LPS로 유발된 우울증에 대한 감초(甘草)의 효과 (Effect of Glycyrrhizae Radix Water Extract on the depression induced by LPS in Rats)

  • Objectives : Investigation of the antidepressant effect of Glycyrrhizae Radix (GR) through the anti-inflammatory effect. Methods : Depression in rats was induced by LPS (i,p.3days). The rats were treated with GR100 mg/kg (GR 100) or GR400 mg/kg (GR 400). The depressive immobility was examined with Tail Suspension Test(TST) and Forced Swimming Test(FST). The expression of nuclear factor-${\kappa}B$(NF-${\kappa}B)$, $I{\kappa}B$ was measured with western blotting. The concentration of corticosterone, cytokine in plasma was measured with ELISA. The expression of c-Fos in the paraventricular nucleus(PVN) and tyrosine hydroxylase(TH) in the locus coeluleus(LC) were measured with immunostaining method. Results : In the TST, GR400 group significantly decreased immobility time compared with the LPS group. In the FST, GR100, GR400 group significantly decreased immobility time comparing with the LPS group. c-Fos expression in GR100 and GR400 group was decreased comparing with the lipoplysaccharide(LPS) group. The $I{\kappa}B$ expression of GR100 and GR400 group was increased comparing with the LPS group. The level of corticosterone of GR100 group was decreased comparing with the LPS group. The concentration of cytokine of GR100 and GR400 group was decreased comparing with the LPS group. TH expression in the LC was increased in LPS group, but in GR100 and GR400 group was not shown significant decrease. Conclusion : According to this results obtained, GR has antidepressant effects by the anti inflammatory action through the suppression of HPA axis activity, not through the action against the catecholaminergic system.

자유로운 신체운동과 멜라토닌이 우울장애 동물모델에 미치는 효과 (Effects of Physical Activity and Melatonin in a Rat Model of Depression Induced by Chronic Stress)

  • Purpose: Stress, depending on its intensity and duration, results in either adaptive or maladaptive physiological and psychological changes in humans. Also, it was found that stressful experiences increase the signs of behavioral despair in rodents. On the other hand, exercise and melatonin treatment is believed to have many beneficial effects on health. Thus, this study was designed to evaluate the anti-depressant effects of physical activity and melatonin against chronic stress-induced depression in rats. Methods: Adult male Sprague-Dawley(SD) rats(200-250g, 7 weeks of age) were subjected to depression induced by chronic stress. Chronic depression was induced with forced-swim stress (FSS) and repeated change of light-dark cycle for 4 weeks. In the last 2 weeks, some rats were confined in a cage enriched with a running wheel, seesaw and chewed a ball from 19:00 to 07:00 every day. Melatonin was injected intra-peritoneally (I.P), and the rats received intraperitoneal injections of melatonin (15 mg/kg). The Forced Swim Test (FST) was performed to evaluate the immobility behaviors of rats for a 5 min test. Results: It was found that, the immobility time in FST was significantly (p<.05) lower in physical exercise ($M=58.83{\pm}22.73$) and melatonin ($M=67.33{\pm}37.73$) than in depressive rats ($M=145.93{\pm}63.16$) without physical activity. Also, TPH positive cell in dorsal raphe was significantly (p<.05) higher in exercise ($M=457.38{\pm}103.21$) and melatonin ($M=425.38{\pm}111.56$) than in depressive rats ($M=258.25{\pm}89.13$). Conclusion: This study suggests that physical activity and melatonin produces antidepressant-like effect on stress-induced depression in rats. So, physical exercise and melatonin may be a good intervention in depression patients.

한약처방 JSB가 구금스트레스로 우울증을 유발한 생쥐의 행동과 생체지표에 미치는 영향 (Effects of JSB, a Korean Medicine, on Behaviors and Biological Factors in a Mouse Model of Depression Induced by Restraint Stress)

  • Objectives: Depression is a mood disorder showing low energy, loss of interest, anhedonia, and anxiety. It affects a patients' daily life. This disease is considered a social problem because the patient may commit suicide in extreme cases. JSB is composed of 12 Korean medicinal herbs. It has been prescribed to patients with depression. The objective of this study is to investigate anti-depressive effects of JSB on restraint stress-induced depression in a mouse model. Methods: Depression was induced by restraint stress. Mice were orally administered JSB at 10, 20, or 40 mg/kg for 14 days. Forced swimming test (FST) and open field test (OFT) were performed. Brain-derived neurotrophic factor (BDNF) mRNA level was measured by real time-PCR. Plasma levels of corticosterone and serotonin were measured by ELISA. Blood levels of AST and ALS were measured using a biochemical analyzer. Results: JSB treatment significantly reduced the immobility time in FST. BDNF mRNA level was increased by JSB treatment in the hippocampi of mice. Although the expression of TNF-α was also increased by JSB, such increase was not statistically significant. The increase of corticosterone level in plasma induced by restraint stress was significantly down-regulated by JSB. JSB reduced blood level of AST, but not ALT. Conclusions: JSB has a potential to manage depression, setting a foundation for clinical application of Korean medicine with safety.

