Antiviral Activity of Glycyrrhetinic Acid on MA-104 Cell Infection of the K-21 Korea Human Rotavirus Isolate

  • The mutability and frequency of genetic reassortment characteristic of rotavirus and resultant antigenic changes make the rotavirus formidable challenges for control efforts such as the vaccine development. An alternative approach to overcome these difficulties in development of the rotavirus vaccine is to develop effective inhibitors of the virus infection. As an effort to achieve this, effects of glycyrrhetinic acid (GA), which is an active component of glycyrrhizin, on MA-14 cell infection were examined by employing the human rotavirus isolated from Korea, K-21. The data obtained showed that MA-104 cell infection of the K-21 rotavirus was greatly influenced by the presence of both $18{\alpha}-Ga\;and\;18{\beta}-GA$. Both types of GA have inhibited more than 60% of the rotaviral infection at the concentration of 7.68mM. This inhibition effect became much more evident at the higher concentrations of GA. However, the type of GA did not make much differences on the inhibition effect of the drug. Although GA has to be used in high concentrations to exhibit anti-viral activity and to be virostatic, a long history of safe and high dose usage of licoriece in clinical settings in the Far East makes the GA as an attractive inhibitor of the rotaviral infection.

Study on Biological Effect of Euiiin-tang(y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) Extracts by Extraction Methods (의이인탕 추출방법에 따른 생리활성 연구)

  • Objectives: The purpose of this study is to investigate biological effects of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts by extraction methods. Methods: To investigate the effects of anti-oxidative and anti-inflammatory activity, we measured DPPH radical scavenging assay, collagenase inhibition assay and COX-2 inhibition assay. Results: In case of measuring anti-oxidative and anti-inflammatory activities of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) by collagenase and COX-2 inhibition activity, it came out that Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) was unable to inhibit collagenase, but was effective to inhibit COX-2. Cytotoxicity analysis of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts by MTT assay principles, there was no cytotoxicity in aqueous and 50% ethanol extracts in any level of concentration. Conclusions: This results suggest that Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts have anti-oxidative and anti-inflammatory activities.

Study on the Intrinsic Defects in Undoped GaSb Bulk and MBE-grown GaSb/SI-GaAs Epitaxial Layers for Infrared Photodetectors (적외선검출소자를 위한 GaSb 결정 및 MBE로 성장한 Gasb/SI-GaAs 박막의 진성결함에 관한 연구)

  • We have investigated the intrinsic defects remaining in epitaxial GaSb layers grown on SI-GaAs substrates compared to those in bulk GaSb crystal substrate, which is a basic material of Sb-based strained-layer superlattice infrared photodetectors. From the functional dependence of the band-to-band transition energy of the photomuminescence (PL) spectra observing up to near room-temperature (250 K), the temperature parameters of [$E_o$, $\alpha$, $\beta$] of undoped GaSb crystal are determined by using the Varshni empirical equation describing the temperature variation of the bandgap energy. Additionally to the antisite-Ga ([$Ga_{Sb}$]) with an ionization energy of 29 meV that is well known to a major intrinsic defect in GaSb, epitaxial GaSb layers show a pair of deep states at the emission energy of 732/711 meV that may be related with a complex of two antisite-Ga and antisite-Sb ([$Ga_{Sb}-Sb_{Ga}$]). Based on the analysis of the temperature and the excitation-power dependences of PL, it suggests that excess-Sb substitutes Ga-site by self-diffusion and two anti sites of [$Ga_{Sb}$] and [$Sb_{Ga}$] could form as a complex of [$Ga_{Sb}-Sb_{Ga}$] in GaSb epilayers grown under Sb-rich condition.

  • In order to increase the efficiency of LED, transparent electrodes should be also developed. also suitable anti-reflection coating (ARC) is necessary for practical device applications. In our paper, Al-doped ZnO (AZO) films were fabricated by sputtering on GaP substrate(wavelength:620nm). Choosing optimum substrate temperature and sputtering rate, high quality AZO films were formed. We confirmed that the surface and electrical properties, which implemented using the methods of AFM, Hall measurement. The properties of AZO thin films especially depended on the thickness. We presumed that the change of the increase the external quantum efficiency of LED according to the AZO thin film of thickness.

  • For the preliminary step to make and characterize the monoclonal antibodies of human alpha-fetoprotein (AFP) was purified from 534g of human fetal tissues through the procedures of tissue extraction, DEAE-cellulose, concanavalin A-Sepharose, Cibacron Blue F3GA-agarose and immunoadsorbent affinity chromatography. The isolated AFP preparation showed a single band on polyacrylamide gel electrophoresis and a single precipitin are against rabbit anti-human cord serum and anti-human AFP on immunoelectrophoresis. Our AFP also displayed a single band on SDS-polyacrylamide gel electrophoresis. The recovery of AFP was 8.76mg total.

  • The growth and characterization of $In_{0.5}Ga_{0.5}As/In_{0.5}Al_{0.5}As$ narrow-gap inverted high electron mobility transistor structures, developed as a candidate material for spin-injection devices, are presented in this study. We have grown samples possessing surface $In_{0.5}Ga_{0.5}As$ channels of different thicknesses (30 nm and 60 nm) both with and without a thin 3 nm $In_{0.5}Ga_{0.5}As$ cap layer by using molecular beam epitaxy. We then investigated the in-plane transport properties as well as the Rashba spin-orbit coupling constant of the two-dimensional electron gas confined at the heterojunction interface.

