Alteration of hepatic anti-oxidant systems by 4-nonylphenol, a metabolite of alkylphenol polyethoxylate detergents, in Far Eastern catfish Silurus asotus

  • Objectives This study aimed to estimate the effects of 4-nonylphenol (NP), a ubiquitously present surfactant in aquatic environments, on the anti-oxidant systems of the liver in the Far Eastern catfish Silurus asotus. Methods Changes in biochemical parameters involved in glutathione (GSH)-related and other anti-oxidant systems were analyzed following 4 weeks of 4-NP administration (0.1 and 1.0 mg/kg diet) via a formulated diet to catfish. Results 4-NP exposure induced an elevation in hepatic lipid peroxide levels and an accompanying decrease in reduced state GSH after 2 weeks, suggesting pro-oxidant effects of the chemical in catfish. This oxidative stress was associated with an inhibition of the GSH-utilizing enzyme glutathione peroxidase at the same time point. This inhibition was restored after 4 weeks. The activities of other anti-oxidant enzymes, i.e., glutathione reductase, superoxide dismutase and catalase were increased after 4 weeks. These enzyme increases occurred more strongly at the higher 4-NP concentration (1.0 mg/kg diet). Conclusions 4-NP given to catfish at 0.1 to 1.0 mg/kg diet, concentrations relevant to environmental levels, depletes the endogenous anti-oxidant molecule GSH and temporarily inhibits GSH-related anti-oxidant enzymes. Such declines in anti-oxidant capacity and elevated oxidative stress seem to be compensated eventually by subsequent activation of various anti-oxidant enzyme systems.

Effects of Oxidative Stress on Growth Performance, Nutrient Digestibilities and Activities of Antioxidative Enzymes of Weanling Pigs

  • This study was undertaken to investigate the effects of oxidative stress on growth performance, nutrient digestibilities and activities of antioxidant enzymes of weanling pigs. In the experiment, 24 male $Landrance{\times}Yorkshire $weanling pigs were allotted to three groups of 8 animals each. Pigs were fed individually. According to a single factorial arrangement, pigs received diets with 5% of either fresh (group 1 and group 3) or oxidized fish oil (peroxide value was 786.50 meq $O_2/kg$ before inclusion in the diet, group 2). At the beginning of the experiment, pigs in group 3 received an intraperitoneal injection of diquat at 12 mg/kg of body weight. The trial lasted for 26 d. A metabolism test was carried out during the last 4 days of the second week. The results showed that feeding diets containing oxidized fish oil or injection with diquat depressed the growth performance and nutrient digestibilities of weanling pigs, decreased activities of antioxidant enzymes and increased concentration of malondialdehyde in plasma and liver. Intraperitoneal injection of diquat would induce more serious oxidative stress than oral intake of oxidized fish oil in the diet. In conclusion, administration of oxidized fish oil or diquat could induce oxidative stress in weanling pigs, and oxidative stress could depress growth performance and impact anti-oxidative ability of young pigs.

김치 용매획분이 고 콜레스테롤 식이를 섭취한 토끼의 심장, 신장 및 폐의 항산화 효소계 활성에 미치는 영향 (Effects of Kimchi Solvent Fractions on Anti-oxidative Enzyme Activities of Heart, Kidney and Lung of Rabbit Fed a High Cholesterol Diet)

  • 김치의 용매획분중 in vitro에서 LDL 산화억제 활성이 강했고 in vivo에서 지질저하효과가 있었던 디클로로메탄 획분, 에틸아세테이트 획분, 수용성 획분이 고콜레스테롤 식이를 섭취한 토끼의 심장, 신장, 그리고 폐에서 항산화 효소의 활성에 미치는 영향을 살펴보았다. 과산화물 함량은 심장이 가장 높고 신장이 가장 낮았으나, catalase와 GSBf-px활성은 심장이 낮았고, 신장은 높은 활성을 나타냈다 Catalase, GSH-px 및 SOD의 활성은 모든 장기에서 1% 콜레스테롤 식이를 섭취한 대조군에 비해 김치 용매획분군에서 낮았고 특히 디클로로메탄 획분군의 항산화효소 활성은 유의적으로 낮았다(p<0.05).이러한 결과는 김치 용매획분은 심장 신장 그리고 폐에 지방축적을 억제하는 효과가 있었고, 그로 인해 지질 유리기의 생성이 적어, 각 장기에서 유리기를 제거하는 항산화 효소의 활성이 낮은 것으로 생각된다.

