Identification of Polyphenol Substances (MP-1) from Seagrass, Phyllospadix Japonica Makino (잘피 게바다말로부터 폴리페놀 물질(MP-1)의 분리 및 동정)

  • BACKGROUND: Phyllospadix japonica Makino is a perennial plant belonging to the family Zosteraceae. This species is native to the eastern coast of the Korean Peninsula, and it is found attached to rocks on the seashore. As with all seagrass species, P. japonica is also known to play a major role in protecting the marine environment, and studies on its physiological activities suggestive of its antioxidant, antibacterial and anti-inflammatory potentials have been reported. In this study, purification and structural analysis were performed to identify the polyphenol substances derived from P. japonica. METHODS AND RESULTS: An polyphenol substance MP-1 was purified from the 70% aqueous methanol extract of P. japonica using Diaion® HP-20 column chromatography, ODS column chromatography, and medium-pressure liquid chromatography (MPLC). The purified MP-1 was identified as rosmarinic acid having a molecular weight of 360 and a molecular formula of C18H16O8 through electrospray ionization (ESI)-mass and nuclear magnetic resonance (NMR) spectroscopic analysis. CONCLUSION(S): This study highlights the processes used for the identification of the polyphenol substance derived from P. japonica. Rosmarinic acid, the polyphenol derived from P. japonica identified by this study, is a kind of bioactive substance mainly present in plants. These findings provide an important starting point and are valuable for future studies on bioactive substances in seagrass.

Integral Antioxidative Capacity of Extracts by Pressurized Organic Solvent from Natural Plants in Jeju (제주 자생식물 고압용매 추출물의 통합적 항산화 능력)

  • Twenty natural plants from Jeju were extracted by pressurized organic solvent (100% methanol, $40^{\circ}C$, 13.6 MPa, 10 min). Extraction yield, total phenolic content (TPC) and integral antioxidant capacity were measured, and each component was identified by GC/MS. Extraction yields were high as 21.8%, 21.5, 21.1, 20.7 and 20.1% in Rhus javanica, Euscaphis japonica, Alnus firma, Sapium japonicum and Sorbus alnifolia, respectively. The extracts containing high TPC (mg GAE/g of dry sample) were obtained from Malus sieboldii (68.3), Sapium japonicum (57.6), Pyrrosia lingua (56.6) and Euscaphis japonica (55.1). Integral antioxidant capacities of water-soluble substances were 598, 394, 293 and $270\;{\mu}mol$ ascorbic acid equivalent/g in Geranium thunbergii, Sapium japonicum, Cornus kousa and Rhus javanica, respectively. Integral antioxidant capacities of lipidsoluble substances were 611, 314, 296 and $242\;{\mu}mol$ trolox equivalent/g in Ardisia crenata, Ostrya japonica, Geranium thunbergii and Quercus acuta, respectively. Fifteen major peaks were identified by GC/MS from the extract of pressurized organic solvent from Sapium japonicum. Two polyphenols (gallic acid (retention time (RT) 19.7 min)) and quercetin (33.5 min)), ascorbic acid (RT 35.3 min), and several fatty acids (retention time 18.6, 21.0, 21.8, 21.9 and 23.6 min) were identified, and gallic acid was the major polyphenol component due to high peak area.

A Study on the Status of Antioxidant Nutrients and Lipid in the Middle-aged Korean Men Living in Taegu (대구지역 중년 남성의 혈중 항산화성 영양소와 지질상태에 관한 연구)

  • Serum levels of antioxidant vitamins and lipids were determined along with anthropometric measurements in 174 healthy male subjects with mean age of 50.3$\pm$6.8 years from Taegu area in Korea. Body mass index (BMI) and waist/hip(W/H) ratio of the subjects were 23.18$\pm$2.46 and 0.88$\pm$0.04, respectively and their systolic and distolic blood pressures were 127.8$\pm$15.5 and 83.9$\pm$10.8mmHg. Twenty one percent of the subject had BMI over 25. Average seum levels of total cholesterol, LDL- and HDL-cholesterol and triglyceride were 187.7$\pm$34.9, 117.6$\pm$33.5, 41.1$\pm$9.0 and 140.7$\pm$83.6mg/dl, respectively. Sixteen percent of the subject had LDL-chole-sterol over 130mg/dl. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subject was 2.01$\pm$0.77 MDA nmoles/ml and those of $\alpha$-tocopherol, retinol. ascorbic acid and sum of $\alpha$- and $\beta$-carotene were 9.59$\pm$3.11$\mu\textrm{g}$/ml, 1.15$\pm$ 0.38$\mu\textrm{g}$/ml, 10.5$\pm$3.8$\mu\textrm{g}$/ml and 64.6$\pm$43.$\mu\textrm{g}$/dl, respectively. About 14% of the sujects had low vitamin E status of less than 7.0$\mu\textrm{g}$/ml,. while 6% had low vitamin C status of less than 4.0$\mu\textrm{g}$/ml, Serum vitamin E showed positive correlations with total cholesterol and LDL-cholesterol and triglyceride, but no correlation with TBARS. Fatty acids of serum total lipid were composed of 42.9% as saturnted. 19.3% as monounsaturated and 36.7% polyunsaturated fatty acids(PUFA). N-6 and n-3 PUFA were 27.7% and 8.3% of total fatty acids. N-6/n-3 PUFA ratios were negatively correlated both with serum total cholesterol and TBARS.

