  • Phytase from Aspergillus species is a very heat unstable enzyme which inactivates to a great extent during the thermal processing of animal feed formulation. Various protein stabilization additives were tested to improve its heat stability. Among them, a basic amino acid, L-arginine remarkably increased the thermal stability of phytase in an aqueous solution state.

  • Effects of yeast extract and ammonium sulfate were investigated on the production of L-or-nithine by an arginine auxotroph, Brevibacterium ketoglutamicum in flask and batch cultures. Yeast extract as an arginine source and ammonium sulfate as an inorganic nitrogen source had significant effects on L-ornithine production and cell growth. L-ornithine production was repressed by the excessive addition of arginine. Reversion of auxotrophic cells to the wild type was observed when the initial yeast extract concenfration was too low. There existed optimum concentrations of yeast extract and ammonium sulfate for L-or-nithine production. The effects of yeast extract and ammonium sulfate concentrations of the Leudeking-Piret model parameters were examined to analyze the relationship between cell growth and L-ornithine production.

  • The effect of some phytohormones and light on the growth, ADC activity and polyamine content in ginseng was studies. In seedlings, the growth, ADC activity and putrescine content were in creased by Ghs treatment. ADC activity ann putrescine content were slightly slightly derreased by ABA, but not changed by kinetin. Light treatment increased ADC activity and putrescine content greatly. In two year ginseng leaves treated by GA3, the ADC actin$.$its reached maximum and the spermidine content reached maximum 2 days faster than in the control. Thtse results suggest the possibility that these regulators are closely related to growth and polyamine cotent. UeVo'ordsEPanaxgineng C.A.   • Radiochemical synthesis of $N^{G}$-mono[$^{14}$ C-methyl]-L-arginine is described. The compound was synthesized from radio-active mono[$^{14}$ C]-methylamine as easily and purified by strong cation-exchange resin (NH form) liquid chromatography using a gradient of ammonium hydroxide, and crystallized as flavianate. The free amino acid was successfully prepared by strirring its flavianate and strong anion-exchange resin (OH- form), which could remove the flavianic acid from its salt in water below room temperature. Purity of the compound was tested by thin-layer chromatography, thin-layer electro-phoresis, and scintillation spectrometry.y.

  • L-arginine phosphate monohydrate (LAP) and tris(hydroxymethyl)aminomethane sulfate (THAMS) are new organic nonlinear optical materials for the device application such as the frequency conversion of laser radiation. In this work the single crystals of LAP and THAMS have been grown by the falling temperature method and the temperature difference method. The conditions of the growth parameters were presented and the grown crystals were characterized by means of optical microscopy, XRD and FTIR.

Effect of Initial pH and L-Arginine on the Composition of Fatty Acids of Streptomyces viridochromogenes

  • Mycelia of Streptomyces viridochromogenes grown under different pH were analysed for the fatty acid composition. The low relative proportion of 12-methyltetradecanoic acid and the high relative proportion of palmitic acid were characteristic for the young culture under slight acidic pH that caused delay of the aerial mycelium formation. The addition of L-arginine to the culture medium enabled an arginine auxotroph with bald phenotype to have the fatty acid composition similar to that of the wild type and to develop aerial mycelium. The ratio of 12-methyltetradecanoic acid to palmitic acid might be used as a parameter to explain the optimum grwoth in the respect of membrane fluidity.

  • Arginine deiminase (ADI, E.C. 3.5.3.6), one of the arginine deprivation enzymes, exhibits anticarcinogenic activities. The present study investigated the anti-inflammatory activities of the purified recombinant ADI originating from Lactococcus lactis ssp. lactis ATCC7962 (LADI). LADI dose-dependently inhibited lipopolysaccharide (LPS)-induced upregulation of inducible nitric oxide synthase and the production of nitric oxide in RAW 264.7 murine macrophages. The induction of cyclooxygenase-2 expression and subsequent production of prostaglandin $E_2$ by LPS was also attenuated by LADI treatment. Moreover, LADI inhibited the production of interleukin-6 in LPS-stimulated RAW 264.7 macrophages. These results indicate that LADI exerts anti-inflammatory effects, which may in part explain its chemopreventive potential.

Soluble Complex Formation of Rifampicin with Arginine (Rifampicin과 Arginine간의 가용성 Complex 형성에 관한 연구)

  • Rifampicin-arginine complex was prepared to increase the solubility and dissolution rate of rifampicin. Solvation method was applied to make the complex and its formation was identified by the solubility method, powder x-ray diffractometry, differential thermal analysis and spectroscopic determination. The complex was formed in the molar ratio of 1 : 1 which was proved by the slope ratio method and continuous variation method. The complex was a non-crystalline form determined by the x-ray powder diffractometric analysis. The solubility of complex in water was significantly higher than that of rifampicin itself. The stability constant and thermodynamical properties of the complex were determined to investigate the solubilization phenomena. The thermodynamic data showed that the complexation between rifampicin and arginine was an exothermic and spontaneous reaction. Simulated absorption studies carried out through the artificial lipid barrier in artificial gastric and intestinal juices. The results showed that the complex had an enhanced absorption rate of rifampicin nearly twice compared with that of rifampicin itself.

Structural insights showing how arginine is able to be glycosylated by pathogenic effector proteins

  • Glycosylation is one form of protein modification and plays a key role in protein stability, function, signaling regulation and even cancer. NleB and SseK are bacterial effector proteins and possess glycosyltransferase activity, even though they have different substrate preferences. NleB/SseKs transfer the GlcNAc sugar to an arginine residue of host proteins, leading to reduced $NF-{\kappa}B-dependent$ responses. By combining X-ray crystallography, NMR, molecular dynamics, enzyme kinetic assays and in vivo experiments, we demonstrated that a conserved HEN (His-Glu-Asn) motif in the active site plays a key role in enzyme catalysis and virulence. The lid-domain regulates the opening and closing of the active site and the HLH domain determines the substrate specificity. Our findings provide evidence for the enzymatic mechanism by which arginine can be glycosylated by SseK/NleB enzymes.

Nitrogenous Constituents in the Extract of Crabs caught in the Korean adjacent Sea (한국 근해산 주요 게 종류의 함질소엑스분에 관한 연구)

  • Nitrogenous constituents in the extract of 3 species of raw and boiled crabs, tenner crab (Chionoeretes opilio elongatus), horsehair crab (Erimacrus isenbecki), and blue crab (Portunus trituberculatus) caught in the Korean adjacent sea, were analyzed and compared their compositions with the sex and the tissues, body meat, leg meat, and viscera. The crabs contained about 3% of the extractive nitrogenous constituents in the tissues, meat and viscera. The free amino acid compositions were commonly featured a high content of arginine, taurine, proline, and glycine regardless of the differences in species and tissues. Free amino acids and quarternary bases were greatly increased in hot water extraction while nucleotides were slightly diminished. Glycine betaine and trimethylaminoxide were accounted for above 30% of the total extractive nitrogenous constituents. Arginine, taurine, proline, glycine, glycine betaine, and trimethylaminoxide which abundant in the meats and viscera of the crabs were estimated to greatly contribute directly or indirectly toward appearing the characteristic crabs taste.
