Objectives: Realgar has been frequently used for skin disorders in history of herbal medicine. However, the efficacy of realgar has not been examined in atopic dermatitis(AD). In this study, the effects of realgar on AD were investigated, especially on pruritus and inflammation. Methods: AD lesions were induced in the shaved backs of BALB/c mice through repeated application of DNCB. The mice were treated for 11 days with 1% realgar ($100{\mu}L/day$). Histological changes in skin thickness were observed. The anti-pruritic effects of realgar were evaluated by the change in numbers of scratching behavior of mice and expression of substance P. The expressions of cytokines IL-4 and IL-6 were measured. Also, anti-inflammatory effects of realgar were examined on expressions of NF-${\kappa}B$, phospho-$I{\kappa}B{\alpha}$ and mitogen-activated protein kinases (MAPKs). Results: Realgar decreased skin thickness (both dermal and epidermal) 38% and 17% respectively, compared to positive control, DNCB group. The scratching behavior of mice was reduced by 42% and expression of substance P was significantly less. Cytokines IL-4 and IL-6 were significantly reduced by 52.6% and 77.6%, respectively. The expressions of NF-${\kappa}B$, phospho-$I{\kappa}B{\alpha}$ and MAPKs (phospho-ERK1/2, -p38 and -JNK) were significantly suppressed with marked effects on phospho-ERK1/2. Conclusions: The collective results suggest that realgar shows anti-pruritic and anti-inflammatory effects on AD. And realgar might be a potential therapeutic candidate for treatment of atopic dermatitis.
Objectives : The present study was undertaken to determine whether the ethanol extact of Citri Pericarpium (CP, Pericarp of Citrus unshiu Markovich, Rutaceae) is effective against atopic dermatitis according to it's storage period. Methods : To evaluate antiallergic effect of CP and OCP (Old Citri Pericarpium) evaluated in vivo their inhibitory effects against passive cutaenous anaphylaxis (PCA) reaction induced by IgE-antigen complex and scratching behaviors by compound 48/80. The Anti-atopic effects were measured by contact dermatitis, prurient animal model and PCA reaction. Contact dermatitis in mice as a model of the Type IV reaction caused by Oxazolone. Results : The results showed that anti-pruritus effects, analgesic effects of CP was depends on its hesperetin contents. And It also showed that keep longer in storage appeared to be higher in hesperetin contents. Both CP and OCP(Old CP) have a dose-dependent analgesic action in acetic acid induced writhing syndrome. OCP Potently inhibited PCA reaction in mice, although OCP weakly inhibited in long term contact dermatitis model in mice. Conclusions : These results suggest that the Proportional to the storage period, Citri Pericarpium possesses analgesic effects and anti-allergic effects.

Objectives : The aim of this study was to investigate the effect of Chunghwatang (CHT) for atopic dermatitis. Methods : CHT, a verified anti-oxidant and anti-inflammatory effect, was treated in atopic dermatitis animal model to investigate cytokine levels and immunoglobulin production. Results : Clinical skin index was 47.1%, suggesting significant efficacy of CHT in atopic dermatitis treatment. Serum IL-4, IL-6, IL-13, TNF-${\alpha}$ and histamine productions were significantly decreased to 52.3%, 61.8%, 68.0%, 37.4%, and 46.6%, respectively. The production of IL-5 was decreased to 33.3%. The increase of Immunoglobulin IgG1 production along with IgE through the interaction of IgE and IL-4 induced by IL-4 and IL-13 were measured as 20.7% and 23.4%, respectively. Conclusions : The results above, along with the in vitro test results, strongly supports the CHT samples as effective immunomodulator in AD treatments, suggesting its use in clinical practice and basic information for EBM.

