Anti-Platelet Aggregation Effect of Extract from Gamisopunghwalheol-tang in Vitro (가미소풍활혈탕의 혈소판 응집억제 작용에 대한 in vitro 연구)

  • The study was designed to test anti-platelet effect and find out anti-platelet mechanism of extract from Gamisopunghwalheol-tang in vitro. The extract was investigated for the inhibition against the aggregation of human platelet suspensions induced from collagen by aggregometer. And also the extract was investigated for the inhibition against the aggregation of human platelet suspensions who is taking aspirin or clopidogrel induced from collagen by aggregometer. In collagen-induced platelet aggregation test, the extract significantly inhibited collagen-induced platelet aggregation in a concentration-dependent manner(p<0.05). The extract significantly inhibited collagen-induced platelet aggregation of human platelet who is taking aspirin or clopidogrel ing 0 mg/ml concentration(p<0.05). And the extract inhibited more ingpatients who is taking aspirin. These results show that the extract from Gamisopunghwalheol-tang has anti-platelet aggregation effect.

  • By increasing trades between countries, importance of harbors is becoming serious, including our country. When it comes to Container Crane Operation, the most important matter is how many containers are loaded in a truck or a ship by given time. This can be a crucial matter of harbors in taking care of materials. The present harbors' crane uses a wire-rope conveyance materials are transported in the air and have high free-angle of location. The sway can cause the delay of time, wrong position of Trolley and the damage of materials. In this study, we obtain the optimal PID parameters with GA(Genetic Algorithm) and apply those parameters to the PID Controller. In the result of the experimentation, we can see how effectively the PID controller, applied with the optimal parameters obtained by GA, can control the sway angle.

The Effect of Bee-venom Acupuncture on Neck Pain (경항통(頸項痛)에 봉약침요법(蜂藥針療法)이 미치는 영향(影響))

  • Objective: To evaluate the treatment effect of Bee-venom Acupuncture on Neck Pain, which is well known for anti-inflammatory effect and function of activating immune system. Methods : We investigated 22 cases of patients with Neck Pain induced by soft tissue injury. One group was treated by Bee-venom Acupuncture therapy (Bee-venom Acupuncture Group :BA), the other group was treated by General Acupuncture therapy (None Bee-venom Acupuncture Group :GA). We evaluated the treatment effect of each group by few statistical analysis such as frequency analysis, Mann Whitney U test and spearman correlation analysis. Results : 1. In the age distribution of both groups, we found the highest frequency at the interval 35 to 45 years old. And in the number of medical treatment, Both groups show the highest frequency at the interval of less than four times. 2. BA was significantly decreased in VAS and clinical grade compared with GA. 3. We found significant correlation between period and number of treatment. So did we between VAS and clinical grade. Conclusion : As a result of thorough investigation, we could come to a conclusion that Bee-venom Acupuncture Group might have a notable effect on VAS and clinic grade.

Studies on Triterpenoid Corticomimetics

  • It was our working hypothesis that introduction of 11-keto groups to 12-oleanene/ursene series of triterpenoids should endow them with corticoid-like activities, since pharmacological actions of glycyrrhetinic acid (GA) are known to be caused by inhibition on $corticoid-{\delta}^4-reductase$. 11-Keto-triterpenoids derived artificially in these studies, such as 11, 19-diketo-18, 19-secoursolic acid methyl ester(I), $11-keto-{\beta}-boswellic$ acid derivatives (IIa-IIc), 11-Keto-presenegenin dimethyl ester (III), II-keto-oleanolic acid derivatives (IVa-IVd) and 11-keto-hederagenin (V) possess the fundamental functions of ${\alpha},\;{\beta}-unsaturated$ ketone on C-11 and hydroxyl group on C-3, as like GA (VI). Additionally, they involve the carboxyl groups on rings A (II, III), D (I, III, IV, V) and E (VI), and the hydroxyl groups on rings A (III, V) and C (III). All the compounds competitively inhibited $corticoid-5{\beta}-reductase$, and the highest inhibitory potency appeared in I. All of them except $3,\;11-diketo-{\beta}-boswellic$ acid methyl ester (IIc) were more effective about five times to twice than GA. On carrageenin-induced edema test, compounds I and IVa-IVd showed anti-inflammatory activities, but III enhanced rather edema. Structure-activity relations were found in the aspects of hydrophilicity of ring A and hydrophobicity of rings C/D. The more they were hydrophilic in ring A and hydrophobic in rings C/D, the more they inhibited the enzyme. And the more they were hydrophobic in rings C/D, the more they exhibited antiiflammatory activities. However, the increased hydrophilicity in ring A resulted in increasing edema, probably due to a nonspecific inhibition on $aldosterone-5{\beta}-reductase$.

