Probiotic Property of Lactobacillus pentosus Miny-148 Isolated from Human Feces (인체분변으로부터 분리한 유산균 Lactobacillus pentosus Miny-148의 생균제 특성 연구)

  • Three hundred lactic acid bacteria isolated from human feces were studied their probiotic characters to develop potential probiotics. The properties were tested on the basis of guideline for probiotic selection protocol such as tolerance for acid or bile salt, thermal stability, antimicrobial, anticancer cell, and antiviral activity. Strain Miny-148 was selected as a potential probiotic bacterium which showed resistance to low pH, bile salts and thermal stability. On the basis of fatty acid profiles and 16S rDNA sequences analysis, the strain was identified as Lactobacillus pentosus (similarity 99.9%). The strain, L. pentosus Miny-148, showed broad antimicrobial spectrum against E. coli O157:H7, Shigella flexneri, Bacillus anthracis, Staphylococcus aureus, E. coli, Vibrio cholerae, V. vulnificus, Salmonella typhimurium, and Methicillin-resistant S. aureus (MRSA). Cell-free culture supernatant of the strain also inhibited against the growth of HT-29 colon cancer cell and transmissible gastroenterits virus.

The Early Detection of the Spore Using Sonication and Fluorescent Dye in the Field (현장에서 초음파 파쇄와 형광시약을 이용한 포자의 조기 탐지)

  • This study was carried out to establish the optimum condition of cell disruption with a sonicator for the detection of the spore, Bacillus anthracis ${\Delta}$-sterne for the purpose of developing automatic fluorometer. The efficiency of sonication on the ${\Delta}$-sterne spore disruption was very weak. The ${\Delta}$-sterne spore with zirconia bead showed greater disruption than the ${\Delta}$-sterne spore alone when sonificated. The volumn of the zirconia bead added in the spore solution has little effect on the disruption efficiency. The detection limit of the ${\Delta}$-sterne spore with zirconia bead and the ${\Delta}$-sterne spore alone was $10^6$ CFU/mL and $5{\times}10^7$ CFU/mL respectively, when sample was sonicated for 20 seconds with a sonicator probe of 13 mm diameter.

A review on the Pathogens and Diseases Associated with Biological weapons (생물무기로 사용된 병원균과 질병에 대한 고찰)

  • Recently, biological weapons (BWs) prepared with pathogenic microorganisms, toxins and biological vectors have been used maliciously for biological warfare, bioterrorism and/or agroterrorism by hostile countries and terrorists. In this review, historical background of disease and malicious use of BWs pathogenicity of microorganisms, advanced methodology involved in laboratory diagnosis, and prevention and control of anthrax (Bacillus anthracis), plague (Yersinia pseudotuberculosis subs. pestis), glanders (Burkholderia mallei), and smallpox (Variola virus) which have been abused for biological warfare or bioterrorism were discussed. In addition, the pathogenicity of microorganisms and the methodology needed to diagnose and control 6 diseases identified by WHO/CDC, ie., smallpox, inhalation anthrax, pneumonic plague, botulism, tularemia, and hemorrhagic fevers that would wreak havoc if terrorists successfully disseminated the germs by air were described.

  • On studying of microbes and appearance frequency which are floating around dwelling space and life environments in Seoul area, the results are summarized as follows ; 1. 1931 strains of 29 bacteria are isolated in dwelling spaces, Among them, there are Staphylococcus aureus, S. anthracis which are pathogenic to human beings. 2. In fungi, 76 strains of 13 species are isolated. That of highest frequency is confirmed to be Aspergillus which is pathogens of Aspergillosis (one of a group of diseases of animals and human being caused by various species of Aspergillus). The places, where the number of kinds and the frequency of appearances have been observed highest, are coffee shops, lunch counters and office rooms, while factory districts are of far lower level than expected. 3. 0.1% $HgCl_2$, Leasking oil, and Telephone disinfectant were better than any other in fungicidal effect.

  • Four bacterial strains having inducible resistance to erythromycin were isolated from soil samples in Korea and characterized. MLS inducibility was checked in each strain. Cloning of inducible resistance gene(s) has been tried. Four isolates were identified as B. anthracis, Micrococcus luteus, Streptococcus faecium and B. licheniformis, in which erythromycin, oleandomycin, cirramycin and carbomycin acted as resistance inducers respectively. The resistance gene cloned from B, licheniformis 597 strain using pBS 42 vector was found to have a 3.2 kb insert.

  • Edema factor(EF) is a portion of anthrax toxin which produces edema when combined with protective antigen. This paper describes about technique for cloning, expression, purification and activity test of EF. Using the E. coli expression system, we could make recombinant EF protein although it's origin is Bacillus anthracis. And also we could culture massively and purify highly pure protein. Finally we confirm a enzyme activity of purified EF to increase intracellular cAMP level. Through establishing this technique, it can be possible to research about EF in depth and apply to expression and purification of many other protein in biology.

  • The purpose of this study is to estimate the movement of microbial contaminant caused by bio-attack using bio-agent such as bacillus anthracis for preventing contaminant diffusion. multizone simulation was carried out in the case of three types of bio-attack scenario in the government building. Simulation results show that severe contaminant diffusion is brought about in all cases of bio-attack scenario in one hour, though pollution boundaries have different mode according to bio-attack scenarios. Simulation results also show that immune building technology such as filter and UVGI technology gives us powerful alternatives to meet the emergent situation caused by unexpected bio-attack.

Development and Validation Study of Biological Agent Detection Kit (생물학작용제 검출 키트 개발 및 성능시험 연구)

  • In biological warfare, it is important to identify biological agents for proper treatment. We focused on developing a real-time RT-PCR kit that can detect multiple species of biological agents. AccuPower(R) Biothreat Real-Time RT-PCR Kit(v3.0) could detect Bacillus anthracis, Yersinia pestis, Vibrio cholerae, Francisella tularensis, Salmonella typhi, Rickettsia prowazekii, Variola virus, Hantaan virus, Yellow fever virus, Brucella spp., Shigella dysenteriae in a single reaction. The results showed that the kit was verified to be able to detect at least 0.005 ng of nucleotide and 10,000 CFU/ml of bacteria. Therefore, the kit is expected to be used as a rapid and sensitive detection kit for 11 species of biological agents within 2 hours.
