Studies on Constituents of Higher Fungi of Korea (LXXIII) Antitumor Components of the Cultured Mycelia of Paxillus atrotomentosus (한국산 고등 균류의 성분 연구(제73보) -좀우단버섯 배양 균사체의 항암 성분-)

  • To find antitumor components from higher fungi, the cultured mycelia of Paxillus atrotomentosus were extracted with hot water. The water soluble fraction was purified and separated by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel filtration method. The separated fractions(Fr.) were designated CR A, B, C and D. Fr. A showed the highest inhibition ratio of 68.51% among the five tractions at a dose of 20 mg/kg/day. When Fr. A was examined for immunopotentiation activity, it increased the amount of the superoxide anion from activated macrophages to 1.1 fold and the number of plaques in hemolytic plaque assay to 2.3 fold, respectively. Otherwise, it did not show direct cytotoxity in sarcoma 180. Delayed type hypersensitiyity reaction showed that the decreased footpad swelling of tumor-hearing was restored to the normal. These results indicate that antitumor activity was exerted through immunopotentiation. Its chemical analysis showed 86.36% polysaccharide, 1.52% protein and 1.64% hexosamine. The polysaccharide consisted of fucose, galactose, glucose, mannose and xylose. This component was named paxillan.

Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Oudemansiella radicata (민긴뿌리버섯(Oudemansiella radicata)의 자실체로부터 추출한 조다당류의 항암 및 면역 활성 효과에 관한 연구)

  • Oudemansiella radicata, edible and medicinal mushroom belonging to Agaricales of Basidiomycota, has been known to exhibit outstanding curative effects on the fungal infection and hypertension caused by high blood pressure. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr, NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity test showed that Fr. NaCl was not cytotoxic against NIH3T3 and Sarcoma 180 at the concentration of $10{\sim}1,000\;{\mu}g/ml$. Intraperitoneal injection with Fr. NaCl exhibited antitumor activity with life prolongation effect of $42.9{\sim}66.7%$ in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.4{\sim}3$ folds compared with controlled and LPS groups, respectively. Intraperitoneal injection with Fr. MeOH increased the numbers of peritoneal exudate cells and circulating leukocytes by 3.5 and 2.5 folds, respectively. Therefore, the antitumor effect exhibited on mouse Sarcoma 180 cells was likely due to immune-modulating activity of crude polysaccharides extracted from fruiting body of O. radicata.

  • Tremella fuciformis, one of edible and medicinal mushroom belonging to Tremellaceae of Basidiomycota, has been known to have a curative effect on sarcoma 180 of mice and lowering high blood pressure of human beings. Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) substances were extracted from Tremella fuciformis. In vitro cytotoxicity tests, Fr. HW and Fr. NaCl were not cytotoxic against cancer cell lines such as NIH3T3, Sarcoma 180, and HT-29 at the concentration of $2000{\mu}g/ml$, while Fr. MeOH was cytotoxic to NIH3T3 and Sarcoma 180. Intraperitoneal injection with Fr. NaCl showed antitumor effect with life prolongation of 53% in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $3.0{\sim}8.1$ folds, respectively. Intraperitoneal injection with Fr. NaCl increased the numbers of peritoneal exudated cells and circulating leukocytes by 7.4 folds and 1.6 folds, respectively, than in the control group. The antitumor effect of T. fuciformis against Sarcoma 180 of mice was likely due to immunopotentiation activity.

  • Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) materials were extracted from Daedaleopsis tricolor. In vitro cytotoxicity tests, Fr. HW was not cytotoxic against cancer cell lines such as Sarcoma 180, HepG2 and HT-29 at the concentration of $0{\sim}2,000\;{\mu}g/ml$, while Fr. NaCl and Fr. MeOH were cytotoxic to the cell lines. Intraperitoneal injection with Fr. NaCl showed antitumor effect with life prolongation of 77.4% in mice inoculated with Sarcoma 180. Fr. NnCl and Fr. HW improved proliferation of spleen cells and the immunopotentiation activity of B lymphocyte by increasing spleen cells and the alkaline phosphatase activity by $1.7{\sim}2.4\;and\;2.2{\sim}8.7$ folds, respectively. Fr, NaCl generated $90\;{\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of $50\;{\mu}g/ml$, while lipopolysaccharide, a positive control, produced $79{\mu}M$. Intraperitoneal injection with Fr. NaCl (50 mg/kg body weight) increased the numbers of peritoneal exudate cells and circulating leukocytes by 10 folds and two folds, respectively, than in the control group. The antitumor effect of D, tricolor was likely due to immunopotentiation activity.

