Candicidal Action of Resveratrol Isolated from Grapes on Human Pathogenic Yeast C. albicans

  • Resveratrol (3,5,4'-trihydroxystilbene) is a naturally occurring, multi-biofunctional chemical existing in grapes and various other plants as a polyphenol type, and it is one of the best known natural anticancer and antiatherosclerosis reagents. In this study, we investigated the antifungal action by resveratrol in Candida albicans, which is a human infectious fungi as an agent of candidiasis. Resveratrol displayed potent fungicidal activity in an energy-dependent manner, without any hemolytic effects against human erythrocytes. It was found that the serum-induced mycelial forms, which playa crucial role in the pathogenesis of C. albicans during host tissue invasion, were disrupted by resveratrol. To understand the correlation between lethal effects and resveratrol action, we examined the physiological changes of C. albicans. A significant accumulation of intracellular trehalose was induced by stress responses to resveratrol action, and a remarkable arrest of cell-cycle processes at the S-phase in C. albicans occured. Therefore, the fungicidal effects of resveratrol demonstrate that this compound is a potential candidate as an antifungal agent in treating infectious diseases by candidal infections.

생칠과 화칠의 우루시올 조성 및 생리활성 비교 (Comparison of Urushiol Composition and Biological Activity between Fresh Sap and Fire Distilled Sap of Lacquer Tree)

  • For the comparison of the urushiol composition and biological acitivity between the fresh sap and fire distilled sap of lacquer tree(Rhus vernicifera), we analysed the urushiol composition by HPLC and EI-MS, and investigated the antioxidative , antimicrobial and anticancer acitivities according to solvent fractionations. There was no difference in the urushiol composition between fresh and fire distilled saps of lacquer tree. The hexane frqctionsof two saps showed a strong DPPH radical scaverging activity (RC50 : 7.0-7.5$\mu\textrm{g}$). They also showed a strong antifungal activity onthe spore germination of Cladosporium herbarum(MIC : 8$\mu\textrm{g}$/ml), whereas they have no or low activity against the bacteria(BAcillus subtilis , Escherichia coli). In addition , hexane and butanol fractions of two saps showed a strong inhibitory activity against cultured tumour cell lines (GI50 : 0.35, 12.29$\mu\textrm{g}$/ml) in vitro. These results confirm that the fresh sap and fire distilled sap might have the similar urushiol compositions and biological activities.

기능성 활엽수종의 생리활성 (Bioactivities of Several Functional Hardwood Trees)

  • 아까시나무의 목질부와 현사시나무, 물푸레나무 및 느릅나무의 수피를 채취하여 아세톤-물의 혼합용액으로 추출한 후 hexane, chloroform, ethylacetate 및 물 분획으로 분류하고 동결건조하며 생리활성 시험용 추출분말을 조제하였다. 각 수종의 목질부에서 채취된 목재 블록에 대하여 수행된 내후성 시험은 아까시나무가 가장 우수하였으며 현사시나무가 가장 낮은 내후성을 나타내었다. 목질부 또는 수피부 추출물에 대한 항진균, 항세균 및 항산화 활성 시험이 수행되었으며 항진균 및 항세균에 대한 활성 시험에서는 모든 분획에서 유의적인 차이를 나타내지 않았다. 항산화 활성 시험은 천연 항산화제인 ${\alpha}$-tocopherol과 합성 항산화제인 BHT를 표준물질로 사용하여 추출물 분획의 항산화 활성과 비교하였다. 이 시험에서 물푸레나무의 ethylacetate 분획이 BHT를 제외하고 가장 높은 활성을 나타내었으며 아까시나무는 모든 분획이 다른 수종보다 높은 활성을 나타내었다.

키틴과 키토산 분해 미생물 유래 효소의 식품에의 이용 (Food application of enzymes derived from microorganisms degrading chitin and chitosan)

  • Most reports demonstrated the substrate specificity-based kinetic properties of chitin or chitosan degrading enzymes. However, there is virtually less information on the high quality and quantity production of chitin or chitosan hydrolysates having a larger than (GlcN)7 from the hydrolysis of high molecular weight chitosan using specific enzymes and their biological activity. Therefore, the production of such molecules and the discovery of such enzyme sources are very important. Fortunately, the author has established a mass production method of chitosan hydrolysates (GlcN)n, n=2-13 that have been characterized as a potent antioxidant substance, as well as antifungal and antibacterial activities against Penicillium species and highly selective pathogenic bacteria. In addition, preclinical studies using (GlcN)n, n=5-25 demonstrated that these molecules played a very important role in maintaining biometric balance. Collectively, it is implicated that the application of these mixed substances to foods with significant biological activity is very encouraging.