Objectives : Atopic dermatitis is a chronic inflammatory skin disease characterized by pruritic and erythematous skin lesions. The purpose of this study was to investigate the suppressive effects of Lonicerae Flos, Forsythiae Fluctus and Hwangryunhaedok Decoction Pharmacopuncture on the development of atopic dermatitis-like skin lesions in NC/Nga Mice. Methods : The Atopic Dermatitis was induced by biostir AD on the mice's back skin. Experimental groups were divided into three including LFP(Lonicerae Flos Pharmacopuncture, EtOH extract), FFP(Forsythiae Fluctus Pharmacopuncture, EtOH extract) and HHP(Hwangryunhaedok Decoction Pharmacopuncture, Hydrodistillation extract). Every second day, the mice of three groups were treated with $0.1m{\ell}$ of pharmacopuncture using a syringe at right and left acupoints ($BL_{13}$), alternatively. On the control group, normal saline was used instead of pharmacopuncture. Subsequently optical observation with a handscope, a clinical skin score, Tissue(general/immune) mast cell, Serum IgE level, Serum histamine level, and Serum lymphokine(IL-2, IL-4, $IFN-{\gamma}{\gamma}$) were measured. Results : FFP and HHP decreased the clinical skin score, the total cell number of mast cells, and the Serum total IgE level and Serum histamine level. In Serum lymphokine levels, all groups were decreased to the IL-4 level, LFP and FFP were increased to the IL-2 level, and LFP was increased to the $IFN-{\gamma}$ level. Conclusions : From the above results, Forsythiae Fluctus Pharmacopuncture (EtOH extract) and Hwangryunhaedok Decoction Pharmacopuncture (Hydrodistillation extract) exerted anti-allergic and anti-inflammatory effects, suggesting a promising agent for improving atopic dermatitis related symptoms.
Oldenlandia diffusa (OD), a member of the Rubiaceae family, has been used for treating inflammatory and infectious diseases. However, the anti-atopic effects of OD are not completely understood yet. Thus, the aim of the present study was to elucidate beneficial effects of OD on atopic dermatitis (AD) in vivo. We examined the pharmacological effects of OD on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlrobenzene (DNCB)-induced AD-like skin lesions in mice. Additionally, we evaluated regulatory effects of OD on the expression of interleukin (IL)-6 and tumor necrosis factor (TNF)-α in DNCB-induced AD-like skin lesions. Results showed that OD inhibited the scratching behavior and AD symptoms. It also decreased serum levels of IgE and histamine in mice. Moreover, OD significantly ameliorated the expression of IL-6 and TNF-α in AD-like skin lesion. These finding suggest experimental evidence of that OD's potential for treating AD.

Objective : This study was carried out to compare Saengmaeksan-gamibang(SG) to Saengmaeksan(SM) on the efficacy of moisturization, skin whitening, anti-inflammation, and anti-allergy in atopic dematitis using NC/Nga mice model. Methods : We assessed the effects of SG and SM on tyrosinase, filaggrin, serine-palmitoyl transferase(SPT), COX-2, AP-1 in vitro, on IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo with NC/Nga mice. Results : 1. SG increased the tyrosinase inhibition activity and the levels of filaggrin and SPT, decreasing the level of COX-2. 2. On the other hand, SM decreased the tyrosinase inhibition activity and the levels of filaggrin and SPT, increasing the level of COX-2. 3. Serum IgE, IL-4, 5, 6, IgM, and IgG1 were decreased in both SG group and SM group compared to the control group. The decrease degree in these factors was higher in SG than in SM. 4. Serum IFN-${\gamma}$ was increased in both SG group and SM group compared to the control group. The increase degree in IFN-${\gamma}$ was higher in SG than in SM. Conclusion : As the result of the above experiments, it was proved that SG has the moisturization, skin whitening, anti-inflammation, and anti-allergy effects, which are greater than those of SM, and provides the significant efficacy on atopic dermatitis treatment.

