Antioxidative Characteristics of Extracts from Aromatic Herb Elsholtzia splendens

  • We investigated the antioxidant activity of ethanol extracts obtained from the flowers of Elsholtzia splendens on Chinese hamster ovary CHO-K1 cells. When cells were treated with E. splendens extracts (ESEs), low concentrations (<12.5 ug/mL) of stimulated cell proliferation via radical generation. Relative mRNA expression of Cu/Zn-superoxide dismutase (SOD) and Mn-SOD in cells exposed to ESEs (1-20 ug/mL) was significantly induced in a dose-dependent manner (p<0.05). In the case of catalase, ESEs had opposing effects; that is, a low-level treatment caused a decrease, and a high-level treatment induced elevated levels (p<0.05). The results demonstrated that components of ESEs exhibit potential antioxidant properties. Also, further studies are required to clarify the active components of Elsholtzia splendens extracts responsible for such biological activities.

  • In order to find the antioxidative components, fractionation of Cyperus amuricus (Cyperaceae) methanol extract was performed by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging effect. Three compounds, 3,4-dimethoxy benzoic acid (1), 4-hydroxybenzoic acid (2), and piceatannol (3) were isolated from the active ethylacetate soluble fraction of C. amuricus through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 3 showed the significant antioxidative effect on DPPH free radical scavenging test. These compounds are reported for the first time from this plant.

Quality Analysis for Recycle of the Drained Soybean Boiling Water Discarded in the Mass Production of Fermented Soy Foods (장류식품 대량제조시 폐기되는 콩 삶은 물의 재활용을 위한 품질특성 분석)

  • Nutritional components and quality characteristics of drained soybean boiling water(DBW), which is discarded in the mass production of fermented soy foods, were compared with raw soybean(Control) and Cheonggukjang(CGJ) to provide the basic data for its recycle. The contents of moisture, crude protein, crude lipid and crude ash of DBW were shown as 87%, 2.2%, 0.15% and 1.42%, respectively. Decreased total amino acid of 1,677.8 mg/100g in DBW was comparable with 29,051.1 mg/100g in control, however, there was no great difference in the proportion of essential amino acid to the total. While the total sugar contents were decreased in both DBW and CGJ with 8.39% and 7.17% each from the control of 11.50%, the reducing sugars were increased with higher amount of 6.44% in CGJ and 8.30% in DBW than 5.60% in control. pH of DBW was lower than both of the control and CGJ. Hunter's color values revealed the increase of redness(a value) and yellowness(b value) of DBW and CGJ suggesting that Maillard reaction products were produced by the heating and fermentation process. Polyphenol compounds were highly abundant in CGJ of 0.74 tannic acid equivalent(mg/g) followed by similar low amounts of 0.33 and 0.29 tannic acid equivalent(mg/g) in DBW and control, respectively. Antioxidative activity determined by Electron Donating Ability(%) using DPPH radical showed that CGJ, of which polyphenols were the highest, has the strongest electron donating ability with the lowest $EC_{50}$ value of 5.91 mg/mL. DBW was much lower but similar with the control. From the above results the drained soybean boiling water was shown to have many nutritional and functional components as much as soybean, therefore, it could be a potent reusable food material.

Characteristics of Propolis Extracts from Ethanol Extraction (에틸 알코올로 추출한 프로폴리스의 이화학적 특성)

  • Propolis was extracted using various concentrations of ethanol at different solvent-sample ratios. Volatile flavor components of propolis were detected by GC/MS, and the principal components were analysed. The content of total flavonoids (TF) in propolis extract increased 2 to 14% with increasing concentration of ethanol in the extraction solvent from 20 to 50%, but deceased slightly between of 60 and 70%. The 70% ethanol extract of propolis had the strongest antimicrobial activities to E. coli, L. plantarum and S. cerevisiae. Propolis ethanol extracts (2 to 14%) showed higher antioxidative activities when added oil to soybean oil during storage at $37^{\circ}C$.

