Ethanolic Extract of Pancake Mixture Powder Supplemented with Helianthus tuberosus Enhances Antidiabetic Effects via Inhibiting Inflammatory Mediator NO Production

  • Helianthus tuberosus is perennial plant as Compositae family and is shown various physiological activities such as analgesic, antipyretic, anti-inflammatory, anti-fungal, anti-spasmodic, aperient, cholagogue, diuretic, spermatogenic, stomachic, and tonic effects. In this study, we investigated the antidiabetic and anti-inflammatory effects of pancake mixture powder (PM) supplemented with H. tuberosus (PMH) in rat skeletal muscle L6 cells and murine macrophage RAW 264.7 cells, respectively. PM and PMH inhibited in vitro α-glucosidase activity. Glucose consumption was increased by PM and PMH without cytotoxicity in rat myoblast L6 cells. Western blot analysis revealed that PM and PMH down-regulated glycogen synthase kinase (GSK)-3β activation in L6 cells. PM and PMH inhibited inflammatory mediator, nitric oxide (NO) production without cytotoxicity in LPS-induced RAW 264.7 cells. The anti-diabetic and anti-inflammatory effects of PMH was more stronger than those of PM. Anti-diabetic and anti-inflammatory effects of PMH would be due to functional characteristics of the supplemented H. tuberosus and the presence of garlic and onion used as ingredients of PM. Taken together, our results that addition of functional materials such as H. tuberosus in product has synergic effects and PMH is potential candidate for treatment of diabetes through inhibiting inflammation.

담쟁이덩굴 추출물과 분획물의 항산화, 항당뇨 및 항염증 효과 (Antioxidant, Antidiabetic, and Anti-Inflammatory Effects of Extracts and Fractions from Parthenocissus tricuspidata Stems)

  • 본 연구에서는 담쟁이덩굴 추출물이 항산화, 항당뇨 및 항염증 효과에 미치는 영향을 조사하고자 수행하였다. 담쟁이덩굴 열수 및 에탄올 추출물의 총 페놀함량은 각각 61.5 mg TAE/g과 122.1 mg TAE/g으로 에탄올 추출물에서 훨씬 높게 나타났다. 에탄올 추출물과 부탄올 분획물의 DPPH 라디칼 소거활성은 1 mg/ml에서 95%와 92%의 radical 소거능을 나타내어 positive control로 사용한 비타민 C와 유사한 항산화력을 보였다. SOD 활성은 에탄올 추출물과 부탄올 분획물 1 mg/ml 농도에서 91%, 97%로 높은 활성을 나타내었다. DPPH radical 소거능과 SOD 활성을 측정한 결과 항산화 활성은 모두 농도 의존적으로 증가하였으며 특히 열수추출물과 비교할 때 에탄올 추출물과 부탄올 분획물에서 높은 항산화 효과를 보였다. 담쟁이덩굴 추출물의 ${\alpha}$-glucosidase 활성억제 효과는 에탄올 추출물과 핵산 및 부탄올 분획층에서 높은 항당뇨 효과를 나타내었다. 이러한 결과는 지금까지 항당뇨 약물로 사용된 것보다도 높은 것으로 나타났다. LPS에 의하여 유도된 NO 합성은 1 mg/ml 농도의 담쟁이추출물을 처리함으로써 NO 합성이 약 40%로 감소하였다. 이러한 결과들은 담쟁이 추출물의 우수한 항산화, 항당뇨, 항염 효과를 시사하고 있으며 특히, 천연 항당뇨 기능성 소재로서 담쟁이 추출물의 활용 가능성이 높을 것으로 사료된다.