The Effect of Jininwhachul-tang-ga-wasong on the Hepatic Cancer in Mouse by N-Nitrosodiethylamine (진인화철탕가와송(眞人化鐵湯加瓦松)이 N-Nitrosodiethylamine으로 유발된 흰쥐의 간암에 미치는 영향)

  • Objective : The purpose of our study was to investigate the effect of JinlnWhaChul-tang-ga-wasong (JIN) on NDEA-induced liver tumorigenesis. Materials and Methods : We investigated the possible protective effects of Jininwhachul-tang-ga-wasong (JIN) as an anticancer against NDEA-induced liver injury in mice. Experimental mice were classified into 3 groups; normal, saline administered group (control group), and JIN extract (0.15g/kg/every other day) administered group (JIN group) after being injected with NDEA over 12 weeks. We examined the state of differentiation of these tumors and the effects of JIN after 6 weeks. To confirm the induction of apoptosis, the cells were analyzed by terminal deorynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) assay, acridine orange staining and flow cytometric analysis. To investigate inhibitory effect on the expression of COX-2 by JIN, we performed COX-2 immunohistochemistry and reverse transcription polymerase chain reaction analysis. Results : Body weights significantly decreased in the control and JIN groups compared with the normal group. The levels of cholesterol, hemoglobin and testosterone decreased in the control compared with the normal group. The level of estradiol significantly increased in the control compared with the normal group. The control group reacted with TUNEL assay more than the normal and JIN groups. Upon naked eye, light and electron microscopic examination, JIN improved the morphological and histopathological changes of the liver caused by NDEA-induced hepatic neoplasm. COX-2 immunoreactivity decreased in the JIN group compared with the control group, mRNA expression of the control group was greater than the normal and JIN groups. Conclusion : these results suggest the possibility that JIN may exert an anti-tumor effect on NDEA-induced liver tumorigenesis.

Effect of Bee Venom Pharmacopuncture and Scolopendra Subspinipes Pharmacopuncture on Functional Recovery and Anti-inflammation after Sciatic Crushed Nerve Injury in Rats (봉독약침과 오공약침이 좌골신경 손상 흰쥐의 기능 회복과 염증 억제에 미치는 영향)

  • Objectives : Peripheral nerve injuries are commonly encountered clinical problems and often result in serve functional deficit. Bee venom and scolopendra subspinipes have been traditionally used in oriental medicine to treat several inflammatory diseases and chronic pain conditions. Methods : In the present study, the effects of bee venom pharmacopuncture and scolopendra subspinipes pharmacopuncture on functional recovery, severity of pain, and expressions of neurofilament, cycloxygenease-2(COX-2), and tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) following sciatic crushed nerve injury in rats were investigated. For this study, walking tract analysis, plantar test, western blot for COX-2 and TNF-${\alpha}$, and immunohistochemistry for neurofilament were performed. Results : In the present results, sciatic functional index(SFI) in walking tract analysis was significantly decreased following sciatic crushed nerve injury, and pain severity in plantar test was significantly increased. COX-2 and TNF-${\alpha}$ expressions were increased whereas neurofilament expression was decreased by sciatic crushed nerve injury. On the other hand, bee venom pharmacopuncture and scolopendra subspinipes pharmacopuncture improved SFI in walking tract analysis and suppressed the pain severity in sciatic crushed nerve injury. Bee venom pharmacopuncture and scolopendra subspinipes pharmacopuncture suppressed COX-2 and TNF-${\alpha}$ expression and enhanced the neurofilament expression in sciatic crushed nerve injury. Conclusions : In the present study, we have shown that treatment with bee venom or scolopendra subspinipes is the effective therapeutic modality to ameliorate the symptoms of sciatic crushed nerve injury. The efficacies of bee venom and scolopendra subspinipes were similar.

  • Wasong(Orostachys japonicus A. Berger) has been used as anti-cancer medicinal plants from ancient time. This experiment was conducted to obtain some fundamental informations concerning anatomical study, germination test, soil analysis of indigenious districks, adaptaion of soil for wasong. The leaf of wasong was xerotype with spine, mesophyl chloroplast is dense surrounded vascular bundle, with a many large water storage tissue without chloroplast, and was bifacial stomata leaf type. The stem of wasong was amphicribral vascular bundle, central cylinder was cylinder type and protostele. Root was polarch type, potoxylem of it's was arranged in ring shape of annulation and metaxylem. The structure of flower was cymose, was caylyx of 5 sepals, corolla of 5 petals, roecium of 10 stames, ovules of five, placentation parietal, ovary superior, axile placenta. The optimum temperature for seed germination was $25^{\circ}C$ under light and germination percentage was 22.5%. Dormancy breaking was effective for 6-8 days at 5$^{\circ}C$ and at 100 ppm level of $GA_3$ The soil analysis of wasong local districks, content of available $P_2O_5$, Ca were higer than optimal level of upland and C.E.C., exchangeable k was not significantly different of that, but exchangeable magnesium was very low. The growth of wasong was affected draining regardless soil combination treatment.

