철분 투여한 돼지에서 비타민 C의 항산화 작용 (Antioxidant activity of vitamin C in iron-overloaded swine plasma)

  • Iron deficient anemia in piglets could be overcome by supplementary iron. Overloaded iron induced peroxidation of cell membrane and increased malonaldehyde (MDA). Antioxidant activity of vitamin C has been studied in iron-overloaded swine plasma. Erythrocyte fragility, MDA, glutathione, vitamin A, and vitamin E were measured in swine plasma with or without iron (0~1mg/dl) and vitamin C (0~10mg/dl). Erythrocyte fragility increased from 8% to 45% in iron group and reduced from 57% to 43% in vitamin C group with dose dependant response. MDA was $0.94{\pm}0.05$ and $1.86{\pm}0.10$ nmol/ml in piglet and pig, respectively, and significantly high in pig (p<0.05). Iron increased MDA from $1.86{\pm}0.10$ to $9.46{\pm}0.04$ nmol/ml in pig, but not in piglet (p<0.05). Vitamin C reduced MDA from $9.46{\pm}0.04$ to $4.80{\pm}0.10$ nmol/ml in pig. Iron increased glutathione from $90.12{\pm}0.10$ to $108.52{\pm}5.29$ nmol/dl in pig, and vitamin C reduced glutathione from $108.52{\pm}5.29$ to $93.52{\pm}2.44$ nmol/dl (p<0.05). Vitamin A and E were $24.86{\pm}2.70$ to $138.29{\pm}6.70{\mu}g/dl$, respectively in iron group, and $35.76{\pm}0.60$ to $177.21{\pm}2.95{\mu}g/dl$, respectively in supplementary vitamin C group (p<0.05). These data indicated an antioxidant activity of vitamin C in iron-overloaded swine plasma.

노인에서 비타민 E 보충이 면역능력과 항산화상태에 미치는 영향 (Effects of Vitamin E Supplementation on Immune Response and Antioxidant Defense Parameters in Healthy Korean Elderly Women)

  • This study investigated the effects of vitamin E supplementation on immune responses and antioxidant status in healthy Korean old and young women. Blood samples were obtained from 15 healthy old women (over 60 years old) and from 15 healthy young women(20 years old) before and 4 weeks after vitamin E( tocopherol acetate) supplementation(400IU/day). Daily nutrient intakes were calculated, and plasma vitamin E concentration, numbers and percentages of white blood cell and their subpopulation, percentages of lymphocytes and subpopulation, NK cell percentages, plasma immunoglobulin A, G, M and C3 concentration, proliferation of PMN with mitogen were measured. Also plasma TBARS concentration and radical scavenger activity of erythrocytes were investigated. Plasma vitamin E concentrations were significantly increased after supplementation in both groups. In elderly women, vitamin E supplementation restored the per centages of neutrophils, lymphocytes, and eosinophils which had been out of normal ranges before supple mentation. And after vitamin E supplementation, helper T cell percentages significantly increased in elderly. Plasma immunoglobulin and complement C3 concentrations were not affected by vitamin E supplementation in both groups. PMN proliferations with mitogen were significantly lower in old women than in young women, and there was no effect of vitamin E supplementation. Vitamin E supplementation significantly decreased plasma TBARS concentrations in old and young women. RSA of erythrocytes was increased in both groups, but the statistical significant was only found in young women group. Therefore, these results suggest that the moderate vitamin E supplementation in old women improves immune responses, especially nonspecific immunity and cell mediated immunity, via protection of oxidant stress.

Induction of Quinone Reductase , an Anticarcinogenic Marker Enzyme, by Vitamin E in Both Hepalclc7 Cells and Mice

  • Induction of NAD(P)H : (quinone-acceptor) oxidoreductase (QR) which obligatory two electron reduction of quinones and prevents their participation in oxidative cycling and thereby the depletion of intracellular glutathione, has been used as a marker for chemopreventive agents. We postulated that vitamin E, an antioxidant, which induces QR as the gene of QR was reported to contain antioxidant reponsive element in the 5'-flanking region. Vitamin E resulted in significant induction of QR in both hepalclc7 cells and mouse tissues. QR induction was observed; to be maximal at 25uM vitamin E for hepalclc7 cells while it was maximal in the level of 2.5∼5 μmoles vitamin E/㎏ BW for mouse tissues. Thus the cancer-preventive effect of vitamin E may be exerted by it induction of intracellular detoxifying enzymes.

