount of vertical opening was somewhat increased when silicone-gypsum molding technique was used. 2. Surface smoothness of the processed denture was makedly by increased when silicone-gypsum molding technique was used.

Horticultural Activity Interventions and Outcomes: A Review

  • The objective of the present study was to review the current research about the outcomes of horticultural activity interventions in order to determine research trends and analyze the characteristics of horticultural activity interventions. We reviewed the literature using search engines such as the Web of Science, Proquest Dissertation and Theses, Academic Search Premier, Research Information sharing Service, and Korean National Assembly Digital Library to locate journal articles that include relevant search terms (e.g., gardening activity, gardening program, allotment garden, urban agriculture, horticultural therapy, and horticultural activity). We initially identified 14,414 journal articles that were published before April 2014 and selected 509 of the papers for review. We found that studies investigating treating children and adults 8 to 64 years old were the most common, and horticultural activities such as gardening outdoors, planting indoors, making crafts with live plants, arranging flowers, making crafts with artificial or pressed flowers, and other activities were usually combined. Short/medium term (11 to 20 sessions) horticultural activity programs were the most frequent, and most interventions were of medium duration (> 60 min to 120 min). Most of the studies focused on the psychological or emotional effects of horticultural intervention, such as its effects on emotional intelligence, self-esteem, stress, and depression. Further studies are needed to analyze the research methodology, specific outcomes, and strengths or weaknesses of studies investigating horticultural activity interventions.

Bacteriological Control of Cyanobacterial Bloom. (시안세균 수화의 세균학적 조절)

  • A Gram (-), rod-shaped bacterium in size of 1.3∼$1.8{\times}0.35{\mu}m$ inhibiting the growth of cyanobacterium (Ana-baena cylindrica) was isolated and designated NG-2 in this manuscript. This isolate showed positive reactions for catalase and oxidase, and optimal growth conditions of 35∼TEX>$40<^{\circ}C$ and pH 9.0. In a mixed-culture of A. cylindrica and the isolate, each microorganism grew inverse-proportionally, and the cyanobacterial vegetative cells almost completely disappeared within 24 hours. NG-2 lysed A. cylindrica only under light, which means that lytic activity of NG-2 was dependent on the photosynthetic activity of host. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas het-erocysts have not been lysed. When cyanobacterial cell walls have been lysed partly, NG-2 attatched around A. cylindrica filament and formed colony, then encouraged complete lysis of cyanobacterial cells. The isolate showed similar lytic activity in natural water as in an artificial medium. And lytic activity of NG-2 was enhanced when attached on expandable polystyrene bead.

Development of bag culture medium of Auricularia auricula (목이버섯 봉지재배 배지 개발)

  • Studies were made to optimize the media composition in bag culture and conducted to determine the possibility of artificial cultivation of Auricularia auricula. Sawdust spawn of media composition for optimal growth were found to be oak-sawdust 80%combination of 20% popla-sawdust were the best of the optimal combination. And optimal substrate combination were found to be cotton-seed meal combination of 10% wheat bran 5% mixed were the best of combination. The duration of spawn run period and primordial formation period on bag(1.2 kg) were 50 days and 7 days, respectively. The weight of fruiting body and the yield(100ea, 1.2 kg) of fresh fruit-body were 24 g and 45,000 g, respectively.

Contraction Behavior of Collagen Gel and Fibroblats Activity in Dermal Equivalent Model

  • We developed a dermal equivalent (DE) which was engineered using human dermal fibroblasts and a matrix of collagen gel. The in vitro construction of the DE was accomplished by casting a porcine collagen type I solution plus concentrated medium with isolated and cultured fibroblasts. These constructs were attached to culture dishes or left floating in culture medium. Contraction of attached gels results in decreased gel thickness without a change in gel diameter, and contraction of floating gels results in decreased gel thickness and diameter. After contraction, there was no increase in cell number in floating gels, but cells in attached gels began to increase after about 4 days of the lag phase in cell growth curve. At this lag phase, addition of fibroblast growth factor (FGF) at a concentration of $0.1{\mu}$/ml promoted cell proliferation in the attached collagen gels, but no effect in floating gels. These results indicate that the method of contraction had an influence on the extracellular matrix (ECM) organization, and this influenced not only cell growth but also fibroblast responsiveness to FGF. This suggests that attached collagen gel is more suitable as a dermal equivalent than the floating gel. And the final contracted area of attached gel is much larger than that of the floating gel since floating gel is contracted in all directions but attached gel is contracted only vertically.

