A Study on the Pharmacetical Characteristics & Analysis of Glycyrrhizin Extract (감초 추출물의 약리적 특성 및 분석)

  • From experiment results on pharmacetical characteristics and analysis of Glycyrrhizin extract, some conclusions are obtained as follows. From results on extract experiment of Glycyrrhizin, it appeared about 8%-extraction ratio as semi-solid state, and after dried in freezing from Glycyrrhizin extract of semi-solid state, it obtained about 70%-Glycyrrhizin extract as solid state of yellow gold color. From results on antimicrobial experiment of Glycyrrhizin extract, number of S-typhimurium and Fungus in microbe decreased more and more according to time passage. This phenomenon shows that Glycyrrhizin extract keeps antimicrobial effect. From results on antioxidation experiment of Glycyrrhizin extract, DPPH scavenging activity of free radical shows that Glycyrrhizin extract appears more remarkable reduction ability than reference samples. This phenomenon means that antioxidation of Glycyrrhizin extract appears higher than Vitamin-C and BHA. From results on instrument analysis, the fatty and aromatic components of 2-pentanone, cyclohexasiloxane, tetrasiloxane, benzoquinoline-2-carboxylic acid etcs from Glycyrrhizin extract was detected with GC/MS and inorganic components of Ca, Mg, Ti, Zn, Fe etcs from Glycyrrhizin extract was detected with ICP/OES.

The Effect of Long-term Exercise of Different Intensity on the Activation of Antioxidation Enzyme and Lipid Peroxidation (장기간 다른 강도의 운동이 항산화 효소의 활성도와 지질 과산화에 미치는 영향)

  • This study was to investigate the effect of 10th weeks Exercise of Different Intensity on the activation of antioxidation enzyme and lipid peroxidation. 15 subjects were divided into the 65$\%$ running exercise(EG; 5), 85$\%$ running exercise(EG: 5), and the control(CG: 5) groups. The exercise group had 10 week of running exercise 4 times a week with ACSM(1995) protocol, 65$\%$ of HRmax from the beginning to 5th week, 85$\%$ of HRmax from 6th to 10th week. The subjects had 60 min exercise for each day. After extracting the blood sample, the activation of antioxidation enzyme and lipid peroxidation were compared between the groups. The one way ANOVA was conducted about pretest, 5th week, and 10th week of the subject data. The results obtained from this study were as follows; 1. The 65$\%$ exercise group showed the significantly greater increase in the activation of SOD after 10 weeks for the rest and all-out exercise. (p < .05). 2. The 65$\%$ exercise group displayed the significantly higher increase in the activation of CAT after 10th weeks for the rest exercise. (p < .05). 3. The 65$\%$ exercise group had the significantly greater decrease in the activation of MDA after 10 weeks for the all-out exercise. (p < .05).

Antioxidative Properties of Amaranth Cauline Leaf and Suppressive Effect against CT-26 Cell Proliferation of the Sausage Containing the Leaf

  • The study investigated antioxidative properties and rectal cancer cell inhibition effect of amaranth (Amaranthus cruentus L.) cauline leaves (ACL) to produce the sausage with ACL powder (ACLP). Antioxidative effects of ACLP prepared with different stem lengths (10-45 cm) were evaluated through DPPH, ABTS, reducing power, total phenol, and total flavonoid. Inhibition effect on rectal cancer cells growth was also examined with CT-26 cell. To determine appropriate ACL amounts in sausage formula, response surface methodology was used. The sausages without ACL (control) and the sausage with ACL (ACLP sausage) were the subjected to the examinations of antioxidation, growth inhibition on CT-26, and physicochemical properties (pH and water content). ACLP made from the leaf with 15 cm length stem generally showed the highest antioxidative effect through results of DPPH, ABTS, reducing power, total phenol, and total flavonoid. ACLP also showed inhibition effect on the proliferation of CT-26, depending on concentration of ACLP. The surface response model showed that 4.87 g of ACLP was optimized amount for sausage production. Physicochemical properties between optimized ACLP and control sausages were not significantly different. Higher antioxidative effect of optimized ACLP sausage extract was observed (p<0.05) in antioxidation tests than control sausage extract except for DPPH. Cell viability of CT-26 cells were higher (p<0.05) in ACLP than in control sausage extracts. These results indicate that ACLP has functional effects on antioxidation activity and growth inhibition on CT-26 cell, and thus, it should be useful as a supplement in sausage, which may some effect as ACLP itself.

