Effect of Three Microalgal Species on Gonadal Development and Sex Maturation of Ark Shell, Scapharca broughtonii (미세조류 3종에 따른 피조개 Scapharca broughtonii의 생식소 발달과 성숙)

  • Even though ark shell, Scapharca broughtonii is commercially important species, its artificial seed production is not yet fully developed. This study was carried out to understand the effect of three microalgal species on gonadal development and sex maturation of S. broughtonii in terms of broodstock management and food organism. Isochrysis galbana, Phaeodactylum tricornutum and Tetraselmis tetrathele were supplied to S. broughtonii broodstock in single or mixed. And condition index, gonadal development, sexual maturation and survival of the broodstock were analysed. After 45 rearing days, frequency of ripe stage of gonadal phases, rate of induced sexual maturation and survival of S. broughtonii broodstock fed mixed diet with 3 microalgal species or single diet of T. tetrathele were the highest.

Utilizability of Shell Powder as Wall Coatings for Thin Textured Finishes (건축용 벽 바름재로서 패각분말의 활용성 연구)

  • 0.4 Million tons of shell powder have been disused as waste in KOREA and caused severe environmental pollution though shell powder can be utilized in real life for many ways. It is impending problem to recycle shell powder as it requires high expense for burying and temporary outside heap and causes severe environmental pollution being a main factor of ocean waste. To suggest the basic data for development of eco-friendly and high-function Wall Coatings Thin Textured Finishes, a wall coating sample was applied to indoor walls of a mock-up and temperature and humidity were measured to assess the thermal performance of it, and a survey of preference for the color sense and feel of the materials with a movie of specimens. The results of the study are following; 1) High insulation performance is shown from the assessment result of the room polystyrene board adhered on the walls then high humidity controlling performance is shown from that of the room polystyrene board coated by shell powder. This point out that shell powder has superiority for humidity controlling. 2) The result of thermal and humidity assessment shows that shell powder makes up for thermal conduction of the polystyrene board and same result can be expected from the assessment with materials which has similar thermal characteristics with polystyrene.3) Ranking of preferred specimens is; 1st Case 13, 2nd Case 17, 3rd Case 16, and 4th Case 12. Preferred shell powder was the ark shell. Preferred powder for plaster was the powder mixed with that sifted by 0.8mm, 100mesh and 40mesh, and for spray was the fine powder mixed with that sifted by 100mesh and 40mesh.

Culture of the Ark Shell, Anadara broughtonii in Yoja Bay (여자만에서의 피조개 양성)

  • A culture experiment of the ark shell, Anadara broughtonii, one of the edible clams, was carried out in Yoja Bay located at the south ern coast of Korea to find the state of growth and any problems which influence them. Seed shells at an average length of 2.5cm and a weight of 2.5g were planted on bottom mud with a density 20 to 40 shells per square meter on March 10 and 11, 1982. Shell length, shell height, shell breadth, meat weight (wet weight), and total weight for six months from August 1982 to February 1983 were measured to find their state of growth. At the same time, the survival rate was investigated during the whole culture period through December 1983 when they were harvested. Growth, on the whole, was fast when water temperature was high. For a year, an average shell length and total weight attained to 6.1 cm and 63.0g respectively and meat weight was gained to 19.5g. It showed that the state of growth was favourable. Shell shape became round from the oval ana meat weight increase was faster than shell growth as time goes by regardless of water temperature while both shells and total weight increased faster during high temperature. A month after seed shells were planted, about $30\%$ of them was dead and a high mortality, say $5\%\;to\;8\%$ per month, was kept till September. It dropped to around $2\%$ aften divers eliminated starfish, Asterias spp., in August. Thereafter, the rate maintained through December. In all, the final survival rate was about $10\%$ at the time of harvest in December 1983. It was caused mainly by starfish predation. Therefore, a best way for satisifactory culture is seemed to find a way to get rid of the starfish.