Antioxidant, Anti-diabetic, Anti-cholinesterase, and Nitric Oxide Inhibitory Activities of Fruiting Bodies of Agaricus brasiliensis (신령버섯 자실체 메탄올 추출물의 항산화, 항당뇨 및 Nitric Oxide의 저해 효과)

  • Agaricus brasiliensis, belonging to the family Agaricaceae of Agaricales, Basidiomycota, has been used for edible and medicinal purposes. This study was initiated to evaluate the antioxidant, anti-diabetic, and nitric oxide inhibitory activities of fruiting bodies of A. brasiliensis extracted with methanol. The HPLC analysis of phenolic compounds from the mushroom extracts identified 10 phenolic compounds including gallic acid, procatechuic acid, chlorogenic acid, (-)-epicatechin, vanillin, rutin hydrate, naringin, quercetin, formononetin, and biochanin-A. The free radical scavenging activities of methanol extract were lower than that of positive control, BHT. The chelating effects of methanol extract were significantly higher than those of the positive control, BHT at the all concentrations tested. The methanol extract exhibited the lower reducing power activities compared with the positive control at the 0.5~6.0 mg/mL concentration. The mushroom extract inhibited the ${\alpha}$-glucosidase activity by 54.48% and 78.43% at the 1.0 and 2.0 mg/mL while acarbose, the positive control, inhibited the ${\alpha}$-glucosidase activity by 51.77% and 81.81% at the same concentrations, respectively. Nitric oxide (NO) production in lipopolysaccahride (LPS) induced RAW 264.7 cells were inhibited by the methanol extracts in a concentration dependent manner. Therefore, it is concluded that fruiting bodies of A. brasiliensis contained natural antioxidant, anti-diabetic, and anti-inflammatory substances which can be useful for human health.

Antineoplastic Effect of Several Herbal Medicine Mixtures on SNU-80 Anaplastic Thyroid Carcinoma Cell Line (수종 한약 복합물의 역형성갑상선암세포 SNU-80에 대한 항암효과)

  • Objectives: The purpose of this study was to investigate the antineoplastic effect of several herbal medicine mixtures (compositions of Astragalus membranaceu, Angelica gigas, Trichosanthes kirilowii, Panax ginseng, Rhus verniciflua Stokes) on the SNU-80 anaplastic thyroid carcinoma cell line. Methods: MTT assay was used to examine whether our herbal medicine mixtures decreased cell growth rate of SNU-80. Wound healing assay and Transwell invasion assay was performed to investigate whether our herbal medicine mixtures affect the migration and invasion of anaplastic cancer cells, SNU-80. ELISA assay was performed to know if our herbal medicine mixtures suppressed the expression of pro-invasive molecules, such as vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2) secreted from SNU-80. Results: MTT assay demonstrated that A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas :T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 strongly suppressed the growth of SNU-80. Conclusions: The results obtained in this study suggest that several herbal medicine mixtures suppresse the growth and inhibit the migration and invasion of SNU-80, which is anaplastic thyroid cancer cells. Especially, A. membranaceus:A. gigas: T. kirilowii=1:1:1 mixture had a stronger anti-cancer effect.

Cha-ga Mushroom Water Extract induces G0/G1 Arrest in B16-F10 Melanoma cells (차가버섯추출물에 의한 흑색종의 세포주기 억제효과)

  • Chaga mushroom extract is well known as immune modulator and anti-cancer agent. However, the molecular mechanism by which Chaga exerts cell cycle arrest and apoptosis of cancer cells is poorly understood. In this study, we demonstrated anti-proliferative effects of Chaga extract on murine melanoma B16 cells. Chaga extract dose-dependently inhibited cell growth along with the arrest of G0/G1 phase and the induction of apoptotic cell death. Treatment with Chaga extract resulted in a decrease of cyclin E, cyclin D1, cdk 2, cdk 4 expression levels. Furthermore, in vivo inoculation study of B16 melanoma cells into Balb/c mice Chaga extract markedly suppressed the metastatic growth of tumor cells (6 folds, p<0.05,). These results indicate that Chaga mushroom extract induces apoptosis of B16 melanoma cells through arrest of G0/G1 phase in cell cycle.
