Tissue-cultured Gymnema sylvestre Leaf Extract Enhance Anti-adipogenic Effects through Inhibition of C/EBP-${\alpha}$ Expression and Lipid Droplet Accumulation (조직배양된 Gymnema sylvestre 추출물의 C/EBP-${\alpha}$ 및 지방축적 억제에 따른 항비만 효과)

  • Gymnema sylvestre, a traditional India medicine called a 'Gagaimogouip' in Korea, has been used to regulate sugar metabolism in patients with diabetes mellitus. First, we induced callus from Gymnema sylvestre leaf and mode extract from cultured callus. In this study, we investigated the anti-obesity effect of tissue-cultured Gymnema sylvestre leaf extract in 3T3-L1 cells. The tissue-cultured Gymnema sylvestre extract inhibited the cytoplasmic lipid accumulation as well as adipogenic differentiation of preadipocytes. The tissue-cultured Gymnema sylvestre extract inhibited insulin-induced predipocyte differentiation through the inhibition of expression of the early adipogenic transcription factor C/EBP-${\alpha}$ that regulate adipogenesis. These results indicate that tissue-cultured Gymnema sylvestre extract may be potential natural ingredient for slimming cosmetic products.

Fumigaclavine C attenuates adipogenesis in 3T3-L1 adipocytes and ameliorates lipid accumulation in high-fat diet-induced obese mice

  • Fumigaclavine C (FC), an active indole alkaloid, is obtained from endophytic Aspergillus terreus (strain No. FC118) by the root of Rhizophora stylosa (Rhizophoraceae). This study is designed to evaluate whether FC has anti-adipogenic effects in 3T3-L1 adipocytes and whether it ameliorates lipid accumulation in high-fat diet (HFD)-induced obese mice. FC notably increased the levels of glycerol in the culture supernatants and markedly reduced lipid accumulation in 3T3-L1 adipocytes. FC differentially inhibited the expressions of adipogenesis-related genes, including the peroxisome proliferator-activated receptor proteins, CCAAT/enhancer-binding proteins, and sterol regulatory element-binding proteins. FC markedly reduced the expressions of lipid synthesis-related genes, such as the fatty acid binding protein, lipoprotein lipase, and fatty acid synthase. Furthermore, FC significantly increased the expressions of lipolysis-related genes, such as the hormone-sensitive lipase, Aquaporin-7, and adipose triglyceride lipase. In HFD-induced obese mice, intraperitoneal injections of FC decreased both the body weight and visceral adipose tissue weight. FC administration significantly reduced lipid accumulation. Moreover, FC could dose-dependently and differentially regulate the expressions of lipid metabolism-related transcription factors. All these data indicated that FC exhibited anti-obesity effects through modulating adipogenesis and lipolysis.

Anti-adipogenic Effects of Vibration with Varied Frequencies on 3T3-L1 Preadipocytes (3T3-L1 지방전구세포에 대한 다양한 주파수 진동의 지방 생성 억제 효과)

  • Vibration is a mechanical cue that can be applied to adipose tissues for the purpose of treating obesity. However, the exact correlation between vibration and other anti-adipogenic pathways, such as development of cytoskeleton and apoptosis, remains unknown. The objective of this study was to investigate the unknown anti-adipogenic effects of vibration with varied frequencies on preadipocytes. 3T3-L1 preadipocytes were cultured in Dulbecco's modified Eagle's medium (DMEM) containing 5% calf serum at 37 ℃ with 5% CO2 in a humidified incubator. Vibration was generated using Arduino Uno microcontroller and vibration motor module with 1 V DC, and applied to preadipocytes for 3 days. Frequency conditions were set to 20, 55, and 90 Hz. Then, the expressions of p38 pathway, ROCK-1, α-actinin, Bax, Bcl-2, caspase-9, 8, and 3 were analyzed with western blot. As a result, p38 pathway was inhibited in 55 and 90 Hz while ROCK-1 and α-actinin were expressed in 20 Hz. Caspase-3, a terminal apoptotic factor, was activated in 20 Hz via extrinsic pathway rather than intrinsic pathway. Results suggest that various frequencies of vibration can inhibit adipogenesis via different pathways which sheds light on future mechanotransduction applications of vibration for the treatment of obesity.