우울증(憂鬱症) 모델 흰쥐에 대한 귀비온담탕(歸脾溫膽湯)의 실험적(實驗的) 연구(硏究) (The Effect of Guibiondamtang(歸脾溫膽湯) in an Animal Model of Depression using Chronic Mild Stress)

  • This study was performed to investigate the anti-depression effect of Guibiondamtang in rat model of depression. The rats in the experiment were stratified into 3 groups, ie, Guibiondamtang, saline, normal (non-stressed) groups. Chronic exposure to mild unpredictable stress such as white noise, flashing lights and restriction of food and water, causes the behavioral symptoms correspondent to depression. Consumption of 1% sucrose solution fell in rats exposed to CMS for 4 weeks. In the open field test , the exploratory activity ie. locomotion and centering decreased after CMS. We then evaluated the sucrose consumption and activity during 4 weeks of treatment with experimental drugs. The results were as follows: 1) There was no relation between sucrose intake and weight. 2) The Guibiondamtang(歸脾溫膽湯) group reinstated sucrose consumption within 5-6 weeks while having no influence on sucrose intake in normal group. 3) The Guibiondamtang(歸脾溫膽湯) group restored some exploratory activity in the open field test. 4) The Guibiondamtang-group had a-reduced potentiated startle response.

우울증(憂鬱症)모델 흰쥐에 대한 조위승청탕(調胃升淸湯)과 fluoxetine의 항우울효과(抗憂鬱效果) 비교 (The effects of Jowiseungchungtang versus fluoxetine in the chronic mild stress model of depression in rats)

  • This study was performed to evaluate the effects of Jowiseungchungtang and fluoxetine in the chronic mild stress(CMS) model of depression in rats. Chronic exposure to mild unpredictable stress was found to depress the consumption of sucrose solution in rats for 5 weeks. These CMS-treated rats were stratified into Jowiseungchungtang group, fluoxetine group and vehicle group. And control rats were also stratified into other CMS-treated group. The change of the consumption of sucrose solution was measured, and open field test were performed to investigate the anti-depression effect of Jowiseungchungtang and fluoxetine. The results were as follows : 1. The consumption of sucrose solution was significantly reversed in Jowiseungchungtang-treated group at 7 week, but there was no significant change in other groups. 2. CMS schedule decreased body weight. Jowiseungchungtang group and fluoxetine group showed significant decrease of body weight after 6 weeks. 3. In open field test, Jowiseungchungtang group and fluoxetine group showed no significant change of exploratory activity.

Saccharomyces Cerevisiae 효모 추출물 SCP-20의 스트레스 반응, 불안 및 우울에 대한 효과 : 이중 맹검 위약 통제 연구 (The Effects of Saccharomyces Cerevisiae Yeast Extract SCP-20 on Stress Response, Anxiety and Depression : A Double-Blind Placebo-Controlled Trial)

  • Saccharomyces cerevisiae 효모추출물인 SCP-20은 동물실험에서 항스트레스, 항불안 및 항우울 효과를 보인 바 있다. 본 연구는 정상인을 대상으로 위약통제 이중맹검 연구를 시행하여 SCP-20의 효과를 검증하였다. 선별과정을 거치고 충분한 순응도를 보인 61명의 자원자의 정보가 최종 분석에 사용되었다. 이들은 4주간 SCP-20이 70% 함유된 캡슐, SCP-20 99.5% 함유 캡슐 그리고 위약캡슐 중 한가지를 하루 4회 복용하였다. 4주 뒤 SCP-20 복용군은 위약군에 비해 SRI, BAI 점수에서 유의한 개선을 보였으나,BDI 점수변화는 군간 유의미한 차이가 없었고 집단별 사후검정에서 SCP99.5군만이 복용 전후 유의미한 BDI 점수감소를 보였다. HRV는 유의미한 변화를 보이지 않았다. 부작용은 관찰되지 않았다. SCP-20은 정상인에게 투여하였을 때 위약에 비해 유의하게 항스트레스, 항불안 효과를 보이나 항우울 효과는 명확하지 않았으며, 부작용은 보이지 않았다. 향후 SCP-20의 항우울기능 추가 검증 및 환자군을 대상으로 한 대규모, 장기적 연구가 필요하다.

