5-Substituted Pyrimidine Acyclis Nucleoside Analogues 1-Cyanomethyl- and 1-(4-Cyanobutyl)-5-substituted Uracils as Candidate Antitumor Agents

  • A number of 5-substituted pyrimidine acyclic nucleosides were synthesized and tested for invitor cytotoxicity against four cell lines (j-82 cell, p-388 cell, FM-3A cell and U-938 cell lines). Synthesis of 1-cyanomethyl-5-substituted pyrimidines (1a-e) and 1-(4-cyanobutyl)-5-substituted pyrimidines (2a-e) was acomplished from the series of alkylation reactions ofl 5-substituted uracils with the corresponding chloacetonitrile and 5-chlorovaleronitile in DMSO under $50^{\circ}C$ temperature. These 5-substituted pyrimidine acylic nucleosides (1a-e and 2a-e) exhibited moderate to significant acitivity aginst four cell lines.

Synthesis of Nonclassical Quinazolinone Antifolates as Thymidylate Synthase Inhibitors and Their Antitumor Activity In Vitro

  • Nonclassical quinazolinone analogs I, II, and III, in which the glutamic acid moiety of the classical antifolates is substituted by phenylglycine, phenylalanine or aminobenzoic acid and their methyl esters, were synthesized and evaluated as lipophilic inhibitors of thymidylate synthase (TS). The target compounds were generally potent inhibitors of L. casei and human TS with $IC_{50}$ values within the narrow range of 0.2-10 ${\mu}$M and 0.003-0.03 ${\mu}$M, respectively. Further, most of the target compounds showed cytotoxicity against tumor cell lines of murine and human origin with $IC_{50}$ values of as low as 0.050 ${\mu}$M. Substitution of another hydroxyl or carboxylic acid/ester group at the phenyl ring further increased the potency of TS inhibition and cell growth inhibition. Most effective were compounds If and Ic in which extra carboxylic acid/ester was present at the phenyl ring with nanomolar $IC_{50}$ values of 0.0044 and 0.0093 ${\mu}$M against human TS and submicromolar cytotoxic growth inhibition against all four tumor cell lines.

수홍화자(水紅花子)의 항암활성(抗癌活性) 및 항전이(抗轉移) 효과(效果)에 관(關)한 연구(硏究) (Study on Antitumor Activity and Antimetastatic Effects of Polygoni Orientalis Fructus(POF))

  • To evaluate the antitumor activity and antimetastatic effects of Polygoni Orientalis Fructus(POF), studies were done experimentally. The results were obtained as follows : 1. In cytotoxicity against A549, and XF498 cell concentration inhibiting cell growth up to below 30% of control was recognized at $200{\mu}g/m{\ell}$ of POF. Also POF inhibited cell growth up to below 30% of control against SK-OV-3, SK-MEL-2, HCT15 and MCF-7 cell at $100{\mu}g/m{\ell}$. 2. The concentration inhibiting adhesion of A549 and SK-OV-3 to complex extracellular matrix up to below 70% of control was recognized at $100{\mu}g/m{\ell}$ of POF. 3. In pumonary colonization assay, a number of colonies in the lungs were decreased significantly in POF treated group as compared with control group. These results suggested that POF extracts might be usefully applied for prevention and treatment of cancer.

  • To evaluate the antitumor activity and antimetastatic effects of Gobongeerlhobang(GG1B), studies were done experimentally. The results were obtained as follows: 1. GG1B extracts exhibited a significant cytotoxicity against HT1080, A549, SK-MEL-2 and SK-OV-3 cell lines. 2. The T/C% was 120.7% in GG1B treated group in S-180 bearing ICR mice. 3. GG1B extracts exhibited inefficient adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 4. GG1B extracts showed a significant inhibition of lung metastasis of B16-BL6 cells in C57BL/6. 5. In CAM assays, angiogenesis was insignificantly inhibited in GG1B treated group than control group. These results suggested that GG1B extracts might be usefully applied for prevention and treatment of cancer.

Immunomodulatory Activity of Protein-Bound Polysaccharide Extracted from Gheiidonium majus

  • In the course of searching immunomodulators from natural sources, the protein-bound polysaccharide, CM-Ala, has been isolated from the water extract of Chelidonium majus L. (Papaveraceae). The immunostimulatory characteristics have been investigated in several experiments such as generation of activated killer (AK) cells, proliferation of splenocytes, activation of macrophages and granulocyte macrophage-colony forming cell (GM-CFC) assay. Of the fractions obtained using Sephacryl S200 column chromatography, CM-Ala was the most effective fraction that augmented the cytotoxicity against Yac-1 tumor cells from 0.88% to 34.18% by culturing with splenocytes for 5 days. CM-Ala also enhanced nitric oxide production by two fold in peritoneal macrophages and exhibited antitumor activity. It showed mitogenic activity on both spleen cells and bone marrow cells. CM-Ala induced proliferation of splenocytes by 84 fold and increased GM-CFC numbers by 1.48 fold over than the non-treated. On the contrary, CM-Ala had cytotoxic activity to a diverse group of tumor cells. From the above results, we proposed that CM-Ala has a possibility of an effective antitumor immunostimulator.

