구강균 Streptococcus sanguis에 대한 황백의 생육 저해 효과 (Antibacterial Activity of Phellodendri Cortex on Dental Caries Bacteria Streptococcus sanguis)

  • To develop the natural antibacterial agents which don't have any toxicity against man, collected several species of medicinal plants were tested for their antibacterial activity from dental caries bacteria Streptococcus sanguis. The result of using paper disc method and the result of viable cell counting method, Phellodendri Cortex was selected as antibacterial agent. The high antibacterial activity was acquired at high extraction temperature and long extraction temperature. The antibacterial of Phellodendri Cortex was not effected by the concentration of ethanol.

냉면용 동치미액 제조를 위한 항균 활성이 높은 젖산균의 선발 (Screening of high Antibacterial Lactic Acid bacteria for the Preparation of Dongchimi-Juice for Naengmyon)

  • In order to screen lactic acid bacterial starter having high antibacterial activity and giving good flavor in Dongchmi for Naengmyon antibacterial activity and acid producing ability of 24 strains of Kimchi lactic acid bacteria were tested. Among 24 strains tested Leuconostoc mesenteroides subsp. mesenteroides C16, Leu. paramesenteroides C18 Lactobacillus bavaricus B01, B06, C19, C32 Lac. homohiochii B21 and B22 showed high antibacterial activity and their antibacterial activities were more active against Listeria monocytogenes and Staphylococcus aureus than against Escherichia coli and Salmonella typhimurium. The odors of Dongchimi-juices fermented with Leu. mesenteroides or Leu. paramesenteroides were more favorable than those with Lac. bavaricus or Lac. homohiochii. But the odors of Naengmyon-broths which were made with each Dongchimi-juice were not significantly different each other. In consideration of antibacterial ac-tivity and flavor of Dongchimi-juice two strains of lactic acid bacteria Lac. homohiochii B21 and Leu. mes-enteroides subsp. mesenteroides C16 were finally screened out as Dongchimi starters.

Antibacterial Activity and Inhibitory Effect of Biofilm Formation by Actinidia polygama against S. mutans and P. gingivalis

  • In oriental medicine, the fruit of Actinidia polygama has long been used to alleviate the symptoms of gout, arthritis, and inflammation. In this study, it was to designed to analyze the antibacterial activity of A. polygama ethanol extract (APEE) against Streptococcus mutans, one of the major strains for dental caries, and Porphyromonas gingivalis, one of the critical strains for periodontal disease. The antibacterial activity of APEE was analyzed by disk diffusion, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) assays. In addition, it was also analyzed the inhibitory effect of APEE on bacterial growth and biofilm formation against both oral pathogens. APEE exhibited its antibacterial effect through the inhibited bacterial diffusion as well as low concentration of MIC and MBC. In addition, APEE significantly inhibited not only bacterial growth but also biofilm formation in a dose-dependent manner. Consequently, APEE showed potent antibacterial activity against both S. mutans and P. gingivalis, which indicates that APEE might be used as a potential antibacterial material for the improvement of oral healthcare.

Lactobacillus helveticus의 자연항생작용에 관한 연구 (Natural Antibiotic Activity of Lactobacillus helveticus)

  • The objective of this study was to extract and purity the antibacterial agent from the fermented milk with Lactobacillus helveticus CH-1. The extraction and purification of antibacterial agent from the Lb. helveticus fermented milk were carried out by methanol extraction, acetone extraction, Sephadex G-200 gel filteration and thin layer chromatography and the results were as followings. The antibacterial activity of methanol-acetone extraction showed antibacterial activity against test organisms, B. subtilis, E. coli, Pseu. fluorescens, Sal. typhimurium, Shi. flexneri, and Sta. aureus. Sephadex G-200 gel chromatography showed only antibacterial activity from 33 to 37th fractions of 60 fractions. The agent purified from TLC plate confirmed the antibacterial activity by the means of bioautography.

Antibacterial and virucidal activity of 28 extracts from plants endemic to Korea against Bacillus cereus, Staphylococcus aureus, and murine norovirus

  • Antibacterial activity against foodborne bacteria (Bacillus cereus, Staphylococcus aureus, Salmonella Enteritidis) and inhibitory activity against murine norovirus, a human norovirus surrogate, of 28 extracts from plants endemic to Korea were investigated in this study. All plant extracts showed antibacterial activity only against gram-positive bacteria, B. cereus and S. aureus. Extracts from Callistemon speciosus and Nymphaea tetragona showed inhibition zones of 16.54 and 24.35 mm against B. cereus and S. aureus, respectively, presenting the highest antibacterial activities recorded in this study. Among all samples, Ardisia japonica extract at concentrations of 100 and 200 ㎍/mL showed the highest virucidal activities of 96.6 and 100.0%, respectively. Ardisia japonica, Duchesnea indica, Polygonum aviculare, and Geum japonicum extracts showed high antibacterial and virucidal activity simultaneously without Raw 264.7 cell cytotoxicity. These plant extracts may serve as potential antimicrobials to control foodborne infections.

Antibacterial Activity of the Phaeophyta Ecklonia stolonifera on Methicillin-resistant Staphylococcus aureus

  • In an effort to discover an alternative therapeutic agent against methicillin-resistant Staphylococcus aureus (MRSA), several medicinal plants and seaweeds were evaluated for its antibacterial activity against MRSA. A methanolic extract of the Phaeophyta Ecklonia stolonifera exhibited significant antibacterial activity against MRSA. To perform more detailed investigation on antibacterial activity, the methanol extract of E. stolonifera was further fractionated with organic solvents such as hexane, dimethylchloride, ethyl acetate, and n-butanol. Among them, the hexane fraction showed the strongest antibacterial activity against MRSA strains with MIC from 500 to $600 {\mu}g/mL$. The fraction also exhibited a bactericidal activity against MRSA, indicating that E. stolonifera contains a bactericidal substance against MRSA.

