약용식물 추출물에 의한 사과 저장병 방제 효과 (Effect of Medicinal Plant Extracts on Apple Storage Diseases)

  • This experiment was conducted to test the control effect of methanol extracts of 10 medicinal plants on apple storage diseases caused by Botryosphaeria berengeriana, Glomerella cingulata and Penicillium expansum. Out of the 10 medicinal plants, methanol extracts of Coptis japonica and Anemarrhena asphodeloides inhibited effectively the mycelial growth of B. berengeriana, G. cingulata and P. expansum in vitro, for which the inhibition ratios of the two plant extracts were 100.0% and 89.3%, 73.7% and 94.1%, and 100.0% and 51.6%, respectively. Spore germination of the three fungi was inhibited 100% only by C. japonica extract, but only P. expansum was inhibited 100% by A. asphodeloides extract. No lesion was formed y the fungi at 5$^{\circ}C$ up to 2 weeks after inoculation. Lesion sizes produced by the three fungi at the temperature ranges of 1$0^{\circ}C$ to $25^{\circ}C$ and infection of B. berengeriana and G. cingulata were inhibited by C. japonica extract, but not by A. asphodeloides extract, while no lesion was formed by the fungi at 5$^{\circ}C$. Infections of the fungi on apples were somewhat stimulated by A. asphodeloides extract.

Anti-inflammatory effect of Malus domestica cv. Green ball apple peel extract on Raw 264.7 macrophages

  • We examined the anti-inflammatory effect of the peel extract of the newly bred Korean apple (Malus domestica Borkh.) cultivar Green ball. To test its possible use as anti-inflammatory functional material, Raw 264.7 macrophages were treated with pro-inflammatory lipopolysaccharide (LPS) in the presence or absence of Green ball apple peel ethanol extract (GBE). Notably, up to 500 ㎍/mL of GBE did not result in any signs of inhibition on cellular metabolic activity or cytotoxicity in Raw 264.7 macrophages. Supplementation with GBE to LPS-treated Raw 264.7 macrophage significantly suppressed various pro-inflammatory responses in a dose-dependent manner, including i) nitric oxide (NO) production, ii) accumulation of inducible NO synthase and cyclooxygenase-2, iii) phosphorylation of nuclear factor-kappa B (NF-κB) subunit p65, and iv) expression of pro-inflammatory biomarker genes, including tumor necrosis factor alpha, interleukin 1 beta, interleukin 6, monocyte chemoattractant protein-1, and prostaglandin E synthase 2.

Investigation of Antioxidative Activity from Apple (Malus pumila Miller) Peel by Various Extraction Solvents

  • Consumption of fruits and vegetables has been conducted to be effective in the prevention of chronic diseases. In this study, 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used as solvents in the extraction of apple peels. The total phenol content, total flavonoid content and antioxidant activity of various extracts were investigated using in vitro assays. The extract obtained by 70% methanol showed the highest total phenol content ($20.87{\pm}0.17mgCAE/g$), ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity and ferric reducing antioxidant power. However, 70% ethanol extract possessed the strongest antioxidant activity assayed by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ${\beta}$-carotene bleaching method. And CM extract was found to show the highest total flavonoid content with the value of $9.26{\pm}0.06mgQE/g$. These results indicated that apple peels can be used in dietary applications with a potential to reduce oxidative stress.

Comparative Study of Antioxidant Activity of Apple and Pear Peel

  • Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

사과 잎(apple leaf) 추출물의 항산화 효과 (Antioxidants of apple leaf extract)