Green perilla leaf extract ameliorates long-term oxidative stress induced by a high-fat diet in aging mice

  • BACKGROUND/OBJECTIVES: Oxidative stress is caused by an imbalance between harmful free radicals and antioxidants. Long-term oxidative stress can lead to an "exhausted" status of antioxidant defense system triggering development of metabolic syndrome and chronic inflammation. Green perilla (Perilla frutescens) is commonly used in Asian cuisines and traditional medicine in southeast Asia. Green perilla possesses numerous beneficial effects including anti-inflammatory and antioxidant functions. To investigate the potentials of green perilla leaf extract (PE) on oxidative stress, we induced oxidative stress by high-fat diet (HFD) in aging mice. MATERIALS/METHODS: C57BL/6J male mice were fed HFD continuously for 53 weeks. Then, mice were divided into three groups for 12 weeks: a normal diet fed reference group (NDcon), high-fat diet fed group (HDcon), and high-fat diet PE treated group (HDPE, 400 mg/kg of body weight). Biochemical analyses of serum and liver tissues were performed to assess metabolic and inflammatory damage and oxidative status. Hepatic gene expression of oxidative stress and inflammation related enzymes were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). RESULTS: PE improved hepatopathology. PE also improved the lipid profiles and antioxidant enzymes, including hepatic glutathione peroxidase (GPx) and superoxide dismutase (SOD) and catalase (CAT) in serum and liver. Hepatic gene expressions of antioxidant and anti-inflammatory related enzymes, such as SOD-1, CAT, interleukin 4 (IL-4) and nuclear factor erythroid 2-related factor (Nrf2) were significantly enhanced by PE. PE also reduced the levels of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in the serum and liver; moreover, PE suppressed hepatic gene expression involved in pro-inflammatory response; Cyclooxygenase-2 (COX-2), nitric oxide synthase (NOS), interleukin 1 beta (IL-1β), and interleukin 6 (IL-6). CONCLUSIONS: This research opens opportunities for further investigations of PE as a functional food and possible anti-aging agent due to its attenuative effects against oxidative stress, resulting from HFD and aging in the future.

  • Antioxidants are compounds that protect cells against the damaging effects of reactive oxygen species (ROS). Some ROS, such as superoxide and hydrogen peroxide, are normally produced in cells as by-products of biochemical reactions or as signaling molecules. When ROS-generating reactions are activated excessively, pathological quantities of ROS are released to create an imbalance between antioxidants and ROS, called as oxidative stress. Oxidative stress, which may result in cellular damage, has been linked to cardiovascular disease, diabetes, cancer, and other degenerative conditions. In humans the first line of antioxidant defence are the antioxidant enzymes, especially SOD, glutathione peroxidase (GPX), and to a lesser extent catalase, as well as the tripeptide glutathione(GSH). These enzymes will help destroy ROS(reactive oxygen species) such as hydroxyl radical, $H_2O_2$ and lipid peroxides, while GSH protects against oxidized protein. Many herbal medicines possess antioxidant properties. Herbal antioxidants may protect against these diseases by contributing to the total antioxidant defense system of the human body. Here, many herbal medicines including Ginseng, Licorice, Ligusticum Chuanxiong, Ginkgo biloba and many others was reviewed in terms of anti-oxidant efficiency related to their components.

백서의 음릉천에 시술한 저주파 침자극의 항산화 효과에 대한 실험적 연구 (Anti-oxidative effect of electroacupuncture to Yinlingquan (SP9) in AAPH induced oxidative stress of rats)

  • Objectives & Methods : The purpose of this study is to observe the anti-oxidative effects of electroacupuncture to Yinlingquan(SP9) in AAPH induced oxidative stress of rats. The author performed several experimental items including measurements of body weight, levels of albumin, total bilirubin, LDL-cholesterol, GOT and GPT in serum, levels of SOD, glutathione, catalase, NO and MDA in liver, histological analysis of liver. The conclusions are as follows. Results : 1. In the SP9-EA group, the level of LDL-cholesterol was significantly decreased in comparison with that of the holder group and control group. 2. In the SP9-EA group, SOD activity, glutathione concentration in liver were increased, and NO concentration was decreased significantly in comparison with those of the control group and the holder group. 3. In the SP9-EA group, the density of liver tissue was maintained more similarly to the normal group in comparison with those of the control group and the holder group. 5. The results of the SP9-NR group showed similar tendencies with those of the SP9-EA group, but the effects were lower than those of the SP9-EA group. Conclusion : These results suggest that electroacupuncture at SP9 has an anti oxidative effect through suppressing both the reduction of anti-oxidative enzymes and production of oxidized substances.

