The present investigation was undertaken to study the status of plasma antioxidant vitamins in normal cycling and $\alpha$-tocopherol supplemented anestrus buffalo heifers. The pre-supplementation plasma levels ($\mu$mol/L) of vitamin E and $\beta$-carotene were significantly (p<0.05) lower and of vitamin C was significantly (p<0.05) higher in anestrus heifers (4.06$\pm$0.07; 4.56$\pm$0.17; 21.04$\pm$0.21) when compared to normal cycling ones (4.92$\pm$0.05; 6.76$\pm$0.12; 14.24$\pm$0.16). The oral supplementation of$\alpha$-tocopherol at 3,000 mg per week per animal in anestrus heifers resulted in a significant (p<0.01) increase in vitamin E and $\beta$-carotene levels and a significant (p<0.01) decrease in vitamin C concentration. Results indicated that supplementation of $\alpha$-tocopherol to anestrus buffalo heifers improved the antioxidant status by mitigating the harmful effects of free radical induced oxidative stress.
The spontaneous frequency of genetic damage and the possible relationship of this damage to total radical-trapping antioxidant potential(TRAP) and antioxidant vitamins, including plasma levels of $\alpha$-carotene, $\beta$-carotene, cryptoxanthin, retinol, $\alpha$-tocopherol and ${\gamma}$-tocopherol in humans were investigated in 57 subjects using two indices of genetic damage, SCE & HFC frequency. The mean of SCE and HFC frequencies were weakly correlated with plasma TRAP(r=-0.305, p<0.1 for SCEs: r=-0.297, p<0.1 for HFCs, respectively), but those were strongly negatively correlated with plasma $\beta$-carotence(r=-0.385, p<0.01 for SCEs : r=-0.392, p<0.01 for HFCs) and cryptoxanthin(r=-0.312, p<0.05 for SCEs : r=0.335, p<0.05 for HFCs, respectively) levels in the subjects. However, those DNA damage markers including SCE and HFC did not correlate with either plasma $\alpha$-carotene, $\alpha$-tocopherol or retinol concentrations. The mean of SCE and HFC frequencies were positively correlated with plasma ${\gamma}$-tocopherol level(r=0.421, p<0.01 for SCEs : r=0.399, p<0.01 for HFCs, respectively). These findings indicate that increased cytogenetic DNA changes, as determined by SCE and HFC frequencies are possibly associated with generation of free radicals in lymphocytes and decreased plasma antioxidant vitamin($\beta$-carotene and cryptoxanthin) status in the subjects. In this study the in vitro protective effects of several antioxidant vitamins (vitamin C, $\alpha$-tocopherol, $\beta$-carotene), fruits and vegetables (strawberry, tangerine, orange and 100% orange juice, carrot juice), on the levels of isolated human lymphocyte DNA damage was measured using Comet assay. Comet assay has been used widely to assess the level of the DNA damage in the individual cells. Lymphocytes were pre-treated for 30 minutes with antioxidant vitamins (10, 50, 100, 500 $\mu$M) or fruits$.$vegetables (10, 100, 500, 1000 $\mu$g/ml), an4 then oxidatively challenged with 100 $\mu$M $H_2O$$_2$ for 5 min at 4$^{\circ}C$. The protective effect of antioxidant vitamins against DNA damage at a concentration of 50 $\mu$M were 50% in vitamin C, 32% in $\alpha$-tocopherol, whereas, fJ-carotene showed a 55% protection at a dose as low as 10 $\mu$M. The inhibitory effects of DNA damage by strawberry, tangerine, orange, orange juices, carrot juices were 50 - 60% with wide ranges of doses. The results of the present study indicate that most the antioxidant vitamins and fruits$.$vegetables juices produced a significant reduction in oxidative DNA damage.
The purpose of this study was to find the extent of lipid peroxidation of erythrocytes in cigarette smokers, and to determine the relative effectiveness of $\beta$-carotene, canthaxanthin , and $\alpha$-tocopherol as antioxidants. Thirty smokers and 30 nonsmokers participated in this study . No significant differences according to age, sex, and height were shown. Cigarette smokers in this study had higher hemoglobin concentrations and more oxidation of hemoglobin than non-smokers. In addition, the erythrocytes of cigarette smokers had significantly higher MDA concentrations than crythrocytes of nonsmokers, which suggests that smokers may have tocopherol were studied in vitro by measuring the concentration of malondialdehyde(MDA) and precent hemolysis of erythrocytes. The addition of any antioxidant to erythrocytes significantly decreased MDA concentrations(p<0.05) while antioxidants showed nonsignificant inhibition of hemolysis. Among the antioxidant used in this study, canthaxanthin showed the greatest inhibition of both lipid peroxidationand hemolysis. Meanwhile, $\alpha$-tocopherol showed potent inhibition of lipid peroxidation, but not of hemolysis.
