Functional Characterization of the Extracts from Nipa Palm, Molokhia, and Finger Root for Cosmetic Ingredients (니파팜, 몰로키아, 핑거루트 추출물의 화장품 소재로서의 기능적 특성 분석)

  • This study was conducted to evaluate the applicability of the extracts from nipa palm, molokhia, and finger root in functional cosmetics as a natural active ingredient. The extracts were obtained through the processes of heating under reflux with ethanol, filtration, concentration, and freeze-drying. UV absorption and blocking effects of the extracts were examined by using the UV-vis spectrophotometer equipped with an integrating sphere. Antioxidant activity and its stability between the extracts were compared using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Also, total polyphenol content in the extracts was determined quantitatively using the Folin-Ciocalteu reagent, with gallic acid as the standard. Antibacterial activity of the extracts was investigated by the disc diffusion test against Staphylococcus aureus (gram-positive) and Escherichia coli (gram-negative). Finally, collagenase inhibitor assay was performed to examine the anti-wrinkle effect of the extracts. From the results of this study, the extract of nipa palm showed the potential for use in cosmetics as an antioxidant and anti-wrinkle agent, and the extract of finger root as a sunscreen and antibacterial agent.

  • All living organisms exhibit the characteristics of aging, such as skin wrinkle formation, muscle degeneration, cataracts, and hair graying as the number of aged cells increases over time. Senescence, which is known as a key cause of aging, is directly related to the aging of living organisms because cells are aged by external and internal factors and eventually cell proliferation is stopped. Senescence is caused by the gradual shortening of the telomere with cell division, and lifespan is determined by the length of the telomere. Recently, it has been found that the histone deacetylase, which can influence gene expression, is not only involved in yeast but also deeply involved in anti-aging mechanisms in both C. elegans and humans. It was also discovered that old cells play a decisive role in the aging phenomenon, and it has been reported that it is possible to promote the proliferation of young cells and delay aging by removing these senescent cells from the inside. Therefore, in order to develop potential anti-aging agents in the future, research should begin with an in-depth study of telomerase activators, sirtuin activators, and senolytics.

  • Fructose-1, 6-diphosphate (FOP), a glycolytic metabolite is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FOP has cytoprotective effects against hypoxia or ischemia/reperfusion injury in brain and heart. In this study, we examined whether FDP has protective effects on UV-induced oxidative damage in skin cell culture system and human skin in vivo. FDP had a protective role in UVB-induced LDH release and ROS accumulation in HaCaT although it did not show direct radical scavenging effect in the experiment using 1, 1-diphenyl-2-picrylhydrazyl (DPPH). FDP also preserved cellular GSH content after UV irradiation in HaCaT and normal human fibroblast culture system. Cellular oxidative stress induces multiple downstream signaling pathways that regulate expression of multiple gene including MMP-1 and collagen, we examined the effects of FDP on UV-induced alteration of these protein expression in fibroblast culture and human skin in vivo. The increased MMP-1 expression in fibroblast and human skin by UV irradiation was significantly decreased by FDP. FDP also prevented the UV-induced decrease of collagen expression in fibroblast and human skin. Moreover, the decreasing the intracellular levels of reducing equivalents in human fibroblast by glutathione (GSH) depletion lowered the UVA dose threshold for reduction of procollagen expression, indicating that the differences of glutathione contents define the susceptibility of fibroblasts towards UV-induced reduction of procollagen expression. FDP also preserved cellular GSH content after UV irradiation, indicating that FDP has protective effects on UV-induced reduction of procollagen expression, which are possibly through maintaining intracellular reducing equivalent. Based on these premises, we examined the effect of daily use of a moisturizer containing FDP on facial wrinkle in comparison with vehicle moisturizer lacking FDP. In the clinical study, FDP significantly decreased facial wrinkle compared with vehicle alone after 6 months of use. Our results suggest that FDP has anti-aging effects in skin by increasing cellular antioxidant system and preventing oxidative signal and inflammatory reaction. Therefore FDP may be useful anti-aging agent for cosmetic purpose.

