Expression of Neurotrophin 4 and Its Receptor Tyrosine Kinase B in Reproductive Tissues during the Follicular and Luteal Phases in Cows

  • The neurotrophins, required for the survival and differentiation of the nervous system, are known to be important for the development of the reproductive tissues. However, the signals initiating the growth of follicles, gamete development, and transport and the development of zygote in the reproductive system of cows remain ambiguous. The purpose of the present study was to identify the transcripts and proteins of Neurotrophin 4 (NT4) and its receptor tyrosine kinase B (TrkB) in bovine reproductive tissues. The transcripts and immunoreactivity of NT4 and TrkB proteins were detected by reverse transcription polymerase chain reaction and western blot analysis. Using immunohistochemistry, the specific immunoreactivity of NT4 and TrkB were detected in the oocytes of primordial follicles and in the growing primary follicles. The NT4 and TrkB immunoreactivity was predominantly observed in granulosa cells, cumulus granulosa cells, cumulus oocyte complexes, theca cells of mature follicles, as well as in the oviduct epithelial cells, uterine gland cell, and epithelium cells of the uterus during the follicular and luteal phases in cows. Expressions of NT4 and TrkB mRNAs were not significantly different among the ovary, oviduct, and uterus of the follicular phase. For the luteal phase, the expression of NT4 mRNA in the ovary was significantly higher than that in the oviduct and uterus, and the expression of TrkB mRNA in the oviduct was significantly higher than that in the ovary and uterus, as determined by fluorescence quantitative reverse transcription polymerase chain reaction. The expression of NT4 mRNA was significantly higher than that of TrkB mRNA in the ovary and uterus, whereas NT4 mRNA expression was lower than that of TrkB mRNA in the oviduct during the luteal phase. The present study hypothesizes that NT4 participates in the regulation of both gonads and extra-gonadal reproductive tissues in cows.

Effect of acute heat stress on feed intake and plasma concentrations of tumor necrosis factor-α and free amino acids in growing pigs

  • Exposure to heat stress (HS) has negative effects on pig production and health. Plasma concentrations of amino acids (AAs) can be used as indicators of HS. Therefore, this study was conducted to evaluate the effect of acute HS on feed intake, water drink, and plasma AAs in pigs. A total of 6 growing pigs (n = 6, 3 boars and 3 gilts) were raised in thermal neutral (TN; 25℃) conditions for the 5-d adaptation period as a control. After the adaptation, pigs were exposed to HS at 33℃ (HS33) for 24 h. All the pigs were fed the same diet formulated to meet or exceed predicted requirements during the whole experimental period. Blood samples were collected after the adaptation and heat treatment to verify the AAs. Measurements were the average daily feed intake (ADFI), average daily water intake (ADWI), water loss, tumor necrosis factor (TNF)-α, nitrate, total nitric oxide, and AAs in the plasma samples. Data were analyzed using the PROC GLM of SAS. HS33 had a lower ADFI (p < 0.05) and a tendency for an increased ADWI and water loss compared to TN. The level of TNF-α was significantly decreased (p < 0.05) in HS33. HS33 had a lower concentration of histidine and sarcosine compared to TN; however, there were no differences in the levels of nitrates and total nitric oxide. In conclusion, the results of this study show the differential effect of HS on the plasma concentration of AAs. We expect that the changed AAs could be potential indicators of HS.

  • Transplantation is considered to be a very useful approach to improve human welfare and to prolong life-span. Heterologous organ transplantation using pig organs which are similar to human beings and easy to make mass-production has known as one of the alternatives. To ensure potential usage of the pig organ for transplantation application, it is essentially required to generate transgenic pig modifying immuno-related genes. Previously, we reported production of heterozygous ${\alpha}1,3$-galactosyltransferase (GalT) knock-out and human membrane cofactor protein (MCP) expressing pig ($GalT^{-MCP/+}$), which is enforced for suppression of hyperacute and acute immunological rejection. In this study, we reported generation of homozygous pig ($GalT^{-MCP/-MCP}$) by crossbreeding $GalT^{-MCP/+}$ pigs. Two female founders gave birth to six of $GalT^{-MCP/-MCP}$, and seven $GalT^{-MCP/+}$ pigs. We performed quantitative real-time PCR, western blot, and flow cytometry analyses to confirm GalT and MCP expression. We showed that fibroblasts of the $GalT^{-MCP/-MCP}$ pig do not express GalT and its product Gal antigen, while efficiently express MCP. We also showed no expression of GalT, otherwise expression of MCP at heart, kidney, liver and pancreas of transgenic pig. Taken together, we suggest that the $GalT^{-MCP/-MCP}$ pig is a useful candidate to apply xenotransplantation study.

축분 혼합 고형연료의 연소성에 관한 실험적 연구 (Experimental Study on the Combustion Characteristics of the Solid Fuels Blended with Domestic Animal Excreta)

  • Rivers of our country are in the serious state of water pollution because of sewages, factory wastes, domestic animal excreta, and so forth. The development of waste water treatment technology applied in a small-scaled farm is urgent because the government regulation becomes strict. In the present study, four types of solid fuels are made by blending domestic animal excreta, rice hulls and briquets, and its combustion characteristics is investigated by analyzing burning pictures. Domestic animal excreta sampled from a farm in Kimhae was dried with sunlight. From experimental results, it is shown that combustion characteristics of solid fuels becomes better by blending rice hulls which have superior ignitability. Since solid fuel made by blending domestic animal excreta with rice hulls can bum continuously, it can be appropriate for the heating fuels.

