Xylitol is a 5-carbons carbohydrate, which can be replaced with sucrose for preventing caries. The replication of Streptococcus mutans and its formation of artificial plaque were studied in the media containing xylitol. The combined effect of xylitol and other carbohydrates on Streptococcus mutans was also studied. The replication of Streptococcus mutans was inhibited according to the increased concentration of xylitol. Streptococcus mutans replicated at the initial stage of incubation in the media containing glucose, fructose or lactose, while replicating from the beginning of incubation in the media containing fructose as combining with xylitol. The formation of artificial plaque by Streptococcus mutans was significantly reduced in the media containing with xylitol and fructose. These results indicated that the replication of Streptococcus mutans was inhibited according to the increased concentration of xylitol, and the formation of artificial plaque by Streptococcus mutans was significantly inhibited in the media containing xylitol and fructose.

Dental caries is a bacterial disease of the dental hard tisssus, characterized by a localized, progressive, molecular disintegration of tooth structure. The action of Leuconostoc lactis 51 about plaque formation and replication by Streptococcus mutans was studied as follows. 1. Lower amount of plaque was produced at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone on the wires in the beaker. 2. Fewer cells of S. mutans were replicated at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone. 3. In M17Y broth, viable cells of S. mutans and L. lactis 51 increased for 12 hours, and decreased for 24 hours. In M17YS broth, viable cells of S. mutans showed time-dependent decrease at mixed culture of S. mutans and L. lactis 51. 4. The culture supernatant of L. lactis 51 didn't inhibit the replication of S. mutans and the formation of artificial plaque. 5. Sucrose and frutose were extracted from the culture supernatant of L. lactis 51 in M17YS broth. These results suggest that L. lactis 51 isolated from the oral cavity inhibits the replication of S. mutans and the formation of artificial plaque.

Objective: The authors have experimented for the artificial plaque removal effect of several kinds of 360 degree rotating head typed tooth-brushes with sonic vibratory actioned by using of automatic machine for horizontal scrub method in order to find the better toothbrush type for plaque removal. Methods: The experiment was conducted on three medium to 360 degree rotating head toothbrushes, a medium sized toothbrush and a medium sized toothbrush, and a flat toothbrush consisting of 30 ordinary toothbrushes. A brushing machine with horizontal scrubbing was manufactured and had variations of the end of the bristle attached to or near the surface of the teeth, a vibrating wave action force of 16,000 or 18,000 cycles per minute, and a working time of 2 or 3 minutes. The tooth removal effect was confirmed by scanning and analyzing images with a computer program after automatic brushing with the machine. The elimination rate results for each group were analyzed using the independent t-test and one-way ANOVA test. Results: It revealed the most in removal effect for the artificial plaque in such conditions as action at near the tooth surface with 18,000 cycle for 3 minutes in case of using A, B, and C tooth-brush. And it has more removal effect rate than for using the plane tooth-brush (p<0.05). Conclusion: It was recommended to develop the 360 degree rotating head and vibratory toothbrush focusing to use near the tooth surface with 18,000 cycles of vibration for 3 minutes at one site of the teeth area.

Due to the attractive benefits with regard to bone health, digestion, and hydration, carbonated water consumption have rapidly grown over the past few years. However, the acidic drink has latent potential for enamel erosion. The most experimental studies about the enamel erosion have focused on the carbonated beverages with sugar and artificial sweeteners. Here, we determined the enamel erosion potential by commercially available carbonated waters with bovine teeth. The erosion was verified by pH value, calcium concentration, and scanning electron microscope. Then plaque accumulation by bacterial adhesion was determined on the enamel erosion surface to measure roughness. In the present study, we observed that the increased calcium content after being immersed in carbonated waters result from the overall enamel erosion. There were no significant differences between general carbonated waters and mineral waters for erosive capacity. Therefore, commercially available carbonated waters are potentially erosive. In addition, oral bacteria strongly adhered to the erosive enamel surfaces thereby facilitating the development of dental plaque. Thus, it is urgently necessary to provide food safety information on the carbonated water as acidic drink to prevent the enamel erosion.
Objectives : This study was carried out to investigate the caries resistant effect of fluoride by dental polishing prior to fluoride application in vitro. Methods : Artificial caries lesion was made on the surface of specimen enamel taken from cow's permanent cuspid on the part of labial surface after resin embedding and polishing. Artificial dental plaque was formed on the 72 dental specimen 25~45 VHN(Vickers Hardness Number) which were divided into three groups(fluoride varnish, APF gel, control) with dental polishing and without polishing respectively. Fluoride varnish and APF gel group with 20 second polishing or without polishing were immersed in the artificial saliva respectively. Control group with or without polishing were immersed in the artificial saliva. Results : 1. Significant difference was not found by fluoride varnish between polishing group and non polishing group (p>0.01). 1) polishing group. The changes of Vickers Hardness Number(VHN) were $14.49{\pm}13.73$. 2) non-polishing group. The changes of VHN were $11.67{\pm}5.39$. 2. Significant difference was not found by APF gel between polishing group and non polishing group (p>0.01). 1) polishing group. The changes of VHN were $8.48{\pm}8.37$. 2) non-polishing group. The changes of VHN were $5.32{\pm}2.59$. Conclusions : Showed no significant difference between polishing group and non-polishing group regardless of fluoride materials (fluoride varnish, APF gel).