인간 폐암세포주 H-460세포에서 팔진탕합화적환과 $As_2O_3$의 병용처리에 의한 항종양 증진효과 (Antitumor Effect of the Cotreatment of Paljintanghabhwajeoghwan and $As_2O_3$ in Human Lung Cancer Cell Line H-460)

  • This study was designed to elucidate the synergistic cytotoxic mechanisms of the cotreatment of Paljintanghabhwajeoghwan (Paljin) and As₂O₃ in human lung cancer cell line, H-460. The combination of Paljin and As₂O₃ synergistically augmented the cytotoxicity of Paljin and As₂O₃ in H-460 cells. The nature of cytotoxicity was revealed as apoptosis which characterized by chromatin condensation and fragmentation in DAPI staining. Mitochodrial membrane potential transition was observed in H-460 cells treated with Paljin and As₂O₃. The apoptotic cytotoxicity of Paljin and As₂O₃ was accompanied by the cleavage of PARP and ICAD. Of note, pro-apoptotic Bak protein was obviously increased. However, the expression of p53 was not affected by the cotreatment of Paljin and As₂O₃. In addition, the expression of DR5 was increased by the cotreatment of Paljin and AS203. This results suggest that the synergistic cytotoxicity of the cotreatment of Paljin and As₂O₃ might be caused by the changes of the expression levels of a lots proteins, such as PARP, ICAD, Bak, DR5, which play pivotal roles in survival or death of cells.

The Cytotoxicity of Kahweol in HT-29 Human Colorectal Cancer Cells Is Mediated by Apoptosis and Suppression of Heat Shock Protein 70 Expression

  • Although coffee is known to have antioxidant, anti-inflammatory, and antitumor properties, there have been few reports about the effect and mechanism of coffee compounds in colorectal cancer. Heat shock proteins (HSPs) are molecular chaperones that prevent cell death. Their expression is significantly elevated in many tumors and is accompanied by increased cell proliferation, metastasis and poor response to chemotherapy. In this study, we investigated the cytotoxicity of four bioactive compounds in coffee, namely, caffeine, caffeic acid, chlorogenic acid, and kahweol, in HT-29 human colon adenocarcinoma cells. Only kahweol showed significant cytotoxicity. Specifically, kahweol increased the expression of caspase-3, a pro-apoptotic factor, and decreased the expression of anti-apoptotic factors, such as Bcl-2 and phosphorylated Akt. In addition, kahweol significantly attenuated the expression of HSP70. Inhibition of HSP70 activity with triptolide increased kahweol-induced cytotoxicity. In contrast, overexpression of HSP70 significantly reduced kahweol-induced cell death. Taken together, these results demonstrate that kahweol inhibits colorectal tumor cell growth by promoting apoptosis and suppressing HSP70 expression.

L1210 및 $P388D_1$에 대한 고삼 추출물의 세포독성에 관한 연구(II) (Studies on the Cytotoxicity of Sophora flavescens Ait. Extract Against L1210 and $P388D_1$ Cells (II))

  • This study was carried out to evaluate cytotoxicity of the extracts from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasms) cells in vitro. We have determined cytotoxicity by MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazo-liumbromide} assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: AM> EASF > CFSF > MTSF > WSF > HXSF and AM> EASF> CFSF> MTSF> HXSF> WSF. These results suggest that the ethyl acetate soluble extract of Sophora flavescens Ait may be a valuable choice for the development of antitumor agents.

  • This laboratory has recently reported the synthesis and in vitro antitumor activity of PT(II) complexes containing ethylenediamine and diphosphine. In view of the reports of others, cisplatin is toxic to the kidney since the kidney's vulnerability to PT(II) complexes may originate in its ability to accumulate and retain platinum to a greater degree than other organs. The in vitro cytotoxicity of these synthetic PT(II) complexes on the primary cultured proximal tubular cells of rabbit kidney and renal cortical cells of human kidney was investigated. Three endpoints for cytotoxicity tests were evaluated:3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), $^3H$-thymidine uptake and the glucose consumption tests. The rank order of sensitivity exhibited $^3H$-thymidine uptake>MTT>glucose consumption test. The agents with diphosphine leaving group were significantly less cytotoxic than cisplatin. Moreover, 1,2-bis(diphenylphosphino)ethane (DPPE) exhibited less cytotoxicity than 1.3-bis (diphenylphosphino)propane (DPPP) against on rabbit and human cultured kidney cells. Based on these results, the decreased nephrotoxicity of these new complexes over cisplatin appeared to be partially attributable to a leaving group of DPPP and DPPE. This novel class of platinum compound represents a valuable lead in the development of a "third-generation" agent.

  • Objectives : This study was designed to elucidate the synergistic cytotoxic mechanisms of the co-treatment of Gagamjengac-tang (GGJAT) and As₂O₃ in human lung cancer cell line, H-157. Methods : The combination of GGJAT and As₂O₃ synergistically augmented the cytotoxicity of GGJAT and As₂O₃ in H­157 cells. The cytotoxicity by the combination of these two drugs was revealed as apoptosis which was characterized by chromatin condensation and fragmentation in DAPI staining. Results : Antioxidant NAC completely blocked the apoptotic death of H-157 cells by GGJAT and As₂O₃. The apoptotic cytotoxicity of GGJAT and As₂O₃ was accompanied by the induction of DR4 and DR5 in RT-PCR. In addition, antioxidant enzymes such as SOD1, GSH synthetase and GSH reductase were also increased in H-157 cells treated with GGJAT and As₂O₃. However, of note, p53, Fas, FasL and TRAIL were not detected in H-157 cells treated with GGJAT and As₂O₃ by RT-PCR. Conclusions : These results suggest that the synergistic cytotoxicity of the co-treatment of H-457 cells treated with GGJAT and As₂O₃ may cause induction of death receptors DR4 and DR5 as well as reactive oxygen species.
