Physicochemical and Functional Properties of Yeast-Fermented Cabbage

  • Microbial fermentation is often used to improve the functionality of plant-based food materials. Herein, we investigated changes in the physicochemical and functional properties of cabbage during yeast fermentation to develop new products using fermented cabbage. Among the 8 types of food-grade yeast, both Saccharomyces cerevisiae and Saccharomyces boulardii fermented 10% cabbage powder solution (w/w) the most effectively, leaving no soluble sugars after 12 h of fermentation. In addition, the yeast fermentation of cabbage resulted in functionally positive outcomes in terms of sulforaphane content, antioxidant properties, and anti-inflammatory activity. Specifically, the yeast-fermented cabbages contained about 500% more sulforaphane. The soluble fraction (5 ㎍/ml) of yeast-fermented cabbage had no cytotoxicity in murine RAW 264.7 cells, and the radical-scavenging capacity was equivalent to 1 ㎍/ml of ascorbic acid. Moreover, cabbage fermented with S. boulardii significantly suppressed both lipopolysaccharides (LPS)-induced nitric oxide production and LPS-induced reactive oxygen species production in RAW 264.7 cells, suggesting a potential anti-inflammatory effect. These results support the idea that yeast fermentation is promising for developing functionally improved cabbage products.

The Effect of Polyphenols Isolated from Cynanchi wilfordii Radix with Anti-inflammatory, Antioxidant, and Anti-bacterial Activity

  • Recently, Cynanchi wilfordii Radix has gained wide use in Asian countries as a functional food effective for relieving fatigue, osteoporosis, and constipation, particularly in menopausal disorders. However, its anti-inflammatory and anti-microbial activities have not been explored in detail to date. The anti-inflammatory, antioxidant, and anti-bacterial properties of the Cynanchi wilfordii Radix extracts obtained with water, methanol, ethanol, and acetone were compared. All 4 polyphenol-containing extracts exhibited anti-inflammatory and antioxidant effects. The ethanol extract was found to elicit the most potent reduction of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and cytokine (IL-$1{\beta}$, IL-6, IL-10, and TNF-${\alpha}$) levels, as well as inhibit the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in a concentration-dependent manner. The evaluation of antioxidant activity also revealed the ethanol extract to have the highest free radical scavenging activity, measured as $85.3{\pm}0.4%$, which is equivalent to 99.9% of the activity of ${\alpha}$ -tocopherol. In the assessment of anti-bacterial activity, only ethanol extract was found to inhibit the growth of the Bacillus species Bacillus cereus and Bacillus anthracis. These results show that polyphenols of Cynanchi wilfordii Radix have anti-inflammatory, antioxidant, and anti-bacterial properties that can be exploited and further improved for use as a supplementary functional food, in cosmetics, and for pharmaceutical purposes.

아까시 꽃 추출물을 첨가한 성게 껍질 필름의 제조 (Preparation of Sea Urchin Skeleton Film Containing Robinia pseudoacacia Flower Extract)

  • 본 연구에서는 폐기되는 성게 껍질을 이용한 생분해성 필름소재를 개발하고자 gelatin을 첨가한 복합필름 및 항산화, 항균성을 부여하기 위해 아까시 꽃 추출물(RFE)을 첨가한 필름을 제조하였다. 그 결과 gelatin 비율이 증가함에 따라 성게 껍질 필름의 인장강도가 증가하였고 신장률이 감소하였으며, 성게 껍질과 gelatin을 8:2 비율로 첨가하였을 때 최적의 물성을 보였다. 또한, 필름에 RFE를 첨가함에 따라 인장강도는 감소하였으나 신장률과 필름 투습도는 증가하였다. RFE 함유 성게 껍질 필름의 ABTS, DPPH 라디칼 소거능 측정과 디스크 확산법을 이용하여 항산화 및 항균 능력을 측정하였는데, RFE 농도가 증가함에 따라 항산화, 항균 능력이 향상하였다. 따라서 본 연구 결과, RFE를 첨가한 생분해 성게 껍질 필름은 적절한 물성과 함께 항산화, 항균 능력이 있기에 가공식품의 저장성을 증대시킬 수 있는 기능성 포장 소재로의 활용이 가능하다고 판단된다.