Inhibition of JAK2/STAT3 and activation of caspase-9/3 are involved in KYS05090S-induced apoptosis in ovarian cancer cells

  • To overcome the poor prognosis of patients with ovarian cancer, attempting to target ovarian cancer with effective antitumor compounds has been conducted for numerous years. Although the 3,4-dihydroquinazoline derivative KYS05090S was known to exert antitumor effects in A549 and ovarian cancer cells by inhibition of T-type Ca2+ channels, the complete underlying antitumor mechanism of this compound remains unclear. Thus, in the present study, the potential apoptotic mechanism of KYS05090S was elucidated in SKOV3 and OVCAR3 ovarian cancer cells. KYS05090S exerted significant cytotoxicity in SKOV3 and OVCAR3 ovarian cancer cells, and also increased the number of apoptotic bodies, and the number of terminal deoxynucleotidyl transferase dUTP nick end labeling positive cells and the sub-G1 population as a feature of apoptosis. Consistently, KYS05090S induced cleavage of poly(ADP-ribose) polymerase and caspase-9/3 in ovarian cancer cells. Notably, KYS05090S attenuated the expression of anti-apoptotic proteins, including cyclin D1 and B-cell lymphoma-2 (Bcl-2), and reduced the phosphorylation of Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) in ovarian cancer cells. Additionally, KYS05090S blocked the nuclear translocation of STAT3 and suppressed the signaling of JAK2/STAT3 in interleukin-6-treated SKOV3 cells, as a STAT3 activator. Overall, these observations indicated that inhibition of JAK2/STAT3 signaling and activation of caspase-9/3 are critically involved in the effects of KYS05090S on apoptosis in ovarian cancer types, and the compound may be beneficial as a potent antitumor agent.

  • All fifteen flavonoids (1-15) have been isolated from the roots of Sophora flavescens (Leguminosae) as active principles of the cytotoxic property toward human tumor cell lines such as A549, SK-OV-3, SK-Mel-2, XF498 and HCT15, in vitro. By means of spectral analyses, particularlyby the aid of various two dimensional NMR experiments, all $^1H-NMR$ ad $^{13}C$ -NMR signals of 1-15 were completely assigned, and thus the structures of 1-15 were established unambiguously.

  • As part of a drug discovery program to develope more effective platinum-based anticancer drugs, a series of platinum complexes trans-diaminocyclohexane platinum bi sdiphenylphosphino - ethane ( KHPC- 002) cis-diaminocyclohexane platinum bi sdiphenylphosphino - ethane ( KHPC- 006) has been evaluated in vitro against 4 human carcinoma cell lines with those of cisplatin using a tetrazolium-based colorimetric assay (MTT assay). The cell lines were two human bladder carcinoma cell lines, HT-1197 and HT-1376, human colon carcinoma cell line, HCT-116, and prostate cancer cell line DU-145. in vitro cytotoxic potential of each platinum complex was expressed as the cytotoxicity index (Cl, %).

  • The experiments tvere carried out to different temperature which affected growth, yield, proximate and antitumor activity in the Codonopsis lanceolata. Growth of aerial part and subterranean partwere best at 2$0^{\circ}C$ and lower 3$0^{\circ}C$. Components of fat, protein and fiber were best at 3$0^{\circ}C$. Wild C.fonceoforo had higher contents of fat, protein, fiber and ash as compared with cultur'ed C.lanceolataArginin was predominant amino acid in both wild and cultivated C.laceolata No significant differ-ence in the mineral contents was found between the wild root and the cultivated at 30'c inbiotronroom. No minerals difference in the contents was found between the cultivated temperature.The content of elements of inorganic metal differs according to the part. C. lanceolata were subjectedto preliminary antitumer screening test with Sarcoma 180 ascites and screening on V-79 cell. Thisexperiments were conducted in accordance with the total packed cell volume method and cytotoxicity method.

  • The activity-oriented fractionation of Psoralea corylifolia led to an isolation of a (+) bakuchio 1 as an activ eprinciple of its antitumoral property in vitro 1 was observed to exhibit a mild cytotoxicity against five kinds of cultured human cancer cell lines, i. e. the A549, SK-OV-3, SK-MEL-2, XF-498 and HCT15. The synthesized 2, 3-epoxide of (+)-bakuchiol 3 showed the similar activity as the (+) bakuchiol 1, whereas the other oxidation derivatives 4 and 5 including the acetyl (+) bakuchiol 2 showed a decreased activity.

  • The cytotoxic activities of methanolic extract and its fractions of Ailanthi Cortex Radicis and column chromatographic eluates of its dichloromethane fraction (DCM fr.) were investigated. DCM fr. Showed the strongest cytotoxicity against hepatoma cells. Furthermore, the active equates 1-3, 8 and 9 were obtained. Effects on free radical generation and the growth of vascular endothelial cells were tested to elucidate the action mechanism of anticancer activity. Eluates 1-3 stimulated free radical generation, while eluates 8 and 9 showed no changes. Especially, eluates 8 and 9 efffectively inhibited the proliferation of vascular endothelial cells in a dose- dependant manner. It is speculated that the anticancer effects of eluates 1-3, 8 and 9 might be due to free radical generation and inhibition of endothelial cell growth, respectively.