제조방법에 따른 제호탕의 항산화 및 항당뇨 활성 (Antioxidant and antidiabetic Activity of Jehotang according to Different Cooking Methods)

  • 제호탕은 오매, 사인, 백단향, 초과 등의 한약재와 꿀을 넣어 만든 여름철 전통음료이다. 본 연구에서는 제호탕의 재료인 오매, 사인, 초과, 백단향의 항산화 활성, Total phenolic content, ${\alpha}$-glucosidase, ${\alpha}$-amylase inhibitory activity를 생리활성을 측정하였다. 또한, 전통적인 조리법(연밀)과 현대적인 조리법(중탕법, 모두 끓이는 방법)으로 제호탕을 만들었을 때 pH, 색도, 당도의 차이를 비교분석하였다. Total phenolic content는 사인이 120.45 mg GAE/g, 오매분말 연밀미비법이 152.66 mg GAE/g를 나타냈으며, DPPH free radical 소거활성은 사인이 93.13% 그리고 오매분말 연밀미비법이 56.44%를 나타내었다. ${\rho}$-nitrophenyl-${\alpha}$-Dglucopyranoside를 기질로 사용한 ${\alpha}$-glucosidase 저해효과는 $100{\mu}g/mL$ 농도에서 사인이 89.51% 저해율을 보였으며, 끓이는 방법이 ${\alpha}$-glucosidase 저해효과 52.38%, ${\alpha}$-amylase 저해활성 72.52%로 높은 저해능을 보였다. 이와 같은 결과, 제호탕 소재로는 사인이 항산화, 항당뇨 효과에 뛰어남을 확인할 수 있었다. 조리방법에 서는 오매분말 연밀미비법이 높은 항산화 활성을 나타내었으며, 항당뇨 효과는 끓이는 방법이 뛰어남을 확인할 수 있었다.

육미지황탕가미방(六味地黃湯加味方)이 C57BL/6JHam-ob/ob mice의 비만(肥滿) 및 제2형 당뇨병(糖尿病)에 미치는 영향(影響) (Antiobese and Antidiabetic Effects of Yookmijihwang-tang-gamibang, a Traditional Polyherbal Formula on the Obese and Type II Diabetic C57BL/6JHam-ob/ob Mice)

  • Objectives : The object of this study was to observe the pharmacological effects of Yookmijihwang-tang-gamibang (Liuweidihuang-tang-jiaweifang, YGB) consisted of 17 types of herbs has been traditionally used in Korean medicine for treating various disorders in clinics, aqueous extracts on the genetically obese and type II diabetic C57BL/6JHam-ob/ob(ob/ob) mice. Methods :Three different dosages of YGB were orally administered, once a day for 28 days to ob/ob mice with ob/ob control and C57BL/6JJms normoglycemic intact mice. Four weeks after treatments of YGB: the changes on the body weight, food consumption, blood glucose levels, leptin and adiponectin contents were observed for monitoring the antiobese and antidiabetic effects of YGB. The effects were compared to those of CLA(conjugated linoleic acid) which improve type II diabetes and inhibit related obesity. Results : After end of 28 days of continuous treatments, ob/ob control showed increases of adipocyte hypertrophy, vasodilated atrophic glomerulus which were detected with marked hyperplasia of pancreatic islets, insulin and glucagon producing cells. These obese and related type II diabetes induced in ob/ob mice were markedly and significantly inhibited by 28 days of continuous treatment of three dosages of YGB. The YGB 50mg/kg showed similar favorable effects on the diabetes and related diabetic complications as compared with CLA 750mg/kg in ob/ob mice of the present study. Conclusions : The results obtained in this study suggest that over 25mg/kg of YGB extracts favorably retarded the obese and type II diabetes in genetically obese and type II diabetic ob/ob mice.

고려인삼의 화학성분에 관한 고찰 (Recent Studies on the Chemical Constituents of Korean Ginseng (Panax ginseng C. A. Meyer))