Establishment of the Extraction Process by Evaluation of Proanthocyanidin Contents and Antioxidative Activities of Pine (Pinus densiflora) Needle Extracts. (적송잎 추출물의 proanthocyanidin 함량 분석 및 항산화 활성 검증을 통한 추출공정 확립)

  • We evaluated proanthocyanidin contents out of total polyphenolic compounds in two pine needle extracts prepared by combined extraction methods and compared their antioxidative activities. Amounts of total polyphenolic compounds of sub-supercritical hotwater precipitate (SSHP) and sub-supercritical hotwater supermatant (SSHS) were 225.4 and 116.6 mg/g, respectively. Amounts of proanthocyanidin of SSHP and SSHS were 9.9 and 3.6 mg/g, respectively. Thus, the ratio of proanthocyanidin contents over total polyphenolic compounds of SSHP and SSHS were 4.3 and 3.9%, respectively. Antioxidative activities ($IC_{50}$) of SSHP and SSHS measured by DPPH radical scavenging were 41.5 and $50.7\;{\mu}g/ml$, respectively. These results indicates that two combined extracts, SSHP and SSHS showed higher antioxidative activities than extract prepared by hot water or sub-supercritical. In conclusion, extraction procedures for SSHP and SSHS could be useful methods for preparing the effective components of functional food products.

Evaluation of Proanthocyanidin Contents in Total Polyphenolic Compounds of Pine (Pinus densiflora) Needle Extracts and Their Antioxidative Activities (적송잎 용매별 추출물의 총 폴리페놀성 성분에 대한 proanthocyanidin의 함량 분석 및 항산화 활성 검증)

  • Flavonoids in pine needles are known to be effective scavengers of free radical. Especially, proanthocyanidin, a kind of flavonoids possesses cardiovascular protection and antioxidative activities. Here, we evaluated proanthocyanidin contents in total polyphenolic compounds of pine needle extracts prepared by using hot water, ethanol, hexane or sub-supercritical $CO_2$. Analyses of total polyphenolic compounds and proanthocyanidin in each extracts indicated that hot water extract contained the highest concentrations, but sub-supercritical extract contained the lowest concentrations. On the other hand, evaluation of proanthocyanidin contents in total polyphenolic compounds in each extracts showed that sub-supercritical extract possessed the highest content, but hot water extract possessed the lowest content. These results indicate that extracts containing high concentrations of both total polyphenolic compounds and proanthocyanidin could be obtained by using hot water or ethanol extractions. Furthermore, extract containing high content of proanthocyanidin out of total polyphenolic compounds could be achieved by using sub-supercritical extraction. Measurement of antioxidative activities of extracts showed that hot water extract possessed the highest activity. In this study, we prepared extracts from pine needles by four different methods and evaluated the antioxidative compounds in extracts that could be used for effective components of functional food products.

Antioxidative and Hepatoprotective Effect of Compounds Isolated from Galla Rhois(Rhus javanica Linne) (오배자 성분의 항산화 및 간보호 효과)

  • The major components were isolated from the n-hexane, EtOAc and BuOH extract of Galla Rhois(Rhus Javanica Linne). Their structures were characterized as syringic acid, gallic acid methylester, protocatechuic acid, gallic acid and 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$. This study was carried out to investigate the biological activities of isolated compounds. Five compounds were tested for hepatoprotective effects on CCl4-induced cytotoxicity in primary cultured rat hepatocytes and antioxidative effect on Ferric-Thiocyanate method and TBA method. As a result, isolated five compounds showed stronger antioxidative activity than tocopherol, and the antioxidative activity of gallic acid methylester, protocatechuic acid and syringic acid were similar to that of BHA on Ferric-Thiocyanate method. Specially 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$ showed stronger effect of lipid-peroxidation inhibition than BHA. Gallic acid appeared stronger inhibitory effect of malondialdehyde on TBA method. Hepatoprotective effect of 1, 2, 3, 4, $6-penta-O-galloyl -{\beta}-D-glucose$ was similar or even higher than that of glycyrrhizin on primary cultured rat hepatocyte cytotoxicity.