  • 본 연구는 사과 잎 추출물의 화장품 소재로 응용하기 위한 가능성을 평가하기 위하여 에탄올 70%로 추출하여 실험을 진행하였다. 사과 잎 추출물을 GC/MS로 분석하였고, 폴리페놀, 플라보노이드 함량과 DPPH radical 소거 활성을 통하여 항산화, 세포생존율(MTT assay) 확인을 통한 독성 평가를 하였으며, Nitric oxide (NO) 생성 저해 측정을 통해 사과 잎 추출물을 처리한 군에 4.6배의 NO 생성량이 감소하는 것을 확인하였고, 항염효과를 알아보았다. 총 폴리페놀 함량은 78.80 ± 0.25 mg/g, 총 플라보노이드 65.25±6.62 mg/g로 나타났다. DPPH radical 소거 활성은 추출물 농도 0.25%에서 79.8± 0.99%, 0.5%에서 88.13±0.89%, 1%에서 96.83±2.00%로 소거능이 증가함을 확인하였다. 세포생존율 평가는 고농도인 1000 ppm 농도에서도 91.19±3.49%로 80% 이상의 생존률을 확인하였다. GC-MS 성분분석 결과 catechol(5.65%), DL-Gluciol(12.05%), Ascorbicacid(2.41%), Phytol(13.88%), Hexanoic acid(5.47%) 등 항산화 화장품 소재로 활용이 사료된다. 이에 실험 결과 사과 잎 추출물을 이용한 천연 기능성 화장품 소재 개발에 있어 중요한 기초 자료로 이용되고자 한다.

Rapid Screening of Apple mosaic virus in Cultivated Apples by RT-PCR

  • The coat protein (CP) gene of Apple mosaic virus (ApMV), a member of the genus Ilarvirus, was selected for the design of virus-specific primers for amplification and molecular detection of the virus in cultivated apple. A combined assay of reverse transcription and polymerase chain reaction (RT-PCR) was performed with a single pair of ApMV-specific primers and crude nucleic acid extracts from virus-infected apple for rapid detection of the virus. The PCR product was verified by restriction mapping analysis and by sequence determination. The lowest concentration of template viral RNA required for detection was 100 fg. This indicates that the RT-PCR for detection of the virus is a 10$^3$times more sensitive, reproducible and time-saving method than the enzyme-linked immunosorbent assay. The specificity of the primers was verified using other unrelated viral RNAs. No PCR product was observed when Cucumber mosaic virus (Cucumovirus) or a crude extract of healthy apple was used as a template in RT-PCR with the same primers. The PCR product (669 bp) of the CP gene of the virus was cloned into the plasmid vector and result-ant recombinant (pAPCP1) was selected for molecule of apple transformation to breed virus-resistant transgenic apple plants as the next step. This method can be useful for early stage screening of in vitro plantlet and genetic resources of resistant cultivar of apple plants.

신육성 품종인 피크닉 (Picnic; Malus pumila Mill.) 사과의 기능성 (Functional properties of newly bred Picnic apple (Malus pumila Mill.))

  • 신육성품종 피크닉 사과로부터 phenolic compounds를 추출 후 기능성 식품 및 기능성 화장품 활성을 검정하여 신육성 품종 피크닉사과의 기능성 소재 활용가능성을 살펴보았다. 피크닉 사과로부터 생리활성으로 작용하는 phenolic 화합물을 추출하기 위하여 water과 ethanol로 추출하였을 때 각각 6.81, 13.25 mg/g의 비교적 높은 phenolic 함량을 나타내었다. 피크닉 사과 추출물의 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical 소거능은 water과 ethanol 추출물 $100{\mu}g/mL$ phenolic 농도에서 각각 87.69, 90.55%와 98.44, 96.95%의 매우 높은 DPPH 및 ABTS 전자공여능을 나타내었다. PF는 water과 ethanol 추출물 $100{\mu}g/mL$ phenolics 농도에서 각각 2.33, 2.40 PF를 나타내었으며, thiobarbituric acid reactive substances는 $100{\mu}g/mL$ phenolics 농도에서 water 추출물은 56.05%, ethanol 추출물은 84.19%의 항산화능을 나타내어, 피크닉 사과는 항산화 효과가 우수하여 노화방지를 위한 천연 항산화제로 활용가능성이 매우 높은 것으로 판단하였다. 피크닉 사과 추출물의 hyaluronidase 저해 효과를 측정한 결과 water과 ethanol 추출물에서 3-4% 정도의 낮은 저해 효과를 나타내었으며, 당분해 억제를 통해 항당뇨 효과를 나타내는 ${\alpha}$-glucosidase 저해 효과를 측정한 결과 ethanol 추출물에서는 $200{\mu}g/mL$ phenolics 농도에서 50.34%의 우수한 저해 효과를 나타내었다. 주름개선 효과를 측정하는 elastase 및 collagenase 저해 효과는 $200{\mu}g/mL$ phenolics 농도에서 water과 ethanol 추출물에서 각각 31.77, 48.71와 44.43, 52.78%의 저해 효과를 나타내었다. 위의 결과에 따라 피크닉 사과 추출물의 항산화, 항염증, 당분해 억제 및 주름개선 효과를 확인하였고, 기능성 식품 및 기능성 화장품의 기능성 소재로 활용가능성이 매우 높은 것으로 판단하였다.

