Changes of Anthocyanidin, Growth Characteristics and Brown Rice Yield of Red Colored Rice at Two Region of Eastern Coast in Gyeongsangbuk-do Province

  • The study was carried out to determine the changes of growth characteristics, anthocyanidin, and brown rice yield grown at coast and inland areas apart from the east coastline of Gyeongsangbuk-do province. Number of spikelets per panicle was much more increased in inland area. Regional difference in number of spikelets per panicle was observed in Jeongjinju cultivar. Among these red rice cultivars, the highest brown rice yield was Jeongjinju rice cultivar having 702 kg in inland area and 692 ㎏ in coast area, respectively. Anthocyanidin content ranged 524 to $610{\mu}g/g$ dry weight basis. Cyanidin content was 11.4 to 14.0 times higher than that of delphinidin under coast and inland area. Anthocyanidin content was higher in rice cultivar grown at coast area. Highest head brown rice rate was only observed in Geonganghongmi to 95.2 at coast area and 95.4 inland area. In considering brown rice yield and pigment content, Hongjinju rice cultivar was recommended in optimal pigment rice cultivar in eastern coast of Gyeongsangbuk-do Province.

오디의 안토시아니딘에 구리를 이용한 착화합물 합성 및 상주실크에 천연염색 (Analysis of Anthocyanidin in Mulberry of Cu Complex Compound and Natural Dyeing on Sangju Silk)

  • This study explores the solution of a variety of conditions of the natural dyeing with the Mulberry. To obtain this, the Cu complex compound was added into the extracts. The Mulberry extract was added the Copper acetate to obtain the Black sediment the metal complex compound of anthocyanidin. According to the result of using the complex compound of the solid material, the dyeing properties were excellent. The metal complex compound acted as a mordant, thus it reduced the process of mordanting in the natural dying.

천연 색소인 안토시아니딘 기반의 인쇄형 시간-온도이력 지시계 개발 (A Novel Printable Time-temperature Integrator with Anthocyanidin, a Natural Coloring Matter)

  • 인쇄형 TTI의 잉크로 천연물질인 안토시아닌의 적용 가능성을 확인하기 위해 실제 인쇄형 TTI를 제작하여 색 변화를 확인하였다. 안토시아닌은 반응속도가 느려 TTI에 적용이 어렵다. 이를 해결하기 위해 ${\beta}$-글루코시다아제를 이용하여 안토시아닌의 탈당화를 유도하여 보다 불안정한 안토시아니딘 잉크를 제작하였다. 그리고 인쇄형 TTI로 제작이 적합한지 확인하기 위해 인쇄적성을 확인하고 적용가능한 식품군을 탐색하였다. 실험결과, 인쇄적성의 경우, 틱소트로피의 히스테리시스 성질을 나타내어 인쇄적성이 적합한 것으로 확인되었다. 색 변화의 경우, 같은 온도와 pH 조건 하에서 안토시아닌 TTI의 색 변화 속도보다 안토시아니딘 TTI의 색 변화속도가 빠른 것을 확인할 수 있었다. 또한, 안토시아닌 인쇄형 TTI의 활성화 에너지는 65.21 kJ/mol, 안토시아니딘 인쇄형 TTI의 활성화 에너지는 86.92 kJ/mol로 확인되었다. 이는 ${\beta}$-글루코시다아제를 처리한 안토시아닌의 탈당화가 안토시아닌의 반응속도를 빠르게 할 뿐만 아니라, 활성화 에너지도 증가시킨 것으로 사료된다. TTI의 활성화 에너지와 식품의 활성화 에너지를 비교한 결과, 안토시아닌 TTI는 냉장 육제품에 한정적으로 적용 가능하였다. 반면에 안토시아니딘 TTI는 냉장 육제품과 냉장 어류에 적용 가능하였다. 하지만, 안토시아닌 TTI는 색 변화 종말점에 도달하는 시간이 길기 때문에 냉장 육제품에 적용이 어려울 것으로 사료된다. 반면 안토시아니딘 TTI의 경우, 색 변화 종말점에 도달하는 시간이 짧기 때문에 냉장 육제품과 냉장 어류에 적용이 가능할 것으로 사료된다.

