Antioxidative and $Lp-PLA_2$ Inhibitory Activities in 29 Fruits and Vegetables (시중 유통 과일 및 채소류 29종의 항산화 효과 및 $Lp-PLA_2$ 활성 저해 연구)

  • Total phenolic content and the antioxidative activity (DPPH radical-scavenging capacity) of 29 fruits and vegetables were determined. The total phenolic contents of plum (1.06 mg GAE/g fresh weight) and cherry (0.95 mg GAE/g fresh weight) were high, whereas those of banana, carrot, and cucumber (each at 0.07 mg GAE/g fresh weight) were low. In the DPPH radical-scavenging assay, apple mango extract showed the highest value(92.8%) among the 29 ethanol extracts tested, followed by paprika (75.4%), beet (75.3%), and cherry (75.1%). In the $Lp-PLA_2$ inhibition assay, cherry (34.95% inhibition) and grape (31.03%) showed the highest $Lp-PLA_2$ activities among the 29 fruits and vegetables examined.

Anti-inflammatory effects of ethyl acetate fraction of unripe astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) on lipopolysaccharide-stimulated RAW 264.7 cells (지방질다당류로 자극한 RAW 264.7 세포에서 청도반시 땡감 에틸 아세테이트 분획물의 항염증 효과)

  • Unripe astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) is a by-product produced when thinning out the superfluous fruit of persimmon. We investigated whether unripe astringent persimmon has antioxidative and anti-inflammatory effects. Unripe astringent persimmon extract was fractionated sequentially in n-hexane, chloroform, ethyl acetate, n-butanol, and water. The ethyl acetate fraction had the highest total phenolic content, total flavonoid content, and antioxidant capacity compared to those of the other fractions. Pretreatment of lipopolysaccharide-stimulated RAW 264.7 macrophages with the ethyl acetate fraction reduced nitric oxide, interleukin-6, and intracellular oxidative stress in a dose-dependent manner. Ultra-high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry analysis revealed gallic acid, protocatechuic acid, 4-hydroxybenzoic acid, quercetin-3-O-glucoside, quercetin, and p-coumaric acid as the phenolic compounds of the ethyl acetate fraction. Collectively, these findings suggest that unripe astringent persimmon is a source of functional materials that can promote antioxidative and anti-inflammatory effects.

Effects of Dandelion (Teraxacum platycarpum) with Various Extracting Method on Antioxidative Capacity, Lipid Metabolism in Diet-induced Obese Rats (초음파추출과 열수추출에 의한 민들레의 항산화 및 지질강하 효과)

  • This study was conducted to investigate the effects of dandelion (Teraxacum platycarpum) extracts obtained by only water and with ultrasonification on antioxidative system and lipid metabolism in high cholesterol-fed rats. Five groups of rats were given high cholesterol diets for 8 weeks. The control group received without dandelion extracts and the other four groups received with one of dandelion extracts for 4 weeks respectively ; TP-N-1(100 mg/kg/day of Teraxacum platycarpum water extract), TP-N-2(200 mg/kg/day of Teraxacum platycarpum water extract), TP-S-1(100 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract), TP-S-2(200 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract). The results are summarized as follows; The hepatic and plasma TBARS levels significantly decreased in the dandelion extracts groups compared to those of no treatment group. Especially the group TP-N-2 was comparatively best among those. TP-N-2 groups had significantly higher levels of glutathione peroxidase (GSH-Px) and catalase activities. There was no significant difference between dandelion extracts groups and no treatment group in SOD levels. In plasma triglyceride level, plasma FFA level, TP-S-2 group had significantly lower levels than that of the other groups. In plasma glucose levels, dandelion extracts group were similar to those of normal rats. Plasma total cholesterol levels significantly decreased in the TP-S-2 group compared to those of the other groups. HDL levels were also significantly higher than those of the other groups. Compared with those of no treatment group, dandelion extract groups had significantly higher levels of LDL. In liver total cholesterol level, TP-S-2 groups had significantly lower levels than that of the other groups. Compared with those of no treatment group, dandelion extracts groups had significantly lower levels of liver triglyceride, but especially the TP-S-2 group showed comparatively the best significant effect among those. TBARS, triglyceride, LDL, FFA levels significantly decreased in TP-S-2 groups compared to the other four groups. HDL levels was also significantly higher than the other four groups. According to the above result, it could be suggested that ultrasonic extraction have the upper hand in lipid metabolism and water extraction have the advantage of antioxidative system.

