Comparison of Urushiol Composition and Biological Activity between Fresh Sap and Fire Distilled Sap of Lacquer Tree (생칠과 화칠의 우루시올 조성 및 생리활성 비교)

  • For the comparison of the urushiol composition and biological acitivity between the fresh sap and fire distilled sap of lacquer tree(Rhus vernicifera), we analysed the urushiol composition by HPLC and EI-MS, and investigated the antioxidative , antimicrobial and anticancer acitivities according to solvent fractionations. There was no difference in the urushiol composition between fresh and fire distilled saps of lacquer tree. The hexane frqctionsof two saps showed a strong DPPH radical scaverging activity (RC50 : 7.0-7.5$\mu\textrm{g}$). They also showed a strong antifungal activity onthe spore germination of Cladosporium herbarum(MIC : 8$\mu\textrm{g}$/ml), whereas they have no or low activity against the bacteria(BAcillus subtilis , Escherichia coli). In addition , hexane and butanol fractions of two saps showed a strong inhibitory activity against cultured tumour cell lines (GI50 : 0.35, 12.29$\mu\textrm{g}$/ml) in vitro. These results confirm that the fresh sap and fire distilled sap might have the similar urushiol compositions and biological activities.

Bioactivities of Several Functional Hardwood Trees (기능성 활엽수종의 생리활성)

  • Wood of black locust(Robinia pseudoacacia) and bark of poplar(Populus alba x glandulosa), ash(Fraxinus rhynchophylla) and elm(Ulmus davidiana var. japonica) trees were collected, extracted with acetone-$H_2O$(7:3. v/v), fractionated with hexane, chloroform, ethylacetate and $H_2O$. then freeze dried to get some dark brown powder for bioactive tests. Decay-resistant activity was tested using wood block specimens from the hardwood trees and expressed by weight loss rate. Black locust specimens indicated the best anti-decaying property and poplar blocks were the worst. Antimicrobial and antioxidant activities were also investigated against each wood or bark extractives. Antifungal and antibacterial activities did not indicate any significant differences among the tested fractions. In antioxidant activity. ${\alpha}$-tocopherol one of natural antioxidants. and BHT, one of synthetic antioxidants, were used as references to compare with the antioxidant activities of the extracted fractions. Ethylacetate fraction of ash bark indicated the highest activity besides BHT in this test and all fractions of black locust extractives also indicated higher activities compared with the other fractions.

  • Most reports demonstrated the substrate specificity-based kinetic properties of chitin or chitosan degrading enzymes. However, there is virtually less information on the high quality and quantity production of chitin or chitosan hydrolysates having a larger than (GlcN)7 from the hydrolysis of high molecular weight chitosan using specific enzymes and their biological activity. Therefore, the production of such molecules and the discovery of such enzyme sources are very important. Fortunately, the author has established a mass production method of chitosan hydrolysates (GlcN)n, n=2-13 that have been characterized as a potent antioxidant substance, as well as antifungal and antibacterial activities against Penicillium species and highly selective pathogenic bacteria. In addition, preclinical studies using (GlcN)n, n=5-25 demonstrated that these molecules played a very important role in maintaining biometric balance. Collectively, it is implicated that the application of these mixed substances to foods with significant biological activity is very encouraging.

Antimicrobial Activity against Food-hazardous Microorganisms, Dermatophytes, and Pytopathogens and Antioxidative Activity of Sancho Oil (식품위해성균, 피부사상균 및 식물성 병원균에 대한 산초유의 항균 활성 및 항산화 활성)