Antioxidative Activity and Related Compounds of Apple Pomace (사과박의 항산화 활성 및 항산화 성분)

  • To enhance the utilization of apple pomace for the functional food resources, we analyzed the useful components and examined the antioxidant activity of apple pomace. The contents of total dietary fiber, total flavonoid, total phenolic acid and vitamine C were 55.56%, 458 mg%, 1048 mg% and 19.8 mg%, respectively. Protocatechuic acid, cinamic acid, caffeic acid, ferulic acid, syringic acid, vanillic acid and phydroxybenzoic acid were identified in the apple pomace extract by GC-MS. Phloridzin and quercetin-3-glucoside were identified in the apple pomace extract by HPLC. Ethyl acetate fraction showed the highest antioxidative ability by ferric thiocyanate method. Malondialdehyde(MDA) formation in normal rat liver tissue also showed the lowest in ethyl acetate fraction.

Comparison of methods for proanthocyanidin extraction from pine (Pinus densiflora) needles and biological activities of the extracts

  • Flavonoids are known to be effective scavengers of free radicals. In particular, proanthocyanidins are flavonoids that possess cardiovascular protection, antioxidative activities, and immunomodulatory activities. Here, we evaluated proanthocyanidin contents in the total polyphenolic compounds of pine needle extracts prepared by hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE). Analysis of each extract indicated that the ethanol extract contained the highest proanthocyanidin concentration. The HWH and hexane extracts also contained relatively high concentrations of proanthocyanidin. On the other hand, proanthocyanidin content analyses out of the total polyphenolic compounds indicated that the HWH extract contained the highest content. These results suggest that HWH extraction is a suitable method to obtain an extract with a high level of pure proanthocyanidins and a relatively high yield. The HWH extract possessed superior activity in diverse antioxidative analyses such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferrous ion chelating (FIC), and ferric-ion reducing power (FRAP) assays. In addition, upon assessing the effects of the pine needle extracts on macrophages (Raw 264.7 cell), the HWH extract exhibited the highest activity. In this study, we discerned an efficient extraction method to achieve relatively pure proanthocyanidins from pine needles and evaluated the biological functions of the resulting extract, which could potentially be used for its efficacious components in functional food products.

Inhibitory Effect of Euscaphic Acid and Tormentic Acid from the Roots of Rosa rugosa on High Fat Diet-Induced Obesity in the Rat (고지방 식이로 유도된 비만 흰쥐에서 해당근에서 분리된 Euscaphic Acid 및 Tormentic Acid의 효과)

  • The roots of Rosa rugosa have been used to treat diabetes mellitus in the folkloric society of Korea. To demonstrate the active component for the rat obesity induced by high fat diet for 6 weeks, the phytochemical fractionation and the pharmacological activity test were performed on this crude drug. It was shown that the methanolic extract and its EtOAc fraction inhibited the weight increase of the rat body, abdominal fat pad and hyperlipidemia at 200 mg/kg dose. Further, the triterpenoids, euscaphic acid and tormentic acid, isolated from R. rugosa roots were active at 30 mg/kg in the same assay. The two components shifted serum total-, HDL, and LDL-cholesterol levels toward the values of the unteated group, suggesting that the active compounds has hypolipidemic effects. The rats fad euscaphic acid and tormentic acid also reduced thiobarbituric acid-reactive substance (TBARS) and hydroxyl radical in the rat blood and increased superoxide dismutase activity compared to the control. TBARS values and carbonyl contest of the hepatic protein were reduced by treatment with the two triterpenoids. Antioxidative enzyme (SOD, glutathione peroxidase, and catalase) activities in hepatic were increased by treatment of rats with the triterpenoids, which suggests that triterpenoids inhibited the reduction of hepatic antioxidative activity caused by high fat diet. Taken together, these results support that euscaphic acid and tormentic acid improve a high fat diet-induced hyperlipidemia via the activation of antioxidative mechanism.
