Recently, we reported that Jeju ground water contains vanadium components and exerts antioxidant effects in vitro and in vivo via the scavenging of reactive oxygen species and enhancement of antioxidant enzyme activities. In the present study, the antioxidant actions of Jeju ground water were compared with those of tap water against gamma-ray radiation in mice. C57BL/6 mice were irradiated with gamma-ray at a dose rate of 2 Gy. The mice were then given tap water or Jeju ground water for 90 days. Jeju ground water compared with tap water enhanced the activities and levels of superoxide dismutase, catalase, and glutathione peroxidase in irradiated liver tissues. Jeju ground water also enhanced the levels of intracellular reduced glutathione, which is vital for normal liver function and repair. These results suggest that vanadium-containing Jeju ground water can safeguard against the harmful actions of gamma-ray radiation through the support of hepatic antioxidant processes.
Astaxanthin is one of many carotenoids present in marine animals, vegetables and fruits. Since carotenoids are known to exert antioxidant actions, we explored to determine if astaxnathin could have such regulatory actions in normal-and $CCl_4$-treated rat liver. Astaxanthin treatment caused a slight increase in $\alpha$-tocopherol levels in the control rat liver. Glucose-6-phosphatase activity was significantly increased by astaxanthin in a dose-dependent manner and its activity decreased in response to $CCl_4$treatment was slightly inhibited by astaxanthin. These results suggest that astaxanthin could protect liver damages induced by $CCl_4$via inhibiting lipid peroxidation and it may have a potential to activate the anti-oxidant system of normal liver by stimulating $\alpha$-tocopherol production.
Oxidative stress suppresses animal health, performance, and production, subsequently impacting economic feasibility; hence, maintaining and improving oxidative status especially through natural nutrition strategy are essential for normal physiological process in animals. Phytochemicals are naturally occurring antioxidants that could be considered as one of the most promising materials used in animal diets in various forms. In this review, their antioxidant effects on animals are discussed as reflected by improved apparent performance, productivity, and the internal physiological changes. Moreover, the antioxidant actions toward animals further describe a molecular basis to elucidate their underlying mechanisms targeting signal transduction pathways, especially through the antioxidant response element/nuclear factor (erythroid-derived 2)-like 2 transcription system.
The heartwood of Caeralpinia sappan L. (Leguminosae) has been used to activate blood flow, remove blood stasis, reduce swelling and relieve pain in Korean folk medicine. In this study. the antioxidant effects of this crude drug and its hepatoprotective activity on CCl$_4$-induced liver injury in rats were evaluated. The levels of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) reduced by CCl$_4$treatment, were recovered by this crude drug. It is suggested that Caesalpinia sappan L. have antioxidant effect. The increased levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) by CCl$_4$were also recovered by treatment with this crude drug. These results can be attributed to the agent's antioxidant and membrane - stabilizing actions.
지방산화는 기본적으로 불포화지방과 산소의 결합이나 이 현상을 제대로 알고 실제 식품에 적용하기 위해서는 지방산화에 미치는 모든 인자들의 화학적 관점(one-electron reduction potential, bond dissociation enthalpy) 뿐만 아니라 물리적 현상(interface, antioxidant polar paradox)을 포괄적으로 이해해야 한다. Invitro실험법에 의해 도출된 산화방지제는 가능성을 보여주는 것이나 실제 식품과 bulk oil, 유화형태, 오르가노젤과 같은고체 상태의 다양한 매트릭스에서는 다른 활성을 나타낸다. 또한 산화방지제의 농도와 다른 물질의 존재에 따라 예상과 다른 활성을 나타내기도 한다. 산화방지제를 활용하여 최종제품의 유통기한을 증가시키기 위해서는 실제 활용될 제품의 매질로 사용해야 한다.
Objectives: To summarize and make a reference number of herbal plant-derived antioxidant researches worldwide. Methods: We surveyed all papers of antioxidant-focused studies using plants in PubMed database as "herbal plant AND antioxidant" of Default Tag "Title". The type of materials used in the studies, formation of experiments, frequency of herbal plants studied and their actions, and main study subjects were analyzed. Results: The number of studies on herbal plant-related antioxidant effects have increased worldwide since 2000. Studies have been performed using mainly single plant and single compounds for medical disorders such as immunity, heart/blood, liver, and central nerve functions. The list of plants frequently includes SalviamiltiorrhizaBge.,GinkgobilobaL., ScutellabaicalensisGeorgi.,and so on. Most of these plants have strong effects against oxidative stress and also against free radicals and increase the activities of antioxidant enzymes. Conclusion: This study produced an overview of previous research on antioxidant-focused herbal plants. This result will provide useful information for the field of Korean traditional medicine for the development of drugs related to antioxidants.

