Syntheses of Pyrrolo[2,1-b]thiazole Carboxanilides and Their Selective Antifungal Activities against Rice Blast and Wheat Leaf Rust (Pyrrolo[2,1-b]thiazole 카르복스아닐라이드 유도체의 합성 및 그들의 벼 도열병균과 밀 붉은녹병균에 대한 선택적인 항균활성)

  • For development of new agrochemical fungicide, syntheses of pyrrolo[2,1-b]thiazole carboxanilide derivatives 9 and antifungal screening against 6 kinds of plant pathogens were carried out. Functionalization of carboxylic acid on C-2 into carboxanilide in the pyrrolo[2,1-b]thiazole resulted in new 20 candidates, in which are ${\alpha},{\beta}$-unsaturated carboxanilide and methyl groups that are in cis relationship. Treatment of acetoxy-1,4-thiazin with magnesium in refluxing methanol gave pyrrolo[2,1-b]thiazole ethyl ester 4 in higher yield than that of the previous report. Hydrolysis of this compound afforded the corresponding acid 5, which reacted with aniline derivatives in the presence of coupling reagent, DIC to give pyrrolo[2,1-b]thiazole carboxanilides 9. As the result of in vivo antifungal assay of 9 against rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew, some compounds showed selectively antifungal activities against the rice blast and wheat leaf rust.

Evaluation on Anti-Dermatophyte Effect of Larix (kaempferi) Essential Oil on the Morphological Changes of Eermatophyte Fungal Hyphae (피부사상균 균사의 형태학적 변화를 통한 일본잎갈나무 정유의 항진균 활성 효과 구명)

  • This study was to investigate the antifungal activity of Larix kaempferi essential oil against dermatophytes, Epidermophyton floccosum, Trichophyton mentagrophytes and Trichophyton rubrum. The active components of L. kaempferi against dermatophytes were determined (characterized by GC-MS), and the morphological changes of the dermatophytes exposed to the L. kaempferi essential oil were observed by electron microscope. Main component of L. kaempferi essential oil was (-)-bornyl acetate. In antifungal activity tests, MIC of L. kaempferi crude oil was 125 ppm on every fungi and 100% (agar dilution method) at more than 500 ppm. By using SEM and TEM, the fungal morphology of E. floccosum exposed to the L. kaempferi essential oil was different from that of normal hyphal morphology. Hyphae exposed to the L. kaempferi essential oil was damaged with distorted and collapsed surfaces. In addition, there were destruction and disorganization of organelles in cytoplasm and collapse of cell membrane. Active antifungal components from L. kaempferi essential oil were identified as terpene alcohol compounds like (-)-${\tau}$-muurolol, (+)-terpinen-4-ol, ${\alpha}$-terpineol, and ${\alpha}$-cadinol.

The Effect of C-4 Substituent of 2-phenylimino-1,3-thiazolines on the Antifungal Activity Against Rice Blast (2-페닐이미노-1,3-티아졸린 유도체의 C-4 치환체가 벼도열병에 대한 항균활성에 미치는 영향)

  • New morpholinylcarbonylmethyl-2-phenylimino-1,3-thiazolines 2(X=O) and piperidinylcarbonylmethyl-2-phenylimino-1,3-thiazolines 3(X=C) to which morpholinyl or piperidinyl functional group were introduced at C-4 side chain of the 2-phenylimino-1,3-thiazoline scaffold were synthesized to investigate the effect of NH hydrogen of 2-phenylimino-1,3-thiazoline-4-acetanilide derivatives on the antifungal property against rice blast. Synthesized 30 compounds were screened against 6 kinds of typical plant fungi. Treatment of ketene dimer with chlorine followed by the reaction of morphorine or piperidine without isolation of the intermediate acetoacetylchloride gave $\gamma$-chloro-$\beta$-keto derivatives. These were reacted with thioureas to give morpholinylcarbonylmethyl-2-phenylimino-1,3-thiazolines and piperidinylcarbonylmethyl-2-phenylimino-1,3-thiazolines respectively in good yield (27-98%). The compound 3j, in which two fluorine atoms are substituted at ortho and para position of phenyl group of 2-phenylimino moiety and piperidinyl group is substituted at C-4, showed the highest antifungal activity (100 ppm, 90%). This result suggested that the substituent at C-4 of the 2-phenylimino-1,3-thiazolines may play a supplementary role to show the antifungal activity against rice blast.