식품위해성균, 피부사상균 및 식물성 병원균에 대한 산초유의 항균 활성 및 항산화 활성 (Antimicrobial Activity against Food-hazardous Microorganisms, Dermatophytes, and Pytopathogens and Antioxidative Activity of Sancho Oil)

  • Background: Although Sancho (Zanthoxylum schinifolium Siebold & Zucc) oil has traditionally been used for its antibiotics properties, there is currently a lack of scientific evidence regarding its biological activities. In this study, we investigated the antimicrobial and antioxidant activities of Sancho oil against food-hazardous microorganisms, phytopathogens, and dermatophytes. Methods and Results: We investiated the antimicrobial activity of Sancho oil against 11 food-hazardous microorganisms, nine phytopathogens, and six dermatophytes. The Sancho oil was found to show the strongest antibacterial activity against Shigella flexneri and Listeria spp. Sancho oil also showed high antifungal activity against plant pathogens, particularly Fusarium oxysporum, and showed antimicrobial activity against dermatophytes such as Trichophyton rubrum, Microsporum canis and Candida albicans. The antioxidant activity of Sancho oil was measured using the DPPH method, and was found to be stronger than that of unrefined oil. Moreover, this activity increased with increasing oil concentration. Conclusions: We found that Sancho oil showed differing antimicrobial activities against food-hazardous microorganisms, dermatophytes, and plant pathogens. The antimicrobial activity spectrum of Sancho oil was not broad and varied among microbial strains. On the basis of our findings, we consider that Sancho oil could be used an antibacterial material for food-borne S. flexneri and Listeria spp., a biopesticide for Fusarium spp., and a treatment for dermatophytes such as T. rubrum.

Functional Properties of Filamentous Fungi Isolated from the Indonesian Fermented Dried Cassava, with Particular Application on Poultry

  • The study aimed to evaluate the probiotic properties, antioxidant activity and fermentative capacity of Acremonium charticola and Rhizopus oryzae isolated from the Indonesian fermented dried cassava, with particular application on poultry. A. charticola inhibited the growth of Escherichia coli and Aspergillus flavus. A. charticola and R. oryzae grew in potato dextrose agar (PDA) adjusted to pH 3 and 8 or in PDA supplemented with bile salt up to 0.8%. After soaking for 8 hr, the survival rate of A. charticola in the simulated gastric juice (pH 2) and bile solutions (2% bile salt) was lower than that of R. oryzae. A. charticola and R. oryzae exhibited strong antioxidant activities. Compared to unfermented cassava pulp (control), the fibre content of cassava pulp tended to be lower after fermentation with A. charticola for 14 days. The populations of A. charticola and R. oryzae were significantly higher in fermented cassava pulp than in unfermented one. Coliform was higher in cassava pulp fermented with R. oryzae or A. charticola + R. oryzae compared to control after 7 days of fermentation, however, the bacteria were not different between A. charticola-fermented cassava pulp and control. Lactic acid bacteria (LAB) were higher in A. charticola- and R. oryzae-fermented cassava pulp than those in control, however, no difference of LAB was observed between A. charticola + R. oryzae-fermented cassava pulp and control. In conclusion, A. charticola exhibited antibacterial, antifungal and antioxidant activity, gastrointestinal persistence and fermentative capacity that may be beneficial for poultry industry.

  • An important aspect of indigenous medicine is the ability of traditional societis to manipulate the environment for their own benefit, for example for their health care. In Indonesia, this indigenous medicine is called Jamu. Jamu mostly consists of a mixture of herbals of the genus Curcuma, Zingiberaceae. There are 19 species of curcuma grown in Indonesia. Eleven of them are popular in the jamu preparations. Ethnopharma-cological surveys have shown that 50% of these species are used for post partum protection, dismenorrhea, 30% are used for the treatment of stomache and as cosmetics, 20% for the treatment of various diseases such as fever, worms, asthma, etc. Chemical studies show that they contain curcuminoids, volatile oils, flavonoids, starch, and resinous substance. Pharmacological studies of extracts and isolated bioactive compounds have shown that they have a broad pharmacological activity such as antibacterial, antifungal, anti-inflammatory, choleretic, antihepatotoxic, antitumor, antioxidant, and antihyperlipidemic activity.

Phytochemical Compounds from the Ethanolic Extract of Gymnema sylvestre, Senna auriculata and Cissus quadrangularis through GC-MS Analysis

  • Plants are a traditional source of many chemicals used as biochemical, flavors, food, color, and pharmaceuticals in various countries, especially India. Most herbal medicines and their derivatives are often made from crude extracts containing a complex mixture of various phytochemical chemical components (secondary metabolites of the plants). This study aimed to identify bioactive compounds from the different parts of the plant from the ethanolic extract of Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis (leaves, flower, stem) by gas chromatography-mass spectroscopy (GC-MS). The gas chromatography - mass spectrometry analysis revealed the presence of various compounds like 3,4-dimethylcyclohexanol, hexanoic acid, D-mannose, and N-decanoic acid. Hence, the Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis may have chemopreventive, anti-cancer, anti-microbial activity, antioxidant, anti-diabetic activity, anti-inflammatory, and antifungal due to the presence of secondary metabolites in the ethanolic extract. These phytochemicals are supported for traditional use in a variety of diseases.

