An Experimental study on the anticonvulsive effects of Jinkyungondamtang (진경온담탕(鎭驚溫膽湯)의 항경련(抗痙攣) 작용(作用)에 관(關)한 실험적(實驗的) 연구(硏究))

  • In order to investigate the anticonvulsive effect of Jinkyungondamtang, the experiment were performed in mice. After solid exract of Jinkyungondamtang was orally administered to mice, four types of convulsion were induced in mice by electric shock, strychnine, picrotoxin, and caffeine. Each of the time to elicit the convulsion and to die was observed. Results were as follows: 1. In the case of convulsion induced by electric shock, the lapse of the time to elicit convulsion and the time to die were delayed by oral administration of Jinkyungondam-tang(P<0.05, P<0.025). 2. The solid extracts of Jinkyungondamtang were revealed rem- arkable effect in delaying the time to die induced by stry-chnine (P<0.05). 3. By oral administration of Jinkyungondamtang, the lapse of the time to elicit convulsion and to die were delayed significantly to elicit convulsion inducing by picrotoxin (P<0.05, P<0.05). 4. Considerable prolongation of the lapse of the time to elicit the convulsion was observed in convulsion induced caffeine due to the anticonvulsive effects of Jinkyungondamtang (P<0.05).

  • In order to investigate pharmacological actions of combined preparations of crude drugs, 'Jakyakgamcho-Tang' were studied. The 'Jakyakgamcho-Tang' using abdominal pain and convulsion of pediatric diseases are composed of Glycyrrhizae Radix and Paeoniae Radix alba. This study was undertaken to investigate the effect of 'Jakyakgamcho-Tang'(S-I) and parched 'Jakyakgamcho-Tang'(S-II) on anticonvulsive, analgesic, antipyretic and sedative actions. The results of this study were summarized as follows; Both 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang' showed anticonvulsive effects against convulsions induced by strychnine, picrotoxine and caffeine. 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang' also showed analgesic and antipyretic effects. Parched 'Jakyakgamcho-Tang' only prolonged the duration of hypnosis induced by sodium pentobarbital.

  • In order to investigate experimentally the effects of Taeumin Handayolsot'ang and Gamihandayolsot'ang, experimental studies were conducted about antipyretic effect, analgesic effect, antiinflammatory effect of edema, sadative effect, anticonvulsive effect experimental animals (mice and rats). Sample A group was a solid extract of Handayolsot'ang treated group. Sample B group was a solid extract of Gamihandayolsot'ang treated group. The results were summarized as fallows. 1. In antipyretic action by yeast method, sample A group was decreased significantly and sample B group showed decreasing tendency, but showed no significance. 2. In the effect of control for writhing syndrome by the acetic acid stimulating method, sample B group was repressed significantly and sample A group showed repressing tendency, but showed no significance. 3. Antiinflammatory action by carrageenine edema method was showed significant effect at all sample groups. 4. Sadative effect by rotor rod method in rats was not noted at all sample groups. 5. In anticonvulsive action by E C T unit method, time to death in mice was significantly prolonged at all sample groups. According to the above results, it is considered that Taeumin Handayolsot'ang and Gamihandayolsot'ang will be effective on pyretic diseases.

Studies on CNS-Depression and Antiinflammatory Action of Ssangwhatang (쌍화탕의 중추억제 및 항염증작용에 관한 연구)

  • Ssangwhatang which has been known to have antipyretic, analgesic and antifatigue effects are composed of Paeoniae Radix, Angelicae gigantis Radix, Astragali Radix, Rehmaniae Radix, Cnidii Rhizoma, Cassiae Cortex, Glycyrrhizae Radix and Puerariae Radix. Effects of some formulations of Ssangwhatang were studied with respect to the analgesic, antipyretic, hypothermic, anticonvulsive and antiinflammatory actions in experimental animals. The results obtained were as follows: 1) Prescription A, B, C and D of Ssangwhatang were showed significant analgesic effect by the acetic acid stimulating method in mice. 2) All Ssangwhatang-prescriptions used in this studies had hypothermic effect in normal mice. 3) Significant antipyretic effect in febrile rabbits were recognized in prescription A and C of Ssangwhatang, while not in prescription B and D of Ssangwhatang. 4) Prescription A, B, C and D of Ssangwhatang showed marked antiinflammatory effect against the edema induced by carrageenin in rats. 5) Prescriptions of Ssangwhatang did not show significant anticonvulsive effect against the convulsion induced by picrotoxin and strychnine in mice. From the above results, analgesic, hypothermic, antipyretic and antiinflammatory effects were recognized in Ssangwhatang. Ssangwhatang A- and C- prescriptions added with Pueraiae Radix showed better effects than prescription B and C of Ssangwhatang.

