The bark of Alnus japonica has been used for the treatment of fever, hemorrhage and diarrehea in oriental traditional medicine. Recently, it was revealed that the diarylheptanoids from the bark of Alnus japonica possess anti-inflammatory activity and are expected to be applicable for atopic dermatitis. In this study, oregonin, one of major active components in the bark of Alnus japonica, was developed in the form of semisolid formulations for topical delivery. Oregonin was incorporated into four ointment bases: O/W cream, W/O cream, hydrophilic ointment and lipophilic ointment. Oregonin release from all formulation prepared was evaluated. Franz cell method and immersion method were employed to characterize the release patterns of drug from each formulation based on solvent availability. O/W cream showed a better release profile than the other formulations when evaluated with Franz cell method with an order of O/W cream, hydrophilic ointment, W/O cream and lipophilic ointment. In the immersion method, hydrophilic ointment showed the greatest release rate at times 1 hour exceeding compared to other bases with an order of hydrophilic ointment, O/W cream, W/O cream and lipophilic ointment. Hydrophilicity and solvent availability of formulation seems to significantly influence the release rate of oregonin from ointment bases. In this study, we successfully characterized the oregon in ointment and found that o/w cream is a promising formulation for the topical delivery of oregonin.
In the present study, we investigated anti-inflammatory activity of artemisinic acid in HaCaT cells and RAW264.7 cells. Artemisinic acid showed inhibitory activity on macrophage-derived chemokines (MDC) expression, a factor related with atopic dermatitis (AD), in interferon (IFN)-${\gamma}$ and tumor necrosis factor (TNF)-${\alpha}$-stimulated HaCaT cells. In the study on action mechanism, pretreated artemisinic acid reduced the phosphorylation of STAT1 and p38 and the degradation of $I{\kappa}B$ by IFN-${\gamma}$ and TNF-${\alpha}$ stimulations. However, artemisinic acid didn't show the inhibitory activity on LPS-induced inflammatory mediators (NO, $PGE_2$, IL-6) in RAW264.7 cell. These results indicate that artemisinic acid inhibits IFN-${\gamma}$ and TNF-${\alpha}$-induced MDC expression through inhibition of signal factors, STAT1, NF-${\kappa}B$, and p38, in HaCaT keratinocytes.

In the present study, baicalin, baicalein, and wogonin, a major flavone isolated from Scutellaria Radix were examined for their effects on PMA-induced Interlukin-6 (IL-6), $interferon-\gamma(IFN-\gamma)$, tumor necrosis factor $(TNF)-\alpha$, IL-4, IL-10, and IL-13 productions in the PMA-stimulated $CD4^+$ Jurkat T cells. These three compounds inhibited PMA-induced Th1 cytokine $(IL-6,\;IFN-\gamma,\;TNF-\alpha)$ and Th2 cytokine (IL-4 and IL-13) productions in a concentration-dependent manner. But wogonin, but not baicalin baicalein, increased PMA-induced IL-10 production. These results suggest that baicalin, baicalein, and wogonin, a major flavone modulate Th1 and Th2 cytokine productions in $CD4^+$ Jurkat T cells and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

Korean red ginseng (KRG) is reported to have anti-allergic properties, including beneficial effects on asthma and atopic dermatitis. However, its effect on allergic rhinitis has not been studied extensively. This study examined how KRG affected allergic inflammation of the nasal cavity in an allergic mouse model. A total of 40 Balb/c female mice were divided into four experimental groups according to treatment and allergic state: group 1 (G1), saline only; group 2 (G2), ovalbumin (OVA); group 3 (G3), OVA+KRG; and group 4 (G4), OVA+dexamethasone. Serum IgE levels were significantly lower in the KRG treatment group (G3) than in the allergic group (G2). However, serum IgG1 levels did not differ between G2 and G3. In the nasal lavage fluid, IL-4 and IL-5 levels were significantly lower in G3 than in G2 (p<0.05). H&E and Luna staining revealed that the eosinophil count was lower in G3 and G4 than in G2 (p<0.05). Immunohistochemical staining revealed that there were fewer IL-4-, IL-5-, and MUC5AC-positive cells in G3 and G4 than in G2 (p<0.05). These results indicate that KRG reduces the nasal allergic inflammatory reaction in an allergic murine model by reducing Th2 cytokines.