Studies on the Lipid and Fatty Acid Compositions of Ark-Shell, Anadara broughtonii (피조개 지질 및 지방산 조성에 관한 연구)

  • The present study was designed to analyze the lipid and fatty acid compositions of ark shell, Anadara broughtonii. The crude lipid was extracted by Bligh and Dyer's method, and then fractionated by TLC and quantitatively analyzed by TLC scanner. Lipid extracted from ark shell was fractionated into neutral and polar lipid by column chromatography with silicic acid. The fatty acid composition of lipid fractions were determined by gas liquid chromatography. Total lipid content of ark shell was 0.83% base on wet weight. The content of unsaponifiable matter was 20.19%, and iodine value was 156.13. The main components of total lipids were triglyceride, diglyceride, hydrocarbon, and sterol ester. The fatty acid composition of total lipid chiefly consisted of $C_{17 : 0}$, $C_{16 : 0}$, $C_{18 : 1}$ and $CT_{16 : 1}$. The main fatty acids of neutral lipid were $C_{16 : 0}$, $C_{18 : 1}$, $C_{22 : 1}$, $C_{18 : 0}$ and $C_{16 : 1}$. The major fatty acids of polar lipid were $C_{16 : 0}$, $C_{18 : 2}$, $C_{20 : 5}$ and $C_{22 : 6}$. In total lipid fractionation, saturated acid contents were high in all (SA>MA> PA), in neutral lipid fractionation, menoenoic acid contents were high in all (MA > SA> PA), and in polar lipid fractionation, saturated acid con-tents were high in all (SA> PA> MA).

Assessment of Bacteriological Safety of the Seawater and Ark shell (Scapharca subcrenata) in Yeoja Bay, Korea (전남 여자만 해수 및 새꼬막(Scapharca subcrenata)의 세균학적 위생 평가)

  • This study evaluated the sanitary state of seawater and shellfish in Yeoja bay from January 2014 to June 2016. The sampling stations for sanitary survey in Yeoja Bay was composed of 50 stations for seawater and 4 stations for ark shell and the samples have been collected monthly at a stated date. The total and fecal coliforms in seawater and Escherichia coli (E. coli) in shellfish were investigated as sanitary indicator bacteria, respectively. The geometric mean and the estimated 90th percentile ranges of total and fecal coliforms for seawater were <1.8-3.9, <1.8-2.6 and <1.8-29.7, <1.8-9.3 MPN/100 mL, respectively. The range of E. coli levels for ark shell were <20~330 MPN/100 g. The sanitary state in Yeoja bay was evaluated clean area according Korea criteria and conditionally approved area according US criteria and Class B area according EU criteria.

Regional Difference in Fatty Acid Content of Korean Shellfish

  • Regional variation in the fatty acid content of shellfish was investigated on 5 species of Korean shellfish including murex shell, ark shell, jack-knife clam, orient hard clam, and little neck clam that were originated from 2 geographically different regions in Korea (Region 1: South coast, $34-35^{\circ}N$, $127-129^{\circ}E$; Region 2: West coast, $36-38^{\circ}N$, $126-127^{\circ}E$). Significant regional difference in total fatty acids content was observed in murex shell and little neck clam (p<0.01), but not in the other species of shellfish. The contents of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids including n-3 and n-6 fatty acids were appreciably higher in murex shell from Region 2 and in little neck clam from Region 1 than the shellfish originated from their counterpart areas (p<0.05). Nevertheless, relative percentages of the fatty acids remained constant within same species regardless of geographic regions or species. Considering the facts of that the fish/shellfish are unique sources of n-3 fatty acids and a little neck clam is the most-consumed shellfish in Korea, n-3 fatty acids intake might vary with the habitat of the shellfish that Koreans consume.

A Study on Survival and Growth of Ark shell, Scapharca broughtonii with differential density during hanging culture

  • Ark shell Scapharca broughtonii length and total weight growth by density, which was hanging-cultured near the sea area of Yeosu-city, Hoenggan-island (below is Yeosu), showed inverse correlation with density. As for the average growth of shell length by density, the test area of 10 shells showed the best result as 5.21 mm, and it became lower as density became higher. Also, the increase of total weight by density showed the same result, thus on the whole, the increase of average shell length and total weight of 10-20 shell test area showed the better result than the test area of 30-40 shells. In addition, even if the parashell's shell length by density and total weight growth in the sea area of Namhae-gun, Mijo-fishing port (below is Namhae) showed inverse correlation with density, it showed the slighter inverse correlation than the sea area of Yeosu. As for the average growth of shell length by density, the test area of 10 shells showed the best result as 4.55mm, but the average growth of shell length by density in the test area of 20-40 shells appeared to be similar as 3.11, 3.36 and 3.27 mm. On the other hand, as for the increase of total weight by density, the test area of 10-20 shells showed the more increase than the test area of 30-40 shells. In conclusion, it is guessed that the culture density of 20 shells/$0.12m^2$ (166 shells / $1m^2$) would be appropriate when hanging-culturing the shells. At the sea area of A, the survival rate of Scapharca broughtonii parashell was 68-87% by density respectively, and it showed the density-dependent result, while the survival rate of sea area of Namhae, where phytoplankton as prey is abundant, showed the much better result as 91-100%. It is guessed that this may be due to the fact that the quantity of phytoplankton in sea area of Yeosu is just 55.89% of sea area of Namhae. When hanging-culturing the shells, the water temperature by sea area ranged from 16.22 to $25.66^{\circ}C$, salinity 30.07-33.48, pH 8.10-8.45, and DO 4.49-9.65 mg/L.

