Fluid Flow Characteristics in the Aquaculture Tank for a Breeding Fish

  • The aquaculture tank is used for breeding fish in sea water which was pumped up to land. The flow characteristics in the aquaculture were investigated with varying the tank geometry and flow rate. The numerical analysis has been employed for calculating the velocity and temperature distributions in a water tank of rectangular type. The finite volume method and SIMPLE algorithm with 3-dimensional standard $\kappa$-$\varepsilon$ turbulence model were used for the numerical analysis. For comparison with experimental results, the PIV system was applied to visualize the flow patterns quantitatively. The numerical results showed good agreements with the experimental results. The mean velocity and temperature versus aquarium depth were represented for various circulating flow rates. Especially, the aquaculture environment is recommended that the aquarium depth has to be d=0.5 m, and this case is the condition of higher velocity and temperature in winter season.

Expression of Prolactin Receptor mRNA and Blood Physiological Responses to Salinity Changes in the Black Porgy Acanthopagrus schlegeli (염분 변화에 따른 감성돔 Acanthopagrus schlegeli의 Prolactin Receptor(PRLR) mRNA 발현 및 생리적 반응)

  • We isolated complementary DNA(cDNA) encoding prolactin receptor(PRLR) from gill of black porgy Acanthopagrus schlegeli. Its PRLR cDNA consists of 1,611 base pairs and encodes the protein of 536 amino acids. To investigate the osmoregulatory abilities of black porgy in different salinities(35, 10 and 0 psu), we examined the expression of PRLR mRNA in osmoregulatory organs(gill, kidney and intestine) using reverse transcription(RT)-PCR. In gill and intestine, PRLR mRNA levels were high in 10 psu, and then decreased in 0 psu, but there is no changes in kidney. Also, plasma osmolality, $Na^+\;and\;Cl^-$ levels decreased during the experimental period. These results suggest that PRLR plays an important role in hormonal regulation in osmoregulatory organs during freshwater acclimation, thereby improving the hyper-osmoregulatory ability of black porgy in hypoosmotic environments.

Biochemical analysis of blood serum from wintering seabream with green liver syndrome (저수온기 돔류의 녹간증 혈청성분)

  • A blood biochemical analysis was carried out for the serum of wintering sea bream (red sea bream Pagrus major, black sea bream Acanthopagrus schlegeli, rock bream Oplegenathus fasciantuas) with green liver syndrome. GOT and GPT were significantaly elavated in the diseased sea bream. The value of $NH_{3}$ was slightly higher but those of TBIL. TP and UA were lower than the control. But there were no differences in albumin. Ca and glucose between the diseased and the control. These results suggent that a green liver syndrome in the sea bream might be related with nutritional defects and the stress of low water temperature during the winter.

Development of Bivalve Culture Management System based on GIS for Oyster Aquaculture in GeojeHansan Bay (거제한산만 굴 양식장에 대한 GIS 기반 어장관리시스템 개발)

  • Oyster production is playing an important role in domestic aquaculture, but facing some problems such as exports decrease, a slowdown in domestic demand and marine environmental deterioration. In order to obtain the suitable and sustainable oyster production, suitable sites selection is an important step in oyster aquaculture. This study was conducted to identify the suitable sites for lunging culture of oyster using Geographic Information System(GIS)-based multi-criteria evaluation methods. Most of the parameters were extracted by Inverse Distance Weighted(IDW) methods in GIS and eight parameters were grouped into two basic sub-models for oyster aquaculture, namely oyster growth sub-model(Sea Temperature, Salinity, Hydrodynamics, Chlorophyll-a) and environment sub-model(Bottom DO, TOC, Sediment AVS, Benthic Diversity). Suitability scores were ranked on a scale from 1(leased suitable) and 8(most suitable), and about 80.1% of the total potential area had the highest scores 5 and 6. These areas were shown to have the optimum condition for oyster culture in GeojeHansan Bay. This method to identify suitable sites for oyster culture may be used to develop bivalve culture management system for supporting a decision making.

Feeding Habits of the Large Yellow Croaker, Larimichthys crocea in the Coastal Waters of Jeju Island, Korea (한국 제주 연안에 출현하는 부세(Larimichthys crocea)의 식성)

  • Feeding habits of the large yellow croaker, Larimichthys crocea, was studied using 816 specimens collected by large pair-trawl fishery from June 2021 to May 2022 in the coastal waters of Jeju Island, Korea. The range of total length was 19.8~57.4 cm. Using the IRI, Pisces were mainly prey component constituted 57.5%. Euphausiacea and Macrura were important prey component constituted 23.2% and 18.7%, respectively. There was a significant difference in the change in diet composition by size group. As the L. crocea grew, the feeding rate of Euphausiacea decreased and the feeding rate of Pisces increased. There were also significant differences in the changes in diet composition by seasonal group. The most important prey species for L. crocea in the winter and spring were Pisces, in the summer it was Macrura, and in the fall it was Euphausiacea. As a result of analyzing the feeding relationship between size groups and season groups, L. crocea showed significant differences in the interaction between size groups and season groups.