Changes of Anthocyanidin Content and Brown Rice Yield in Three Pigmented Rice Varieties Among Different Transplanting and Harvesting Times

  • Anthocyanin pigments from three pigmented rice varieties, Hongjinju, Sintoheugmi and Josaengheugchal pigmented by black color, were quantified to evaluate effects of transplanting and harvesting time during seed development. Hongjinju, a rice variety of grains pigmented by red color, contains only two kinds of anthocyanins, delphinidin and cyanidin. Meanwhile, delphinidin, cyanidin, and pelargonidin were identified in Sintoheugmi and Josaengheugchal. Anthocyanidin contents in pigmented rice variety Hongjinju under different planting times were decreased with prolonged harvesting times. The Sintoheukmi showed that cyanidin content was the highest among analyzed pigments during seed development. Two anthocyanins (cyanidin and pelargonidin) in Josaengheugchal recorded highest contents at 20 days after heading in three transplanting times. Cyanidin content was also slightly increased with prolonged transplanting time. Total anthocyanidin contents in Hongjinju were always lower than that of Sintoheukmi and Josaengheukchal. In two rice varieties, Sintoheugmi and Josaengheugchal, $a^*$ values were commonly slightly decreased by late transplanting and harvesting times. Changes of $a^*$ and $b^*$ values in waxy rice varieties, Sintoheugmi and Josaengheugchal varied more than those in Hongjinju. In Josaengheugchal, early harvesting before maturity showed lower brown rice yield and late harvesting in the each transplanting times resulted in highest brown rice yield.

오디 추출액을 이용한 상주실크의 천연염색 (Natural Dyeing of Sangju Silk with Mulberry Extract Solution)

  • Natural dyeing of Sangju silk for graveclothes with mulberry extract solution was studied. The anthocyanidin dye in the extracted mulberry solution was markedly influenced by pH condition, which resulted in the changed maximum light absorption from a $\lambda$max of 525nm in acidic condition to 380nm in alkaline condition possibly due to the irreversible transformation of anthocyanidin to cyanidin form of the dye. The color fastness properties to both light and washing were good when the fabrics were dyed at $80^{\circ}C$ with the mulberry-extracted solution upto twice extraction.

고구마 끝순 및 괴근의 안토시아니딘 추출 조건 최적화 (Optimization of anthocyanidin extraction conditions from sweet potato tips and tuber)

  • 본 연구에서는 고구마 끝순과 괴근으로부터 자색색소인 안토시아니딘을 추출하기 위한 최적조건을 확립하였다. 에탄올 농도(10-90%), citric acid 농도(0.1-0.5%) 그리고 추출온도(30-70℃)를 변수로 중심합성계획법으로 설계하고 반응표면분석법으로 최적화 하였다. 세가지 변수 모두 안토시아니딘 추출에 유의적인 영향을 주었으며, 산 농도나 추출 온도보다는 에탄올 농도의 영향력이 컸다. 각각의 추출조건 중 에탄올 농도 50%, citric acid 농도 0.3%, 추출 온도 50℃에서 끝순은 26.58±0.07 mg/100 g을 나타내었고 괴근은 117.32±0.39 mg/100 g으로 가장 높았다. 반응표면분석에 의한 최적추출조건은 끝순은 에탄올 농도 56.10%, citric acid 농도 0.25% 그리고 추출 온도 52.24℃이었으며, 괴근은 각각 55.08, 0.30, 53.06℃이었다. 최적추출조건에서의 예측값은 각각 27.45, 119.61 mg/100 g이었으며, 실측값은 각각 27.09±0.11, 118.72±0.67 mg/100 g이었다. 예측값과 비교할 때 각각 98.69, 99.26%로 나타나 추출조건의 유효성이 확인되었다.