VSV-G Viral Envelope Glycoprotein Prepared from Pichia pastoris Enhances Transfection of DNA into Animal Cells

  • Vesicular stomatitis virus G glycoprotein (VSV-G) has been widely used for pseudotyping retroviral, lentiviral, and artificial viral vectors. The objective of this study was to establish a potential approach for large-scale production of VSV-G. To this end, VSV-G was cloned with an N-terminal His-tag into Pichia pastoris expression vector pPIC3.5K. Three clones ($Mut^s$) containing the VSV-G expression cassette were identified by PCR. All clones proliferated normally in expansion medium, whereas the proliferation was reduced significantly under induction conditions. VSV-G protein was detected in cell lysates by western blot analysis, and the highest expression level was observed at 96 h post induction. VSV-G could also be obtained from the condition medium of yeast protoplasts. Furthermore, VSV-G could be incorporated into Ad293 cells and was able to induce cell fusion, leading to the transfer of cytoplasmic protein. Finally, VSV-G-mediated DNA transfection was assayed by flow cytometry and luciferase measurement. Incubation of VSV-G lysate with the pGL3-control DNA complex increased the luciferase activity in Ad293 and HeLa cells by about 3-fold. Likewise, incubation of VSV-G lysate with the pCMV-DsRed DNA complex improved the transfection efficiency into Ad293 by 10% and into HeLa cells by about 1-fold. In conclusion, these results demonstrate that VSV-G could be produced from P. pastoris with biofunctionalities, demonstrating that large-scale production of the viral glycoprotein is feasible.

The Artificial Cultivation of Pleurotus eryngii (I) -Investigation of Mycelial Growth Conditions- (Pleurotus eryngii(큰느타리버섯) 균의 인공재배(I) -균사배양 조건에 관하여-)

  • A study was conducted to obtain optimal mycelial growth conditions of Pleurotus eryngii. Mycelial growth was best on medium Lilly, temperature $25{\sim}30^{\circ}C$, and pH 6.0. Optimal carbon sources were $3{\sim}4%$ glucose and 5% dextrin. Casamino acid 0.12% in medium was good for mycelial growth as a nitrogen source.

Study on the Nutrient Solution Content and Growth of Cherry Tomato in Scoria Culture (제주 송이를 이용한 방울토마토 양액재배시 양액성분 및 생육에 관한 연구)

  • The main purpose of these studies were to clarify differences in Cheju - scoria and other solid media on quantity and quality of cherry tomatoes and on shift of component of the nutrient solution, and to use Practically Cheju-scoria as an excellent solid culture medium. The results obtained were summarized as follows ; 1. Among scoria plots, the rates of dry weights, fruits and their sugar-acid ratio were higher in the plot that was drained well with deep flow for one hour once a day. 2. Fresh fruit weights were lighter in rockwool and deep flow technique, but larger in scoria, Hyugashi (artificial gravel, $\Phi$10-12mm) and perlite in moving to higher flower cluster. 3. The results of analysis on microelement among solution components showed decrease of concentrations of P and K in the period of growth and development. 4. Yields and brix of cherry tomato showed a tendency to increase in rockwool and Hyugashi than anothers. 5. The concentration of fertilizer base was increased in general solution culture. Transpiration and absorption were similar in scoria plot and other media. 6. More studies of the Cheju-scoria development is required in order to use it as a solid medium for solution culture.
