Comparative Study on Anti-inflammatory Effects and Compound Contents of Multi-herbal Extracts HT008 produced by Different Manufacturing Methods (한약복합추출물 HT008의 제조방법에 따른 항염증 효능 및 성분함량 비교연구)

  • Objectives : The biological activities and compound contents of herbal medicine vary depending on manufacturing processes. In this study, we compared anti-inflammatory effects and compound contents of three kinds of multi-herbal extract HT008 produced by different manufacturing processes in order to determine chemical and biological equivalence. Methods : HT008 was produced by three different manufacturing methods: 1. Freeze dried extract of Eleutherococcus senticosus, Scutellaria baicalensis and Angelica sinensis (HT008 FD), 2. Spray dried extract of E. senticosus and S. baicalensis combined with reflux extract of A. sinensis (HT008 SD), 3. Spray dried extract of E. senticosus and S. baicalensis combined with supercritial fluid extract of A. sinensis (HT008 SF). Anti-inflammatory effects were evaluated using acetic acid induced pain model and ${\lambda}$-carageenan induced paw edema model. Compound contents were evaluated by HPLC quantitative analysis of standard compounds of HT008, eleutheroside E, baicalin, z-ligustilide. Results : HT008 FD, HT008 SD and HT008 SF significantly decreased acetic acid induced pain index and ${\lambda}$-carrageenan induced paw edema volume compared with that of control group. There was no significant difference in efficacy among the HT008 FD, HT008 SD and HT008 SF. Standard compound contents of HT008 FD, HT008 SD and HT008 SF were quantified within the range of Korean pharmacopoeia or other research. Conclusions : Three different manufacturing methods of multi-herbal extracts have been developed without noticeable difference in the efficacy or compound contents. The results might be used to establish manufacturing process and industrialization of herbal extracts.

Anti-oxidation, anti-inflammation, anti-wrinkle, and pore-tightening effects of phenolic compounds from Aeonium sedifolium leaves (소인제(Aeonium sedifolium) 잎 유래 phenolic 성분의 항산화, 피부주름생성 억제, 항염증 및 모공 수축 효과)

  • The succulent plant Aeonium sedifolium leaves contain several compounds that are of interest for their cosmetic uses on the skin. This study measured the inhibitory effects of enzyme production and antioxidant, astringent effects and skin wrinkles using Aeonium sedifolium leaves (ASL). The total phenolics compounds (TPC) content of ASL under optimal extraction conditions was 34.49 mg/g for hot water extract (ASLW) and 61.64 mg/g for 50% ethanol extract (ASLE). The ASLW and ASLE extracts were freeze-dried, powdered, and used as solids. TPC content, 2,2-diphenyl-1-picrylhydrazy (DPPH) radical scavenging activity, and 2,2'-azinobis (3-ethylben-zothiazoline 6-sulfonate) (ABTS) radical inhibition of the ASL phenolics were tested. The DPPH radical scavenging activities of ASLW and ASLE were tested at a TPC of 100 ㎍/mL. ABTS radical inhibition showed antioxidant activity of 100.00% in ASLW and ASLE, and the antioxidant protection factor of ASLW and ASLE was 1.07 and 1.22, respectively. The thiobarbituric acid-reactive substance (TBARS) inhibitory activity of ASLW and ASLE was 77.00%. The elastase inhibitory activity of ASLE was 69.03%, and collagenase inhibition activity for ASLW and ASLE was 29.82% and 54.76%, respectively. The astringent effect of ASLE was 89.82% at a TPC of 200 ㎍/mL. Thus, we concluded that ASL has the potential as a functional cosmetic ingredient with anti-aging effects on the skin.