Isolation and Structural Identification of Antioxidant Substances from Ethyl Acetate Extract of Conyza canadensis (망초(Conyza canadensis) Ethyl Acetate 추출물의 항산화성 물질의 분리와 동정)

  • Background: As a result of analyzing the components of wild Conyza canadensis, it contains physiologically active ingredients, so it is necessary to identify the compound. Purposes: It was to study the compound's molecular structure; a previous study showed that C. canadensis contains antioxidant substances. Methods: The ultrasonic pulverized lysate of C. canadensis stem and leaves was first extracted with 90% methanol and then five organic solvents. Next, the extracts was fractionated by HPLC, LC/MS chromatography, and NMR analyzers identified the molecular structure. Results: 100 g of dry C. canadensis was sonicated in 90% methanol and concentrated under reduced pressure to 11.96 g of a crude extract. Then, this crude was extracted with five types of solvents to obtain 123.8 mg of n-hexane, 448.2 mg of dichloromethane, 1047.7 mg of ethyl acetate (EA), 2563.8 mg of butanol, and 7.04 g of water. The EA extracts were fractionated by LC-MS and then re-fractionated to obtain F1 to F20. Next, the F15 was further fractionated to obtain nine fine fractions. Finally, the F17 fraction was re-fractionated to obtain ten fine fractions. As a result of LC-MS and NMR spectrometer analysis of the F15-7, the structure of this compound was confirmed as 3,5-dicaffeoylquinic acid. As a result of examining the structures of the F17-4 and F17-5 fractions, Quercetin-3-o-β-galactose was identified. In addition, the form of the F17-10 was confirmed to be 1,3,4-tri-caffeoylquinic acid. Conclusions: This study demonstrated that C. canadensis contained phenolic antioxidants, and its utilization may be expected.

Effects of Dietary Cadmium on the Respiratory Burst of Phagocytes and the Antioxidant Defense in Cultured Red Seabream (Pagrus major)

  • To examine effects of cadmium on the respiratory burst of kidney phagocytes and antioxidant defense in liver, juvenile red seabream Pagrus major were fed a cadmium-incorporated diet $(1g\;CdC1_2/kg\;diet)$. The respiratory burst activity measured by chemiluminescence (CL) was significantly reduced by oral intake of cadmium. Lipid peroxidation in liver expressed as thiobarbituric acid reactive substances (TBARS) was significantly higher in the fish fed a cadmium-incorporated diet than that of the fish fed a control diet both on Day 3 and Day 9. Liver Glutathione S-transferase (GST) activitiy was significantly increased both on Day 3 and Day 9 by feeding a cadmium-incorporated diet, when compared with the controls. From the present results, it can be concluded that oral intake of cadmium in red seabream is associated with marked reduction of respiratory burst capacity of kidney phagocytes which can elevate susceptibility of fish against infecting pathogens. Cadmium administration also elicits significant increment of lipid peroxidation in liver, and fish try to detoxify cadmium by increasing GST activity.

Anti-Oxidant and Anti-Aging Effect of Supercritical Fluid Extraction of Seed of Euphorbia lathyris L. as a Pharmacopuncture Material (한방약침소재로써 속수자 초임계추출물의 항산화 및 항노화에 대한 연구)

  • Objectives : The purpose of this study was to investigate the anti-oxidant and anti-aging effect of the seed of Euphorbia lathyris L. extracted by supercritical CO2. Methods : Human dermal fibroblast cells dosed with the extract from Euphorbia lathyris L. were harvested and the intracellular proteome was analyzed to examine the expression of proteins related collagen synthesis pathway, metalloproteinases (MMPs), extracellular matrix (ECM)-cell interaction, cytokines, and antioxidant enzymes by 2-dimensional gel electrophoresis. Results : Fatty acid analysis of the extract from Euphorbia lathyris L. showed oleic acid was 84% and linoleic acid was 4.1%. Antioxidative effect was about 53% by beta carotene bleaching assay. In 2-dimensional polyacrylamide gel electrophoresis (2-D PAGE) analysis, fifteen protein changes in five mechanisms which were collagen synthesis pathway, MMPs, ECM-cell interaction, cytokines, and antioxidant enzymes were analyzed. Conclusions : This study suggests the supercritical extraction from the seed of Euphorbia lathyris L. could be used as anti-oxidant substances for pharmacopuncture.