Paraquat Toxicity in Weed Species : Difference in Physiological Responses between Tolerant and Susceptible Species (잡초종(雜草種)에 대한 Paraquat 독성(毒性) : 내성종(耐性種)과 감수성(感受性) 종간(種間)의 반응(反應) 차이(差異))

  • Paraquat, the representative bipyridilium herbicide, has high phytotoxic activity through generating toxic oxygen species such as superoxide, hydrogen peroxide and hydroxy radical. The response patterns of plants to paraquat were various. It was assumed that the different response was derived from different antioxidative mechanisms including antioxidative enzymes and antioxidant. Paraquat treatment increased reducing sugar content and malondialdehyde formation at 35 days after treatment in a dose-dependent manner but chlorophyll content decreased. Glutathione content increased by paraquat treatment and tolerant species showed more glutathione content than susceptible species. Superoxide dismutase activity increased with the increase in paraquat concentration and that was higher in tolerant species than susceptible species. Photosynthetic activity(PSII activity) was affected by paraquat, so the susceptible species showed more reduced oxygen evolving capacity than tolerant species. Catalse, NADPH-cytochrome C reductase, and malate dehydrogenase, the enzymes tested in this study, showed that the activities decreased by paraquat treatment. Further studies are necessary to determine whether antioxidative system cause the tolerance to paraquat.

Changes of Antioxidative Components and Activity of Fermented Tea during Fermentation Period (미생물을 이용한 후발효차의 발효기간별 항산화 성분 및 활성의 변화)

  • Changes of antioxidative components and activity of fermented tea manufactured by Bacillus subtilis, Saccharomyces cerevisiae, and Lactobacillus bulgaricus were evaluated during the fermentation period. The ascorbic acid content in the fermented tea was relatively lower (43.62~62.84 mg%) than that of green tea (66.74 mg%) during the entire fermentation period. The tea fermented by L. bulgaricus, which had the least contact with air, showed less change in ascorbic acid content. The polyphenol content of green tea was 14.88%, whereas that of fermented tea was 11.54~14.12% and it decreased during the fermentation period. The amount of flavonoids in green tea was 7.78 mg%, whereas that of fermented tea was 4.33~7.88 mg%. DPPH radical scavenging activity and ABTS reducing activity of green tea were 87.47% and 203.22 AEAC mg% (ascorbic acid equivalent antioxidant capacity), respectively, whereas those of fermented tea were lower than green tea. Results indicated that the antioxidative components and activity of fermented tea were lower than those of green tea during the fermentation period. But, when the sensory and hygienic quality are considered, fermented tea can be one of the higher quality tea products on the market.

Antioxidative Activities and Antiproliferation Effects on Oral Carcinoma KB Cell of the Brazilin from Caesalpinia sappan L. (소목(Caesalpinia sappan L.)에서 분리한 Brazilin의 항산화 활성과 구강상피암 KB 세포주의 증식억제효과)

  • Caesalpinia sappan L. has long been commonly used in oriental folk medicines and as dyes materials. To investigate the antioxidative activities and antiproliferation effects of brazilin from C. sappan heart wood, the MeOH soluble extract was successively fractionated by using hexane, $CHCl_{3}$, EtOAc, BuOH, MeOH, and $H_{2}O$. In these fractions, we were purified brazilin from EtOAc fraction which partitioned to 3.94% of the highest yields. The effects of brazilin and the extracts on human oral carcinoma cells (KB) by MTT assay and their antioxidant activities by DPPH, TCA assay and Fenton reaction were tested. The results showed that the brazilin could inhibits the proliferation of KB cells and obviously decreased the production of nitric oxide of the cells. When the concentration of the brazilin reached to $100\;{\mu}g/ml$, the inhibition percentage of the cell growth was about 60%. In assay on antioxidant activities, The results showed that brazilin exhibit the highest capacity of DPPH free radical scavenging effects among tested extracts. When the concentration of brazilin reached to 1 mg/ml, the lipid peroxide inhibition and radical inhibition activities were determined to be 65.0% and 85.8%, respectively. These results are suggest that brazilin have stronger antiproliferation effect on KB cell and antioxidant properties.

Changes in Free Sugar, Coixol Contents and Antioxidant Activities of Adlay Sprout (Coix lacryma-jobi L. var. ma-yuen Stapf.) according to Different Growth Stage (생육시기 별 새싹율무의 유리당, 기능성 성분 및 항산화활성 변화)

  • Background: Adlay (Coix lacryma-jobi L. var. ma-yuen Stapf.) is an important medicinal and cereal crop that contains high levels of protein, fatty acids and crude fiber, as well as calcium, phosphorus, zinc, minerals, and essential amino acids. Despite its economic significance, little is known about biological activity in adlay sprouts. This study investigated the total free sugar, polyphenol and flavonoid content, and antioxidative activity in adlay sprouts at different growth stages. Methods and Results: The total free sugar and coixol content of germinated adlay sprouts were analyzed by high performance liquid chromatography. The total phenolic content was measured by the Folin and Denis, and flavonoid content by the Davis method. Antioxidative activity was tested by the 2,2-diphenyl-1-picryl hydrazyl method. All measured component, including total phenolic and coixol content ($2.71{\pm}0.02mg/g$ and $59.70{\pm}0.01mg/g$), and antioxidant capacity ($IC_{50}$; $453.93{\mu}g/m{\ell}$) were highest in 3 day old adlay sprouts. In contrast, total free sugar and flavonoid content (3.02% and $0.60{\pm}0.02mg/g$) were highest in sprouts at 5 days after sowing. In addition, there was a strong positive correlation between antioxidative activity and total polyphenol content (r = 0.902, p < 0.001). Conclusions: Adlay sprouts between 3 days and 5 days following germination contain high levels of functional components and free sugar. This study therefore suggests that adlay sprouts can be developed as a valuable health-promoting food and a good source of natural antioxidants.