  • Background: Although Sancho (Zanthoxylum schinifolium Siebold & Zucc) oil has traditionally been used for its antibiotics properties, there is currently a lack of scientific evidence regarding its biological activities. In this study, we investigated the antimicrobial and antioxidant activities of Sancho oil against food-hazardous microorganisms, phytopathogens, and dermatophytes. Methods and Results: We investiated the antimicrobial activity of Sancho oil against 11 food-hazardous microorganisms, nine phytopathogens, and six dermatophytes. The Sancho oil was found to show the strongest antibacterial activity against Shigella flexneri and Listeria spp. Sancho oil also showed high antifungal activity against plant pathogens, particularly Fusarium oxysporum, and showed antimicrobial activity against dermatophytes such as Trichophyton rubrum, Microsporum canis and Candida albicans. The antioxidant activity of Sancho oil was measured using the DPPH method, and was found to be stronger than that of unrefined oil. Moreover, this activity increased with increasing oil concentration. Conclusions: We found that Sancho oil showed differing antimicrobial activities against food-hazardous microorganisms, dermatophytes, and plant pathogens. The antimicrobial activity spectrum of Sancho oil was not broad and varied among microbial strains. On the basis of our findings, we consider that Sancho oil could be used an antibacterial material for food-borne S. flexneri and Listeria spp., a biopesticide for Fusarium spp., and a treatment for dermatophytes such as T. rubrum.

Functional Properties of Filamentous Fungi Isolated from the Indonesian Fermented Dried Cassava, with Particular Application on Poultry

  • The study aimed to evaluate the probiotic properties, antioxidant activity and fermentative capacity of Acremonium charticola and Rhizopus oryzae isolated from the Indonesian fermented dried cassava, with particular application on poultry. A. charticola inhibited the growth of Escherichia coli and Aspergillus flavus. A. charticola and R. oryzae grew in potato dextrose agar (PDA) adjusted to pH 3 and 8 or in PDA supplemented with bile salt up to 0.8%. After soaking for 8 hr, the survival rate of A. charticola in the simulated gastric juice (pH 2) and bile solutions (2% bile salt) was lower than that of R. oryzae. A. charticola and R. oryzae exhibited strong antioxidant activities. Compared to unfermented cassava pulp (control), the fibre content of cassava pulp tended to be lower after fermentation with A. charticola for 14 days. The populations of A. charticola and R. oryzae were significantly higher in fermented cassava pulp than in unfermented one. Coliform was higher in cassava pulp fermented with R. oryzae or A. charticola + R. oryzae compared to control after 7 days of fermentation, however, the bacteria were not different between A. charticola-fermented cassava pulp and control. Lactic acid bacteria (LAB) were higher in A. charticola- and R. oryzae-fermented cassava pulp than those in control, however, no difference of LAB was observed between A. charticola + R. oryzae-fermented cassava pulp and control. In conclusion, A. charticola exhibited antibacterial, antifungal and antioxidant activity, gastrointestinal persistence and fermentative capacity that may be beneficial for poultry industry.

  • An important aspect of indigenous medicine is the ability of traditional societis to manipulate the environment for their own benefit, for example for their health care. In Indonesia, this indigenous medicine is called Jamu. Jamu mostly consists of a mixture of herbals of the genus Curcuma, Zingiberaceae. There are 19 species of curcuma grown in Indonesia. Eleven of them are popular in the jamu preparations. Ethnopharma-cological surveys have shown that 50% of these species are used for post partum protection, dismenorrhea, 30% are used for the treatment of stomache and as cosmetics, 20% for the treatment of various diseases such as fever, worms, asthma, etc. Chemical studies show that they contain curcuminoids, volatile oils, flavonoids, starch, and resinous substance. Pharmacological studies of extracts and isolated bioactive compounds have shown that they have a broad pharmacological activity such as antibacterial, antifungal, anti-inflammatory, choleretic, antihepatotoxic, antitumor, antioxidant, and antihyperlipidemic activity.

Phytochemical Compounds from the Ethanolic Extract of Gymnema sylvestre, Senna auriculata and Cissus quadrangularis through GC-MS Analysis

  • Plants are a traditional source of many chemicals used as biochemical, flavors, food, color, and pharmaceuticals in various countries, especially India. Most herbal medicines and their derivatives are often made from crude extracts containing a complex mixture of various phytochemical chemical components (secondary metabolites of the plants). This study aimed to identify bioactive compounds from the different parts of the plant from the ethanolic extract of Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis (leaves, flower, stem) by gas chromatography-mass spectroscopy (GC-MS). The gas chromatography - mass spectrometry analysis revealed the presence of various compounds like 3,4-dimethylcyclohexanol, hexanoic acid, D-mannose, and N-decanoic acid. Hence, the Gymnema sylvestre, Senna auriculata, and Cissus quadrangularis may have chemopreventive, anti-cancer, anti-microbial activity, antioxidant, anti-diabetic activity, anti-inflammatory, and antifungal due to the presence of secondary metabolites in the ethanolic extract. These phytochemicals are supported for traditional use in a variety of diseases.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Aerial Bulbils of Dioscorea batatas Decne (영여자의 항균, 항혈전 및 항산화 활성 평가)