The Anti-Inflammatory Effect of Gabyeobda Tea in High Fat Diet-Induced Obese Mice (가볍다차(茶)가 고지방식이로 유도된 비만 마우스에서 항염증에 미치는 효과)

  • Objectives: The purpose of this study was to investigate the effects of Gabyeobda tea (GT) on anti-inflammation in ice induced high fat diet (HFD). Methods: The C57BL/6 mice fed HFD were administrated with GT once daily for 8 weeks. The changes of body weight, calorie intake levels were measured in mice. The level of serum total cholesterol, triglyceride, high density lipoprotein cholesterol, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPT) were measured in mice by enzyme-based assay. It was also observed the histological changes of liver, and fat tissues with hematoxylin and eosin staining. Further real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay were employed to detect inflammatory cytokine levels such as tumor necrosis factor (TNF)-𝛼, interleukin (IL)-6, and IL-1𝛽. Results: HFD+GT group, which was administered with GT with HFD, showed no body weight gain compared with HFD group. However, levels of GOT, GPT, and inflammatory cytokines such as TNF-𝛼, IL-6, and IL-1𝛽 in the blood of HFD+GT group were significantly reduced compared with HFD group. In addition, the messenger RNA (mRNA) expression level of the IL-12 gene was significantly reduced and the mRNA expression level of the IL-10 was increased in the liver. Conclusions: It suggests that Gabyeobda tea can alleviate inflammatory responses induced by high fat diet by inhibiting inflammatory cytokines production.

Identification of anti-adipogenic withanolides from the roots of Indian ginseng (Withania somnifera)

  • Background: Withania somnifera (Solanaceae), generally known as Indian ginseng, is a medicinal plant that is used in Ayurvedic practice for promoting health and longevity. This study aims to identify the bioactive metabolites from Indian ginseng and elucidate their structures. Methods: Withanolides were purified by chromatographic techniques, including HPLC coupled with LC/MS. Chemical structures of isolated withanolides were clarified by analyzing the spectroscopic data from 1D and 2D NMR, and HR-ESIMS experiment. Absolute configurations of the withanolides were established by the application of NMR chemical shifts and ECD calculations. Anti-adipogenic activities of isolates were evaluated using 3T3-L1 preadipocytes with Oil Red O staining and quantitative real-time PCR (qPCR). Results: Phytochemical examination of the roots of Indian ginseng afforded to the isolation of six withanolides (1-6), including three novel withanolides, withasilolides GeI (1-3). All the six compounds inhibited adipogenesis and suppressed the enlargement of lipid droplets, compared to those of the control. Additionally, the mRNA expression levels of Fabp4 and Adipsin, the adipocyte markers decreased noticeably following treatment with 25 µM of 1-6. The active compounds (1-6) also promoted lipid metabolism by upregulating the expression of the lipolytic genes HSL and ATGL and downregulating the expression of the lipogenic gene SREBP1. Conclusion: The results of our experimental studies suggest that the withasilolides identified herein have anti-adipogenic potential and can be considered for the development of therapeutic strategies against adipogenesis in obesity. Our study also provides a mechanistic rationale for using Indian ginseng as a potential therapeutic agent against obesity and related metabolic diseases.

The Korean Traditional Anti-obesity drug Gyeongshingangjeehwan Stimulates $AMPK{\alpha}$ Activation in Skeletal Muscle of OLETF Rats

  • Our previous study demonstrated that the Korean traditional medicine Gyeongshingangjeehwan (GGEx) inhibits obesity and insulin resistance in obese type 2 diabetic Otsuka Long-Evans Tokushima Fatty (OLETF) rats. We investigated whether GGEx may affect AMP-activated protein kinase ${\alpha}$ ($AMPK{\alpha}$) since $AMPK{\alpha}$ activation is known to stimulate fatty acid oxidation in skeletal muscle of obese rodents. After OLETF rats were treated with GGEx, we studied the effects of GGEx on $AMPK{\alpha}$ and acetyl-CoA carboxylase (ACC) phosphorylation, and the expression of $AMPK{\alpha}$, $PPAR{\alpha}$, and $PPAR{\alpha}$ target genes. The effects of GGEx on mRNA expression of the above genes were also measured in C2C12 skeletal muscle cells. Administration of GGEx to OLETF rats for 8 weeks increased phosphorylation of $AMPK{\alpha}$ and ACC in skeletal muscle. GGEx also elevated skeletal muscle mRNA levels of $AMPK{\alpha}1$ and $AMPK{\alpha}2$ as well as $PPAR{\alpha}$ and its target genes. Consistent with the in vivo data, similar activation of genes was observed in GGEx-treated C2C12 cells. These results suggest that GGEx stimulates skeletal muscle $AMPK{\alpha}$ and $PPAR{\alpha}$ activation, leading to alleviation of obesity and related disorders.