  • Panax ginseng C.A. Meyer(Araliaceae) has been traditionally used as an expensive and precious medicine in oriental countries for more than 5, 000 years. Ginseng saponin isolated from the root of Panax ginseng have been regarded as the main effective components responsible for the pharmacological and biological activities. Such as antiaging effects. antidiabetic effects anticancer effects. Protection against physical and chemical stress. Analgesic and antipyretic effects. Effects on the central nervous system, tranquilizing action and others. Thirty kinds of ginsenosides have been so far isolated from ginseng saponin and their chemical structures have been elucidated since 1960's. Among which protopanaxadiol type is 19 kinds. protopanaxatriol type. 10 kinds and oleanane type, one. Since ginsenosides are generally labile under acidic conditions ordinary acid hydrolysis is always accompanied by many side reactions, such as epimerization. hydroxylation and cyclization of side chain of the sapogenins Especially. it is well known that C-20 glycosyl linkage of ginsenoside was hydrolysed on heating with acetic acid to give an equilibrated mixture of 20(S) and 20(R) epimers. And also, the chemical transformations of the secondary metabolites have appeared during the steaming process to prepare red ginseng. Indicating demalonylation of malonyl ginsenosides, elimination of glycosyl residue at C-20 and isomerization of hydroxyl configuration at C-20. But these studies have not provided a comprehensive picture in explaning how these ginsenosides showed val'iotas pharmacological activities of ginseng. Though some of them have been involved in the mechanism of pharmacological actions. Recently, non-saponin components have received a great deal of attention for their antioxidant, anticancer antidiabetic, immunomodulating. anticomplementary activities and so on. To meet the demand for such wide applications, studies on the non-saponin components play an important role in providing a good evidence of pharmacological and biol ogical activities. Among the non-saponin constituents of Korean ginseng, polyacetylenes, phenols. Sesquiterpenes, alkaloids. polysaccharides oligosaccharides, oligopeptides and aminoglycosides together with ginsenosides of terrestrial part are mainly described.

Anti-hyperglycemic and Anti-hyperlipidemic Effects of the Triterpenoid-Rich Fractions from Rubus coreanus and Rubus crataegifolius and Their Main Component, Niga-ichigoside $F_1$, in Streptozotocin-induced Diabetic Rats

  • To find the antidiabetic activity of the tripterpenoid-rich fractions of Rubus coreanus (TRF-cor) and R. crataegifolius (TRF-cra) leaves or its main component niga-ichigoside $F_1$ (Niga-$F_1$), anti-hyperglycemic and antihyperlipidemic effects were investigated in the diabetic rat model induced by streptozotocin (STZ). Treatments of rats with 200 mg/kg of the TRF-cor, TRF-cra (each, p.o.) or 20 mg/kg of Niga-$F_1$ significantly inhibited the increase of blood glucose concentration about 44.8%, 28.7% or 20.6%, respectively, in the diabetic rats. In addition, treatments with those fractions inhibited the increase of serum concentrations of triglyceride, total cholesterol or LDL-cholesterol caused by STZ. The inhibitory rate on atherogenic index (AI) values of the TRFcor (200 mg/kg), TRF-cra (200 mg/kg) or Niga-$F_1$ (20 mg/kg)-treated groups were decreased about 55.7%, 36.3% or 22.6%, respectively, comparable to STZ-treated group. In the oral glucose tolerance test, treatment of TRF-cor or TRF-cra inhibited the increase of blood glucose concentration in the STZ-induced rats. Administration of 20 mg/kg of Niga-$F_1$ (p.o.) also exhibited similar effects with the effects of both TRFs at 200 mg/kg dose (p.o.). These results support that the triterpenoids, in particular Niga-$F_1$, are contributed to the antidiabetic effects of R. coreanus or R. crataegifolius.

고지방식이(HFD)/stereptozotocin(STZ) 유도 당뇨모델에서 여주가 글루카곤 분비에 미치는 영향 (Effect of Momordica charantia on Glucagon Secretion in High-fat diet(HFD)/Streptozotocin(STZ)-induced Diabetic Rat)

  • In present study, we investigated the antidiabetic effect of Momordica charantia(as well known "bitter melon"). This study was conducted to determine antidiabetic mechanism of Bitter Melon Extract (BME). We measured blood glucose, insulin, glucagon level in a Sprague-Dawley rat model of high-fat diet/streptozotocin(HFD/STZ)-induced diabetes. Five experimental groups were used: normal, HFD/STZ, BME 62.5 mg/kg HFD/STZ, BME 125 mg/kg HFD/STZ and BME 250 mg/kg HFD/STZ. BME was orally administered to the rats every other day for 9 weeks. Results showed that fasting blood glucose levels were significantly lower in the BME 125 mg/kg(150.17 ± 20.22 mg/dL) and 250 mg/kg(124.17 ± 22.17 mg/dL) groups than in the vehicle group(188.83 ± 26.63 mg/dL)(p<0.05). In addition, glucagon levels were lower in the three BME treatment groups than in the vehicle group(p<0.05). Oral glucose tolerance tests revealed that the BME 250 mg/kg group had significantly(p<0.05) reduced 120-minute blood glucose levels and areas under the curve. Our results suggest that BME induces antidiabetic effects via the reduction of glucagon and blood glucose levels.