Chlorhexidine dihydrochloride and sodium fluoride have been used as agents inhibiting the replication of oral bacteria and the formation of dental plaque. There are various kinds of bacteria with different sensitivity against these agents. In this study, chlorhexidine dihydrochloride and sodium fluoride were studied about their effects on the replication of Streptococcus mutars, Streptococcus sobrinus, Streptococcus oralis and Streptococcus salivarius at their concentrations inhibiting the formation of artificial plaque by Streptococcus mutars. When Streptococcus mutans was cultured for 8 hours in the media added with $1.0{\mu}M$ chlorhexidine dihydrochloride, the weight of formed artificial plaque was decreased to $5.1{\pm}1.5mg$ compared with $106.1{\pm}18.1mg$ of the control(p<0.05). At the same time, the optical density of cultured media was decreased. The optical density of cultured media was slightly decreased in Streptococcus sobrinus, but was not decreased in Streptococcus oralis. The optical density of Streptococcus salivarius was decreased at 8 hours-incubation, was not decreased at 24 hours-incubation. When Streptococcus mutars was cultured for 8 hours in the media added with 3.0 mM sodium fluoride, the weight of formed artificial plaque was decreased to $26.7{\pm}8.3mg(p<0.05)$. At the same time, the optical density of cultured media was decreased. The optical density of cultured media was decreased in Streptococcus sobrinus and Streptococcus oralis, but was slightly decreased in Streptococcus salivarius. These results suggest that at the concentration of chlorhexidine dihydrochloride and sodium fluoridehe inhibiting the formation of artificial plaque by Streptococcus mutans, different species of genus Streptococcus show the different sensitivity against these agents.
The study was to expand the choice of electric toothbrush by number of operations of the plaque removal effect on the tooth surface. Three different toothbrushes were selected for oscillations and pulsations type of electric toothbrush. A is Oral-B D12 (7,600 pulsations), B is Oral-B D16 (20,000 oscillations and a 7,600 pulsations), C is Oral-B D20 (40,000 oscillations and an 8,800 pulsations). In each trial, each tooth was brushed for 2 seconds. For the 10 trials. And each tooth was coated with 1layer, 2layers and 3layers. After the experiment, the plaque removal effect was calculated and analzed using the Image J program(p<0.05). According to the study, C toothbrush had the highest rate of plaque removal effect. Based on this results of the study, dental worker will need to evaluate oral hygiene and recommend appropriate toothbrushes according to oral conditions.

This study was performed to evaluate the effect of Streptococcus oralis on the formation of artificial plaque and the replication of Streptococcus mutans. S. mutans was incubated alone and in the combination with S. oralis in the beaker with wires. The produced plaque weight and the viable cells of S. mutans were compared between those cultures. Various factors were studied about the effect on the formation of plaque and the replication of S. mutans. Followings are the results. 1. Lower amount of plaque was produced and fewer cells of S. mutans were replicated at the mixed culture of S. mmutans and S. oralis than S. mutans alone. 2. When 10 mM glucose was added, the plaque weight was increased in the culture of S. mutans alone. But in the mixed culture of S. mutans and S. oralis, the plaque weight was not increased when 10 mM of glucose was added. 3. When 10 mM fructose was added, the plaque weight was increased in the culture of S. mutans alone or combined S. mutans and S. oralis. 4. In the mixed culture of S. mutans and S. oralis with different concentration, the more S. oralis exist, the less plaque and the fewer viable cells of S. mutans were observed. 5. The plaque weight and the viable cells of S. mutans were more decreased in the mixed culture of S. mutans and S. oralis than S. mutans alone after 12 hours. 6. When Staphylococcus epidermidis consuming hydrogen peroxide was added to the mixed culture of S. mutans and S. oralis, the plaque weight and the viable cells of S. mutans were increased. These results indicate that S. oralis inhibited the formation of plaque and the replication of S. mutans, and this may result from the formation of hydrogen peroxide S. oralis.

