Dose-dependent, immune modulatory effects of aconitine and heated aconitine were studied in mice. Mice, administered aconitine and heated aconitine intraperitoneally every other day for 4 weeks, were sensitized and challenged with sheep red blood cells. Serum antibody titer, foot pad swelling and rosette forming cell number were measured to evaluate hurmoral and cell mediated immune responses. The results show that Humoral immune response was suppressed by aconitine 0.05 mg/kg and heated aconitine but increased by aconitine 0.10 mg/kg administration. Cell mediated immune response was suppressed in all groups. Especially heated aconitine administration significantly suppressed the cell mediated immune response. The number of peripheral circulating white blood cell was reduced by aconitine but was not affected by heated aconitine.
This study was conducted to evaluate the effect of inclusion of propolis extraction residue in the feed of broilers from 1 to 21 d of age on phagocytic activity of macrophages, cutaneous basophil hypersensitivity response to phytohemagglutinin, antibody production against Newcastle disease, lymphoid organ weight and hematological profile and to determine the optimal level of inclusion. 120 chicks, reared in metabolism cages until 21 days of age, were distributed in a completely randomized design, with five treatments (0%, 1%, 2%, 3%, and 4% of propolis residue) and six replications. The relative weight of thymus and monocyte percentage were affected by propolis residue, with a quadratic response (p<0.05) and lowest values estimated at 2.38% and 2.49%, respectively. Changes in relative weight of cloacal bursa and spleen, percentage of lymphocyte, heterophil, basophil, eosinophil, and heterophil:lymphocyte ratio, antibody production against Newcastle disease, phagocytic activity of macrophages and the average number of phagocytosed erythrocytes were not observed. The nitric oxide production with regard to positive control (macrophages+erythrocytes) decreased linearly (p<0.05) with increased doses of propolis residue. The remaining variables of nitric oxide production (negative control - macrophages, and difference between the controls) were not affected by propolis residue. The cutaneous basophil hypersensitivity response to phytohemagglutinin as determined by the increase in interdigital skin thickness exhibited a quadratic response (p<0.05), which predicted a lower reaction response at a dose of 2.60% of propolis residue and highest reaction response after 43.05 hours of phytohemagglutinin injection. The inclusion of 1% to 4% of propolis extraction residue in broiler diets from 1 to 21 days of age was not able to improve the immune parameters, despite the modest changes in the relative weight in thymus, blood monocyte percentage, nitric oxide concentration, and interdigital reaction to phytohemagglutinin.

The study was undertaken to evaluate the effect of naked neck gene on mortality, cell mediated and humoral immune response in white plumage broiler population. The mortality of homozygous naked neck (Na/Na) broilers (11.71%) was comparatively lower than that of heterozygous naked neck (Na/na) (12.28%) and normally feathered (na/na) (13.59%) broilers. The humoral immune response was measured against (1% v/v) sheep red blood cells (SRBC) for total haemagglutinin (HA) antibody, 2-mercaptoethanol resistance (MER) or (IgG) antibody and 2-mercaptoethanol sensitive (MES) or (IgM) antibody titre on 7 days post-immunization. The titre was expressed as log2 of the highest dilution which shows complete haemagglutination. Total HA titers of Na/Na and Na/na (11.05$\pm$0.53 and 11.09$\pm$0.38) were comparatively higher than that of na/na (10.26$\pm$0.42). The MES antibody titre of Na/Na (8.50$\pm$0.53) and Na/na (7.63$\pm$0.45) broilers were significantly higher as compared to na/na (6.11$\pm$0.32) broilers. The MER titre of na/na genetic group (4.15$\pm$0.42) was significantly higher than Na/Na (2.55$\pm$0.37) and comparatively higher than Na/na (3.45$\pm$0.38) broilers. In vivo cell response to phytohaemagglutinin-P (PHA-P), measured as Foot Index (FI) in mm expressed significantly higher response in Na/na (0.473$\pm$0.05) and Na/Na (0.413$\pm$0.04) broilers as compared to na/na (0.304$\pm$0.03) broilers. The result of present study suggested that white plumage naked neck broilers had better immune response as compared to normally feathered broilers.
The present experiment was designed to investigate the effects of behavioral, response to immune function in response to electric footshock in mice. Mice were subjected to electric footshock for 3 days(two sessions a day, 11 times of shock for about 31 minutes a session). The humoral immune response was measured using mice immunized with rat RBC. The cell-mediated immune responses were evaluated by contact hypersensitivity to 2, 4-dinitrofluorobenzene(DNFB) and by phytohemagglutin(PHA)-stimulated splenocytes proliferation assay. In stressed group, electric footshock suppressed significantly anti-rat RBC antibody production(p<0.05), but enhanced significantly $T_{48}$ relative to $T_{24}$ in contact hypersenstivry (P<.01) and T-cell proliferation response(P<.05) by PHA stimulation elative to control group. T-cell proliferation response by PHA stimulation was significantly correlated to the movement than the sensitivity and coping behavior in the mouse, in response to the electric footshock. These data supper the importance of behavioral response in stress-induced changes of immune functions.
