  • In the hope of identifying anti-leukemic agents from traditional herbal medicines. this study was designed to investigate the anti-leukemic effects of the herbal medicine Jipaesan, which is composed of Angelica Dahurica and Fritillariae Verticillata. in acute promyeloid leukemia HL-60 cells. Jipaesan showed anti-proliferative effect through the induction of differentiation and apoptosis in HL -60 cells. Verticinone as a major differentiating agent and imperatorin as major apoptosis-inducing agent were isolated from the water extracts of F. Verticillata and A. Dahurica, respectively. Combined treatment of HL-60 cells with two major compounds showed synergy in the induction of differentiation. Since the induction of differentiation and/or apoptosis has therapeutic values in curing acute leukemic diseases. Jipaesan could be useful as an anti-leukemic agent.

  • This study was carried out to investigate the anti-oxidant effect of Wasabia japonica(WJ). The anti-oxidant activities of various extracts from WJ were investigated. The water and ethanol extracts of WJ leaf were found to cause significant free radical scavenging effects on DPPH and nitric oxide(NO). Sprague-Dawley male rats were randomly assigned to one normal diet and three high cholesterol diet groups which contained 1%(w/w) cholesterol. After 4weeks cholesterol diet, the groups of high cholesterol diet were classified to control(normal diet only), WJR5(normal diet and 5% WJ root) and WJL5(normal diet and 5% WJ leaf). Liver xanthine oxidase(XO) activity in the rats of high cholesterol diet was decreased up to the levels of normal diet group according to WJ diet. The results indicated that WJ possesses antioxidant effects through free radical scavenging effects on DPPH, NO and the decreasing of XO activity. In view of these results, WJ is expected to be an effective material for the anti-oxidant.

  • The crude methanolic extract of the leaves of Amoora cucullata Roxb. was investigated for its possible anti-inflammatory activity using carrageenin induced rat paw edema model and cotton pellet implantation method in rat. The extract was also studied for its antinociceptive activity using acetic acid induced writhing model in mice. At the doses of 200 and 400 mg/kg body weight, the extract showed significant anti-inflammatory activity in both models. At the same doses, the extract also significantly reduced the number of acetic acid-induced abdominal constriction (writhing) in mice. The crude extract also showed significant diuretic activity in albino mice.

  • Pharmacological investigations were carried out with aqueous methanolic extract (AME) of Swietenia mahagoni (L.) Jacq. (Meliaceae) leaves. Acute toxicity studies revealed that the $LD_{50}$ dose of AME was 600 mg/kg, i.p. AME was found to possess significant anti-inflammatory activity in acute, sub-chronic and chronic models of inflammation. AME selectively inhibited cyclooxygenase (COX)-2 activity, which is involved in arachidonic acid metabolism and biosynthesis of prostaglandins under inflammatory conditions. Treatment with AME significantly enhanced total peritoneal cell count and the number of macrophages in normal mice, which revealed that AME may also alter the immune response along with its anti-inflammatory effect. The saponins or the alkaloids present in AME may be responsible for the anti-inflammatory activity.

  • Corylopsis gotoana (Hamamelidaceae) has been used as a traditional medicine for the treatment various diseases including cold, edema and vomiting. However, previous studies regarding component analysis and pharmacological actions of C. gotoana are extremely limited until now. In this study, we investigated anti-inflammatory activities of the methanolic extract of the twigs of Corylopsis gotoana (MCG) both in vitro and in vivo. MCG effectively inhibited excessive NO production in IFN-${\gamma}$ and LPS-stimulated mouse peritoneal macrophages without notable cytotoxicity. In addition, we also found that MCG could attenuate the expression of inflammatory mediators such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). We further tested in vivo anti-inflammatory activity of MCG using paw edema mouse model. Herein, MCG demonstrated significant suppression on the paw edema induced by both of trypsin and carrageenan. These results indicated that MCG has potent anti-inflammatory potential and may be useful for prevention and treatments of inflammatory diseases.

  • Ohyaksungisan, combined preparation of crude drugs, has been used for hemiplegia, arthralgia and paralysis in traditional Korean medicine. The anti-inflammatory activity of the aqueous extract from Ohyaksungisan(OSSE) was investigated on acetic acid-induced edema and adjuvant arthritis in rats. Acute toxicity and analgesic action in mice were also examined. Its anti-inflammatory activity on 5% acetic acid-induced edema and adjuvant arthritis was observed with oral administration. The acute toxicity showed 10% mortality at 2400 mg/kg (p.o), but was not showed at 1200 mg/kg (i.p). OSSE was showed to have significant analgesic action (P<0.05) at 150 mg/kg and this action was strengthened at 300, 600 mg/kg. The anti-inflammatory effect was showed significant preventive effect on the hind paw edema from 90 min. and the adjuvant arthritis, when orally administered for 19 days. showed significant inhibitory effect on the hind paw edema from the 5th day.

