Sol-Gel Encapsulation as Matrix for Potentiometric Nitrite-Selective Membranes Doped with Chloro (5, 10, 15, 20-Tetraphenylporphyrinato) Cobalt (III)

  • Organic-inorganic hybrid sol-gel matrices were used as hosts for chloro (5, 10, 15, 20-tetraphenylporphyrinato) cobalt (III) (Co[TPP]Cl), a known ionophore for nitrite. The sol-gel precursor was prepared by the reaction of (3-isocyanopropyl) triethoxysilane with 1,4-butanediol. An appropriate amount of the anion-exchanger, tridodecylmethylammonium chloride (TDMAC) and the plasticizer, tributylphosphate (DBP) were used as membrane additives. On mixing with an acidic catalyst, the sol-state precursors slowly gelled, yielding a membrane in which the active components, Co[TPP]Cl and TDMAC, were encapsulated. The performances of the sol-gel membrane-based electrodes were compared to those of Co[TPP]Cl-based poly(vinyl chloride) (PVC) membrane electrodes. Membranes with a molar ratio of Co[TPP]Cl: TDMAC (1 : 0.1) showed reasonable response slopes toward nitrite. The response slopes were typically 53 mV/decade between $10^{-5.4}$ and $10^{-1.0}\;M$. Selectivities toward nitrite over hydrophilic and small anions such as chloride were somewhat inferior to those observed with PVC-based membranes, but selectivities over lipophilic anions were quite similar. Reduced asymmetry potentials due to protein adsorption were found to occur with the sol-gel matrix relative to PVC-based films when the sensors were employed as a detector in flow-through configuration.

  • 해안가 및 폐염전 등에 서식하며 각종 미네랄, 아미노산 및 유용 생리활성 물질을 함유하고 있는 함초로부터 혈중 동맥경화 및 심장 질환 예방에 유효한 효능을 보일 것으로 예상되는 성분인 betaine을 HPLC를 이용하여 분석하였다. 함초는 양쪽성 물질로 같이 존재하는 다른 이온성 물질들에 의해 방해를 받으며 UV 흡수가 매우 낮아 일반적인 정량방법으로는 분석이 매우 까다롭다. 따라서 본 연구에서는 이러한 문제를 해소하기 위하여 2단계 이온교환수지 컬럼을 이용하여 방해물질을 제거한 후 betaine을 4-bromophenacyl bromide (PBPB)로 유도체화시켜 UV-labelling 한 후 분석하였다. 이온교환수지를 통과한 함초추출물의 회수율은 83.6%이며 함초추출액 mL당 4.85mg의 betaine이 함유되어 있는 것으로 나타났다.

유로키나제 분리를 위한 아민화 HPP-g-GMA 중공사 이온교환체의 합성에 관한 연구 (Synthesis of Aminated Hollow PP-g-GMA Fibrous Ion-Exchanger for Separation of Urokinase)

  • E-beam 전조사법을 이용하여 HPP-g-GMA 공중합체와 아민화 반응을 통한 아민화 HPP-g-GMA 이온교환체를 합성하였다. 그라프트율은 GMA 단량체 농도가 증가함에 따라 증가하였으며 GMA 단량체 농도가 1.46 M에서 그라프트율이 130%로 최대를 나타냈다. 아민화율은 그라프트율이 증가함에 따라 증가하는 경향을 나타내었으며, 그라프트율이 100%일 때 37.4%로 최대값을 나타내었다. 아민화 HPP-g-GMA 섬유이온교환체의 이온교환용량은 약 3.78 meq/g으로써 흡착 성능이 매우 우수한 소재임을 확인하였다. BET 분석결과 아민화 HPP-g-GMA의 비표면적은 54.83 $\m^2/g$, 기공크기는 $26\AA$으로 반응전보다 비표면적은 감소하였고 기공크기는 약간 증가하는 경향을 보였다. 또한 SEM 분석 결과, 반응 후 섬유의 두께가 굵어짐을 관찰하였으며 기공 막힘현상이 관찰되지 않았으며 이로부터 본 연구에서 합성한 섬유이온교환체가 음이온 흡착.분리에 적합함을 확인하였다.