영여자의 항균, 항혈전 및 항산화 활성 평가 (Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Aerial Bulbils of Dioscorea batatas Decne)

  • 영여자는 마의 주아를 말하며, 년간 생마 생산량 5,000 톤의 절반인 2,500 톤 정도가 생산되고 있다. 그러나 영여자는 다음해 마의 종자로 일부 이용될 뿐 대부분이 폐기되고 있는 실정이다. 본 연구에서는 영여자의 새로운 생물소재 가능성을 검토하고 생마에서 보고된 항균, 항혈전 및 항산화 활성을 평가하고자, 영여자로부터 메탄올 추출물을 조제하고 이로부터 헥산, 에틸아세테이트, 부탄올의 다양한 유기용매 분획물 및 물 잔류물을 조제하여 각각의 항균, 항혈전 및 항산화 활성을 평가하였다. 영여자 메탄올 추출물은 마의 그것과 유사하게 다량의 수용성 성분을 함유하고 있었으나 총 폴리페놀 및 총플라보노이드 함량이 각각 12배, 3.4배 높았다. 항균 활성의 경우, 헥산 및 에틸아세테이트 분획에서 항세균 활성이 나타나 전반적으로 마와 유사하였으나, 부가적으로 S. aureus에 강한 생육저해를 나타내어 마와 차별되었다. 항혈전 활성의 경우, 헥센 분획물이 매우 강력한 활성을 나타내어 4.8 mg/mL 농도에서 300초 이상, 2.4 mg/mL 농도에서 67초의 트롬빈 타임을 나타내어 마의 경우와 비교할 때 4배 이상의 강력한 활성을 나타내었다. 메탄올 추출물 및 유기용매 분획물의 항산화 활성 평가 결과, 에틸아세테이트 분획에서 매우 우수한 DPPH 소거능($IC_{50}=38.1{\mu}g/mL$), SOD 유사활성(vitamin C의 36%) 및 환원력(BHT의 59.4%)을 나타내었다. 본 연구결과는, 대부분 폐기되고 있는 영여자의 새로운 생물소재 개발 가능성을 제시하며, 영여자가 마의 지하괴근이 가진 항세균, 항혈전 및 항산화 활성과 거의 동등한, 또는 보다 우수한 활성을 가지고 있음을 나타내고 있다.

수종의 한약재 추출물의 항산화 활성 및 항진균 활성 (Biological and Antifungal Activity of Herbal Plant Extracts against Candida Species)

  • 본 연구에서는 여성의 질염을 일으키는 칸디다증 병원균에 대한 항진균 성분을 한약재에서 조사하였다. 한약재에 ethanol을 이용하여 phenol 성분을 추출하고, 추출물의 phenol 함량을 출정하였다. Ethanol농도에 따른 phenol성 물질의 용출량은 시료마다 다른 양상을 보이고 있는데, 40% ethanol 농도에서 가장 높은 용출량은 어성초, 지모, 금은화 등이며, 60% ethanol에서 가장 높은 용출량은 포공영, 작약 등이다. 황련은 80% ethanol 농도에서는 낮은 용출량을 보이나 다른 농도에서는 비슷한 용출량을 보이고 있고, 연교는 80% ethanol에서 높은 용출량을 보였다. 전자공여능은 ethanol 농도별 추출물에 따라 다르게 나타나고 있는데, 포공영은 80% ethanol 추출물 이상에서 전자공여 능을 보이며, 황련, 지모, 금은화는 100%의 ethanol추출물에서 전자공여능을 보였다. 한약재 추출물의 항균활성은 C. albicans는 황련 추출물 $200{\mu}g/mL$농도로 주입하였을 때 3mm의 저해환을 나타내었으며, C. tropicalis는 포공영 추출물을 $200{\mu}g/mL$ 농도로 주입시 2mm, 어성초 추출물을 $200{\mu}g/mL$ 농도로 주입시 2mm, 작약 추출물을 $200{\mu}g/mL$ 주입시 3 mm 저해환을 나타내었다. 황련 추출물은 50, 100, 150, $200{\mu}g/mL$ 농도로 주입시 2, 3, 6, 9mm의 저해환을 나타내었다. 추출물을 이용한 C. glabrata, C. parapsilesis, C. utils 3개 종에 대한 항균활성은 나타나지 않았다. 황련 추출물에서 항균성분은 TLC와 HPLC 분석을 통하여 berberin인 것으로 나타났다.