A Modified Cryopreservation Method of Psychrophilic Chlorophyta Pyramimonas sp. from Antarctica

  • Polar psychrophiles which thrive under extreme conditions such as cold temperature, high salinity, and high dose ultraviolet light, emerge as novel targets for biotechnology. To prevent genetic drift and the possibility of contamination by subculturing, cryopreservation was employed for two psychrophilic microalgae, Porosira sp. (KOPRI AnM0008) and Pyramimonas sp. (KOPRI AnM0046), which have anti-freeze activities. Five cryoprotectants (dimethyl sulphoxide, ethylene glycol, glycerol, methanol and propylene glycol) showed toxicity at 20-30% (v/v). The optimal cryoprotectant concentration and equilibration time were less than 20% and 10 min, respectively. Cryopreservation was carried out in the presence of cryoprotectants either by direct freezing in liquid nitrogen ($LN_2$) or controlled freezing using a controlled rate freezer followed by storage in the $LN_2$ tank. As a result, Pyramimonas sp. (KOPRI AnM0046), a psychrophilic chlorophyta was revived. Cryopreserved Porosira sp. was not revived from either freezing protocols probably due to the silicic cell wall and its relatively large cell size. In the case of Pyramimonas sp. (KOPRI AnM0046), the controlled freezing method showed higher revival yield than the direct freezing method.

  • This paper was analized the thermal conductivity of polystylene (TENSAR- GEOGRID) embeding into the subbase through frost penetration depth, frost heave, change of bearing capacity, and soil moisture movement due to freezing, thawing and icing actions, and their results were as follows : 1.The change of temperature into the sub-base was much increased by the Tensar-Geogrid insertion, and the frost penetration and frost heave were decreased as the thinner of the insulation thickness but the thawing velocity of melting period was appeared to be faster in case of non-insulated. 2.The frost heave had a close relationship with the thickness of insulations which was reasonably included anti-frost effects. 3.The moisture content during the freezing period of upper layer of the insulation insertion was increased by 15 per cent but it was returned to initial state of the thawing period, and at the down layer temporarily increased by 10 per cent and returned to the original state at once. 4.The insulation was acted as a function of distribution of surcharge, and the settlement of the sub-base was about 1.5 mm under 15 tonnage of load and which was included within the allowable limits. 5.The sliding resistance due to the icing which was induced by the insulation insertion into the sub-base was appeared as more 40 per cent than noninsulation area, so that the insulations should be restricted on the place such as mountains, curved and cross area which were required the braking power under the traffics.

  • The automotive exhaust gases generated by the vehicles containing carbon monoxide, hydrocarbons and lead, is a large concern because of their harm to human health or the living environment. To reduce exhaust gas, it is important to develop a variety of techniques that are currently being used by elemental analysis to determine the optimal conditions. In this study, the anti-freeze coolant contained in the exhaust gas was examined, which can affect the emissions. The effects of the commercially available coolant from five domestic companies on the HC, NOx and $CO_2$ emissions were analyzed to determine the optimal amount of antifreeze. In addition, antifreeze products from the five companies were analyzed with respect to driving time of the cooling fan and the correlation of the NOx emission analysis. The temperature of the engine oil was matched using a manual gear of small passenger inspection standard speed $40{\pm}2Km/h$ so the vehicle could meet the specifications for inspection $90{\sim}93^{\circ}C$. The Company D fan operation time resulted in the shortest antifreeze, $CO_2$ and NOx emissions.

  • Sunjeonhwadok-Tang was a drug that treated carbuncle and cellulitis. So, the purpose of this study was to investigate effects of Sunjeonhwadok-Tang on the proliferation of cancer calls and immunocytes focusing around combined effects of anticarcinogen. We used Sunjeonhwadok-Tang extract(SHT) with freeze-dried, 8wks-old male balb/c mice and cancer cell lines(L1210, Sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results : 1. SHT was significantly showed cytotoxicity on the L1210 cell lines. 2. SHT was significantly increased proliferation of thymocytes and splenocytes in vitro. 3. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly inhibited proliferation of L1210 cell lines, but was not inhibited proliferation of Sarcoma-180 cell lines compared with positive control group. 4. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly increased proliferation of thymocytes and splenocytes compared with positive control group. 5. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of thymocytes and splenocytes in normal mice. 6. In combined effects of SHT and vincristine, SHT was significantly inhibited proliferation of L1210 cells in L1210 cells transplanted mice 7. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of L1210 cell in L1210 cells transplanted mice. The present author thought that SHT had action of anti-cancer and immuno-activity, and in combined effects of vincristine, SHT had recoverable effects on damage by anticarcinogen.