서귀포 백년초의 기능성 원료 제조 특성 (Characteristics of Opuntia monacantha Haw. for the Functional Raw Material Production)

  • 백년초(Opuntia) 손바닥 선인장은 우리나라 남쪽 지방인 거제도, 제주도, 남해안 등지에서 자생하며 매년 4~5월경에 꽃이 피고 11~12월경에 자주색 열매가 익는다. 백년초는 예로부터 민간요법에 흔히 사용되었으며 혈액순환을 원활하게 하고 해독, 진통, 항산화 작용 등에 큰 효과가 있다. 본 연구에서는 서귀포 백년초의 항산화성 기능, 항당뇨, 항균활성 효능을 입증하여 기능성 원료로서의 가치를 파악하고자 하였다. 연구 결과 백년초 내 항산화성을 갖는 물질인 택시폴린을 추출하여 강력한 항산화 효과를 확인하였고 항당뇨성 기능 실험에서 시판되고 있는 당뇨치료제와 비교하였을 때 높은 수준의 항당뇨 기능을 확인하였다. 또한 항균 효과 측정 결과 대장균과 살모넬라균 등에서 강한 항균활성을 보여 서귀포 백년초의 기능성 원료로서의 우수한 가치를 입증하였다.

기능성 식품 복합처방의 항산화 및 항당뇨 효과 (The Antioxidant and The Antidiabetic Effects of Ethanol Extract from Biofuntional Foods Prescriptions)

  • To investigate the antioxidant activitys of Biofuntional Foods Prescriptions (BFP), we were measured radical scavenging activity with 1,1-diphenyl-2-picryl-hydrazyl (DPPH) method and anti-lipid peroxidative efficacy on human LDL with thiobarbituric acid reactive substances (TBARS) assay. The DPPH and TBARS assay were revealed that Two BFP had significantly activity when compared to that of contrast drug. The antidiabetic effect of Dicollow-B (Rx B) were investigated in streptozotocin-induced diabetic Spraque-Dawley (SD) rats. When each 100,200 kg/mg fraction of Rx B was administrated. the blood glucose showed 155.7 mg/dl, 124.7 mg/dl and inhibitive activity of the serum total cholesterol showed 67.8 mg/dl, 61.5 mg/dl and the serum triglyce of hyperglycemic rats revealed 66.3 mg/dl, 62.3mg/dl. Those were decreased do dependently when compared with control group. The effect of Rx B on The body weight was not dose-dependently decreased dosedependently when by the administration of 100 mg/kg or 200 mg/kg fractions. But it was dose-dependantly decreased from $22.2{\times}4.9$ g to $18.9{\times}4.4$ g by the administration of 400 mg/kg fraction.

오가나무 잎, 줄기의 항산화 및 항당뇨 효능 분석 (Antioxidant and antidiabetic effects of leaves and stems of Acanthopanax sieboldianum (Makino) Koidz)

  • The aim of this study was to investigate the potential of Acanthopanax sieboldianum (Makino) Koidz (ACS) as a potent antioxidant and antidiabetic agent. The antioxidative and alpha-glucosidase inhibitory activities were examined using the methanol extracts and solvent fractions from ACS-leaf and ACS-stem. Antioxidative activities were measured by in vitro methods such as DPPH and ABTS radical scavenging activity and superoxide dismutase (SOD) activity. When the chloroform and ethyl acetate fractions of ACS-leaf and ethyl acetate fractions of ACS-stem were compared with the control, the SOD-like activity was impaired even at the low treatment concentrations. In addition, the ethyl acetate fractions of ACS-leaf and ACS-stem showed alpha-glucosidase inhibition activities at low treatment concentrations. Analysis of the major components in the fractions of ACS-leaf and ACS-stem was also performed using HPLC. Finally, astragalin, isoqurecetin, chlorogenic acid and caffeic acid contents were measured. Based on this work, we propose that ACS-leaf and ACS-stem have great potential as natural antioxidant and antidiabetic materials related to health benefits.