  • Caragana sinica (Leguminosae) is a plant, which has been used as a traditional medicine for the treatment of lots of diseases including neuralgia, goat, hypertension and eczema. However, scientific studies of C. sinica in pharmacological aspects are not carried out. In this study, the anti-nociceptice effect of methanolic extract of C. sinica (MCS) was evaluated using various pain models. Our data represented that MCS significantly delayed the latency time under central pain condition which are arose from thermal stimuli, indicating MCS possess analgesic potential against central nociception. In addition, MCS showed strong and dose-dependent anti-nociceptive activities on acetic acid-induced peripheral pain, compared to positive control such as indomethacin. Further combination studies using naloxone, a non-selective opioid receptor antagonist, have revealed that analgesic activity of MCS was not changed in the presence of naloxone, indicating MCS exerts anti-nociceptive activity independent of opioid receptor. These results suggest that MCS may be an effective medicine in managing pain.

  • Simotang oral liquid (SMT) is a traditional Chinese medicine (TCM) consisting of four natural plants and is used to alleviate gastrointestinal side effects after chemotherapy and functional dyspepsia (FD). However, the mechanism by which SMT helps cure these gastrointestinal diseases is still unknown. Here, we discovered that SMT could alleviate gastrointestinal side effects after chemotherapy by altering gut microbiota. C57BL/6J mice were treated with cisplatin (DDP) and SMT, and biological samples were collected. Pathological changes in the small intestine were observed, and the intestinal injury score was assessed. The expression levels of the inflammatory factors IL-1β and IL-6 and the adhesive factors Occludin and ZO-1 in mouse blood or small intestine tissue were also detected. Moreover, the gut microbiota was analyzed by high-throughput sequencing of 16S rRNA amplicons. SMT was found to effectively reduce gastrointestinal mucositis after DDP injection, which lowered inflammation and tightened the intestinal epithelial cells. Gut microbiota analysis showed that the abundance of the anti-inflammatory microbiota was downregulated and that the inflammatory microbiota was upregulated in DDP-treated mice. SMT upregulated anti-inflammatory and anticancer microbiota abundance, while the inflammatory microbiota was downregulated. An antibiotic cocktail (ABX) was also used to delete mice gut microbiota to test the importance of gut microbiota, and we found that SMT could not alleviate gastrointestinal mucositis after DDP injection, showing that gut microbiota might be an important mediator of SMT treatment. Our study provides evidence that SMT might moderate gastrointestinal mucositis after chemotherapy by altering gut microbiota.

  • Objectives : This study aims to analyze the terms describing anti-aging effects in Dongeuibogam, and to propose the methodology for selecting medicinal herbs related to anti-aging effects. Methods : We considered aging or anti-aging theories by reviewing the major literature in korean medicine or traditional chinese medicine, and then we deduced the concepts describing anti-aging effects and the medicinal herbs for anti-aging effects. We investigated the medicinal herbs based on the terms describing anti-aging effects in Dongeuibogam and drew the methodology for selecting and classifying the medicinal herbs related to anti-aging. Results & Conclusions : The following conclusions could be drawn. 1. A set of causes and signs of aging in Dongeuibogam and a set of therapies for alleviating the aging signs in Chengumikbang, Yanglobongchinseo, and Nolohangeon was found. 2. Anti-aging effects should not be confused with disease treatments. Therefore, we defined the medical terms of anti-aging effects as "the terms expressing the effect of delaying or alleviating the subjective or objective signs of aging concretely". Based upon the terms of anti-aging effects, we defined the medicinal herbs for anti-aging effects as "the medicinal herbs whose effects are described in medical terms expressing the effects of delaying or alleviating the subjective or objective signs of aging concretely". 3. We considered Lycii Fructus, Asparagi Tuber in Dongeuibogam as examples of medicinal herbs whose effects are described in medical terms expressing anti-aging effects. It shows that we can select medicinal herbs for anti-aging effects by comparing the effects of 937 single therapies in Dongeuibogam to the terms describing anti-aging effects defined in this study. Futhermore, the results could be utilized as the basic materials of experimental study on anti-aging.