  • The annual productions of yam and its aerial bulbils are estimated to 5,000 and 2,500 ton, respectively. But the majority of bulbils had been discarded without specific use. In this study, methanol extract and its organic solvent fractions were prepared from bulbils of Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activities were evaluated, respectively. The methanol extract contained 58.98% of water-soluble materials as like yam's extract. But the bulbils's extract contained 12-folds of total polyphenol and 3.4-folds of total flavonoids compared than yam's extract, respectively. For antimicrobial activity the hexane and ethyl acetate fraction showed strong antibacterial activity at $500{\mu}g/disc$ concentration against gram-negative and gram-positive bacteria. Antifungal activity was not observed in any fractions. Strong antithrombin activity was found in the hexane fractions. At 4.8 mg/mL concentration thrombin time (TT) was over 300 sec, which is 4-folds extended than the TT of yam. In a while, the ethyl acetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $38.1{\mu}g/mL$), SOD-like activity and reducing power, which are comparable to vitamin C or BHT. Our results suggest that the bulbils of yam as yam tuber have useful bio-activities, such as antibacterial, antithrombosis, and antioxidant activity.

Biological and Antifungal Activity of Herbal Plant Extracts against Candida Species (수종의 한약재 추출물의 항산화 활성 및 항진균 활성)

  • Anticandidial activity of seven herbal extracts, Taraxacum Platycarpum, Houttuyniae Herba, Lonicerae Flos, Anemarrhena Rhizome, Forsythia Fruit, Paeoniae Ratix, and Coptidis Rhizoma, were determined against five different Candida sp. by agar diffusion assay. The concentration of total phenolic compounds of seven herbal extracts ranged from 0.6 to $2.5{\mu}g/mg$. The total antioxidant activities showed that Taraxacum Platycarpum and Houttuyniae Herba were 60% in 80% ethanol extract and Lonicera Flower and Paeoniae Ratix were 70, 75%, respectively, in 100% ethanol extract. Coptidis Rhizoma extract showed antifungal activity against non-Candida albicans, C. tropicalis and C. glabrata. The MIC values of a compound separated in TLC from Coptidis Rhizoma extract were 24, and $48{\mu}g/mL$ against C. tropicalis and C. glabrata. The above compound showed the same retention time with berberin in HPLC analysis.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Dioscorea batatas Decne (마(Dioscorea batatas Decne)의 항균, 항산화 및 항혈전 활성 평가)

  • Yam (Dioscorea batatas Decne) has been recognized as healthy food due to its various biological activities, such as anti-obesity, anti-constipation, anti-proliferation, anti-mutagenic activity, and decrease of blood glucose and cholesterol level. In this study, the methanol extract and its organic solvent fractions were prepared from dried Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activity were evaluated, respectively. The 66.7% of methanol extract was fractioned into water residue, and most of total-polyphenol/total-flavonoids were found in ethylacetate fraction. This ethylacetate fraction at $500\;{\mu}g/disc$ concentration showed strong antibacterial activity against gram-negative and gram-positive bacteria, except Staphylococcus aureus. Antifungal activity was not observed in methanol extract and its fractions. But, strong antithrombin activity was found in methanol extract. At 4.8 mg/mL concentration, thrombin time was 99.5 sec, which is coincides with 30% activity of aspirin, a commercial available antithrombosis agent. The ethylacetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $80.5\;{\mu}g/mL$), compared to vitamin C ($IC_{50}$ of $15.2\;{\mu}g/mL$). Also, the ethylacetate fraction showed strong SOD-like activity and reducing power, which are coincide with 43% of vitamin C and 82.7% of butylated hydroxytoluene activity, respectively.