Allergic inflammatory disease has been increased by abnormal lifestyle and food habits. Especially, prevalence of atopic dermatitis (AD) has been elevated and treatment of AD has not been unclear. Red sweet pepper (RSP), named as Capsicum annuum L, has been known as having pharmacological effects such as antioxidant, detoxification and antibacterial effects. However, the beneficial effect of ethanolic extract of RSP on AD has not been partly examined yet. Therefore, the aim of this study was to investigate anti-inflammatory effects of RSP on AD in vitro and in vivo models. The treatment of RSP inhibited the secretion of inflammatory cytokine such as interleukin (IL)-6 and IL-8 in tumor necrosis factor (TNF)-${\alpha}$ and interferon (IFN)-${\gamma}$-stimulated human keratinocyte (HaCaT cell). Also, RSP extract regulated 2,4-dinitroflorobenzene (DNFB)-induced AD-like skin lesions in BALB/c mice. Oral administration of RSP ameliorated DNFB-induced AD-like symptoms. In presented results indicated that RSP inhibited inflammatory cytokines in HaCaT cell and ameliorated AD-like skin lesion through suppression of symptom of DNFB-induced skin inflammation. Thus, RSP might be a potential therapeutic agent for AD.

Objectives : Cudrania tricuspidata (C. tricuspidata) is well-known traditional herbal remedy and its root, leaf and fruit were used for treatment of inflammation, tumor and painkilling. However, effect of C. tricuspidata fruit on tight junction is still unknown. The aim of this research was to determine the effect of C. tricuspidata fruit extract on human keratinocyte HaCaT cells. Methods : The antioxidant effects of water extract of C. tricuspidata (WECT) and ethanol extract of C. tricuspidata (EECT) were analyzed by using an ABTS assay. To confirm the cytotoxicity of WECT and EECT, MTS assay was performed. The mRNA expression levels of tight junction related genes were analyzed using quantitative RT-PCR analysis. Furthermore, dispase assay was used to investigate the alteration of cell-cell adhesion strength of EECT treated HaCaT cells. Results : WECT and EECT showed strong antioxidant activity. No obvious cytotoxicity was observed in both WECT and EECT until $2.0mg/m{\ell}$ concentration. The mRNA expression level of Claudin 6 were significantly increased by EECT treatment, whereas the WECT did not affect the expression of Claudin 6. Furthermore, EECT treatment enhances cell-cell adhesion strength. Conclusions : In this study, we investigated the physiological activities of the extracts of Cudrania tricuspidata fruit extracts on human keratinocytes by two different extraction methods. EECT might have an anti-aging activity on the skin by reducing oxidative stress. Moreover, it may be a useful ingredient in atopic dermatitis and skin-moisturizing, given its effects of altering Claudin 6 gene expression and enhancing cell-cell adhesion strength.

In this study, oregano was extracted by supercritical extraction and hydrothermal extraction method. In vitro experiments such as antimicrobial and antioxidant activity test were performed. As a result of the disc diffusion method, only the supercritical extracts formed a clear zone. The MIC for S. aureus was found only in the supercritical fluid extracts and it was 1000 ㎍/mL. The hydrothermal extract's MIC is 125 ㎍/mL for C. acnes. Through biofilm inhibition assay, we found that the supercritical fluid oregano extracts inhibit the biofilm of S. aureus by more than 70% even at low concentrations of 125 ㎍/mL. On the other hand, the antioxidant ability of the hydrothermal extract was better than that of the supercritical fluid extracts. Furthermore, we tried to make a skincare ingredient for atopic dermatitis by utilizing the S. aureus biofilm inhibitory ability of oregano supercritical fluid extracts. Liposome was used to overcome the low solubility of the oregano supercritical fluid extracts and increase stability.