The Study on Antioxidation of Retinal (Retinol에 대한 항산화 연구)

  • In an attempt to compare the antioxidation effects of constrain the oxidation and improve the structural stability, retinol and various antioxidants were together encapsulated by liposome. Four water soluble and four oil soluble antioxidants were tested for performance. The influence of tertiary butylhydroquinone(TBHQ), ${\alpha}$-glycosyl rutin(${\alpha}$-G rutin), licorece, pycnogenol as water soluble antioxidants and butylated hydroxytoluene(BHT), ${\alpha}$-lipoic acid, ferulic acid, natural concentrated tooopherol(no-tocopherol) as oil soluble antioxidants on the constraint of oxidation of retinol were investigated. Additional study was conducted to compare the synergic effect of antioxidation for retinol with licorice, pycnogenol, ${\alpha}$-lipoic acid and BHT. All the antioxidant used at the study constrained oxidation of retinol. The effect of antioxidation for retinol increased in order of licorice, pycnogenol, TBHQ, ${\alpha}$-G rutin as water soluble antioxidants and ${\alpha}$-lipoic acid, BHT, no-tocopherol, ferulic acid as oil soluble antioxidants. In conclusion, ${\alpha}$-lipoic acid is more effective retinol antioxidants than BHT. And the combination of ${\alpha}$-lipoic acid and BHT gave best synergic among six combinations.

Investigation of the effect of Lithospermi Radix on tight-junction related genes in HaCaT cells (자초(紫草) 열수추출물이 각질형성세포 HaCaT의 세포 연접 관련 유전자의 발현에 미치는 영향 연구)

  • Objectives : The aim of this research was to determine the diverse effects of Lithospermi Radix Water Extract (LR) on human keratinocyte HaCaT cells, and to examine whether those effects could be applied to the human skin. Methods : We examined effect of LR on the cell viability of using the MTS assay in human keratinocyte HaCaT cells. The antioxidation effect of LR was analyzed relative to the well-known antioxidant resveratrol, using an ABTS assay. Quantitative RT-PCR analysis revealed that, in HaCaT cells, LR influenced the mRNA expression of tight-junction genes associated with skin moisturization. Furthermore, a wound-healing assay demonstrated altered cell migration in LR-treated HaCaT cells. Result : The cytotoxicity was confirmed to be higher in LR at a concentration of $800{\mu}g/m{\ell}$ using the MTS assay in HaCaT cells. In comparison to $100{\mu}M$ resveratrol, $1,600{\mu}g/m{\ell}$ LR showed either a similar or superior antioxidation effect. LR treatment in HaCaT cells reduced the mRNA expression levels of claudin 3, claudin 4, claudin 6, claudin 8, and ZO-2 to less than 0.80-fold, whereas JAM-A and Tricellulin mRNA expression level increased more than 1.33-fold. In addition, HaCaT cells migration was decreased to 83.9% by LR treatment. Conclusions : LR of antioxidation activity will have an anti-aging effect on the skin by reducing oxidative stress. Further studies are required to address the implications for human skin, given LR's effects of altering mRNA expression of tight junction-related gene and decreasing cell migration of HaCaT cells.

Study of antioxidation activity and melanocyte effect of Pueraria Lobata Root Extract (갈근추출물의 항산화 활성 및 멜라닌세포 효과에 관한 연구)

  • This study investigated antioxidation activity through the content of total polyphenol, that of flavonoid and DPPH radical scavenging activity and measured the cytotoxicity against B16F10 melanoma and inhibiting function of melanin biosynthesis to evaluate antioxidation activity and melanocyte effect of pueraria lobata root extract. As the results of study, it was recognized that the toxicity did not show against B16F10 melanoma and the increase of generating melanin was inhibited as the results of measuring the inhibition function of melanin biosynthesis after inducing the generation of melanin by ${\alpha}$-MSH against B16F10 melanoma cell. The high contents of polyphenol and flavonoid was found as the contents of pueraria lobata root extract increases and DPPH radical scavenging activity as the results of antioxidation activity. Through this study, it was recognized that pueraria lobata root extract has the feasibility that can be used as the material of cosmetics as it has the excellent effect of antioxidation activity and inhibiting the generation of melanin against melanocyte, low toxicity against skin cell and its safety against melanocyte of skin was found.

Antioxidative Effects of Ethanol Extract Obtained from Rooibos Tea(Aspalathus linearis) and It's Application of Food (Rooibos Tea(Aspalathus linearis)에탄올 추출물의 항산화 효과 및 식품에 대한 응용)

  • This study was designed the antioxidative effect of the ethanol extract obtained Rooibos tea (Aspalathus linearis) on various kinds of oil, and examined the synergistic effect of Rooibos tea extract by addition of citric acid to the antioxidation activity and also investigate to antioxidation effect of the extract in food production and storage peroids. The antioxidative activity of 0.2mg of Rooibos tea extract was showed similar to same doses of $\alpha$-tocopherol, BHA and BHT in linolieic acid-ethanol system. The antioxidative effect of the mixture with 0.1% Rooibos tea extract on lard was more effective than that of the mixture with same doses of $\alpha$-tocopherol, BHA and BHT. The antioxidative effect of Rooibos tea extract was showed slightly effects on lard or soybean oil. Antioxidative effects of Rooiboe tea extract in addition of citric acid as synergist showed more effective in linoleic acid-ethanol system, but did not showed in the other oils. In the application of Rooibos tea extract to food prodctuction and storage period, the antioxidative effect was more effective in biscult, preparation added lard mixed with 0.1% Rooibos tea extract.