給餌器具의 色彩變化에 따른 産卵用 育成鷄의 行動形態에 미치는 影響 (Effect of Colors of Feed-trough on the Behaviour of Pullets)

  • The purpose of this study was to investigate the effect of colors of feed-trough on the eating behaviour of pullets. A total of 64 12wks old pullets were assigned to a $4{\times}4$ Latin square design with 4 treatments(control, blue, yellow, red colored feed-troughs). The results of the study are summarized as follows: 1. Average daily gain and eed intake of the pullets were significantly(P<0.05) decreased with red color feed-troughs. 2. Feed conversion ratio appeared to be improved in the yellow color group compared to the others. 3. Total time spent for eating were not significantly different, among treatments.

Selection Responses for Milk, Fat and Protein Yields in Zimbabwean Holstein Cattle

  • One way of evaluating the effectiveness of a dairy breeding program is to measure response to selection. This may be direct or indirect. The objectives of this study were to estimate expected progress for direct selection on milk, fat and protein yields; to estimate the expected correlated responses on indirect selection for milk, fat and protein yields in Zimbabwean Holstein cattle and to establish the effect of selection intensity on responses. The Animal Model contained fixed effects of herd, year of calving, calving month, dry period, milking frequency and additive effects pertaining to cows, sires and dams. AIREML software package was used to analyse the data. The genetic and phenotypic parameters obtained in this study were used to compute direct and correlated responses to selection. Because of the higher heritabilities in first parity, genetic progress was found to be greater when selection was practised on first parity cows as compared to later lactations. It is therefore recommended that older cows in the herd be replaced with improved heifers so as to enhance genetic progress.

Ovarian Activity of Dromedary (Single Humped) Camel (Camelus dromedarius) in North-Eastern Nigeria

  • A total of 69 ovaries were collected from 345 camels slaughtered in Maiduguri abattoir, North-Eastern part of Nigeria, to study the influence of season on ovary weight and corpus luteum count in the dromedary (single humped) camel (Camelus dromedaries). Right ovary was significantly heavier (p<0.01) and had more follicular fluid than the left. Although corpus luteum count increased gradually throughout the study period, no camel had 3 active corpora lutea and only 53 out of the total number had two. Ovarian activity seem to be higher in the cold dry harmattan season, November/December, than during the warmer post rainy season, September/October. These periods can be exploited by farmers, especially peasants who rely heavily on natural mating, to obtain greater conception rates and reduce the chance of failure during mating.

Effects of the multimodal intervention program including animal-assisted therapy on depression and self-esteem among university students

  • This study aimed to investigate the effects of the multimodal group intervention that combined animal-assisted therapy (AAT) and integrated play therapy (IPT) on depression and self-esteem in undergraduate university students. The subjects were 40 students attending animal-related and social welfare departments of universities located in a metropolitan city. The multimodal intervention program was conducted for eight sessions (twice a week, 60 min each) in the experimental group. Data analysis was conducted using the independent sample t-test, ANCOVA, and paired sample t-test for pre- and post-test. Therefore, the multimodal intervention program applying AAT and IPT showed positive effects on depression and self-esteem in university students. Based on these results, this study proposed the operation of multidisciplinary education and practical and policy utilization methods to reduce depression among university students and help improve their self-esteem.

A Study on the Use of Animal Symbols for Healing of the Korean Traditional Medicine - Focusing on "Donguibogam(東醫寶鑑)"

  • Animal symbols exert a profound influence on humans. This can be likened to homeopathic magic, with its source tied to the symbol. It has been believed that consuming an animal or crafting items from its parts would imbue humans with the creature's powers, thereby enhancing its symbolic significance over time. An animal can have various symbolic meanings, and among them, those related to healing have been adopted as folk remedies. Therefore, in this study, we would like to examine an example in which animals were used in folk therapy as a symbol of healing in traditional Korean medicine through "Donguibogam(東醫寶鑑)". In this context, 'healing' not only pertains to direct cures but also encompasses the use of amulets to fend off afflictions and bolster one's immunity. By looking at animals that have traditionally been used in folk remedies, it is possible to reaffirm the role of animals as healing symbols and to find ways to effectively use animal symbols to enhance the healing effect in the future by recognizing their value.

The use of animal models in rheumatoid arthritis research

  • The pathological hallmark of rheumatoid arthritis (RA) is a synovial pannus that comprises proliferating and invasive fibroblast-like synoviocytes, infiltrating inflammatory cells, and an associated neoangiogenic response. Animal models have been established to study these pathological features of human RA. Spontaneous and induced animal models of RA primarily reflect inflammatory aspects of the disease. Among various induced animal models, collagen-induced arthritis (CIA) and collagen antibody-induced arthritis (CAIA) models are widely used to study the pathogenesis of RA. Improved transplantation techniques for severe combined immunodeficiency (SCID) mouse models of RA can be used to evaluate the effectiveness of potential therapeutics in human tissues and cells. This review provides basic information on various animal models of RA, including CIA and CAIA. In addition, we describe a SCID mouse coimplantation model that can measure the long-distance migration of human RA synoviocytes and cartilage destruction induced by these cells.