음이온 교환수지에서의 희토류-EDTA 용리액에 의한 희토류 원소의 분리 (Separation of Rare Earth Elements Using Ln-EDTA Eluent in an Anion Exchanger)

  • $Cl^-$형 음이온 교환수지에서 희토류-EDTA 용리액을 사용하여 희토류 원소 혼합물 중의 원하는 한 원소를 분리 회수하는 연구를 하였다. 음이온 교환수지에 $Sm^{3+}$, $La^{3+}$, $Ce^{4+}$의 EDTA-착물을 흡착시키고, Sm-EDTA 용액으로 용리시키면, 수지에 흡착된 희토류 원소 중 $Sm^{3+}$ 외의 희토류 원소는 용리된다. 수지에 남아 있는 $Sm^{3+}$는 단계적으로 1M HCl 용액으로 용리시켜 분리하였다. Sm-EDTA 용액 대신 La-EDTA 용액으로 용리시키면 $Sm^{3+}$$Ce^{4+}$$La^{3+}$과 같이 용리되어 완전한 분리가 되지 않았고, Ce-EDTA 용액으로 용리시킬 때도 분리가 불가능하였다. Sm-EDTA 용액으로 용리시킬 때 용리 구조는 다음과 같다. 흡착과정 : $RCl+Ln-Y^-{\leftrightarrows}RLnY+Cl^-$, Sm-EDTA 용리 : $RLnY+Sm-Y^-{\leftrightarrows}RSmY+Ln-Y^-$, HCl 용리 : $RSmY+HCl{\leftrightarrows}RCl+Sm-Y^-$.

Altered Regulation of Renal Acid Base Transporters in Response to Ammonium Chloride Loading in Rats

  • The role of the kidney in combating metabolic acidosis has been a subject of considerable interest for many years. The present study was aimed to determine whether there is an altered regulation of renal acid base transporters in acute and chronic acid loading. Male Sprague-Dawley rats were used. Metabolic acidosis was induced by administration of $NH_4Cl$ for 2 days (acute) and for 7days (chronic). The serum and urinary pH and bicarbonate were measured. The protein expression of renal acid base transporters [type 3 $Na^+/H^+$ exchanger (NHE3), type 1 $Na^+/{HCO_3}^-$ cotransporter (NBC1), Na-$K^+$ ATPase, $H^+$-ATPase, anion exchanger-1 (AE-1)] was measured by semiquantitative immunoblotting. Serum bicarbonate and pH were decreased in acute acid loading rats compared with controls. Accordingly, urinary pH decreased. The protein expression of NHE3, $H^+$-ATPase, AE-1 and NBC1 was not changed. In chronic acid loading rats, serum bicarbonate and pH were not changed, while urinary pH was decreased compared with controls. The protein expression of NHE3, $H^+$-ATPase was increased in the renal cortex of chronic acid loading rats. These results suggest that unaltered expression of acid transporters combined with acute acid loading may contribute to the development of acidosis. The subsequent increased expression of NHE3, $H^+$-ATPase in the kidney may play a role in promoting acid excretion in the later stage of acid loading, which counteract the development of metabolic acidosis.

아민화 PP-g-AAc 초극세 이온교환섬유의 산성가스(SOx, NOx) 흡착거동 (Behaviour of Acidic Gases(SOx, NOx) Adsorption on Aminated PP-g-AAc Ultrafine Fibrous Ion Exchanger)

  • 본 연구는 아민화 polypropylene grafted acrylic acid(이하 PF-g-AAc로 표기) 초극세 이온교환섬유의 $SO_2$, $NO_2$에 대한 흡착거동을 고찰하였다. $SO_2$에 대한 흡착량은 초기농도가 높을수록 증가하였으며 농도가 낮을수록 흡착파과 시간이 짧아졌다. 흡착파과 평형은 60분 이내에 일어났으며 초기농도 100 ppm 이하에서는 약 80%정도 흡착이 이루어졌고, $SO_2$ 농도 100 ppm 이상에서 90% 이상 흡착되었다. $NO_2$의 흡착량은 $SO_2$에 비해 낮은 선택흡착성을 나타내었다. 한편, 유속이 증가함에 따라 $SO_2$의 흡착률은 낮아졌으며 60분 이내에 흡착파과 평형에 도달하였고, $NO_2$의 흡착량은 60%로 $SO_2$에 비해 낮았다. 함수율 250 mL/g에서 $SO_2$의 흡착량은 92%이었다. 아민화 PP-g-AAc 초극세 이온교환섬유의 $SO_2$등온흡착모델은 Freundlich 모델보다 Langmuir 모델에 근접하였다.