The bursa of Fabricius (BF) is a central humoral immune organ unique to birds. Four bursal peptides (BP-I, BP-II, BP-III, and BP-IV) have been isolated and identified from the BF. In this study, the immunoadjuvant activities of BPs I to IV were examined in mice immunized with H9N2 avian influenza virus (AIV) vaccine. The results suggested that BP-I effectively enhanced cell-mediated immune responses, increased the secretion of Th1 (interferon gamma)- and Th2 (interleukin-4)-type cytokines, and induced an improved cytotoxic T-lymphocyte (CTL) response to the H9N2 virus. BP-II mainly elevated specific antibody production, especially neutralizing antibodies, and increased Th1- and Th2-type cytokine secretion. BP-III had no significant effect on antibody production or cell-mediated immune responses compared to those in the control group. A strong immune response at both the humoral and cellular levels was induced by BP-IV. Furthermore, a virus challenge experiment followed by H&E staining revealed that BP-I and BP-II promoted removal of the virus and conferred protection in mouse lungs. BP-IV significantly reduced viral titers and histopathological changes and contributed to protection against H9N2 AIV challenge in mouse lungs. This study further elucidated the immunoadjuvant activities of BPs I to IV, providing a novel insight into immunoadjuvants for use in vaccine design.
We determined the effects of temperature shifts on the kinetics of the numbers of antibody-secreting cell (ASC) in the olive flounder Paralichthys olivaceus immunised with formalin-killed Edwardsiella tarda. When fish that were acclimated to $22^{\circ}C$ and immunised at that temperature were transferred to a lower temperature ($12^{\circ}C$) at a various times (immediately, 1, 2 or 4 weeks) after immunisation, both further differentiation of B cells and secretion of antibody from the ASC developed at $22^{\circ}C$ were suppressed at $12^{\circ}C$. However, in the converse experiment ($12^{\circ}C$ to $22^{\circ}C$), the magnitude of the humoral immune response was recovered independent of the time of the transfer after immunisation at low temperature, even though the peak levels of each transferred group did not reach the level found in $22^{\circ}C$ control group. The results were confirmed by counting the number of specific antibody secreting cells (SASC) in the spleen. This study provides the evidences of the immune reaction that the potential for antibody production in B cells of flounder, the most important species in aquatic industry of Korea, immunized at high temperature is suppressed by subsequent exposure to low temperature and that low temperature-induced humoral immuno suppression can be reversed by exposure to a higher temperature.

Mice immunized with equine herpesvirus type-1(EHV-1) L-particles skewed a significant increase (p<7.75) in serum antibody titers. Upon a booster dose four weeks lateral antibody titers increased significantly. Interestingly, immunization via intravenous or intramuscular route induced significantly higher (p<0.75) antibody titers. However, mice iummunized with UV-treated L-particles, visions or immunization via intranasal route induced lower antibody titers. Upon challenge inoculation with wildtype EHV-1, our data showed there was a poor correlation between antibody titers and protection against virus replication. Therefore, the role of cell-mediated immunity Inwards protection was investigated. As predicted, the strongest cell-mediated immunity, as measured by delayed-hypersensitivity test, was detected in mice immunized with live virus particles. The magnitude of cell-mediated immune response correlated with the efficacy of L-particles as immunizing agent. The highest efficacy, as indicated in mice immunized via intranasal routed was highly correlated with cell-mediated immunity. A similar phenomenon was also demonstrated in mice immunized intranasally with UV-treated L-particles. However, the degree of protection was reduced when mice immunized intravenously or intramuscularly with UV-treated L-particles. In conclusion, protection conferred in these animals was highly implicated by immune cells and the least by antibodies. The route of immunization and the nature of the antigen also contributed to the efficacy of L-particles as immunizing agent. In contrast to that of herpes simplex virus type 1, our data showed EHV-1 non-infectious L-particles are highly suitable for immunization of the host against EHV-1 disease.
In this paper, we propose an adaptive PID controller using a cell-mediated immune response to improve a PID control performance. The proposed controller is based on the specific immune response of the biological immune system that is cell-mediated immunity. The immune system of organisms in the real body regulates the antibody and the T-cells to protect an attack from the foreign materials like virus, germ cells, and other antigens. It has similar characteristics that are the adaptation and robustness to overcome disturbances and to control the plant of engineering application. We first build a model of the T-cell regulated immune response mechanism and then designed an I-PID controller focusing on the T-cell regulated immune response of the biological immune system. We apply the proposed methodology to building structures to mitigate vibrations due to strong winds for evaluation of control performances. Through computer simulations, system responses are illustrated and additionally compared to traditional control approaches.
The effect of rancid perilla oil on the immune response in mice was studied. ICR male mice were divided into 5 groups and were fed on the experimental diets for 4 weeks. Mice were sensitized and challenged with sheep red blood cell. Immune responses were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette forming cell and macrophage activity. Biochemical items were measured by serum protein and serum albumin. The weight of spleen, thymus and liver were measured. The rancid perilla oil diets decreased humoral and cellular immune responses, the number of peripheral circulating white blood cells and total protein and serum albumin. These results showed that the high rancid perilla oil diet decreased more humoral and cellular immune response, the number of peripheral circulating white blood cells, and total protein and serum albumin than the low rancid perilla oil diet did.
Effect of the butanol fraction of Astragali Radix (BFAR) on the humoral immune response were investigated in ICR mice. Mice were divided into 4 groups and BFAR at doses of 5,25 and 125 mg/kg were administered orally to mice daily for 3 weeks, and the normal animals were given vehicle. The results of this study are summarized as follows; the relative weight of spleen was markedly increased by BFAR treatment, compared with that in normal mice. However, the body weight gain and the relative weight of liver were not affected. Splenic plaque forming cells and hemagglutination titers to sheep red blood cells, and the secondary IgG antibody response to bovine serum albumin were also dose-dependently enhanced by BFAR treatment. In these mice, BFAR did not increase serum alanine aminotransferase total protein, sect albumin and albumin/globulin ratio when compared with those in normal mice. Thus, these findings indicate that BFAR significantly enhances humoral immune response to antigen in concentrations that do not affected liver function.