4'-O-β-D-Glucosyl-5-O-Methylvisamminol Attenuates Pro-Inflammatory Responses and Protects against Oxidative Damages

  • We attempted to examine anti-inflammatory and anti-oxidant effects of 4'-O-${\beta}$-D-glucosyl-5-O-methylvisamminol (GOMV), the first epigenetic inhibitor of histone phosphorylation at Ser10. While GOMV did not affect the viability of murine macrophage RAW 264.7 cells, it significantly suppressed lipopolysaccharide (LPS)-induced generation of prostaglandin $E_2$ ($PGE_2$) and nitric oxide (NO) through transcriptional inhibition of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). GOMV also scavenged free radicals in vitro, increased NF-E2-related factor 2 (NRF2), and activated antioxidant response element (ARE), thereby resulting in the induction of phase II cytoprotective enzymes in human keratinocyte HaCaT cells. Finally, GOMV significantly protected HaCaT cells against 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced oxidative intracellular damages. Together, our results illustrate that GOMV possesses anti-inflammatory and anti-oxidant activity.

Anti-oxidative and Anti-inflammatory Effects of Protulaca Oleracea on the LPS-stimulated AGS Cells

  • Protulaca oleracea, a widely distributed weed, has been reported to exhibit different health promoting effects. The objective of this study was to evaluate the anti-oxidative and anti-inflammatory effects of P. oleracea on LPS-stimulated AGS cells. The cytotoxicity of P. oleracea in AGS cells was examined by MTT assay. The anti-oxidative effects of P. oleracea were examined by DPPH assay. RT-PCR was carried out to examine the effect of P. oleracea in the mRNA expression of different inflammatory mediators. MTT assay revealed that P. oleracea have almost no cytotoxity in AGS cells. DPPH radical scavenging activities were better than butylated hydroxyl toluene (BHT). The mRNA expression of different endogenous anti-oxidative enzymes (SOD2, GPx3 and catalase) were preserved by P. oleracea in AGS cells. The nitric oxide production and expression of iNOS in LPS stimulated RAW264.7 were suppressed in P. oleracea treated groups. Based on these findings, P. oleracea has protective anti-oxidant and anti-inflammatory effects.

Anti-apoptotic Effects of Red Ginseng on Oxidative Stress Induced by Hydrogen Peroxide in SK-N-SH Cells

  • Ginseng (Panax ginseng C.A. Meyer) has been shown to have anti-stress effects in animal studies. However, most studies have only managed to detect altered levels of biomarkers or enzymes in blood or tissue, and the actual molecular mechanisms by which ginseng exerts these effects remain unknown. In this study, the anti-oxidative effect of Korean red ginseng (KRG) was examined in human SK-N-SH neuroblastoma cells. Incubation of SK-N-SH cells with the oxidative stressor hydrogen peroxide resulted in significant induction of cell death. In contrast, pre-treatment of cells with KRG decreased cell death significantly. To elucidate underlying mechanisms by which KRG inhibited cell death, the expression of apoptosis-related proteins was examined by Western blot analysis. KRG pre-treatment decreased the expression of the pro-apoptotic gene caspase-3, whereas it increased expression of the anti-apoptotic gene Bcl-2. Consistent with this, immunoblot analysis showed that pre-treatment of the SK-N-SH cells with KRG inhibited expression of the pro-inflammatory gene cyclooxygenase 2 (COX-2). RT-PCR analysis revealed that the repression of COX-2 expression by KRG pre-treatment occurred at the mRNA level. Taken together, our data indicate that KRG can protect against oxidative stress-induced neuronal cell death by repressing genes that mediate apoptosis and inflammation.

The hepatoprotective effects of silkworm: Insights into molecular mechanisms and implications

  • The liver, a multifunctional organ, plays a vital role in maintaining overall health and well-being by regulating metabolism, detoxification, nutrient storage, hormone balance, and immune function. Liver diseases, such as hepatitis, cirrhosis, fatty liver disease, and liver cancer, have significant clinical implications and remain a global health concern. This article reviews the therapeutic potential of silkworm larvae (Bombyx mori) and explores their underlying molecular mechanisms in protecting against liver diseases. Silkworm larvae are rich in proteins, vitamins, minerals, and n-3 fatty acids, making them a promising candidate for therapeutic applications. The anti-inflammatory mechanisms of silkworm larvae involve modulating the production of cytokine such as TNF-α and interleukins, inflammatory enzymes including cyclooxygenase-2 and macrophage polarization, thereby attenuating liver inflammation. Silkworm larvae also exhibit anti-oxidative effects by scavenging free radicals, reducing intracellular reactive oxygen species and enhancing the liver's antioxidant defense system. Moreover, silkworms have been reported to decrease the serum alcohol concentration and lipid accumulation. Understanding the therapeutic properties of silkworm larvae contributes to the development of innovative strategies for liver injury prevention and treatment. Further research is warranted to elucidate the precise signaling pathways involved in the anti-inflammatory and anti-oxidative effects of silkworm larvae, paving the way for potential therapeutic interventions in liver diseases.
