Influence of Culture Comditions on Acid Tolerance of Lactobacillus casei YIT 9018 (배양조건에 따른 Lactobacillus casei YIT 9018의 니산성 변화)

  • We studied the influence of culture conditions on the acid tolerance of Lactobacillus casei YIT 9018 in artificial gastric juice with respect to relative amount of membrane bound ATPase and their biochemical characteristics. With raising incubation temperature from 30.5$\circ$C to 40.5$\circ$C and lengthening incubation time from exponential phase to late stationary phase, the acid tolerance of L. casei YIT 9018 was increased. As acid tolerance enhanced, C$_{18:1}$ content of membrane fatty acid was reduced and C$_{19:0 cyclo}$ was enriched but the others were not changed greatly. At high ATPase activity, proton permeability was relatively low but this phenomenon did not correspond to acid tolerance. In conclusion, it was considered that changes of C$_{18:1}$ and C$_{19:0 cyclo}$ were closely related to the acid tolerance of L. casei YIT 9018.

  • The stability of ftorafur in artificial gastric juice and its major decomposition product were studied quantitatively by the combination of TLC technique and UV-recording spectrophotometry. The decomposition proceeded in first-order reaction. The decomposition rate was accelerated by the increase of temperature. The major decomposition products were identified as 5-fluorouracil. The apparent activation energy for the decomposition was found to he approximately 24.3kcal/mole from Arrhenius plot.

Characterization of Bacillus polyfermenticus KJS-2 as a Probiotic

  • The identification and characterization of Bacillus polyfermenticus KJS-2 (B. polyfermenticus KJS-2) was conducted using TEM, an API 50CHB kit, 16S rDNA sequencing, a phylogenetic tree, and catalase and oxidase testing. The conversion rate of glucose to lactic acid by B. polyfermenticus KJS-2 was found to be $60.7{\pm}4.9%$. In addition, treatment of B. polyfermenticus KJS-2 with artificial gastric juice (pH 2.0) and bile acid (pH 6.5) for 4 h resulted in a final viability of $140{\pm}7.9%$ and $108{\pm}3.5%$, respectively. Finally, the results of adhesion experiments using Caco-2 cells revealed that the adherence of B. polyfermenticus KJS-2 to Caco-2 cells was approximately $65{\pm}0.6%$.

  • For the isolation of probiotics which may be used for the human beings and animals, we have screened the microorganisms from chicken intestines which have acid and bile tolerance and the growth inhibition of pathogenic E. coli and Salmonella typhimurium. Among them, a strain which was identified as Lactobacillus salivarius had around 66% of survival after 2h incubation in the artificial gastric juice and 9% of survival after 24h incubation in the presence of 0.3% bile salts, and showed complete inhibition against both path ogenic E. coli and Salmonella typhimurium after 24 h coincubation. Its storage stability after lyophilization could be improved by adding polyvinylpyrrolidone.

  • In order to compare the iron concentrations of the marketed ferruginous hematinics with the sustained-releasedosage forms and to prepare the good hemantinic which can be absorbed effectively without forming "iron block" in the gastrointestinal tract, gelatinized micropellets containing medicament were prepared and hardened in 10% formalin-isopropanol in the time of 72 hours. These micropellets were digested with artificial gastric juice and plotted a curve of released iron concentration. As a result, it is found that gelatinized micropellets sustained-release dosage form is an excellent ferruginous hemantinic and it is shown that several marketed preparations can be improved in their hemantinic actions by forming the sustained-release dosage.se dosage.