Effect of the Ether Fraction of Gastrodia elata Methanol Extract on the Pentylenetetrazole-induced Seizures (천마의 항경련작용기전 연구)

  • Gastrodia elata is a medicinal plant which has been used as anticonvulsant in Korea, Japan and China. This study was conducted to examine the action mechanism of Gastrodia elata centering around the change of GABA and glutamic acid level in brain while observing the anticonvulsive effect in PTZ-induced seizure model. Seizures were reduced effectively by pretreatment of ether soluble part of methanol extract of Gastrodia elata. The pretreatment of ether soluble part inhibited not only the decrease of brain GABA level but also the increase of brain glutamic acid level observed in PTZ model of convulsive dose. Although there was not any change in glutamic acid level, the same development was also observed in the model of subconvulsive dose. From above results, it seems that the anticonvulsive component of Gastrodia elata is lipophilic, and its action mechanism is originated from both control action of GABA level and inhibition of glutaminergic neurotransmission.

The effect of Youngyanggaksan on the anticonvulsive, antipyretic, analgesic, sedative and GABAergic system (영양각산(羚羊角散)이 진경(鎭痙), 해열(解熱), 진통(鎭痛), 진정(鎭靜) 및 GABAergic system에 미치는 영향)

  • This study has been carried out to investigate the effects of Youngyanggaksan (YGS) extract on the anticonvulsive, antipyretic, anlgesic, sedative and GABAergic system of experimental animals. The results of this study were as follows : 1. YGS extract prolonged significantly the beginning time to convulsion and time to death induced by strychnine. 2. YGS extract prolonged significantly the time to death induced by electrical shock of ECT unit (3sec, 200F, 25mA). 3. On the experiment of hypothermic effects of YGS extract on the rectal temperature of rats, YGS extract decreased significantly the rectal temperature of rats. 4. On the experiment of antipyretic effects of YGS extract on the febrile induced by the subcutaneous injection of $150{\mu}g/kg$ endotoxin in rats, YGS extract decreased significantly the rectal temperature of rats. 5. On the experiment of analgesic effects of YGS extract on the writhing syndrome induced by intraperitoneal injection 0.7% acetic acid 1ml/100g in rats, the writhing syndrome was reduced significantly by administration of YGS extract. 6. On the experiment of sedative effects of YGS extract on spontaneous motor activity measured by wheel cage method in mice, the spontaneous motor activity was reduced significantly by administration of YGS extract. 7. On the experiment of effects of YGS extract on the activity of GABA-transaminase(GABA-T) in rat brains after 21 days of oral administration of YGS extract, the activity of GABA-T was reduced significantly by administration of YGS extract. 8. On the experiment of effects of YGS extract on the activity concentration of GABA in rat brains after 21 days of oral administration of YGS extract the activity concentration of GABA was reduced significantly by administration of YGS extract. 9. On the experiment of effects of YGS extract on the activity of GAD in rat brains after 21 days of oral adminstration of YGS extract, the activity of GAD was reduced significantly by administration of YGS extract. According to the those results, Youngyanggaksan extract reveals the effects on the anticonvulsive, antipyretic, anlgesic, sedative and GABAergic system.

  • The sedative, antipyretic, analgesic and anticonvulsive action, action on the isolated ileum in mice, retricting action on edema, action on blood pressure and respiration of Sipmidojuksan were evaluated. The results were as follows; sedative effects were recognized by the unbalanced effects of spontaneous momentum by wheel cage method, muscle relaxing action by rotor rod method and prolongation of sleeping hours. In mice, a significant antipyretic effect to endotoxin was recognized. Significant anagesic effects by acetic acid and Randall-Sellito method were recognized. Significant anticonvulsive effects to strychnine and picrotoxin were recognized. Spontaneous momentum of isolated ileum in mice was restricted, and relaxing effects on smooth muscle of digestive organ were noted by anti-acetylcholine, anti-barium chloride and anti-histamine effects. Significant antiedemic effects to carrageenin and histamine were recognized. Dilatation of blood vessels and decrease of blood pressure were noted.

  • In order to investigate the pharmacological action of combined preparation of crude drugs, 'Oyo-Tang' were studied. 'Oyo-Tang' consist of Glycyrrhizae Radix, Ephedrae Herba, Armenicae Semen, Platycodi Radix and Nepetae Herba. They have widely been prescribed in Oriental Medicine for controlling of cough, asthma, nasal abstraction, pharygeal pain and hoarseness. Experimental studies were implemented on analgesic, sedativc, anticonvulsive and ileum relaxing actions. The results of the studies were as follows; Significant anticonvulsive action was recognized by fraction IIII. Sedative and analgesic actions were noted by fraction I and III. Relaxing action was shown on the extracted ileum in mice, antagonistic actions were seen on $BaCl_2-induced$ contraction of the ileum of mice and Histamine-induced contraction of the ileum of guinea-pigs that the relaxing effect of the intestinal smooth muscle was recognized.

  • The recombinant gamma-interferon (LBD-001) which was produced by yeast as host system was investigated on the pharmacological activities. This gamma-interferon exhibited potent inhibitory effect on adjuvant induced arthritis, but no effect on carrageenin induced paw edema in rats. It did not show any sedative, anticonvulsive, analgesic and hypothermic activities in animals. It also had no influences on isolated tracheal muscle and ileum of guinea pig, isolated uterus and fundus strip of rats, and on blood pressure and respiration in situ experiments of rabbits.