Lactobacillus rhamnosus CBT-LR5 Improves Lipid Metabolism by Enhancing Vitamin Absorption

  • Probiotics provide a symbiotic relationship and beneficial effects by balancing the human intestinal microbiota. The relationships between microbiota changes and various diseases may predict health abnormalities and diseases. Treatment with vitamins and probiotics is one therapeutic approach. To evaluate the effect of probiotics on vitamin absorption, we chose Lactobacillus rhamnosus CBT-LR5 treatment, which has resistance to vitamin C-inducible toxicity, with vitamins in high-fat diet (HFD)-induced obesity models. CBT-LR5 affected the absorption of micronutrients, such as ionic minerals and water-soluble vitamins. An increase in vitamin C absorption by CBT-LR5 enhanced the antioxidant response in HFD-induced obesity models. Increased vitamin B absorption by CBT-LR5 regulated lipid metabolism in HFD-induced obesity models. These favorable effects of CBT-LR5 on the absorption of vitamins should be investigated as candidate therapeutic target treatments for metabolic diseases.

Aflatoxin $B_1$ 투여 마우스의 간 기능 효소 및 간 손상에 미치는 항 산화비타민의 효과 (The Effect of Antioxidant Vitamins on Liver Function Enzymes and Hepatic Damage of Aflatoxin $B_1$ treated mice)

  • Aflatoxin $B_1(AFB_1)$ is a potent hepatotoxic and hepatocarcinogenic mycotoxin in human beings. It is accumulated in animal tissues and injured cell through variable metabolic pathway. This study was conducted to determine the effect of antioxidant vitamins on liver function enzymes and hepatic damage of $AFB_1$ treated mice. The 6 weeks old male ICR mice were randomly separated 6 groups, vehicle solvent or vitamin C(10 mg/kg/day) and vitamin E(63.8 mg/kg/day) were administered by intraperitoneal(i.p.) injection and 1 hr later, vehicle solution(DMSO) or $AFB_1$(0.4 mg/kg) were injected. The results obtained as follow ; The levels of liver function enzymes such as GOT, GPT, LDH, and alkaline phosphatase, in sera of mice were remarkably elevated by treatment with $AFB_1$ only. However, those enzymes were significantly alleviated by co-treatment with antioxidant vitamins(p<0.01). Especially the levels of LDH and ALK phosphatase were similar to those of control groups(p<0.01). The transmission electron microscopy(TEM) image of intracellular microrganelles on the liver cell of mice was also degenerated extremely by treatment with $AFB_1$, but vitamin C and vitamin E gave good effects on cellular deformation. The intracellular microrganelles such as mitochondria, endoplasmic reticulum, nucleus and nucleic membrane were nearly disappeared the cellular deformation by antioxidant vitamins co-administration. With above results, we could estimated that antioxidant vitamins blocked AFB1 induced hepatic cell damage.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

Antioxidant, antibacterial, and antiinflammatory effects of yoghurt made with vitamin tree (Hippophae rhamnoides L.) fruit powder

  • It is well known that the fruit of the vitamin tree (Hippophae rhamnoides L.) provides excellent anti-diabetic, antibacterial, immune regulation, anti-inflammatory, and anti-aging effects. In some countries including Europe the fruit has been added to certain foods to develop functional foods. The present research was carried out to elucidate the biological function of vitamin tree fruit powder added to fermented milk. It was found that there was an antioxidant effect of yoghurt made with vitamin tree fruit powder, and this effect was greater with increased incubation time and amount of vitamin powder, as shown by 1,1-diphenyl2-picrylhydrazyl (DPPH) and 2,2-anziobis (3-ehtylbenzothiazoline-6-sulfonic aicd) (ABTS) radical scavenging activities. The antibacterial effect of yoghurt containing vitamin tree fruit powder was shown to be effective against four pathogenic microorganisms, Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonela Typhimurium. In particular, yoghurt supplemented with 5% of vitamin tree fruit powder showed the best antibacterial effect. The yogurt containing the vitamin tree fruit powder significantly inhibited the expression of the anti-inflammatory cytokines interleukin (IL)-6 (yogurt [Y] + Hippophae rhamnoides L. powder [HP] and yogurt containing 5% Hippophae rhamnoides L. powder [HPY]) and IL-1β (HP, Y + HP and HPY) in a concentration-dependent manner among tumor necrosis factor (TNF)-α, IL-6, IL-1, and induced nitric oxide synthase (iNOS). Our results clearly demonstrated that vitamin tree fruit powder could be a good functional ingredient for improving health through yoghurt manufactured with vitamin tree.