The development of a new type of functional fresh apple juice using prebiotic fibers, ginger extract, and cardamom essential oil: Antioxidant capacity and chemical analysis

  • The formulation of a novel functional fresh apple juice enriched with dietary prebiotic fiber (inulin or polydextrose), ginger extract (GE), and cardamom essential oil (CEO) was carried out based on a combined D-optimal design. In the first stage, sensory evaluation was performed to screen and select the optimum sample for further experiments. The sensory evaluation showed that the sample containing inulin 0.25 g/100 g GE and 0.03 g/100 g CEO had the highest organoleptic score. In the second stage, various chemical experiments, including pH, acidity, formalin index, total phenol, flavonoids, antioxidant capacity, and vitamin C content, were evaluated on the selected enriched apple juices. The addition of GE and CEO caused changes in nutritional characteristics, including antioxidant capacity, total phenol, flavonoids, vitamin C, and IC50, from 35 g/100 g, 350 mg GAE/g, 17 mg/L, 370 mg/kg, and 1,800 mg/kg to 45 g/100 g, 460 mg GAE/g, 21 mg/L, 420 mg/kg, and 1,200 mg/kg respectively. The steady shear flow and dynamic oscillatory shear rheological tests were also performed on the screened samples, and results showed that the addition of dietary fiber in apple juices increased the apparent viscosity, storage modulus, loss modulus, and complex viscosity. In general, adding plant extracts and processed essential oil to apple juice increased the nutritional-nutraceutical value and sensory attributes of apple juice.

A facile green reduction of graphene oxide using Annona squamosa leaf extract

  • A highly facile and eco-friendly green synthesis of Annona squamosa (custard apple) leaf extract reduced graphene oxide (CRG) nanosheets was achieved by the reduction of graphene oxide (GO). The as-prepared CRG was characterized with X-ray diffraction (XRD), transmission electron microscope (TEM), Fourier-transform infrared spectroscopy (FT-IR), ultraviolet-visible (UV-Vis), X-ray photoelectron spectroscopy (XPS) and Raman spectroscopic techniques. Removal of oxygen containing moieties from the GO was confirmed by UV-Vis, FT-IR and XPS spectroscopic data. The XRD and Raman data further confirmed the formation of the CRG. TEM images showed the sheet structure of the synthesized CRG. These results show that the phytochemicals present in custard apple leaf extract act as excellent reducing agents. The CRG showed good dispersion in water.

액아배양에 의한 호접란 대량번식시 배지조성의 영향 (Effect of Medium Composition on Phalaenopsis Micropropagation Using Lateral Buds From Flower Stalks)

  • 호접란의 화경액아배양을 호르몬의 첨가없이 대량번식에 이용할 수 있도록 PLB 형성과 증식, 유식물분화를 위한 배지조성을 구명한 결과는 다음과 같다. PLB 형성을 위해서는 1.2X VW 배지에 사과추출물과 감자추출물, sucrose 1%, PVP 1.5g/l 또는 활성 탄소 2.5g/l, gellan gum 4g/l을 첨가하는 것이 가장 양호하였다. PLB 증식을 위해서는 sucrose 2%와 사과추출물과 감자추출물을 첨가한 VW에 cotton plate를 지지물로 사용한 것이 가장 양호하였다. 유식물의 재분화를 위해서는 Hyponex 3g/l에 사과와 감자추출물을 각각 3%, 바나나추출물은 4%를 혼용첨가한 배지가 가장 효과적이었다.