The protective actions of ambroxol, rutin, glutathione and harmaline on oxidative damages of various tissue components were compared. The mechanisms by which they prevent oxidative tissue damages were explored. Lipid peroxidation of liver microsomes induced by combinations of $Fe^{2+}$ and ascorbate or $Fe^{+3}$, ADP and NADPH was inhibited by $50\; \muM$ of rutin, ambroxol, harmaline and glutathione. Ambroxol ($100\; \muM$) inhibited the degradation of hyaluronic acid by $Fe^{2+}$, $H_2O$_2$ and ascorbate, and it was greater than that of harmaline, whereas hyaluronic acid degradation was not prevented by rutin and glutathione. The compounds used ($100\; \muM$) did not protect the degradation of cartilage collagen by xanthine and xanthine oxidase. Rutin, glutathione and harmaline decreased the degradation of IgG by xanthine and xanthine oxidate, while ambroxol did not attenuate degradation of IgG. Glutathione showed a scavenging action on $H_2O_2$. The compounds all showed scavenging actions on hydroxyl radical. Ambroxol and harmaline exhibited quenching effects en singlet oxygen. In conclusion, ambroxol, rutin, glutathione and harmaline may exert protective effects differently on tissue components against oxidative attack depend on kind of tissue component and free radical.
In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.
EGCG[(-)-epigallocatechin gallate], is a major component of green tea has been considered as a major antioxidant constituent. It has been considered as potential chemopreventive and chemotherapeutic agents. However, very little is known about the cellular actions by which EGCG mediates its therapeutic effects. Various aspects of antioxidant activity of EGCG were evaluated in this study. EGCG itself did not show significant cytotoxicity. Significant 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was observed in all ranges of concentration ($0.8-100{\mu}g/ml$) used in this study. Protective effect of EGCG against hydrogen peroxide induced cell death was observed. Relatively high lipid peroxidation inhibitory activity were detected ($IC_{50}$ was about $20{\mu}g/ml$). EGCG also dose-dependently enhanced the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) in V79-4 cells. In concentrations of $100{\mu}g/ml$ of EGCG, activities of SOD, CAT and GPX were measured as 36.9 U/mg of protein, 22.9 U/mg of protein and 17.8 U/mg of protein, respectively. When these values were compared with those of the control groups (24.9 U/mg of protein, 14.9 U/mg of protein and 11.7 U/mg of protein), the relative increases were calculated as 48, 54 and 52%, respectively. Taken together, our findings suggest that EGCG can act as an antioxidant by scavenging radicals and enhancing antioxidant enzyme activities.
Mitochondrial dysfunction is closely associated with reactive oxygen species (ROS) generation and oxidative stress in cells. On the other hand, modulation of the cellular antioxidant defense system by changes in the mitochondrial DNA (mtDNA) content is largely unknown. To determine the relationship between the cellular mtDNA content and defense system against oxidative stress, this study examined a set of myoblasts containing a depleted or reverted mtDNA content. A change in the cellular mtDNA content modulated the expression of antioxidant enzymes in myoblasts. In particular, the expression and activity of glutathione peroxidase (GPx) and catalase were inversely correlated with the mtDNA content in myoblasts. The depletion of mtDNA decreased both the reduced glutathione (GSH) and oxidized glutathione (GSSG) slightly, whereas the cellular redox status, as assessed by the GSH/GSSG ratio, was similar to that of the control. Interestingly, the steady-state level of the intracellular ROS, which depends on the reciprocal actions between ROS generation and detoxification, was reduced significantly and the lethality induced by $H_2O_2$ was alleviated by mtDNA depletion in myoblasts. Therefore, these results suggest that the ROS homeostasis and antioxidant enzymes are modulated by the cellular mtDNA content and that the increased expression and activity of GPx and catalase through the depletion of mtDNA are closely associated with an alleviation of the oxidative stress in myoblasts.