Analysis on the Efficacy of Cosmetic Application of Lijang Snow Tea (Nekemias grossedentata) (리장 설차 (Nekemias grossedentata )의 화장품적 적용 효능 분석)

  • In this study, we analyzed the cosmetic applicability of extract from snow tea, native to Lijiang, Yunnan-province, China. After confirming the species as N. grossedentata through DNA analysis of Lijiang snow tea, experiments were conducted using representative tea, green tea, and a representative control group for each efficacy analysis. Both teas were extracted using 70% (v/v) ethanol aqueous solution. The polyphenol content in the Lijiang snow tea extract (gallic acid equivalent, 23.9 ± 3.2 mg/mL) was higher than that in green tea extract (16.4 ± 2.3 mg/mL). In contrast, the antioxidant (Radical scavenging, IC50 104 ㎍/mL), tyrosinase enzyme inhibitory (whitening agent, IC50 40.7 ㎍/mL), and Escherichia coli growth inhibitory (preservative) activities (IC50 2.85 mg/mL) were analyzed based on the solid content in the extract, and it was confirmed that the activities of Lijiang snow tea extract were superior to those of green tea extract (radical scavenging, IC50 234 ㎍/mL. It also showed similar efficacy to previously used active substances such as antioxidants (vitamin C, IC50 108 ㎍/mL), whitening agents (vitamin C, IC50 80㎍/mL), and preservatives (methylparaben, IC50 4.35 mg/mL). However, green tea was found to be better in collagenase inhibition activity (anti-wrinkle). Through this study, the cosmetic application potential of Lijiang snow tea is high.

Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening (왕겨초액의 대량생산과 항노화 및 미백 효과)

  • Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.

  • Chitosan and CA/BTCA were employed in order to induce crosslinking in the fiber for the improvement of wrinkle recovery of the cotton fabrics and the endowment of anti-microbial functions to the fabric. The treated fabrics were dyed by using reactive dyestuff and their dyeing behaviors were investigated. As a result, the fabric treated with chitosan only exhibited more dye-uptake amount than the untreated fabric, and the treated and untreated fabrics together showed excellent light-fastness and wash-fastness. In the cases that CA or BTCA was added to the chitosan, the CA-treated showed better dye-uptake and dyeing properties as light-fastness than the BTCA-treated. In conclusion, it is possible to maintain the dye-uptake level at reduced treatment cost when the CA is employed as a substitute cross-linking agent for BTCA.

The Melanin Inhibition, Anti-aging and Anti-inflammation Effects of Portulaca oleracea Extracts on Cells (쇠비름 추출물의 미백 및 항노화, 항염증 효과)

  • The Portulaca oleracea (P. oleracea) is a popular herbal medicine in East Asia that was known to possess detoxification, antifebrile and antifungal effects. In the present study, we examined the biological activities of ethanol extracts of P. oleracea under various conditions with NIH3T3, B16F10, and MCF-7 cell line model systems. Extracts of P. oleracea (0.5 mg/ml) showed inhibition of expression of tyrosinase, but does not suppress either of TYRP-1 or DCT expression on B16F10 cells. Extracts of P. oleracea (2 mg/ml) showed anti-inflammatory effects on TNF-$\alpha$-stimulated NIH3T3/$NF{\kappa}B$-Luc cells and increase of the synthesis of collagen on NIH3T3 (wild type) cells. These results suggest that extracts of P. oleracea could be used as a functional biomaterial in developing a skin whitening agent and having the anti-inflammatory, anti-wrinkle, and anti-aging activities.