Antifungal Activity of Bacillus Subtilis HK2 against Trichothecium Roseum Causing Pink Rot of Melon and White Stain Symptom on Grape (멜론 분홍빛썩음병과 포도 흰얼룩병의 원인균인 Trichothecium Roseum에 대한 Bacillus Subtilis HK2의 항균활성)

  • Pink Rot on melon and White Stain Symptom on grape are caused by Trichothecium roseum, one of the most important diseases of grape and melon. These diseases have been occurred in national-wide in Korea and causes irreversible damage on the grape and the melon at harvest season. This research presents the evaluation of the capacity of Bacillus subtillis HK2 to protect both melon and grape against T. reseum and establishes its role as a biocontrol agent. In this study, we isolated a Bacillus strain HK2 from rhizosphere soil, identified it as Bacillus subtillis by 16S rRNA analysis and demonstrated its antifungal activity against T. roseum. Under I-plate assay it was observed that the effect of hyphal growth inhibition was not due to production of volatile compounds. The optimum culture condition of HK2 was found at 30℃ and initial pH of 7.0. Application of HK2 culture suspension reduced 90.2% of white stain symptom on grape as compared to control, resulting in greater protection to grape against T. roseum infestation. Butanol extract of HK2 culture purified using flash column chromatography. The antifungal material was a polar substance as it showed antifungal activity in polar elute. Therefore, our results indicated a clear potential of B. subtilis HK2 to be used for biocontrol of Pink rot in melon and white stain symptom on grape caused by T. roseum.

Antifungal Activity against Trichoderma spp. of Water Soluble Essential Oil extracted from Pinus densiflora and Chamaecyparis obtusa (소나무 및 편백나무 수용성 정유를 이용한 Trichoderma spp.의 생장억제 활성)

  • This study was carried out to investigate the antifungal activity of the water soluble essential oil against Trichoderma spp. Water soluble essential oils from Pinus densiflora and Chamaecyparis obtusa were obtained from GAP (Gas assisted process) extraction apparatus. When the water soluble essential oil was treated, the growth of the conidial germination was maximum under the cultivation condition at $25^{\circ}C$ and in the culture medium adjusted to pH 5.0. The yield of water soluble essential oil was 3.9% and 3.7% in P. densiflora and C. obtusa, respectively. 24 and 15 kinds of compounds were identified in water soluble essential oils of P. densiflora and C. obtusa, respectively. The major components in the essential oil of P. densiflora were $\alpha$-Terpineol acetate and Terpinen-4-ol and those of C. obtusa were Terpinen-4-ol and $\alpha$-Terpineol. Antifungal activity was the best in the 5000 ppm of P. densiflora against Trichoderma harzianum and in 5000 ppm of C. obtusa against Trichoderma atroviride.

  • Endophytic fungi are microorganisms inhabiting living plant tissues without causing apparent harm to the host. They are drawing increasing attention due to their ability to produce various bioactive compounds as well as their effects on host growth and resistance to biotic and abiotic stresses. As a first step to assess biodiversity of plant associated fungi in Korea and the following evaluation on diverse biological activities, we are collecting endophytic fungi from plant in wild followed by systematic long-term storage in liquid nitrogen. Molecular identification using ITS sequences was also incorporated for pure culture by hyphal tip isolation. As of April 2015, about 1,400 fungal strains had been isolated from about 170 plant taxa. Fungal isolates belonging to Pleosporales, Diaporthales, Glomerellales, Hypocreales, and Xylariales were the most abundant. These collections are being used for several complementary researches, including screening of isolates with novel bioactive compounds or conferring drought stress resistance, phylogenetic and genomic study. Genome sequencing was performed for 3 isolates, one Xylaria sp. strain JS573 producing griseofulvin, an antifungal compound, and two Fusarium spp. strains JS626 and JS1030, which are assumed to be new species found in Korea. More detailed analysis on these genomes will be presented. These collections and genome informations will serve as invaluable resources for identifying novel bioactive materials in addition to expand our knowledge on fungal biodiversity.