Residues of Antibiotics in Wild and Cultured Fishes Collected from Coast of Korea (시중 유통 자연산 및 양식산 활어의 항생제 잔류)

  • Wild and cultured fish including olive flounder, sea bass, rock bream, yellowtail, gray mullet, gizzard shad, black rockfish, red seabream and squid were collected from a fish market located on the coast of Korea, and the antibiotic content of their muscle was investigated. Tetracycline group antibiotics were not detected in the 108 individuals of 9 species of wild fish. However, oxytetracycline (OTC) and tetracycline(TC) were detected in some samples of the 111 individuals in 7 cultured live fish species. The detected ranges of OTC and TC were ND~ 0.06 and ND~ 0.03, respectively. Five different fluoroquinolone antibiotics were also tested for, but were not detected in the wild fish species. Only small amount of criprofloxacin(ND~0.029 mg/kg) were detected in a few cultured fish samples. Oxolinic acid was not detected in either wild and cultured fish samples. Results showed that even very low levels of antibiotics could be detected by the testing methods used. Antibiotics were identified in a few fish samples but levels were far below the maximum allowable limits of the Korean Food Code, and the safety of fish being sold in markets, with regard to antibiotic levels, was confirmed.

Erythromycin Withdrawal Time in Olive Flounder (Paralichthys olivaceus) after Oral Administration (양식 넙치(Paralichthys olivaceus)에 대한 Erythromycin 휴약기간 설정)

  • The withdrawal time of erythromycin in cultured olive flounder was investigated to ensure the food safety of the fish treated with erythromycin. The oral administration of erythromycin was carried out using the experimental diet containing erythromycin (200 mg/kg) daily dose of 40 mg/kg body weight. The 45 day experimental period was broken into 7 days of habituation, 8 days of medication and 30 days of additional feeding without antibiotics. The erythromycin concentration in the flounder muscle had been increased gradually with medication. After 5 days of medication, the concentration increased to its maximum level of 6.05 mg/kg. After discontinuing the antibiotic, the erythromycin concentration decreased drastically and day 9 was below 0.1 mg/kg. The erythromycin concentration had slowly declined from the 6th to the 20th day after medication and disappeared completely after 25 days. From these results, the time needed to reduce the erythromycin level to the 0.2 mg/kg limit adopted by the ED and Japan was suspected to be 4-6 days. Therefore, a reasonable withdrawal time following ED and Japanese regulatory guidelines for erythromycin in the cultured flounder could be estimated to be 10 days.

Toxicity of Puffer Fish, Arothron firmamentum (Byeolbok) and Lagocephalus spp. in Korea (연안 어시장에서 유통되는 별복(Arothron firmamentum)과 밀복류(Lagocephalus spp.)의 독성)

  • The toxicity of five species of puffer fish, Arothron firmamentum (Byeolbok), Lagocephalus gloveri (Heukmilbok), L. wheeleri (Eunmilbok), L. inermis (Minmilbok) and L. lunaris (Milbok), collected from fish markets in Korea, was determined using a mouse bioassay. In A. firmamentum, the proportion of toxic specimens containing >10 MU/g was 87.5% in the ovaries, and 10.0% in the skin; no toxicity was detected in the muscle, fin, liver, intestine and gallbladder using the mouse bioassay. The highest toxin levels were found to be 87 MU/g in the ovaries, and 13 MU/g in the skin. Toxic specimens containing >10 MU/g were not detected from samples taken from any of the organs in L. wheeleri and L. inermis. In L. gloveri, most specimens were found to be non-toxic, but toxin levels of 11-72 MU/g were detected from within the skin, fins, and intestines in one specimen. In L. lunaris, the proportion of toxic specimens was 50.0% in the ovaries, and 7.1% in the gallbladder; no toxicity was detected in the other organs by the mouse bioassay. The highest toxin levels were 75 MU/g in the ovaries, and 14 MU/g in the gallbladder. Therefore, the toxicities of edible muscle and skin in the five species of puffer fish marketed in Korea were found to be within acceptable levels for human consumption.

Horizontal Distributions of Salinity and the Concentrations of DIN and DIP After Heavy Rainfall Events in Areas of Cochlodinium Polykrikoides Bloom Occurrence (Cochlodinium Polykrikoides 적조가 발생하는 해역에서 호우에 의한 담수 유입 범위와 질소, 인의 농도변동)

  • We examined the horizontal distribution of salinity and the concentrations of DIN and DIP after heavy rain-fall events in coastal areas of South Korea from Yeoja Bay to Narodo and from Gwangyang Bay to Geomodo to determine whether fresh water actually flows into areas of Cochlodinium polykrikoides red tides and to observe its effect on the growth of this organism after heavy rainfall. Following heavy rainfall (155 mm) in the Yeosu and Suncheon regions, the average salinity was 21 and 29 psu at Yeoja Bay and in the coastal waters of Narodo, respectively. After 126 mm of rainfall, the values were 19 and 25 psu in the coastal waters of Yeosu and Geomodo, respectively. This may have been caused by an influx of fresh water, after the rainfall event, into the open sea coastal areas around Narodo and Geomodo from the Dong and Seomjin Rivers, which are about 3540 km away. After the rainfall, the concentrations of $NH_4-N,\;NO_2-N$, and $PO_4-P$ were slightly increased; however, the concentration of $NO_3-N$ was greatly increased and diffused throughout the coastal areas of Narodo and Geomodo, which frequently experience C. polykrikoides blooms. The influence of $NH_4-N,\;NO_2-N$, and $PO_4-P$ on the occurrence of C. polykrikoidesred tides in coastal areas around Narodo and Geomodo after heavy rainfall does not appear to be great. Instead, the occurrence C. polykrikoides red tides in the coastal areas of Narodo and Geomodo seems to be facilitated by $NO_3-N$.