Spin-polarization and x-ray magnetic circular dichroism in GaAs

  • The combination of angular spin momentum with electronics is a promising successor to charge-based electronics. The conduction bands in GaAs may become spin-polarized via optical spin pumping, doping with magnetic ions, or induction of a moment with an external magnetic field. We investigated the spin populations in GaAs with x-ray magnetic circular dichroism for each of these three cases. We find strong anti-symmetric lineshapes at the Ga $L_3$ edge indicating conduction band spin splitting, with differences in line width and amplitude depending on the source of spin polarization.

Anti-inflammatory and Anti-bacterial Activities of Artemisia iwayomogi Kitamura Extract Fractions (한인진 분획물의 항염증, 항균 효과)

  • This study investigated the anti-bacterial and anti-inflammatory activities of Artemisia iwayomogi Kitamura for medical and food additive applications. A 70% methanol extract of A. iwayomogi Kitam. was fractionated with a series of solvents in order of increasing polarity; these fractionated extracts showed high 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) scavenging activity in the ethyl acetate fractions and high 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity in both the ethyl acetate and chloroform fractions. Levels of total phenolics and flavonoids were highest in the ethyl acetate fraction extracts. Eight bacteria were then treated with the A. iwayomogi Kitam. extract fractions and most showed similar or lower levels of anti-bacterial activity when compared to the control group, except for Staphylococcus aureus. In addition, the anti-oxidant activities of the ethyl acetate fractions of the A. iwayomogi Kitam. extract were higher than those of the other fractions. To confirm the anti-inflammatory activity of the extract fractions, we applied them to RAW 264.7 cells using LPS, and the results indicate that an ethyl acetate fraction at 250 ㎍/ml concentration inhibits nitric oxide production. This study demonstrates that an ethyl acetate fraction of A. iwayomogi Kitam. extract inhibits bacterial and inflammatory activities and could be useful as a potential source of bioactive compounds.

Effect of $18{\beta}$-Glycyrrhetinic Acid on Septic Arthritis Caused by Candida albicans ($18{\beta}$-Glycyrrhetinic Acid의 항 감염성관절염효과)

  • A polymorphic fungus, Candida albicans, causes various forms of infections such as disseminated candidiasis and vaginitis. Recent reports indicate that the fungus is a main etiological agent for the arthritis. In search of new sources for treatment of the fungal arthritis, we examined $18{\beta}$-glycyrrhetinic acid ($18{\beta}$-GA) against C. albicans-caused septic arthritis. The compound is isolated from Glycyrrhizae Radix that is known to have various immunomodulating activities and is one of the most popular herbal medicines. For induction of animal model of a septic arthritis, mice were given an emulsion form of C. albicans cell wall mixed with Complete Freund's Adjuvant (CFA) via footpad-injection. To determine prophylactic and therapeutic effects, the component was given to the animals before or after the induction of the arthritis, respectively. Data showed that intraperitoneal administration of $18{\beta}$-GA resulted in reduction of the inflammation, indicating the component had both prophylactic and therapeutic activities. For investigation of mechanism of the $18{\beta}$-GA, inhibitory effects on NO (nitiric oxide) and on T-lymphocyte proliferation were determined. Results demonstrated that $18{\beta}$-GA suppressed NO production from LPS (lipopolysaccharide)-treated macrophages and also inhibited proliferation of Con A (concanavalin A)activated T-cells. Taken together, $18{\beta}$-GA, a pentacyclic triterpene, has anti-arthritic activity against C. albicans-caused septic arthritis, possibly by blocking NO production and T-cell suppression.

Effect of Applied $GA_3$ and Paclobutrazol, an Inhibitor of GA Biosynthesis, on the Growth of Internodes and Panicle of the Rice Plants ($GA_3$와 GA 생합성 억제제 처리가 수도의 절간신장 및 수의 발육에 미치는 영향)

  • Recently several synthetic chemicals inhibiting biosynthesis of gibberellic acids were found to be effective in preventing cereal crops from lodging by reducing culm length. The present study aimed to clarify the extent of practical manipulation of the culm length of the rice crop by changing the endogenous level of gibberellic acids with exogenous application of GA$_3$ or GA biosynthesis inhibitor. Three rice cultivars, 'Jinheung', 'Minehikari' (Japonica) and 'Hangangchal' (semi-dwarf Indica x Japonica) were treated with GA$_3$ (0, 20, 50 and 100 ppm solution) and an anti-gibberellin, paclobutrazol (0, 100, 200 and 300 g ai/ha) at three growth stages (maximum tillering, panicle initiation and meiotic stage), respectively. The application of GA$_3$ (50, 100 ppm) at maximum tillering stage and panicle initiation stage promoted the elongation of the 5th internodes from the top and GA$_3$ applied at meiotic stage promoted remarkably the elongation of the 3rd internodes. Culm was most elongated when GA$_3$ was applied at maximum tillering stage in 'Jinheung', at panicle initiation stage in 'Minehikari' and at meiotic stage in 'Hangangchal'. Paclobutrazol shortened the 4th and 5th internodes from the top when it was applied at maximum tillering stage and panicle initiation stage and the 2nd and 3rd internodes when it was applied at meiotic stage in the three cultivars. The semi-dwarf Indica x Japonica variety responded to a greater extent to the GA$_3$ and to a less extent to the anti-gib berellin than the Japonica varieties. The effect of GA$_3$ and paclobutrazol on the panicle length and the number of spikelets per panicle varied with time and dose of their application as well as with variety.