One-step phyto-mediated fabrication of silver nanoparticles and its anti-microbial properties

  • This manuscript describes the one-step eco-friendly green fabrication of silver nanoparticles (AgNPs) through the in-situ bio-reduction of an aqueous solution of silver nitrate using Syzygium aromaticum leaf extract. UV-vis spectroscopy shows a characteristic SPR peak around 442 nm. FTIR spectroscopy showed that the AgNPs were capped with bioactive phyto-molecules. TEM images revealed oval and spherical particles with a mean diameter of ~12.6 nm. XRD analysis revealed crystalline and face-cantered cubic AgNPs. The phytosynthesized AgNPs showed broad-spectrum anti-microbial activity against two foodborne pathogenic bacteria, Listeria monocytogenes and Staphylococcus aureus. The AgNPs showed a prominent ability to inhibit biofilms formed by L. monocytogenes and S. aureus in laboratory conditions through a crystal violet assay. The results suggest that the AgNPs could be a novel nanotool to develop effective antimicrobial and anti-biofilm agents in food preservation.

Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.

Elicitor 처리에 의한 삼백초 추출물(Saururus chinensis)의 미용식품활성 및 피부상재균 억제효과 (Functional cosmetic properties and antimicrobial activity of extracts from Saururus chinensis by elicitor treatment)

  • Elicitor를 처리한 삼백초의 물과 ethanol 추출물들의 기능성화장품활성을 살펴보았다. Elicitor 처리한 삼백초 추출물의 항산화 효과는 1.5 mg elicitor/leaf 1 g (group A)에서 3.0 mg elicitor/leaf 1 g (group B)으로 처리 농도가 높아질수록 항산화 효과는 높아지는 것으로 확인되었으며, elicitor 처리에 의해 물 추출물보다 70% ethanol 추출물의 항산화 효과가 더 우수한 효과를 나타내었다. Elicitor 처리한 삼백초 추출물의 미용식품활성은 elastase 억제 효과와 미백효과가 비처리군인 대조구와 유사한 효소 억제 효과를 나타내어 elicitor 처리가 삼백초 추출물의 미백 및 주름개선 효과의 증대에는 영향을 미치지 않는 것으로 확인되었으나, 염증 억제 효과가 비처리군인 대조구에 비해 우수한 염증 억제 효과를 나타내었으며, grpoup A와 group B 모두에서 대조구에 비해 억제율이 큰 폭으로 상승하는 효과를 나타내어 elicitor 처리가 유용물질의 산업화라는 전제를 충족시키기 위한 biomass 기법으로 매우 적당하다는 것을 시사해주었다. 따라서 삼백초 재배 시에 본 연구에서 사용한 elicitor를 처리하면 수확되는 삼백초의 단위 무게 당 생산량을 높일 수 있고, 생리활성도 증대시킬 수가 있어 산업화를 위한 원료생산에 효율적인 재배기법에 활용할 수 있을 것이라 판단되었다.

Estrogenic activity of Pomegranate extract in MCF7-ERE cells

  • Pomegranate, a small tree originating in Orient, belongs to Punicaceae family. The seeds contain an oil of which about 80% is rare trans 18 carbon fatty acid (punicic acid), and have highest botanical concentration of a sex steroid, estrone. Pharmacological properties of pomegranate extract have been studied, with anti-microbial, anti-parasitic, ant-viral, and anti-cancer effects. We have examined the estrogenic activity of the pomegranate extracts using MCP-7-ERE cells. MCF-7-ERE cells, stably transfected with pERE-Luc were treated various amount of pomegranate extract and after overnight treatments, luciferase activity were measured. Estradiol(E2) dose dependently induced luciferase activity in this cell and maximal response is obtained at 100pM E2.