Diet and Metabolic Disease Risk by Perceived Stress Level in Korean Adult Women (성인 여성에서 스트레스 인지 정도에 따른 식생활과 대사성 질환 위험)

  • Dietary components can modulate stress, inflammatory indicators, and health risk. This study examined the relationship among diet, metabolic disease risk, and perceived stress in Korean adult females using the 2017-2018 Korea National Health and Nutrition Examination Survey. A total of 4,353 adult women aged 19-64 years were classified into four groups according to perceived stress level: very high stress group (VHSG, n=225), high stress group (HSG, n=1,079), moderate stress group (MSG, n=2,532), and low stress group (LSG, n=517). Data collection included the sociodemographics, anthropometrics, blood profile, and dietary survey. After adjusting for covariates, those in the VHSG had a higher body mass index (p=0.013) and obesity rate (p=0.053) with a shorter sleep time than the LSG group. The VHSG also tended to have a higher plasma LDL-cholesterol, hsC-reactive protein and lower levels of HDL-cholesterol, vitamin A, and vitamin E than the low stress group. High stress subjects demonstrated increased breakfast skipping frequency (p<0.0001), decreased fiber intake (p=0.001), potassium (p=0.041), and vitamin A (p=0.011) than the low stress ones. Therefore the perceived stress level was associated with the inflammatory indicators, obesity, and lack of anti-inflammatory or antioxidant nutrients. The dietary components may be an important mediator of stress and metabolic disease.

Effect of Silybin on Body Weight and Glucose Tolerance in High-fat-diet Induced Obese Mice (고지방 식이로 유도된 비만 쥐에서 실리빈 (Silybin)이 체중 및 내당 능에 미치는 영향)

  • In this study, we investigated the effects of silybin on body weight and glucose tolerance in mice fed high fat diet mice. We found that body weight, plasma TG contents, fat size, glycerol, 3-hydroxybutyrate and total cholesterol were significantly decreased in silybin (500 mg/kg) supplemented groups compared to high fat diet group. Whereas, total food intake was not changed between high fat diet group and high fat diet plus silybin group. Futhermore, supplement of high fat elevated the glucose intolerance and was improved in silybin supplement group. Finally, we examined the effect of silybin on circulating adipocytokine level to explore the possible mechanism by which silybin improves high fat diet-induced obesity and diabetes. The silybin supplement significantly reduced the level of adipocytokine, such as leptin, resistin, IL-6, and MCP-1 induced by high fat diet. These results suggest that silybin can be used to improve obesity and diabetes.

Development of an anti-obesity dietary supplement inhibiting the digestion of carbohydrate and lipid (비만 개선 효과를 지닌 탄수화물 및 지방 흡수 억제 기능성 식이조성물 개발)

  • In a previous study, we developed a new food additive as an egg yolk antibody (IgY) against carbohydrate digestion enzymer for the regulation of blood glucose level and weight control. The IgY delayed and decreased the increment if blood glucose level after administration of sucrose in human being by 30% in 20∼30 min. We also developed a lipase inhibitor as a water extract of two kinds of herb, Platycodon grandiflorum and Solanum Melongena, Twenty three volunteers were subjected to the intake of the egg yolk IgY Plus the herbal extracts for 50 days. In average, the treated subjects appeared to lose 1.96 kg of body weight and 3.4 kg of body fat mass during the treated period. Furthermore, Panniculus adiposus and breech size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of egg yolk IgY and natural herbal extract improve the obesity by the decrement of body weight and body fat mass.

Evaluation for Anti-Inflammatory Activities of Rice and Rice Snack Extracts: An In-vitro Study (쌀과 쌀과자 추출물의 in-vitro test를 통한 항염증 활성 효과 평가)

  • Atopic dermatitis (AD) is usually caused by foods such as wheat, egg, milk, and peanuts, leading to common health problems in early childhood with complications like urtication. The aim of this study was to evaluate ethanol extracts of rice and rice snacks concentrated until the ethanol was completely eliminated and hot-air dried. In vitro analyses were carried out using murine macrophage RAW 264.7 cells. We measured cytotoxicity, nitric oxide (NO) production, and inflammatory cytokine level. The NO level of the cells exposed to lipopolysaccharide (LPS) was significantly reduced by rice and rice snack extracts. $TNF-{\alpha}$ level decreased in contrast to the LPS group, although a significant difference was not observed. On the other hand, IL-6 significantly decreased in both rice and rice snack extracts in a dose-dependent manner. The results of the present study suggest that rice and rice snack decreased NO and inflammatory cytokine levels. Therefore, rice could be useful as a raw material for relieving child atopic dermatitis caused by snacks made from wheat.