Changes of Facial Wrinkle after Topical Application of On Emulsion Containing Medimin A (Medimin A를 함유한 O/W 에멀전의 주름 개선 효과)

  • Medimin A is a derivative of vitamin A which has been developed by coupling retinoic acid with polyethylene glycol(PEG) to enhance skin permeability and stability. We carried out the collagen synthesis and clinical test to examine the reducing effect of wrinkles by Medimin A. In vitro collagen synthesis was evaluated by quantitative assay of ($^3$H)-proline incorporation into collagenase sensitive protein in fibroblast cultures. Clinical test was evaluated by image analysis of skin replica, visual observation and self-estimated response of volunteers for 10 weeks. Medimin A stimulated about 40% in collagen synthesis. The area of main deep wrinkle on the skin replica was reduced 38.4% with topical application of O/W emulsion containing 0.2% Medimin A. The wrinkles on the eye region was also reduced 25.4%-44.1% by the visual observation and 93% of all volunteers responded that topical application of the O/W emulsion was showed some reducing effect of wrinkles after 10 weeks. From these results, we suggest that Medimin A is a potent anti-wrinkle agent by objective evaluation methods(in vitro collagen synthesis and image analysis of skin replica) and subjective evaluation methods(visual observation and self-estimated response of volunteers).

Antioxidant and Skin Whitening Effect of Graviola (Annona muricata) Leaf Extracts (Graviola (Annona muricata) 잎 추출물의 항산화 및 미백 효과)

  • In this study, graviola leaf extracts (GLE) was investigated for the effect of antioxidant, antibacterial, whitening, anti-wrinkle. The antioxidant effect of GLE was measured by an electron donating ability assay. As a result, GLE increased the electron donating ability in a concentration-dependent manner. The antibacterial effect of GLE was found to show the higher antibacterial effect in Methicillin-resistant Staphylococcus aureus CCARM3115 compared with that of ampicillin by a paper disc method. The whitening effect of GLE was also measured by tyrosinase inhibition assay, and it was found that the tyrosinase activity of GLE decreased as the concentration increased. The inhibition activity of tyrosinase involved in hydroxylation reaction which is related to converting L-tyrosine to (DOPA) was higher than that of arbutin's at the concentration ranging from 125 to $250{\mu}g/mL$. In addition, GLE reduced melanin contents of B16-F10 melanoma cells in a dose-dependant manner and decreased to about 76.7% at a concentration of $100{\mu}g/mL$ Regarding wrinkling formation of GLE, an elastase inhibition assay was performed. As a result, GLE and ursolic acid were 10.5% and 56.5%, respectively under the identical concentration. These results suggest that GLE has significant antioxidant and whitening activities, and also may be potentially used as a therapeutic agent for hyperpigmentation treatment as an ingredient of whitening cosmetics.

Lamellar-bio nano-hybrid; The Study for Stability of Catechin (Green Tea: EGCG) Using 3-Dimensional Liposome (라멜라-바이오 나노하이브리드: 3 Dimension-liposome을 이용한 카테킨(EGCG)에 안정화에 대한 연구)

  • In these several years, as many people have been attracted by the functional cosmetics, there are a lot of study to enhance the stability of active ingredients for light, heat, oxygen, etc. in the academic and industrial field. Especially, catechin is well known as strong anti-oxidant, anti-inflammatory and reducing agent for oxidative stress but it is very unstable for light, heat, oxygen. etc. In this study, the stability and skin penetration of catechin are improved by 3-dimensional method. As I-dimension, porous silica is prepared using sol-gel method, and then catechin is adsorbed in pores of silica. As 2-dimension, solid lipid nanoparticles (SLN) are obtained using non-phospholipid vesicles. Finally 3-dimension is completion through lamellar phase self-organization that combines SLN catechin with skin lipid matrix. We used laser light scattering system, cyro-SEM, chromameter, HPLC and image analyzer to analyze our 3-dimentional systems. According to chromameter date, the color stability of 3-dimensional catechin is enhanced by 5-10 times compared with general liposome systems. We also confirmed through HPLC analysis that 3-dimensional catechin is more long lasting. The effect of skin penetration and wrinkle reduction are improved, too.