Trioctylamine을 이용한 뇨중 크롬의 원자흡수 분광분석에 관한 연구 (Studies on Atomic Absorption Spectrophotometric Analysis of Chromium in Urine by Trioctylamine)

  • As chromium in urine remains as oxychromic acid ions, chromium was extracted by using ion pair formed by anion exchanger trioctylamine. then after it is noted whether this TOA-MIBK method is effective or not for the chromium analysis by using flame method of Atomic absorption Spectrophotometric Analysis. The result is as following. 1. Effects of various acids on the extraction of Cr with MIBK including TOA are good in order $NHO_3$, $H_2SO_4$, HCl in distilled water sample and its proper concentration of HCl is 0.2 N. 2. For the analysis of urine sample, the best result can be achieved by following condition. After finished pretreatment adjusted to pH 6.5-7.5 by NaOH and again controlled pH 0.5-0.6 by HCl. 3. Though TOA concentration slightly affects the analytic value, best result is noted in 1-3% concentration. 4. Recovery rates of urine samples made by $0.3mg/l{\cdot}urine$, $0.6mg/l{\cdot}urine$, $0.9mg/l{\cdot}urine$ are shown from 96.7% to 104.8%. 5. Recovery rates of urine samples made by $0.01mg/l{\cdot}urine$, $10.03mg/{\cdot}urine$, $0.05mg/l{\cdot}urine$ are shown from 89.3% to 98.6%.

Distribution of Chitinases in Rice (Oryza sativa L)Seed and Characterization of a Hull-Specific Chitinase

  • The uneven distribution of acidic and basic chitinases in different parts of rice seed, and also the characterization of hull-specific chitinases, are reported here. After extraction of chitinases from polished rice, bran, and rice hulls, the chitinases were separated into acidic and basic fractions, according to their behavior on an anion exchanger column. Both fractions from different parts of rice seed showed characteristic activity bands on SDS-PAGE that contained 0.01% glycol chitin. The basic chitinases from rice hulls were further purified using chitin affinity chromatography. The chitinase, specific to rice hulls (RHBC), was 88-fold purified with a 1.3% yield. RHBC has an apparent molecular weight of 22.2 kDa on SDS-PAGE. The optimal pH and temperature were 4.0 and $35^{\circ}C$, respectively. With [$^3H$]chitin as a substrate, RHBC has $V_{max}$ of 13.51 mg/mg protein/hr and $K_m$ of 1.36 mg/ml. This enzyme was an endochitinase devoid of ${\beta}$-1,3-glucanase, lysozyme, and chitosanase activities.

Partial Purification and Properties of a Cysteine Protease from Citrus Red Mite Panonychus citri

  • Several studies have reported that the citrus red mites Panonychus citri were an important allergen of citrus-cultivating farmers in Jeju Island. The aim of the present study was to purify and assess properties of a cysteine protease from the mites acting as a potentially pathogenic factor to citrus-cultivating farmers. A cysteine protease was purified using column chromatography of Mono Q anion exchanger and Superdex 200 HR gel filtration. It was estimated to be 46 kDa by gel filtration column chromatography and consisted of 2 polypeptides, at least. Cysteine protease inhibitors, such as trans poxy-succinyl-L-leucyl-amido (4-guanidino) butane (E-64) and iodoacetic acid (IAA) totally inhibited the enzyme activities, whereas serine or metalloprotease inhibitors did not affect the activities. In addition, the purified enzyme degraded human IgG, collagen, and fibronectin, but not egg albumin. From these results, the cysteine protease of the mites might be involved in the pathogenesis such as tissue destruction and penetration instead of nutrient digestion.

Partial Purification and Characterization of a Cysteine Protease Inhibitor from the Plerocercoid of Spirometra erinacei

  • Helminthic cysteine proteases are well known to play critical roles in tissue invasion, nutrient uptake, and immune evasion of the parasites. In the same manner, the sparganum, the plerocercoid of Spirometra mansoni, is also known to secrete a large amount of cysteine proteases. However, cysteine protease inhibitors regulating the proteolytic activities of the cysteine protease are poorly illustrated. In this regard, we partially purified an endogenous cysteine protease inhibitor from spargana and characterized its biochemical properties. The cysteine protease inhibitor was purified by sequential chromatographies using Resource Q anion exchanger and Superdex 200 HR gel filtration from crude extracts of spargana. The molecular weight of the purified protein was estimated to be about 11 kD on SDS-PAGE. It was able to inhibit papain and 27 kDa cysteine protease of spargana with the ratio of 25.7% and 49.1%, respectively, while did not inhibit chymotrypsin. This finding suggests that the cysteine protease inhibitor of spargana may be involved in regulation of endogenous cysteine proteases of the parasite, rather than interact with cysteine proteases from their hosts.