There are normal inhabitants doing medically useful functions in the body. There are many kinds of bacteria performing specific functions in the oral cavity. Two strains of lactic acid bacteria were isolated from inhabitants of caries-free children's oral cavity, which inhibited the formation of artificial plaque by Streptococcus mutans and the production of volatile sulfur compounds by anaerobic bacteria. The isolates were identified by the test using API 50 CHL medium kit and 16S rDNA partial sequencing. 1. Two isolates were Gram-positive bacilli and produced hydrogen peroxide. 2. When Streptococcus mutans was cultured in the media, the mean weight of formed artificial plaque on the orthodontic wires was $124.4{\pm}30.4\;mg$, whereas being reduced to $5.2{\pm}2.0mg$ and $10.6{\pm}6.6mg$ in the media cultured with Streptococcus mutans and each isolate, respectively (p<0.05) 3. The number of viable cells of Streptococcus mutans was $3.4{\times}10^9$ per ml in the cultured solution, whereas those of Streptococcus mutans in the combined culture with each of isolates were $4.6{\times}10^8\;and\;2.4{\times}10^8$ per ml. 4. The optical density was 1.286 in the supernatant of Fusobacterium nucleatum after vortexing for 30minutes, whereas in the supernatant of combined Fusobacterium nucleatum and each isolate, they were reduced to 0.628 and 0.497, which the percentages of coaggregation between them were 29.4% and 57.8%, respectively 5. The optical density of Fusobacterium nucleatum precipitate was 1.794 in the culture media containing cysteine and $FeSO_4$, being reduced to 1.144 and 0.915 in the coaggregated precipitates of Fusobacterium nucleatum and each isolate. The optical density of Porphyromonas gingivalis precipitate was 1.932 in the culture media, being reduced to 1.170 and 1.266 in the coaggregated precipitates of Porphyromonas gingivalis and each isolate. 6. When two isolates were tested with API 50 CHL medium kit, those were identified Lactobaciallius salivarius and Lactobaciallius delbrueckii subsp. lactis. 7. The similarity values of 16S rDNA sequence between each of isolates and Lactobaciallius salivarius subsp. salicinius were 99.60% and 99.73%, respectively, meaning that isolates were Lactobaciallius salivarius subsp. salicinius. These results indicated that two strains isolated from caries-free children's saliva, which inhibited the formation of artificial plaque and the production of volatile sulfur compounds, were identified as Lactobaciallius salivarius subsp. salicinius.

The main component of dental plaque is the mutan containing the a-1,3 bond. The following results were obtained by using a blue mutan to assess the factors affecting the mutan-digesting activity of Streptomyces exfoliatus isolated from soil. A clear zone was produced by mutanase-producing Streptomyces exfoliatus on the minimal essential agar containing blue mutan. Streptomyces exfoliatus digested more blue mutan in the minimal essential broth at pH 7.0 than at pH 5.5 or 8.5. Streptomyces exfoliatus digested more blue mutan at $37^{\circ}C$ than at $32^{\circ}C$ or $42^{\circ}C$ (P<0.05). When the concentration of $CaCl_2$ was increased in the minimal essential broth, the digestion of blue mutan was increased (P<0.05). The optimal concentration of KCl was 10mM to digest blue mutan, but a similar amount of blue mutan was digested at the range of 0.1mM to 6.4mM of $MgCl_2$. When the culture supernatant of Streptomyces exfoliatus was mixed with 2X brain heart infusion broth containing 0.5% yeast extract and 10% sucrose, less artificial plaque was formed by Streptococcus mutans on the orthodontic wire (P<0.05). These results indicated that the secretion of mutanase was identified in culture supernatant of mutan-digesting Streptomyces exfoliatus, suppressing the formation of artificial plaque by Streptococcus mutans.