화피의 충치균과 치주질환균에 대한 항균활성 및 항염효과 (Antibacterial Activity against S. mutans or P. gingivalis and Anti-inflammatory Effect of Betulae Cortex)

  • Betulae Cortex of Betula platyphylla Suk. var. japonica Hara (Betulaceae) has long been used for treatment of various inflammation, fever and cough in Eastern Asia. In order to investigate antibacterial activity of the Betulae Cortex against Streptococcus mutans or Porphyromonas gingivalis, MIC (minimal inhibitory concentration) and pH were checked, and for anti-inflammation activity, the experiments about trypsin-induced paw edema, vascular permeability and myeloperoxidase activity in rat's hind-paw tissue, were carried out with various extracts of Betulae Cortex (BCXs) respectively. The BCXs showed significant antibacterial activity, and at the dose of over 50 mg/kg, BCX showed significant inhibition on the paw edema, vascular permeability and myeloperoxidase activity. These results indicate that BCXs have antibacterial activity against oral cariogenic bacteria and anti-inflammatory effect.

한약재 추출물의 피부 염증 유발 세균에 대한 항균활성 (Antibacterial Activity of Oriental Medicinal Herb Extracts against Skin Pathogens)

  • 한약재 70종의 메탄올 추출물을 이용하여 4종의 Staphylococcus epidermidis와 S. aureus에 대한 항균효과를 조사한 결과 모든 공시균주에 대하여 항균성을 나타내는 한약재는 황련, 소목, 오배자, 대황과 단삼으로 조사되었다. 이 중 우수한 항균성을 나타낸 오배자와 황련을 물, 에탄올, 메탄올과 에틸아세테이트를 추출 용매로 하여 추출물을 제조하고 항균성을 조사한 결과 황련 열수 추출물에서 가장 우수한 항균성을 확인할 수 있었다. 황련의 항균성은 berberine에 의한 것으로 가정하고 열수 추출물 중 berberine의 함량을 조사한 결과 13.88%로 조사되었다. Berberine에 의한 항균성과 다른 항생제와의 항균성을 비교하기 위하여 berberine chloride와 3종의 항생제를 이용하여 조사한 결과 600 mg/l 이상의 berberine chloride 농도에서 KCCM 35494 S. epidermidis에 대한 항균성만 나타내었으며 다른 공시균주에 대한 항균성은 확인되지 않았다. 이에 본 연구에 사용한 공시균주에 대한 C. japonica 열수 추출물의 항균성은 고 농도의 berberine 또는 다른 물질에 의한 것으로 판단할 수 있었으며 이 물질에 대한 연구가 필요할 것으로 판단되었다.

Influence of the N- and C-Terminal Regions of Antimicrobial Peptide Pleurocidin on Antibacterial Activity

  • Pleurocidin, a 25-mer antimicrobial peptide, has been known to exhibit potent antibacterial activity. To investigate the functional roles in N- and C-terminal regions of pleurocidin on the antibacterial activity, we designed four truncated analogs. The antibacterial susceptibility testing showed that pleurocidin and its analogs exerted antibacterial effect against various bacterial strains and further possessed specific activity patterns corresponding with their hydrophobic scale [pleurocidin > Anal 3 (1-22) > Anal 1 (4-25) > Anal 4 (1-19) > Anal 2 (7-25)]. Fluorescence experiments using 1,6-diphenyl-1,3,5-hexatriene (DPH) and 3,3'-dipropylthiadicarbocyanine iodide [$diSC_3(5)$] indicated that the differences in antibacterial activity of the peptides were caused by its membrane-active mechanisms including membrane disruption and depolarization. Blue shift in tryptophan fluorescence demonstrated that the decrease in net hydrophobicity attenuates the binding affinity of pleurocidin to interact with plasma membrane. Therefore, the present study suggests that hydrophobicity in the N- and C-terminal regions of pleurocidin plays a key role in its antibacterial activity.

Antibacterial activity of Dioscorea batatas, Morus alba, and Tagetes erecta against E. coli and S. aureus

  • In this study, the anti-bacterial activity of three medicinal herbs such as Dioscorea batatas, Morus alba, and Tagetes erecta, have been applied in oriental medicine against Escherichia coli and Staphylococcus aureus. The antibacterial activity of Dioscorea batatas ethanol extract (DBEE), Morus alba ethanol extract (MAEE), and Tagetes erecta ethanol extract (TEEE) was analyzed by disk diffusion, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) assays. As a result, three medicinal herbs exhibited antibacterial activity in a dose-dependent manner as well as MAEE and TEEE showed the most potent antibacterial activity followed by disk diffusion assay against E. coli and S. aureus, respectively. MIC against E. coli and S. aureus was 1.6 mg/mL of MAEE and TEEE as well as 0.2 mg/mL of TEEE. MBC against both strains was 5 mg/mL of DBEE and 2.5 mg/mL of TEEE. Consequently, three medicinal herbs in this study showed potent antibacterial activity through the inhibited growth of E. coli and S. aureus, which is considered as a potential candidate for cosmeceutic agents to attenuate the pruritus. In addition, antibacterial and antihistamine effects followed by the extraction solvents should be analyzed in future studies.