Assessment of Phytochemicals, Quality Attributes, and Antioxidant Activities in Commercial Tomato Cultivars

  • To assess South Korean commercial tomato cultivars, regular and cherry tomato cultivars were grown in the greenhouse and evaluated for color attributes, titratable acidity, pH, total soluble solids, carotenoids (lycopene and ${\beta}$-carotene), total phenols, flavonoids, vitamin C, and antioxidant activity. Significant differences (p < 0.05 using Duncan's multiple range test (DMRT)) were observed in the levels of most phytochemicals, quality parameters, and antioxidant activity among the twenty South Korean tomato cultivars tested. Lycopene and ${\beta}$-carotene contents varied significantly (p < 0.05 using DMRT), from $0.95mg{\cdot}100g^{-1}$ to $5.12mg{\cdot}100g^{-1}$ and $0.65mg{\cdot}100g^{-1}$ to $3.56mg{\cdot}100g^{-1}$ of fresh weight, respectively. ${\beta}$-carotene contents exhibited the highest genetic variation (59.2%), followed by naringenin (52.8%) and other phytochemicals. Most of the cherry tomato cultivars had statistically higher levels (p < 0.05 using DMRT) of carotenoids, phenols, flavonoids, vitamin C, and antioxidant activity compared to the regular tomato varieties, suggesting their higher nutritional value. Lycopene content was highest in the cultivars YoYo, Jicored, Titi-Chal, TY-Endorphin, and Rubyking. Cultivars Rubyking, TY-Endorphin, and Titi-Chal also showed relatively higher antioxidant activities in three assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric ion reducing antioxidant power (FRAP) assays. All the antioxidants, except luteolin, were positively correlated with antioxidant activities; the highest correlation was observed between total phenol and antioxidant activities, followed by the correlation between rutin and vitamin C. Cultivars identified to have superior nutritional status would be useful in tomato breeding programs to further improve quality and health benefits of tomatoes for the fresh and processed markets.

Antioxidant Potential in the Fruits of Pyrus Species (Pear) in Korea

  • Five samples (Bakwoonbae, Chungsilbae, Sandolbae, Dolbae, and Chuwhangbae) of Pyrus species differing in their origin of production were analyzed for total phenolic content, vitamin C content, and antioxidant activities. Two antioxidant activities were measured by the free-radical scavenging activity and reducing power method. Total phenolic content of the Chungsilbae extract (20.35 mg/g) was higher than those of other pears. Vitamin C content in five Pyrus species ranged from $420.32\;{\mu}g/g$ in Sandolbae to $659.75\;{\mu}g/g$ in Bakwoonbae. Chungsilbae also had the highest free radical scavenging activity (76.56%) and reducing power (0.35).

비타민 C 보충이 제 2형 당뇨병 환자의 혈당 및 항산화상태에 미치는 효과 (Effect of Vitamin C Supplementation on Blood Sugar and Antioxidative Status in Types II Diabetes Mellitus Patients)

  • Purpose: This study was to determine the effect of oral vitamin C supplements on blood sugar and antioxidative status in Types II diabetes mellitus patients. Method: Data for the study were collected from June 24 to August 31, 2001. Participants(31) took 1g/day vitamin C for 4 weeks, after a 1 - week taking no Vitamin C, followed by Vitamin C 3g/day for 4 weeks. A baseline blood sample was obtained following a 12hour overnight fast and at the end of each 4week Vitamin C administration. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c, superoxide scavenging activity and hydrogen peroxide scavenging activity. The data were analyzed by SPSS for repeated measures ANOVA. Result: Plasma vitamin C concentration was significantly increased over dose(F=3.316, p=.043). Fasting blood sugar and HbA1c was significantly decreased over dose(F=13.192, p=.000; F=11.995, p=.000). Superoxide scavenging activity and hydrogen peroxide scavenging activity was significantly increased over dose(F=486.138, p=.000; F=177.704, p= .000). Conclusion: The results suggest that megadose vitamin C supplementation may have a beneficial effect in diabetes mellitus patients on both glycemic control and antioxidant status. Thus dietary measures to increase plasma vitamin C may be an important health strategy for reducing the compliance of diabetic patients