분심기음의 우울증 모델 흰쥐에 대한 실험적 연구 (The Effects of Punsimgi-eum(fenxinqi-yin) in the Chronic Mild Stress Model of Depression in Rats)

  • Objectives : The aim of this study was to evaluate the effects of Punsimgi-eum(fenxingi-yin) in rat model of depression induced by chronic mild stress (CMS). Methods : The change of the consumption of sucrose solution was measured, and the open field test and passive avoidance conditioning test were performed to investigate the anti-depressive effect of Punsimgi-eum(fenxingi-yin). Results : The results were as follows; I. The consumption of sucrose solution was significantly reversed in the test group at 5, 7 and 8 weeks, but there was no significant change in the control group. 2. There was no significant change of body weight after 8 weeks in each group 3. In the open field test, there was significant increase of exploratory activity in the test group. 4. In the passive avoidance test, the test group showed differences in the maintenance of retention from the control group at the 2-week training-testing interval. Conclusions : The present results suggest that Punsimgi-eum(fenxingi-yin) has an anti-depressive effect.

노인의 종교활동과 신앙태도가 우울 및 자아통합감에 미치는 영향 (Effects of Religious Activities and Faith Attitudes on Depression and Ego-Integrity of the Elderly)

  • 본 연구는 노인의 인구사회학적 특성에 따른 종교활동, 신앙태도, 우울 및 자아통합감의 차이를 분석하고, 종교활동과 신앙태도가 우울 및 자아통합감에 미치는 영향을 분석하였다. 설문조사는 서울 수도권 지역에서 기독교 신앙을 가진 60세 이상의 노인을 대상으로 2013년 5월 4일에서 25일까지 실시하였으며, 총 408부의 자료를 분석하였다. 연구결과를 요약하면, 첫째, 노인의 인구사회학적 특성에 따라 종교활동, 신앙태도, 우울 및 자아통합감에서 부분적인 차이가 분석되었다. 둘째, 주관적 종교활동과 신앙태도는 우울 요인에 부(-)적인 영향을 미쳤고, 노인의 주관적, 조직적 및 비조직적 종교활동은 비활력 요인에 부(-)적인 영향을 미치고 있었다. 셋째, 주관적 종교활동은 자아통합감의 삶에 대한 태도와 죽음의 수용에, 조직적 종교 활동은 노령의 수용에 정(+)적인 영향을 미쳤고, 종교활동의 모든 요인은 현재 생활 만족에 정(+)적인 영향력을 나타냈다.

Mild stressed rat을 이용한 수종한약의 항우울효과에 대한 실험적 연구 (Anti-depressant Effect of Herbal Extracts in the Chronic Mild Stressed Rat)

  • This study was performed to investigate the anti-depression effect of herbal extracts (Rehmanniae Radix Preparat, Corni Fructus, Lycii Fructus, Pinelliae Rhizoma, Nelumbinis Semen) in the chronic mild stressed rat. After adaptation period for 2 weeks, each herbal extract was administered to the rats divided 5 groups. For comparison, St. John's wart was administered to another group in the same environment. The chronic mild stress was induced to rats by overnight illumination for 2 days before the forced swimming test. Struggling time, first latency and rest duration were calculated during the forced swimming test to evaluate the anti-depressant effect. The results were as follows: 1. In the result of evaluation of the struggling time, there was no significance in Pinelliae Rhizoma (-1.1%), Corni Fructus (-3.9%), Rehmanniae Radix Preparat (15.2%), Lycii Fructus (4.9%), St. John's wart (25.2%). There was a significant increase only in Nelumbinis Semen (43.9%) (P=0.0037). 2. In the result of evaluation of the first latency time, there was no significance in Lycii Fructus (-21.37%), Rehmanniae Radix Preparat (38.4%), Pinelliae Rhizoma (65.5%), Corni Fructus (29.2%), St. John's wart (75.8%). There was a significant increase only in Nelumbinis Semen (90.2%) (P=0.0116). 3. In the result of evaluation of the first rest duration, there was no change in St. John's wart, and Decrease was shown in Pinelliae Rhizoma (62.4%), Corni Fructus (31.6%) Rehmanniae Radix Preparat (63.1 %), Lycii Fructus (12.4%), Nelumbinis Semen (59.0%), but there was no significance in all. 4. Nelumbinis Semen is considered to have a superior anti-depression effect than the other herbal extracts, and to have a superior and-depression effect than St. John's wart being used commonly.

  • Intensive lifestyle modifications and anti-obesity medications are essential for obesity treatment. Antiobesity medications should be selected according to the patient's comorbidities, symptoms, and preferences. This case report describes the treatment of a morbidly obese patient with a history of depression, who complained of tingling and numbness after total thyroidectomy for papillary thyroid cancer. Very low-dose controlled-release phentermine/topiramate was prescribed and intensive lifestyle modifications were encouraged. As a result, the patient effectively lost weight and reached a near-normal weight without adverse drug effects. This implies that even an off-label anti-obesity medication low dose may be better for some patients, and the most important factor in obesity treatment is patient-tailored treatment.