Antioxidative and antimelanogenic effects of ethyl acetate fractions of Korean domestic honeys from different floral sources (화종에 따른 국내산 벌꿀 에틸 아세테이트 분획물의 산화방지능 및 멜라닌 생성 억제 효과)

  • Honey is known to have a variety of health-promoting effects. Ethyl acetate fractions (EAFs) of four Korean domestic honeys from basswood, Korean raisin, chestnut, and acacia as well as a New Zealand manuka honey were analyzed comparatively to evaluate their antioxidative and antimelanogenic effects. The EAFs of basswood, Korean raisin, and chestnut honey had higher antioxidant capacities and tyrosinase inhibition activities than those of manuka honey. Pretreatment of B16F1 melanoma cells with EAFs at $100{\mu}g/mL$ resulted in relative retention of melanin contents as follows: acacia honey (141.0%)>basswood honey (134.3%)>manuka honey (131.5%)>Korean raisin honey (107.3%)>chestnut honey (88.0%). Intracellular tyrosinase activities of B16F1 melanoma cells were significantly (p<0.05) lowered by EAFs of Korean raisin and chestnut honey than by EAF of manuka honey. Consequently, these results suggest that Korean domestic honeys from different floral sources serve as good sources of antioxidative and antimelanogenic agents.

Antioxidant effect of ethanol extract from Poria cocos depending on cultivation methods (재배방법을 달리한 복령 에탄올추출물의 항산화 효과)

  • Objectives : Poria cocos Wolf has been widely used in Korean medicine as a medicinal fungus. In this study, we investigated that comparative anti-oxidant effects of ethanol extract from wild Poria cocos (WP) and plastic bag-cultivated Poria cocos (PBP).Methods : Total polyphenol and flavonoid contents in WP and PBP extract were monitored. And DPPH, ABTS, hydroxyl (·OH) free radical scavenging capacity, reducing power and xanthine oxidase inhibition activities of them were determined at 5, 1, 0.5 mg/ml concentrations.Results : Higher total polyphenol contents were found in the PBP extract (52.07±0.6 mg/TAEg) than in the WP extract (28.44±0.26 mg/TAEg). The flavonoid contents in WP and PBP extract were 17.29±0.30 mg/ RUEg, 21.36±0.40 mg/ RUEg, respectively. Also, DPPH, ABTS and ·OH free radical scavenging capacity of PBP extract in treated concentrations (5, 1, 0.5 mg/ml) significantly increased compared to those of WP-treated group. In particular, DPPH, ABTS free radical scavenging capacity and reducing power of PBP extract at 5 mg/ml concentration were similar to positive control (BHA or vit. C). Xanthine oxidase (XO) inhibition rate in both extract increased dose dependently. But it was significantly increased in PBP-treated group, only at 5 mg/ml, compared to those of WP-treated group. Then, their inhibition rate by PBP was similar to positive control (BHA).Conclusions : These results suggest that PBP extract is superior to WP extract in anti-oxidant capacity thus PBP can be applied in variable antioxidative products as a substitute for WP.

Free Radical Scavenging and Lipid Peroxidation Inhibition Potential of Hygrophila auriculata

  • Hygrophila auriculata (K. Schum.) Heine is a wild herb commonly found in moist places on the banks of tanks, ditches and paddy fields throughout India and is one of the main sources of Ayurvedic 'Rasayana' drug. The free radical scavenging potential of 50% aqueous alcoholic extract of H. auriculata (HAEt) was studied for DPPH scavenging activity, nitric oxide, hydroxyl radical and ferryl bipyridyl complex scavenging activity along with lipid peroxidation and total antioxidant capacity inhibition using Thiobarbituric acid reactive substances (TBARS) in rat liver homogenate. Integral antioxidative capacity was determined by photochemiluminescence assay. HAEt showed good radical scavenging activity at various concentrations $(200-1000\;{\mu}g/ml)$ against 1,1 diphenyl, 2-picrylhydrazyl (DPPH) (32.32-77.02%) with moderate scavenging activity against Nitric oxide (12.46-52.84%), hydroxyl radical (11.69-55.26%), ferryl bipyridyl complex (17.66-58.67%) and lipid peroxidation (0.829-0.416nmoles/mg protein). The above results indicate HAEt to be very effective antioxidant. It was suggested that the varied therapeutic activities claimed for the plant in the Indian indigenous systems of medicine may be in the part due to the free radical scavenging and/ or potent antioxidant activity.