A large body of epidemiologic evidence suggests inverse relationships between ischemic heart disease and plasma vitamin C and E concentrations. Smokers have lower plasma concentrations of these vitamins than do nonsmokers. Smokers therefore need antioxidant vitamin supplementation . The purpose of study was to investigate the effect of antioxidant vitamin supplementation on blood composition in smoking college men. 24 subjects were divided into 3 groups of which were the vitamin C supplementation group(n=8), the vitamin E supplementation group(n=8), and the vitamin C+E supplementation group(n=8). The vitamin supplementation group consumed 500mg of ascorbic acid, the vitamin E supplementation group consumed 200IU of D-$\alpha$-tocopherol , and the vitamin C+E supplementation group consumed 500mg of ascorbic acid +200IU of D-$\alpha$-tocopherol for 4 weeks. We examined the blood compositions of the volunteers bofore and after vitamins were supplemented . The results obtained were as follows ; intakes of energy , carbohydrate , fat protein , vitamin C and vitamin E were not significantly affected by vitamin supplementation in all groups. Blood glucose concentrations were not significantly affected by vitamin supplementation in all groups. Concentrations of plasma uric acid and alkaline phosphatase activity were decreased significantly (p<0.05) with vitamin E supplementation. The results of this study show that antioxidant vitamin supplementation in smokers has a tendency to decrease coronary heart disease risk.
This study was done to evaluate the antioxidant status of female college students by determining their intakes and plasma levels of antioxidnt vitamins (vitamin C, A and E) and total antioxidant status (TAS). Subjects were 46 healthy female college students aged 20 - 29 years. Body composition was determined by a multifrequency bioelectrical impedance analysis. Dietary intakes were examined by 24hr record method and nutrients intakes were analyzed by the Computer Aided Nutritional analysis program for professional (CAN-pro). Plasma vitamin C level were measured by spectrophotometric method and retinol, ${\beta}$-carotene, ${\alpha}$-tocopherol were measured by HPLC. Plasma TAS was measured with a Randox kit using the trolox equivalent antioxidant capacity (TEAC) method. Daily energy and protein intakes of the female college students were 1670.5㎉ (83% of RDA) and 63.3g (115.1% of RDA), respectively. However their intakes of Ca and Fe were below 75% of RDA. Their intakes of vitamin A and C were 596.6 ${\mu}$ gRE (85.2% of RDA) and 71.0mg (101.4% of RDA), respectively. Plasma levels of vitamin C, retinol, ${\beta}$-carotene and ${\alpha}$-tocopherol were 14.7mg/L, 0.7mg/L, 0.2mg/L and 9.1mg/L, respectively which were within normal range. There was no subject with deficiency or marginal level in plasma vitamin A and C. However 1.6% of the subjects had below adequate level in vitamin E. Plasma TAS level was 1.2mmol/L. Correlation data showed that all plasma antioxidant vitamins were positively correlated with plasma TAS. Overall data indicate that the antioxidant status of female college students were pretty good. However it might be necessary to educate them to eat more fruits and vegetables for preventing many chronic diseases in a later life. (J Community Nutrition 5(1) : 13∼20, 2003)
This study was performed to investigate the change of biologically functional compounds and antioxidant activities in Ecklonia cava with blanching times. As biologically functional compounds, the contents of minerals(K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins($vitamin\;C,\;{\beta}-carotene\;and\;{\alpha}-tocopherol$) and total polyphenol were analyzed. And antioxidant activity was determined through free radicals(DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and linoleic acid peroxidation inhibitory activity. As the blanching time increased, the contents of minerals, vitamin C, ${\beta}-carotene$ and total polyphenol were decreased however ${\alpha}-tocopherol$ was not affected by blanching time, and antioxidant activities were decreased with blanching time.
The aim of this study was to investigate the possible relationship between antioxidant vitamin levels and malignant pleural mesothelioma (MPM). For this purpose, we measured the serum levels of 4 antioxidant vitamins, ${\beta}$-carotene, ${\alpha}$-tocopherol, retinol, and ascorbic acid, in patients with environmentally induced MPM and in healthy controls from one tremolite village (Kureysler), the biggest erionite village (Tuzkoy) and Ankara. A total of 160 subjects were enrolled in the study, 42 (26.3%) diagnosed with MPM and 118 (73.7%) healthy subjects. A comparison was made between the MPM group and three control groups of which two were exposed and one was unexposed to mineral fibers. The study population consisted of 82 males (51%) and 78 females (49%) with a mean of age of $44.8{\pm}14$ years (range; 20-65 years). Lowest levels of ${\beta}$-carotene, ascorbic acid, and ${\alpha}$-tocopherol were found in MPM patients (MPM vs control groups combined, p<0.0001 for each antioxidant vitamin), without any relation to age or sex. There was no significant difference between the antioxidant levels of healthy controls of Tuzkoy and Ankara. In conclusion; our findings suggested an increased risk of MPM being associated with low levels of ${\alpha}$-tocopherol and ascorbic acid in patients with MPM.
This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. This study examined the changes in the chemical components and antioxidant activity of Ecklonia cava according to the drying methods. As chemical components, the concentrations of minerals(K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins(vitamin C, ${\beta}$-carotene and ${\alpha}$-tocopherol) and the total polyphenols were analyzed. In additions, the antioxidant activity was determined by measuring the free radical(DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and the linoleic acid peroxidation inhibitory activity. The mineral content was not affected by the drying methods. However, more vitamins were lost and the total polyphenol concentration was reduced as a result of sun-drying than by the other drying methods used. More of the total polyphenol was preserved by freezing-drying than by any of the other drying methods, which meant that there was a higher antioxidant activity after freeze drying.
