Periplanetasin-2 Enhances the Antibacterial Properties of Vancomycin or Chloramphenicol in Escherichia coli

  • Periplanetasin-2 from cockroach exhibits broad-spectrum antimicrobial activity. The underlying antibacterial mechanisms rely on the stimulation of reactive oxygen species overproduction to induce apoptotic cell death. A promising strategy to increase the bioavailability of periplanetasin-2 involves reducing the dose through combination therapy with other antibacterials that show synergistic effects. Thus, the synergistic antibacterial activity of periplanetasin-2 with conventional antibacterial agents and its mechanisms was examined against Escherichia coli in this study. Among the agents tested, the combinations of periplanetasin-2 with vancomycin and chloramphenicol exhibited synergistic effects. Periplanetasin-2 in combination with vancomycin and chloramphenicol demonstrated antibacterial activity through the intracellular oxidative stress response. The combination with vancomycin resulted in the enhancement of bacterial apoptosis-like death, whereas the combination with chloramphenicol enhanced oxidative stress damage. These synergistic interactions of periplanetasin-2 can help broaden the spectrum of conventional antibiotics. The combination of antimicrobial peptides and conventional antibiotics is proposed as a novel perspective on treatments to combat severe bacterial infection.

Antibacterial properties of composite resins incorporating silver and zinc oxide nanoparticles on Streptococcus mutans and Lactobacillus

  • Objectives: Recurrent caries was partly ascribed to lack of antibacterial properties in composite resin. Silver and zinc nanoparticles are considered to be broad-spectrum antibacterial agents. The aim of the present study was to evaluate the antibacterial properties of composite resins containing 1% silver and zinc-oxide nanoparticles on Streptococcus mutans and Lactobacillus. Materials and Methods: Ninety discoid tablets containing 0%, 1% nano-silver and 1% nano zinc-oxide particles were prepared from flowable composite resin (n = 30). The antibacterial properties of composite resin discs were evaluated by direct contact test. Diluted solutions of Streptococcus mutans (PTCC 1683) and Lactobacillus (PTCC 1643) were prepared. 0.01 mL of each bacterial species was separately placed on the discs. The discs were transferred to liquid culture media and were incubated at $37^{\circ}C$ for 8 hr. 0.01 mL of each solution was cultured on blood agar and the colonies were counted. Data was analyzed with Kruskall-Wallis and Mann-Whitney U tests. Results: Composites containing nano zinc-oxide particles or silver nanoparticles exhibited higher antibacterial activity against Streptococcus mutans and Lactobacillus compared to the control group (p < 0.05). The effect of zinc-oxide on Streptococcus mutans was significantly higher than that of silver (p < 0.05). There were no significant differences in the antibacterial activity against Lactobacillus between composites containing silver nanoparticles and those containing zinc-oxide nanoparticles. Conclusions: Composite resins containing silver or zinc-oxide nanoparticles exhibited antibacterial activity against Streptococcus mutans and Lactobacillus.

In vitro Antimicrobial Activities of Edible Seaweeds Extracts Against Cutibacterium acnes (여드름균(Cutibacterium acnes)에 대한 해조류 추출물의 항균효과)

  • The study was aimed at developing an alternative therapeutic agent against acne vulgaris, one of the most common skin diseases, to meet the continuing demand for new therapies. Acne vulgaris is often associated with the acne-causing bacteria such as Cutibacterium acnes. To investigate the safety of agents against acne vulgaris, we evaluated the potential antibacterial activities of edible seaweeds against C. acnes in Korea. Forty-one edible seaweeds, including the brown, green, and red varieties, were selected for the antibacterial test. In comparison with other seaweeds, 70% ethanolic extracts of brown seaweeds, such as Cladophora wrightiana var. minor, Eisenia bicyclis, Ecklonia cava, Ishige foliacea, Ishige okamurae, Sargassum filicinum, and Sargassum miyabei Yendo, exhibited potential antibacterial activity against C. acnes with minimum inhibitory concentrations ranging between 64 and 128 ㎍/mL. To investigate the active anti-acne agents and to enhance our understanding of the antibacterial activities against C. acnes, further solvent-fractionation experiments are warranted. The findings imply that brown seaweeds can be a potential source of natural agents against acne vulgaris.

A Study on the Durability and Synthesis of Sulfa Agents Using Glutaraldehyde (Glutaraldehyde를 이용한 설파제의 합성과 지속성에 관한 연구)

Dual-action agents are unique chemical entities comprised of two different type of antibacterial compounds covalently linked together in a single molecule in such a way that both components are able to exert their bactericidal properties. Crosslinked sulfadiazine-sulfanilamide such as antibiotics is synthesized by synthetic handle with glutaraldehyde. As a result, New synthetic antibacterial agent exhibited the broad antibacterial activities against Gram(+) and Gram(-) of 4 strains and a long durability supposing that the stomach and blood.

Cathodic Coloration of Silk Fabric Treated with Silver Nitrate (질산은 처리된 실크의 환원 발색)

  • In this article, silk fabric was treated with silver nitrate ($AgNO_3$) as oxidizing agent, with conditions such as concentration, and treating time, and subsequently treated with reducing agents such as sodium boron hydride ($NaBH_4$) and sodium sulfide ($Na_2S$) to obtain cathodic coloration. After coloration of silk fabric, dyeing properties (K/S value), colorfastnesses such as wash, rubbing and light, and antibacterial activity property were examined. $AgNO_3$ treating time and reducing time did not influence K/S value, whereas the pH value at alkaline region showed a high K/S value of silk fabric. The cathodic colorations of silk fabric with both of reducing agents at $30^{\circ}C$ have excellent color fastnesses. Also the high antibacterial activities were obtained by the treatment with silver nitrate even at 1% of lower concentration.

  • To develop the natural antibacterial agents which don't have any toxicity against man, collected several species of medicinal plants were tested for their antibacterial activity from dental caries bacteria Streptococcus sanguis. The result of using paper disc method and the result of viable cell counting method, Phellodendri Cortex was selected as antibacterial agent. The high antibacterial activity was acquired at high extraction temperature and long extraction temperature. The antibacterial of Phellodendri Cortex was not effected by the concentration of ethanol.

  • Antibacterial finishing of nylon 66 fabrics was carried out with sericite and medilite which are a cheap price antibacterial agents and had excellent antibacterial effects. The particle size of sericite and medilite was 15 ${\mu}m$ and 30 ${\mu}m$. The antibacterial and deodorant ratio, tensile and tear strength, peel strength were examined to investigate the change of physical properties and antibacterial effect. The results are as follows. 1. Peel strength is increased with increasing adhesive content, and satisfied standard value of peel strength when adhesive content is 20 $g/m^2$. Also peel strength was decreased with increasing antibacterial agent content and particle size in the adhesives. 2. Tensile and tear strength were not related with antibacterial agent content in the adhesives. 3. Laundering nylon 66 fabric treated with antibacterial agent, the optimum content satisfying Korean Standard(KS) is 8%. The antibacterial and deodorant ratio were not affected by several types of adhesives.
