나무딸기 Anthocyanin에 관한 연구 (Studies on Identification of the Anthocyanins in Raspberries)

  • 나무딸기 과즙색소(果汁色素)를 포화염기성 초산 납처리를 하여 색소(色素)를 추출(抽出)하고 polyvinylpyrroldione을 충전(充塡)시킨 column에 색소를 유입(流入)시켜 fraction collector로 분획(分劃)하여 4 개의 개별색소(個別色素)를 분리(分離), 진공건조(眞空乾燥) 시켜 건고(乾固)시켰으며 각(各) 개별색소에 대해 당(糖), aglycone 산부분(酸部分) 가수분해(加水分解), acyl기, 분광학적(分光學的)인 성질 등의 실험결과에 의하여 anthocyanin의 구조를 확인(確認)하였다. 개별색소 1 은 cyanidin-3, 5- digalactoside with caffeic acid 2 번은 cyanidin -3- digalactoside with caffeic acid 3 번은 cyanidin-3,5-diglucoside with caffeic acid 4 번은 cyanidin-3-monoglucoside with caffeic acid로 고정(固定)하였다.

  • Anthocyanins are water-soluble glycosides and acylglycosides of anthocyanidins, having different color variations due to its substitution patterns. Anthocyanins, present in various fruits, vegetables and crops as natural colorant, have been well characterized for its bioactive properties, anti-oxidant, anti-cancer, anti-proliferative and anti-inflammatory properties. During extraction and purification, the factors, such as pH, temperature, oxygen, light, enzymes, nucleophilic agents, sugar derivatives and co-pigments, have affected on anthocyanin stability. For this reason, the extraction method should be thoroughly checked for the qualitative/quantitative analysis of anthocyanin in particular plant material. To identify the optimum extraction method of cyanidin-3-glucoside, major anthocyanin of dark purple-colored grains, Oryza sativa cv. Heugjinjubyeo, Phaselous vulgaris, Phynchosia gngularis, Sesamum indium, Rhynchosia nulubilis and Lablab purpureus, reversed-phase HPLC analysis using solvent system of acetonitrile, methanol and water were accomplished.

Comparison of the Antioxidative Effects and Content of Anthocyanin and Phenolic Compounds in Different Varieties of Vitis vinifera Ethanol Extract

  • This study was a quantitative HPLC analysis of four anthocyanins and five phenolic compounds contained in the skins, vines and seeds of the Campbell Early, Muscat Bailey A and Neo Muscat grape varieties. In the phenolic analysis, the seeds of the Campbell Early were found to contain 1.9, 1.8 and 1.6 times higher quantities of gallic acid, catechin and epicatechin relative to other grape seeds. Three anthocyanins, cyanidin, peonidin and pelargonidin, were also found to be higher in the skins of the Campbell Early relative to other grape skins. Therefore, the Campbell Early is the most useful grape variety with regard to the extraction of these six compounds from these grape seeds and skins. The free radical scavenging effects of grape seeds were also compared, and the results indicated that the Campbell Early seeds were most effective among them.

The Anthocyanin Components and Cytotoxic Activity of Syzygium cumini (L.) Fruits Growing in Egypt

  • Four anthocyanins were isolated from the acidic alcoholic extract of Syzygium cumini fruits growing in Egypt: Pelargonidin-3-O-glucoside, pelargonidin-3,5-O-diglucoside, cyanidin-3-O-malonyl glucoside, and delphenidin-3-O-glucoside. They were identified by the chromatographic, TLC and PC, and spectral analyses, UV, $^1$H-NMR and FAB/MS. The fruits were found to contain 0.03 gm % anthocyanins calculated on fresh weight basis calculated by spectrophotometric assay. Cytotoxic activity of total alcoholic extract of the fruits was performed against several types of tumor cell lines using the SRB assay. The tested extract exhibited significant cytotoxic activity for MCF7 (breast carcinoma cell line) (IC$_{50}$= 5.9 ${\mu}$g/mL), while the IC$_{50}$ was > 10 ${\mu}$g/mL for both Hela (Cervix carcinoma cell line), HEPG2 (liver carcinoma cell line), H460 (Lung carcinoma cell line) and U251 (Brain carcinoma cell line).