Survival of the Ark Shell, Scapharca subcrenata and Physiological and Histological Changes at Decreasing Salinity

  • We examined physiological and histological responses related to the survival, oxygen consumption, excretion, and O/N ratio of the ark shell, Scapharca subcrenata, as a result of salinity changes. The 20-day $LS_{50}$ (median lethal salinity) at $15^{\circ}C$ was 13.87 practical salinity units (psu; confidence limits 10.30-18.74 psu), whereas the 14-day $LS_{50}$ at $25^{\circ}C$ was 12.59 psu (confidence limits 8.03-18.16 psu). In conditions of decreasing salinity, the osmolarity of individuals acclimated within 5 h above 26.4 psu but required more than 60 h below 13.2 psu. Oxygen consumption and ammonia excretion rates varied irregularly as salinity decreased. The O/N ratio was 19 and 27 at water temperatures of $15^{\circ}C$ and $25^{\circ}C$, respectively, but decreased to 1-10 as salinity declined. The effects of decreasing salinity were observed in the histological changes to each organ of S. subcrenata. As salinity decreased, cilia fell off, the epithelial layer underwent necrosis and vacuolation, the connective tissue layers of the mantle and visceral mass were destroyed, and hemocytes increased in the gills. The results of this study could prove important in investigating causes of mass mortality and managing shellfish aquaculture farms.

Reproductive Cycle of the Ark Shell, Scapharca subcrenata, on the West Coast of Korea

  • Monthly changes in the gonad index (GI), egg-diameter composition, gonadal development, reproductive cycle of the ark shell, Scapharca subcrenata, were investigated by histological method and morphometric data. This species is dioecious and oviparous. The gonad is located among the subregion of mid-intestinal gland, digestive diverticula and the outer fibromuscular layers compacted by the fibrous connective tissues and muscle fibers. The gonad index sharply increased in May, reached the maximum value in June, and then gradually decreased from July to December. The reproductive cycle of this species can be divided into six successive stages: early active stage (January to May), late active stage (June to July), ripe stage (June to August), partially spawned stage (July to September), degenerative stage (August to December), and resting stage (January to April). S. subcrenata spawns once a year between July and early September, and the main spawning occurred between July and August when the water temperatures were above $20^{\circ}C$. This evidence suggest that timings of maturation and spawning are closely related to water temperatures. Even though the spawning period was once a year, it is assumed that the number of spawning frequencies (broods) may occur more than twice during the spawning season.

Quality Characteristics of Got Mul-Kimchi during Storage by Type of Kimchi Containers Using Plastic Ark Shell (꼬막껍질을 이용한 김치 저장 용기의 종류별 저장 기간에 따른 갓 물김치의 품질 특성)

  • This study was conducted to evaluate the quality characteristics of got mul-kimchi during storage by type of kimchi container using a plastic ark shell. The pH level, acidity, hardness, color, sensory evaluation, and microbiological activity were performed. Kimchi containers were prepared with 0%, 2%, and 3% plastic ark shell. The pH level of 2% container samples showed no significant difference compared with that of the control, whereas pH levels of 3% container samples were significantly (P<0.05) higher than those of the control and 2% sample. Acidity was not different among treatments up to 4 weeks, whereas acidity of 3% container samples was significantly (P<0.05) lower than those of the control and 2% samples after 8 weeks. Hardness significantly decreased (P<0.05) upon 0, 2, and 3% treatments with increasing storage time, but there was no significant difference among the treatments. Hunter color L values increased in the order of 0, 3, and 2% with increasing storage time. In sensory evaluation of crunchiness, 3% container samples had significantly (P<0.05) higher crunchiness than the control. Total viable cells in the 3% container with got mul-kimchi were significantly (P<0.05) lower than those of other samples at 12 and 16 weeks of fermentation. Numbers of lactic acid bacteria in the 2% and 3% container samples were significantly (P<0.05) lower than those of the control samples after 16 weeks of fermentation. These results show that the plastic ark shell kimchi container extended storage compared with the control.