Objectives : Atopic Dermatitis is a recurrent, chronic, allergic eczema with severe itching sensation. This study designed to research Sihocheonggan-tang & Sihocheonggan-san to find out treatability for atopic dermatitis. Methods : We got 14 search results with searching word '시호청간탕', '시호청간산' in 'OASIS', 'NDSL', 'RISS'. We classified these studies according to the type of study, diseases, treatment methods and treatment outcomes. Results : In the analysis of the diseases, Sihocheonggan-tang & Sihocheonggan-san was given for Eczema nummulare, Ermatitis seborrheica, Erythema multiforme, Eczema and Atopic dermatitis. In the analysis of the treatment methodes, the dosage form of Sihocheonggan-tang & Sihocheonggan-san was herbal medication, pill, granules and herbal medication & pill. In the analysis of the treatment outcomes, the symptoms of all patients were improved. In the experimental researches, it was proved that Sihocheonggan-tang & Sihocheonggan-san has effects that affect anti-inflammatory, skin protection and recovery of keratinocyte. Conclusions : For this study, we can consider Sihocheonggan-tang & Sihocheonggan-san is effective for atopic deramatitis. So, it is necessary to carry out clinical trial for Sihocheonggan-tang & Sihocheonggan-san in atopic dermatitis to evaluate safety and efficacy.
BACKGROUND/OBJECTIVES: Grifola frondosa, commonly referred to as the maitake mushroom, has been studied extensively to explore its potential health benefits. However, its anti-inflammatory effects in skin disorders have not been sufficiently elucidated. This study aimed to elucidate the anti-inflammatory role of the ethanol extract of G. frondosa in atopic dermatitis (AD) using in vivo and in vitro models. MATERIALS/METHODS: We investigated its impact on skin and spleen inflammatory responses in Dermatophagoides farinae extract (DFE)/1-chloro-2,4 dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in a mouse model. Additionally, we determined the immunosuppressive response and mechanism of G. frondosa by inducing atopic-like immune reactions in keratinocytes through tumor necrosis factor (TNF)-α/interferon (IFN)-γ stimulation. RESULTS: Our study revealed that G. frondosa ameliorates clinical symptoms in an AD-like mouse model. These effects contributed to the suppression of Th1, Th2, Th17, and Th22 immune responses in the skin and spleen, leading to protection against cutaneous inflammation. Furthermore, G. frondosa inhibited the production of antibodies immunoglobulin (Ig)E and IgG2a in the serum of AD mice. Importantly, the inhibitory effect of G. frondosa on inflammatory cytokines in TNF-α/IFN-γ-stimulated AD-like keratinocytes was associated with the suppression of MAPK (Mitogen Activated Protein Kinase) pathway activation. CONCLUSIONS: Collectively, these findings highlight the potential of G. frondosa as a novel therapeutic agent for AD treatment and prevention.

Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of Hyunggaeyunkyotanggamibang(HYGB) on the recovery of dermatitic symptoms through its influence on the immune related factors. The results are as below: 1. HYGB treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of dermatitis index was observed after 14 weeks. 2. HYGB treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and CD11b+/Gr-1+ immune cells in dorsal skin by 41.7%, 21.5%, and 23.8%, respectively. 3. HYGB treated group showed an increase of CD19+ immune cells by 10.3% in PBMC, whereas CD3+, CD3+/CD69+, NKT+ immune cells were decreased by 4.3%, 42.9%, and 21.7%, respectively. 4. HYGB treated group showed an increase in the expression of IFN-$\gamma$ in the serum by 514.3%. However, the expressions of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 21.2%, 69.8%, 90.5%, 28.7%, 72.2%, 26.1%, and 19.9%, respectively. Also, the expression of IgE was decreased by 44.3%. 5. HYGB treated group showed a decrease of the expression of IL-4 and IL-5 by 43% and 44.3%. The results above indicated that HYGB clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for EBM(Evidence based medicine) construction for anti-allergic and anti-inflammatory studies.
Lactococcus lactis is a lactic acid bacterium and used in the dairy food industry. The ameliorating effects of Lactobacillus species on atopic dermatitis (AD) have been extensively studied, but the specific effect of L. lactis strains has not yet been investigated. In this study, the efficacy of L. lactis LB 1022, isolated from natural cheese, was evaluated using RAW 264.7, HMC-1 and HaCaT cell lines and an ovalbumin-sensitized AD mouse model. L. lactis LB 1022 exhibited nitric oxide suppression and anti-allergy and anti-inflammatory activity in vitro. Oral administration of L. lactis LB 1022 to AD mice significantly reduced the levels of IgE, mast cells, and eosinophils, and a range of T cell-mediated T helper Th1, Th2, and Th17-type cytokines under interleukin (IL)-10, transforming growth factor-β (TGF-β), thymus and activation-regulated chemokine (TARC), and thymic stromal lymphopoietin (TSLP). In addition, L. lactis LB 1022 treatment increased the concentration of short-chain fatty acids. Overall, L. lactis LB 1022 significantly modulated AD-like symptoms by altering metabolites and the immune response, illustrating its potential as candidate for use in functional food supplements to alleviate AD.