  • Though protein transduction domains (PTDs) are well known for the delivery of exogenous therapeutic proteins into living cells, the overall low efficiency of transduction is a serious obstacle. We investigated the effect of bog blueberry anthocyanins (BBA) on protein transduction efficiency and found that BBA enhanced the transduction efficiencies of Tat-SOD fusion protein into HeLa cells and mice skin. The enzymatic activities in the cells and skin tissue in the presence of BBA were markedly increased compared to controls. Further, BBA did not demonstrate any cell toxicity at various concentrations. Although the mechanism is not fully understood, we suggest that BBA might alter the conformation of the membrane, which would indicate that BBA can be used as a protein transduction enhancer for the efficient delivery of therapeutic proteins for a variety of disorders.

Rapid Isolation of Cyanidin 3-Glucoside and Peonidin 3-Glucoside from Black Rice (Oryza sativa) Using High-Performance Countercurrent Chromatography and Reversed-Phase Column Chromatography

  • Anthocyanins are water soluble plant pigments which are responsible for the blue, red, pink, violet colors in several plant organs such as flowers, fruits, leaves and roots. In recent years, anthocyanin-rich foods have been favored as dietary supplements and health care products due to diverse biological activities of anthocyanins including antioxidant, anti-allergic, anti-diabetic, anti-microbial, anti-cancer and preventing cardiovascular disease. High-performance countercurrent chromatography (HPCCC) coupled with reversed-phase medium pressure liquid chromatography (RP MPLC) method was applied for the rapid and efficient isolation of cyanidin 3-glucoside (C3G) and peonidin 3-glucoside (P3G) from black rice (Oryza sativa L., Poaceae). The crude black rice extract (500 mg) was subjected to HPCCC using two-phase solvent system composed of tert-butyl methyl ether/n-butanol/ acetonitrile/0.01% trifluoroacetic acid (TBME/B/A/0.01% TFA, 1 : 3 : 1 : 5, v/v, flow rate - 4.5 mL/min, reversed phase mode) to give enriched anthocyanin extract (37.4 mg), and enriched anthocyanin extract was sequentially chromatographed on RP-MPLC to yield C3G (16.5 mg) and P3G (8.7 mg). The recovery rate and purity of isolated C3G were 76.0% and 98.2%, respectively, and those of P3G were 58.3% and 96.3%, respectively. The present study indicates that HPCCC coupled with RP-MPLC method is more rapid and efficient than multi-step conventional column chromatography for the separation of anthocyanins.

산성처리가 애기장대의 엽록소, 카로티노이드, 안토시아닌 등의 색소 함량에 미치는 영향 (Effects of Acid Treatments on Chlorophyll, Carotenoid and Anthocyanin Contents in Arabidopsis)

  • 산성비 수준의 고농도 수소이온 스트레스인 pH 5.6-4.0 처리는 애기장대 생장에 크게 해를 주지 않았으며 강산성인 pH 3.5 이하에서 수소이온 농도가 높아질수록 식물 뿌리 및 줄기의 생장이 위축되었으며 특히 뿌리가 줄기보다 더 민감하게 반응하였다. 엽록소 함량은 pH 5.6-4.0까지 pH가 낮아질수확 증가히였다. pH 4.0에서 내려갈수록 점진적으로 엽록소 함량이 감소되는 경향을 보였으며 카로티노이드도 엽록소와 같은 경향을 보였으나 pH 3.5에서 함량이 가장 높았다. 안토시아닌은 pH가 낮아질 수록 계속 증가하여 pH 3.0에서 가장 많이 축적되었으며 pH 2.5에서는 급격히 감소되었다. 스트레스 강도가 세어 질수록 엽록소 대비 식물체내 카혹티노이드 및 안토시아닌 함량이 증가한다는 것은 엽록소를 보호하기 위한 식물의 생리적 반응으로 여겨진다. 그러나 안토시아닌과 카로티노이드는 수소이온 농도에 따른 식물체내 함량의 변화가 상이하여서 고수소이온 스트레스에서 식물의 보호하는 기작이 다를 것으로 에상된다.
