Cryptosporidium parvum has been recognized as a significant cause of life-threatening diarrhea in Acquired ImmunoDeficiency Syndrome (AIDS) patients. Clinical diagnosis of cryptosporidial infections has been primarily based on the detection of infective stage, oocysts, in stools. Anti-Cryptosporidium oocyst monoclonal antibody (mAb), IgG2a, recognizing an antigen of 97 kDa was generated to be used for diagnosis of Cryptosporidium infection in AIDS patients using an immunofluorecence. It appeared to react with the surface antigens. Transmission electron micrographs of the infective stage of Cryptosporidium recognized by this mAb demonstrated sporolulated oocysts, which measure $4~6{\mu}m$, and sporozoites excysting from oocysts.
To study the efficacy monoclonal antibody(MAb) against bovine rotavirus(BCV) in treatment of calf diarrhea, the MAb was fed to 166 calves with diarrhea from Chung-buk area. The results were summarized as follows. 1. Among the 1,049 calves investigated, 166(16%) calves were infected with BCV. 2. The monthly rate of BCV infection were higher in October to December compared with other months of the year. 3. Among the 166 calves with diarrhea, 137(83%) calves were recovered. 4. Young calves within 7-day-old were more effective in treatment than other ages and the rate of treatment was 86% 5. Most effective period for treatment of rotavirus was at the first stage of infection.
Eight monoclonal antibodies(MAbs) against bovine coronavirus(BCV) were produced and characterized. Three MAbs(1G9, 4H12, 5C1) specific to the S glycoprotein and two HE glycoprotein-specific MAbs(2A5, 5G4) were found to neutralize the BCV in fluorescence focus neutralization(FFN) test. Two HE-specific MAbs from the neutralizing MAbs inhibited the hemagglutinating activity of the BCV. None of the N protein-specific MAbs(1C1, 5A12, 6H1) neutralized the virus infectivity. Bovine coronavirus and mouse hepatitis virus, which belong to group II coronaviruses, were differentiated from other groups of coronaviruses(porcine transmissible gastroenteritis virus, porcine epidemic diarrhea virus, canine coronavirus) by all MAbs in fluorescence antibody test(FA), but not in FFN test.

Acute Megakaryoblastic leukemia is a rare and rapidly fatal disease characterized by proliferation of megakaryocyte series and atypical megakaryocytes in the bone marrow. Acute Megakaryoblastic leukemia is suspicious when 1) megakaryocyte in peripheral blood, mixture of large and small mononuclear megakaryoblast in the bone marrow 2) cytoplasmic budding in blast 3) myelofibrosis (dense medullary overgrowth of reticulin fibers) 4) PAS(+), ANAE(+), SBB(-), peroxidase(-) and which is confirmed by platelet peroxidase oxidation on electromicroscope or monoclonal antibody. A case of aute megakaryoblastic leukemia was studied morphologically and monoclonal antibody.
For a immunohistochemical diagnosis of the frozen and paraffin-embedded tissues against rabies virus, mice were intracerebrally inoculated with challege virus standard(CVS) rabies virus and then were used to detect the rabies viral antigen by the immunoperoxidase(IP) and the avidin-biotin complex(ABC) method. In this study, the results confirmed that ABC and IP methods, although the former showed more specific and sensitive than the latter, were reliable and effective for the demonstration of rabies virus in both frozen and paraffin-embedded brain tissues prepared from rabies-infected mice. Additionally, IP technique using the monoclonal antibody against rabies virus could be recommended as a standard diagnostic tool instead of the present immunofluorescent method for the local veterinary services in Korea.
The water soluble long-chain crosslinker, sulfo-succinimidyl-6-[3'-(2-pyridyldithio)-propion-amido]hexanoate (S-LC-SPDP) was used to prepare ricin A chain (RAT) immunotoxins constructed with whole igG and Fab fragments of the anti-common acute lymphoblastic leukemiz antigen (CALLA)monoclonal antibody. In this study, a) S-LC-SPDP modification efficiencies immunoreactivity and cytotoxicity of immunotoxins constructed were examined. IgG-RTA and Fab-RTA immunotoxins were prepared with 67.3% and 57.0% conjugation yields, respectively. These long spacer intemolecular linked immunotoxins were selectively immunoreactive and to antigen K562 cells. Both IgG-RAT and Fab-RAT immunotoxins were 210-and 45-fold more active than intavt RAT in vitro, respectively.