Biological activities of ethanol extract from the seawater algae, Chlorella elliposidea C020 (해수클로렐라 [Chlorella elliposidea C020] 에탄올 추출물에 대한 생리 활성)

  • We investigated the biological activities of ethanol extract from the seawater algae, Chlorella elliposidea C020 such as antibacterial activity, anti-oxidant activity, tyrosinase inhibitory activity and hemolytic activity against human erythrocytes. Extract was obtained from various solvent, methanol, ethanol, acetone and ethanol + acetone (1:1, v/v%), 95% ethanol proved to be best extraction solvents. The contents of ethanol extract were higher in freeze-dried sample than that in frozen-thawing. Antibacterial activities of ethanol extract showed strong inhibitory effect against Bacillus subtilis PM125, Bacillus licheniformis and fish pathogenic bacteria, Vibrio parahaemolyticus KCTC2471 and Edward tarda NUF251. However, this extract didn't worked against antifungal activity against Candida albicans KCTC1940. And, ethanol extract was without hemolytic activity against human erythorocytes. The ethanol extract showed 75% of free radical scavanging effect on 2.0 mg/mL using DPPH method. In tyrosinase inhibition assay of ethanol extract, $IC_{50}$ (Inhibition Concentration) was measured as 10.87 mg/mL. Conclusionally, ethanol extract of Chlorella elliposidea C020 has good candidate for bioactive materials.

Numerical Analysis of Unsteady Heat Transfer for Location Selection of CPVC Piping (CPVC 배관 동파방지용 열선의 위치 선정을 위한 비정상 열전달 수치해석)

  • In this paper, a numerical experiment was conducted to find out the optimal location of electrical heat trace for anti-freeze of water inside the CPVC pipe for fire protection. The unsteady incompressible Navier-Stokes equations coupled with energy equation were solved. Since the conduction equation of pipe was coupled with the natural convection of water, the analysis of conjugate heat transfer was conducted. A commercial code (ANSYS-FLUENT) based on SIMPLE-type algorithm was used for investigating the unsteady flows and temperature distributions in water region. From the present numerical experiment, it has been found that the vector field of water inside the PVC pipe is opposite to the case of steel because of the huge difference of material properties of the two pipes. Furthermore, it was found that the lowest part of the pipe was an optimal position for electrical heat trace since the minimum water temperature of the case was higher than those of the other cases.

  • Frost damage of a storage tank for chemical admixtures caused by a low temperature in winter and quality deterioration of chemical admixtures have been often reported. However, there have been few regulations and researches related to the frost damage of chemical admixtures and facilities. Therefore, this paper is intend to investigate not only, the freezing points of chemical admixtures such as AE admixtures and water-reducing AE admixtures, etc., but also the quality variation of concrete used with chemical admixtures before freezing and after freeze-thaw cycles. According to the results of experiments, most chemical admixtures except anti-freezing agent and accelerating water-reducing AE admixtures should be kept from being frozen in low winter temperatures. However, full agitation of frozen chemical admixtures dose not cause the problems of concrete related to the quality of chemical admixtures.

Development Ultra Rapid Hardening Construction Materials on Cold Weather Environment Considering Curing Temperature (양생온도를 고려한 극한지용 초속경 건설재료 개발)

  • Because ordinary concrete cannot be hardened well under sub-zero temperatures, anti-freeze agents are typically added to prevent the frost damage and to ensure the proper hardening of concrete. With the advantage of a rapid exothermic reaction property, jet set concrete may be used as a cold weather concrete because it can reach the required strength before being damaged by cold weather. Recent studies are reported that magnesia-phosphate composites can be hardened very quickly and hydrated even in low temperature, which can be used as an alternative of severe cold weather concrete in arctic regions. This study developed the magnesia-phosphate composites that can be used in severe cold regions and suggested an appropriate mixture design from the experimental results.