Identification of Anthocyanin from The Extract of Soybean Seedcoat

  • Anthocyanins are naturally occuring phytochemicals and the main components of the coloring of plants, flowers and fruits. They are known to elicit antioxidative, anti-inflammatory and cancer preventive activity. In this study, we investigated anthocyanins in black / yellow soybean seedcoats using different methods of detection - thin layer chromatography (TLC), capillary zone electrophoresis (CZE) and HPLC analysis. The anthocyanins in soybean seedcoats were extracted by five independent methods of extraction and the aglycons (anthocyanidins) of the corresponding anthocyanins were prepared by acid mediated hydrolysis. The anthocyanin / anthocyanidin in black soybean seedcoat showed characteristic TLC mobility, CZE electrophoretic retention and HPLC migration time while little of anthocyanins were detected from yellow soybean seedcoat. The extracted anthocyanins showed pH dependent retention time in CZE and spectral change in UV-Vis spectrum. HPLC analysis of the hydrolyzed extract of black soybean seedcoat identified the presence of four anthocyanidins. The major anthocyanin in black soybean seedcoat was cyanin (cyanidin-3-O-glucoside), with the relative order of anthocyanidin in cyanidin > delphinidin > petunidin > pelargonidin.

Molecular Cloning and Function Analysis of an Anthocyanidin Synthase Gene from Ginkgo biloba, and Its Expression in Abiotic Stress Responses

  • Anthocyanidin synthase (ANS, leucoanthocyanidin oxygenase), a 2-oxoglutarate iron-dependent oxygenase, catalyzed the penultimate step in the biosynthesis of the anthocyanin class of flavonoids, from the colorless leucoanthocyanidins to the colored anthocyanidins. The full-length cDNA and genomic DNA sequences of ANS gene (designated as GbANS) were isolated from Ginkgo biloba for the first time. The full-length cDNA of GbANS contained a 1062-bp open reading frame (ORF) encoding a 354-amino-acid protein. The genomic DNA analysis showed that GbANS gene had three exons and two introns. The deduced GbANS protein showed high identities to other plant ANSs. The conserved amino acids (H-X-D) ligating ferrous iron and residues (R-X-S) participating in 2-oxoglutarate binding were found in GbANS at the similar positions like other ANSs. Southern blot analysis indicated that GbANS belonged to a multi-gene family. The expression analysis by real-time PCR showed that GbANS expressed in a tissue-specific manner in G. biloba. GbANS was also found to be up-regulated by all of the six tested abiotic stresses, UV-B, abscisic acid, sucrose, salicylic acid, cold and ethylene, consistent with the promoter region analysis of GbANS. The recombinant protein was successfully expressed in E. coli strain with pET-28a vector. The in vitro enzyme activity assay by HPLC indicated that recombinant GbANS protein could catalyze the formation the cyanidin from leucocyanidin and conversion of dihydroquercetin to quercetin, suggesting GbANS is a bifunctional enzyme within the anthocyanidin and flavonol biosynthetic pathway.

  • The tallest culm length was observed in Geonganghongmi cultivar grown in coast area, and culm length was always taller in coast area than that of inland area among these three red rice cultivars. However, number of tiller per rice plant was not affected by different cultural area. Number of spikelets per panicle was much more increased in inland area. Regional difference in number of spikelets per panicle was only observed in Jeongjinju cultivar. Among these red rice cultivars, the highest brown rice yield was Jeongjinju rice cultivar having 702kg in inland area and 692kg in coast area, respectively. The $L^*$ values, which expresses the brightness, were in the range of 36.3 ~ 36.7, 36.7 ~ 37.0, and 38.7 ~ 39.6 in Jeogjinju, Hongjinju, Geonganghongmi rice cultivars, respectively. The $a^*$ values, which expresses the redness positively and greenness negatively were in the range of 12.3 ~ 12.8, 12.2 ~ 12.3, and 12.1 ~ 11.9 in Jeogjinju, Hongjinju, Geonganghongmi rice cultivars, respectively. In addition, the b values, which expresses the yellowness positively and blueness negatively were in the range of 13.8 ~ 14.0, 13.6 ~ 13.5, and 14.9 ~ 14.7 in Jeogjinju, Hongjinju, Geonganghongmi rice cultivars, respectively. Anthocyanidin content ranged 524 to $610{\mu}g/g$ dry weight basis. Cyanidin content was 11.4 to 14.0 times higher than that of delphinidin under coast and inland area. Among these rice cultivars, anthocyanidin content was always higher in rice cultivar grown at coast area. The lowest protein content was measured in Jeogjinju and amylose content was relatively increased in inland area compared to that of coast area. Consequently, in considered with brown rice yield and pigment content, Hongjinju rice cultivar was recommended in optimal pigment rice cultivar in eastern coast of Gyeongsangbuk-do Province.