Isolation and Identification of Lactic Acid Bacteria Isolated from a Traditional Jeotgal Product in Korea

  • Seventeen lactic acid bacterial strains (LAB) were isolated using MRS agar medium from Jeotgal, a Korean fermented food, purchased at the Jukdo market of Pohang. To identify the strains isolated, they were tested by examining their cell morphologies, gram-staining, catalase activity, arginine hydrolase activity, D-L lactate form and carbohydrate fermentation. According to the phenotypic characteristics, three strains were tentatively identified as Lactobacillus spp., ten were Enterococcus spp. (or Streptococcus spp., or Pediococcus spp.) and the rest were Leuconostoc spp. (or Weissella spp.). Five strains among 17 were chosen by preliminary bacteriocin activity test. Four bacterial strains which inhibited both indicator microorganisms were identified by 16S rRNA sequencing. The results are as follows; Leuconostoc mesenteroides (HK 4), Leuconostoc mesenteroides (HK 5), Leuconostoc mesenteroides(HK 11), Streptococcus salivarius(HK 8). In order to check LAB which are showing a high survival rate in gut, we investigated three strains inhibiting both indicator microorganisms in artificial gastric acid and bile juice -all except HK8. The three strains mentioned above grew in extreme low acid conditions.

Bioavailability of Commercially Available Norfloxncin Tablets (시판 노르플록사신 정계의 생체내 이용률)

  • This study was attempted to investigate the dissolution rate and the bioavailability after oral administration of commercially available norfloxacin tablets in rabbits. The dissolution test was conducted in artificial gastric juice using basket method with for norfloxacin preparations (A, B, C and D) which were chemically equivalent. The results were as follows ; The dissolution rate was increased in the order of four different brand A>D>B>C. Area under the plasma concentration curve and peak plasma concentration were increased in the order of brand A>D>B>C. Absorption rate constant and peak time were increased in the order of brand B>A>C>D, and there was a little difference in elimination rate constant and biological half-life. The correlation of the dissolution rate and relative bioavailability showed significant linear relationship. From the results of this experiment, the bioavailability of norfloxacin tablets in rabbits may be predicted from the results of dissolution rate studies.

  • Polymorphs of piperacillin monohydrate were prepared by various sedimentation methods from piperacillin base for the purpose of developing piperacillin preparations for oral use. Solubility and dissolution rate of each form of the polymorphs were compared with each other. It was found that Form IV showed the highest solubility and the fastest dissolution rate among four forms of polymorphs. In general, the solubility and dissolution rate of two amorphous forms, Form IV and II were higher than those of two crystal forms, Form I and III. The apparent dissolution rates in the artificial gastric juice within 60 minutes were $0.65{\times}10^{-6}mole{\cdot}min^{-1}\;cm^{-2}$ for From IV, $0.36{\times}10^{-6}mole{\cdot}min^{-1}{\cdot}cm^{-2}$ for From II, $0.30{\times}10^{-6}mole{\cdot}min^{-1}{\cdot}cm^{-2}$ for From III and $0.18{\times}10^{-6}mole{\cdot}min^{-1}{\cdot}cm^{-2}$ for From I, respectively.

  • Sustained-release dosage form of ferruginous hematinics which can be absorbed effectively without forming "iron block" that caused by high concentration of Iron in the gastrointestinal tract, was studied. Gelatinized micropellets containing medicament were prepared according to the method of N. Tanaka et al. and hardened in 10% formalin-isopropanol in various time. Gelatinized micropellets were digested with artificial gastric juice and prepared a graph of iron concentration determined by Hong's method. As a result, it is assumed that gelatinized sustained-release dosage form can be a good ferruginous hematinics and it is shown that the preparation of 72-hour-hardening in 10% formalin-isopropanol has a suitable sustained release.d release.

  • To investigate the possible usage as probiotics, we isolated and identified one strain of bacillus from Korean traditional food, Jeotgal. The isolate was identified as Bacillus megaterium KS12 by examining its biochemical properties and 16S rRNA sequence analysis (99% similarity). B. megaterium KS12 exhibited high viability in artificial gastric juice for 3 h and in artificial bile salts for 24 h, about 14.4 and 14%, respectively. The herbal digestive medicine containing the isolate showed high starch hydrolytic activity and proteolytic activities (about 6-fold) compared to traditional herbal digestive medicine. The viable cells in the herbal digestive medicine containing the isolate were higher than those of traditional herbal digestive medicine.