Effect of Persimmon Peel (Diospyros kaki Thumb.) Extracts on Lipid and Protein Oxidation of Raw Ground Pork During Refrigerated Storage

  • The inhibition effect of persimmon peel extracts (PPE) (0.05(PPE-0.05), 0.1(PPE-0.1), and 0.2 g(PPE-0.2) per meat sample) on lipid and protein oxidation of pork patties during chilled storage for 12 days were investigated and compared to ascorbic acid (As-0.05) and butylhydroxytoluene (BHT) (BHT-0.01). The meat samples treated with PPE had greater (p<0.05) $a^*$ values comparing control in raw pork patties meat from day 4 of storage. The addition of PPE at all concentrations on meat samples effectively inhibited the formation of oxidation products as shown by decreasing conjugated dienes (CD), peroxide values (POVs), thiobarbituric acid reaction substances (TBARS), and carbonyl content during chilled storage for 12 d. The PPE-0.2 and BHT-0.01 had the lowest in decrease rate of free thiol content (0.24 and 0.22 times) during chilled storage. Therefore, results of this study suggest that PPE can be considered a potential antioxidant against lipid and protein oxidation of raw meat products.

Biological Parameters for Assessing Radioprotective Effects in ${\gamma}-irradiated$ Mice (감마선에 조사된 생쥐에 있어서 방사선방어효과 평가를 위한 생물학적 파라메타)

  • This study deals with the biological changes in mice after ${\gamma}-irradiated$. Four weeks old BALA/c mice were irradiated with 6.5Gy of ${\gamma}-ray$ on the fifth day after oral administration of radioprotectants such as ascorbic acid, tocopherol and cysteine. Control group was irradiated with 6.5Gy without pre-administration of radioprotectors. Blood cells and sperm cells were counted and body, testis and spleen were weighed 3 days after irradiation. And also liver antioxidant activity and range of spleen immune cells were measured. Differences in most biological parameters were not clearly distinguished between experimental groups. However, the relative spleen weight, the relative testis weight and the population size of spleen immune cells such as T helper cells, B cells and macrophages measured by means of FACS showed significant difference between irradiated and radioprotectant administered group. It is concluded that the relative spleen weight, the relative testis weight and the population size of spleen immune cells are easy and useful parameters for assessing the effect of radioprotective substances and for quantifying biological damage of radiation, as well.

  • This study purposed to look for its uses by analyzing useful substances in kenaf (Hibiscus cannabin us L.) leaves based on the total polyphenol and flavonoid contents in the leaves, DPPH radical scavenging activity of leaf extract, SOD activity, etc. by different growth stage and variety. According to the results of experiment, the total polyphenol content was highest in Everglade-41 (27.9 mg/g dw.), the total flavonoid content was highest in Tainung-2 (42.3 mg/g dw.), and SOD activity was highest in Dowling (96.1 %). However, DPPH radical scavenging activity was similar among the three varieties.

Cell Proliferation and Antioxidative Effects of Ultrasonic Coffee Extracts

  • Recently, coffee is the most popular beverage for modern people. A great number of substances are found in coffee beans and have been studied for many years such as aliphatic and aromatic compounds. However, studies on the physiological activity of coffee extracts are insufficient. This study was performed to determine the contents of caffeine and chlorogenic acid in coffee extracts according to the solvent and to investigate the physiological activity of coffee extracts. Coffee extracts were extracted by ultrasonication method with various types of solvents including distilled water, ethanol, and other organic solvents under $50^{\circ}C$ and $80^{\circ}C$. The contents of caffeine and chlorogenic acid in coffee extracts were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Also, cytotoxic and antioxidative effects of coffee extracts were evaluated with MTT and DPPH assays to analyze the physiological activity. As a result, it was confirmed that caffeine and chlorogenic acid contents were extracted in distilled water with the highest rate. Antioxidative activity was observed below 10-fold dilute of coffee extracts, however cytotoxicity was not observed. In conclusion, distilled water was the best solvent for extracting caffeine and chlorogenic acids from coffee bean with ultrasonication and these coffee extracts are less cytotoxic in human skin cell lines and have antioxidant effect.