Crystal Growth, Spectral, Magnetic, Antibacterial and Antifungal Studies of Co(II) and Ni(II) Complexes of 4-Nitrobenzoic Acid

  • Single crystals of cobalt(II) and nickel(II) complexes of 4-nitrobenzoic acid have been successfully grown by gel diffusion technique. The grown crystals were characterized by elemental analysis, FT-IR and UV-Visible spectroscopy. The structure of the grown crystals was elucidated using single crystal X-ray diffraction studies. The title compounds $[Co(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 1 and $[Ni(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 2 where PnbaH=4-nitrobenzoic acid, crystallize in centrosymmetric triclinic space group P-1. Magnetic susceptibility measurements reveal that the compounds are paramagnetic in nature. The mechanical strength of the grown crystals was determined by Vicker's microhardness studies. The ligand (4-nitrobenzoic acid) and the complexes have been screened for their biological activity against various bacteria and fungi. The activity data show that the biological activity of the complexes is higher than that of the ligand.

Identification of an antagonistic bacteria and its antibiotic substance against Colletotrichm orbiculare causing anthracnose on cucumber

  • A bacterial strain YC4963 with antifungal activity against Colletotrichum orbiculare, a causal organism of cucumber anthracnose was isolated from the rhizosphere soil of Siegesbeckia pubescens (Siegesbeckia pubescens Makino;Family:Compositae) in Korea. Based on physiological and biochemical characteristics and 16S ribosomal DNA sequence analysis, the bacterial strain was identified as Pseudomonu aureofaciens. The bacteria also inhibited mycelial growth of several plant fungal pathogens such as Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani on PDA and 0.1 TSA media. The antibiotic activity was found from the culture filtrate of TSB(tryptic soy broth) and its active compounds were quantitatively bound to XAD adsorber resin. The antibiotic spectrum was broad and growth of C. orbiculare and F. oxysporum, B. cinerea were inhibited at very low concentration. The chemical data from various chromatographic procedures showed that active fraction consisted of at least two phenazine derivatives. However, the metabolites had no inhibitory effect on Pythium ultimum which was reported to be sensitive to phenazine antibiotics. The compounds responsible for the activity are now under investigation.

  • Cinnamomi Cortex (Lauraceae), the dried bark of Cinnamomum cassia BLUME, has been used as traditional Chinese medicine for its stomachic, astringent, carminative, antispasmodic, antibacterial, antifungal properties. Four compounds were isolated from the MeOH extract of Cinnamomi Cortex, and their structures were identified as trans-cinnamic acid (1), ${\beta}-sitosterol$ (2), bis(2-methylheptyl)phthalate (3), coumarin (4) by comparison of their physical and spectral data with those reported in the literature. These compounds were tested melanogenesis inhibitory effect on B-16 mouse melanoma cell lines. Among them, trans-cinnamic acid (1) showed the most potent inhibitory effect on melanogenesis with $IC_{50}$ value of $13{\mu}g/ml$. Arbutin, positive control, exhibited an $IC_{50}$ value of $29{\mu}g/ml$.

Potential of the Volatile-Producing Fungus Nodulisporium sp. CF016 for the Control of Postharvest Diseases of Apple

  • In vitro and in vivo mycofumigation effects of the volatileproducing fungus Nodulisporium sp. CF016 isolated from stem of Cinnamomum loureirii and the role of its volatile compounds were investigated against phytopathogenic fungi. The volatile compounds produced by Nodulisporium sp. CF016 inhibited and killed a wide range of plant and storage pathogens including to Pythium ultimum, Rhizoctonia solani, Fusarium oxysporum, Phytophthora capsici, Sclerotinia sclerotiorum, Colletotrichum coccodes, Magnaporthe oryzae, Alternaria panax, Botrytis cinerea and Penicillium expansum. Mycofumigation with wheat bran-rice hull cultures of Nodulisporium sp. CF016 showed in vivo antifungal activity against gray mold caused by B. cinerea and blue mold caused by P. expansum of apple. The most abundant volatile compound produced by Nodulisporium sp. CF016 was $\beta$-elemene followed by 1-methyl-1,4-cyclohexadiene, $\beta$-selinene and $\alpha$-selinene. Nodulisporium sp. CF016 could be an attractive mycofumigant in controlling postharvest diseases of various fruits including apple.