The effect of media feeding rate on cell growth and monoclonal antibody production in the fed-batch culture ot hybridoma A4W was studied. In the batch culture, the highest specific antibody production rate was observed at the begining of the culture period but its value tended to decrease rapidly with the culture time. The final antibody concentration and volumetric productivity was 65 $\mu g$/ml and 13 mg Mab/l/day, respectively. In the fed-batch culture, the specific antibody production rate, $q_p$ rebounded sharply within a few hours after the media feeding was started and it remained high until the end of culture if the media feeding was continued. The final antibody concentration was 220 $\mu g$/ml and the volumetric productivity was 45.1 mg/l/day. Further increase in final antibody concentration was achieved by applying a modified media of which component was fortified with glucose and glutamine, hence the final antibody concentration in this case was 270 $\mu g$/ml and the volumetric productivity was 51.8 mg/lday, which is as four tinlcs as high cuixparinf! to that of batch culture.

We have developed four chicken hybridomas secreting monoclonal antibodies to induce a protective immune response against the chicken disease avian coccidiosis, caused by the intestinal parasite Eimeria acervulina. Huwever, since the amount of antibodies secreted from these hybridomas is too low or sometimes they lost their ability to produce antibodies, the hybridoma method is not satisfactory in the production of large amounts of chicken monoclonal antibodies. To bypass these problems, we applied the antibody engineering technology using polymerase chain reaction. We cloned and determined the sequences of variable domains of the four chicken monoclonal antibodies, namely, 2-1, 5D11, 13C8 and 8C3. The sequences comparison to germline sequences skewed that the gene con version mechanism might contribute to developing diversification of heavy and λ-light chains in chicken antibodies. Several pseudogene families regarded as donors in gene conversion were identified at each framework region and the complementarily determining region of λ-light chains. In addition, as expected, numerous changes of nucleotide sequences such as nucleotide substitution, insertion and deletion were found predominantly in complementarity determining regions, which are likely to be somatic hypermutations as a result of affinity maturation in antibody-producing cells.
The Langerhans cells are dendritic nonkeratinocytes found suprabasally in most stratified squamous epithelia, such as human epidermis and the epithelium of the oral mucosa including that of gingiva. After Paul Langerhans found it in the skin in 1968, there have been sturdies of it's function and distribution . Stingle et al. reported that the Langerhans cells seem able to present antigens and to stimulate T-lymphocytes. Shelley et al. discovered that they can take up contact allergens. Accordingly it has been suggested that Langerhans cells are important elements of p Peripheral cell mediated immune system. In this study, the gingival tissue of a adult periodontitis patient was taken and freeze dried. In one specimen, we used the CD1 monoclonal antbody to staining the Langerhans cell. The other specimen, we embedded in paraffin and staining it with S-100 monoclonal antibody. The purpose of this study was to use these specimens to find out the distribution, orientation, morphology of the Langerhans cell and to discover the increase or decrease of Langerhans cell in an increased inflammatory state. The results were obtained as follows : 1. Langerhans cells were distributed between the basal cell layer and spinous cell layer against the CD1 & S-100 monoclonal antibody. 2. Langerhans cessl were plentiful in the oral eptihelium, and there was very little in the sulcular epithelium. 3. There were no Langerhans cell in the junction epithelium and pocket lining epithelium. 4. The number of Langerhans cells that responsed to the CD1 & S-100 monoclonal antibody had a statistically difference. 5. As the infiltration of the lymphocyte into the connective tissue were increased, the number of Langerhans cells in the epithelium were increased. 6. As the inflammation was increased, Langerhans cells in the spinous cell layer were more increased than those of the basal layer.
Relatively little is known about the antigenic relatedness of the different developmental stages of Cryptosporidium. A monoclonal antibody (mAb), an IgG3, was produced against the Cryp-tosporidium merozoite stage by immunizing mice with merozoite preparation. This monoclonal was reacted with sporozoite antigens in Western blotting resulting in recognition of an epitope on a 3.5-kDa antigen. An immunoelectron microscopic technique was used to investigate the antigenic relatedness of Cryptosporidium Sporozoites and merozoites. Mouse intestine was fixed with 1 % glutaraldehyde and embedded in LR White. Thin sections were then sequentially treated with murine IgG3 mAb and anti-mouse IgG conjugated to 15-nm diameter colloidal gold. This mAb showed similar (sur-face/cytoplasmic) immunoelectron microsropic colloidal gold labeling patterns with sporozoites and merozoites, indicalting epitope sharing between these two stages. This information might be useful for identifying possible epitopes to which a vaccine could be developed.