Santalum album Linn wood and its oil: An aromatic Unani traditional medicine with versatile pharmacological activities

  • Santalum album Linn. [Family: Santalaceae] is commonly known as white sandalwood, sandal safaid and safed chandan. It is one of the most valuable trees and second costliest wood in the world. Sandalwood and its oil is extensively used in the Unani and other traditional systems of medicine as it has blood purifier, anti-inflammatory, analgesic, exhilarant, cardiotonic, antiseptic, nervine tonic and expectorant properties. It is used in skin, cardiac, liver, gastrointestinal, respiratory, integument and urogenital disorders. These uses are supported and proven by many in vitro or in vivo studies. The proven pharmacological activities of S. album are antimicrobial, anti-oxidant, anti-inflammatory, antimutagenic and anti-fatigue. The research has proven that sandal oil or its constituents have anti-microbial activity. Sandalwood oil showed skin cancer preventive effect in mice and its constituent alpha santalol showed the anticancer property. The methanolic extract of wood was confirmed for antioxidant, free radical scavenging, analgesic and anti-inflammatory activities. ${\alpha}$ and ${\beta}$ santalols present in sandal oil showed sedative effects. Sandalwood tea had a significant effect on heart muscles of frog and showed increased myocardial contractility. Its oil showed significant changes in hepatic xenobiotic metabolizing enzymes. Sandalwood oil and its major constituents showed less acute oral and dermal toxicity in laboratory animals. Hence, the aforementioned studies justify the uses of sandalwood and its oil mentioned in the classical Unani literature. However, further clinical trials are suggested to confirm its efficacy and safety in humans.

Fisetin Suppresses Macrophage-Mediated Inflammatory Responses by Blockade of Src and Syk

  • Flavonoids, such as fisetin (3,7,3',4'-tetrahydroxyflavone), are plant secondary metabolites. It has been reported that fisetin is able to perform numerous pharmacological roles including anti-inflammatory, anti-microbial, and anti-cancer activities; however, the exact anti-inflammatory mechanism of fisetin is not understood. In this study, the pharmacological action modes of fisetin in lipopolysaccharide (LPS)-stimulated macrophage-like cells were elucidated by using immunoblotting analysis, kinase assays, and an overexpression strategy. Fisetin diminished the release of nitric oxide (NO) and reduced the mRNA levels of inducible NO synthase (iNOS), tumor necrosis factor (TNF)-${\alpha}$, and cyclooxygenase (COX)-2 in LPS-stimulated RAW264.7 cells without displaying cytotoxicity. This compound also blocked the nuclear translocation of p65/nuclear factor (NF)-${\kappa}B$. In agreement, the upstream phosphorylation events for NF-${\kappa}B$ activation, composed of Src, Syk, and I${\kappa}B{\alpha}$, were also reduced by fisetin. The phospho-Src level, triggered by overexpression of wild-type Src, was also inhibited by fisetin. Therefore, these results strongly suggest that fisetin can be considered a bioactive immunomodulatory compound with anti-inflammatory properties through suppression of Src and Syk activities.

Anti-inflammatory, Antioxidant and Antimicrobial Effects of Artemisinin Extracts from Artemisia annua L.

  • The anti-inflammatory, antioxidant, and antimicrobial properties of artemisinin derived from water, methanol, ethanol, or acetone extracts of Artemisia annua L. were evaluated. All 4 artemisinin-containing extracts had anti-inflammatory effects. Of these, the acetone extract had the greatest inhibitory effect on lipopolysaccharide-induced nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and proinflammatory cytokine ($IL-1{\beta}$, IL-6, and IL-10) production. Antioxidant activity evaluations revealed that the ethanol extract had the highest free radical scavenging activity, ($91.0{\pm}3.2%$), similar to ${\alpha}$-tocopherol (99.9%). The extracts had antimicrobial activity against the periodontopathic microorganisms Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum subsp. animalis, Fusobacterium nucleatum subsp. polymorphum, and Prevotella intermedia. This study shows that Artemisia annua L. extracts contain anti-inflammatory, antioxidant, and antimicrobial substances and should be considered for use in pharmaceutical products for the treatment of dental diseases.