Objectives : The purpose of this study is to analyze the current trends of various herbal medicine research on allergic disease with dysbiosis. Methods : Electronic searches were performed using Pubmed, Research Information Sharing Service(RISS), Korean studies Information Service System(KISS), Oriental medicine Advanced Searching Integrated System(OASIS). Results : We analyzed ten studies on the effect of herbal medicine on allergic disease with dysbiosis. Eight studies were animal experimental studies, and two were randomized clinical trial(RCT) study and one-group pretest-posttest research, respectively. Among the studies, three studies were on atopic dermatitis, two on allergic rhinitis, and five on asthma. All different herbal medicines were used in the studies. Changes in gut microbiota composition were observed in nine studies except for 1 RCT study. In eight animal experimental studies, there was significant reduction in allergy-related inflammatory markers. Six studies evaluated the change of metabolites related to gut microbiota and three of them showed significant increase in short-chain fatty acids(SCFA). Conclusion : This study provides current trends of studies on herbal medicine research on allergic disease with dysbiosis. Most research is conducted using animal experiments, and this is a relatively recent trend. These studies offer basic knowledge on the correlation between herbal medicine, gut microbiota, and anti-inflammatory effects in allergic disease.
Poly-$\gamma$-glutamic acid ($\gamma$-PGA) was mixed natural flora of Bacillus subtilis, contaminated from cooked soybeans. Also, it was performed to find out the antiallergic activity by using NC/Nga mice, in vitro. The $\gamma$-PGA (PGA-HM : PGA-high molecular weight), Molecular weight 300 kDa, was decomposed and made PGA-LM (PGA-low molecular weight) which has molecular weight below 30 kDa by sonication. Therefore, it was same result between PGA-HM and PGA-LM, and reported PGA-LM as basic result. We found that PGA-LM contains antiallergic efficacy that inhibit B cells and Th2 cells activation from isolated CD4+T cells in NC/Nga atopic dermatitis model mice, and not show a cytotoxicity in the hFCs. To investigate the effects of these PGA-LM in vitro, isolation of splenic B cell and CD4+ T cells in atopic dermatitis mice were used. To elucidate the role of PGA-LM in anti-CD40+ interleukin-4 (IL-4)-mediated B-cell activation, showed that the capacity of B cells to expression IL-$1\beta$, IL-6, and TNF-$\alpha$ mRNA down-regulated, and IL-10 mRNA up-regulation by PGA-LM treatment, but it had no effect on TGF-$\beta$ expression. In addition to CD4+IFN-$\gamma$+ and CD4+CD25+foxp3+, the functions of PGA-LM in the development of the CD4+CD25+foxp3+ and CD4+IFN-$\gamma$+cells, the phenotype and functions of PGA-LM induced CD4+CD25+foxp3+, and CD4+IFN-$\gamma$+cells in CD4+T cells. These results suggested that PGA-LM could change cytokine production and generate CD4+CD25+foxp3+ Tregs in NC/Nga mice, and may be effective for immunotherapy in patients with AD.
Objective : This experimental study was performed to examine the anti-allergic infiammatory effects of Mori Folium. Method : Macrophage 264.7 cells were pretreated for 1hour with Sangyup. After pretreatment, macrophage were incubated with lipopolysaccharide(LPS) $100ng/m{\ell}$ 12h and media collected and $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, IL-10 concentrations in supernatants were measured each by Enzyme linked immuno-sorbent assay. Sangyup were used $1mg/m{\ell}$, $500{\mu}g/m{\ell}$, $250{\mu}g/m{\ell}$, $100{\mu}g/m{\ell}$, $50{\mu}g/m{\ell}$. Hydrocortisones were used $10^{-4}M,\;10^{-5}M,\;10^{-6}M,\;10^{-6}M,\;10^{-6}M$. Results : Macrophage 264.7 cells were pretreated with Hydrocortisones and Sangyup. After pretreatment, macrophage were incubated with lipopolysaccharide(LPS). To investigate the anti-inflammatory effect of Sangyup, we measured the amount of cytokines, and the results are as follows; 1. Mori Folium. showed statistically significant inhibitory effect on antiinflammation in $TNF-{\alpha}(p<0.02)$ in all five concentrations compared with the (-)controls treated with LPS. Mori Folium showed inhibitory effect on antiinflammation in $TNF-{\alpha}$ in similarpattern in all five concentrations compared with the (+)control pretreated with hydrocortisone. 2. Mori Folium. showed statistically significant inhibitory effect on antiinflammation in IL-6(p<0.01) in all five concentrations compared the (-)controls treated with LPS. Mori Folium. showed inhibitory effect on antiinflammation IL-6 similar pattern in all five concentrations compared with the (+)control pretreated with hydrocortisone. 3. Experimental Group pretreated with Mori Folium. showed statistically significant difference of antiinflammation in $IL-1{\beta}$ in concentrations of $50{\mu}g/m{\ell}(p<0.01)$, $250{\mu}g/m{\ell}(p<0.05)$ compared with the (-)control treated with LPS. Experimental Group pretreated with Mori Folium. showed increased level of $IL-1{\beta}$ in all concentrations compared the (+)control treated with hydrocortisone. 4. Experimental Group pretreated with Mori Folium. did not show statistically significant effect on IL-10 compared with the (-)control. Conclusion : By the findings of this experiment, Mori Folium is observed to have antiallergic and antiinflammation effect.
