급성 부동 스트레스 후 척수 회색질에서 Peroxiredoxin I 및 III의 발현 변화 (Characterization of Peroxiredoxins in the Gray matter in the spinal cord after Acute Immobilization Stress)

  • Purpose: Many stresses produce reactive oxygen species and bring about mechanism of antioxidant reaction. Cytokine and a neurotransmitter through the cell membrane, as well as signal transduction through the cell membrane, are used for various pathological condition of the brain, such as neurodegenerative disease. There are several antioxidant enzymes in cells (superoxcide dismutase, glutathion peroxidasae, peroxiredoxin catalase, etc.) Methods: This study used single- or double-label immunohistochemical techniques to analyze mouse spinal neuron cells expressing Prx I and Prx III after acute mobilization stress. Results: Prx I was observed in dendritic cell of the gray matter of the spinal cord, and Prx III was observed in the cytoplasm of the GM of the spinal cord. Conclusion: The results of this study will help to explain differences of expression in the distributions of the peroxiredoxin enzymes of the spinal cord.

Effects of land-based fish farm effluent on the morphology and growth of Ascophyllum nodosum (Fucales, Phaeophyceae) in southwestern Nova Scotia

  • Phenotypic plasticity was examined in the economically and ecologically important brown alga Ascophyllum nodosum in southwestern Nova Scotia, considering specifically how nutrient loading affected its vegetative and reproductive features. To determine this, we examined morphometric changes in A. nodosum from two sites receiving direct effluent impacts from a land-based finfish aquaculture facility and from two control sites, approximately 2 km away from the aquaculture facility in opposite directions. Fronds from test sites were significantly younger than from control sites (5 y vs. 8 y); however, fronds from farm sites were significantly larger (219 g vs. 90 g) because of their higher growth rates. Thalli from farm sites had greater reproductive potential, as shown by numbers of receptacle initials (797 initials vs. 281 initials). These results suggest limited nutrient inflows from land-based aquaculture may positively affect adjacent Ascophyllum populations by inducing higher growth rates. We conclude that the coordination of effluent management from land-based aquaculture with natural resource harvesting of A. nodosum may be beneficial. Further study is necessary to determine the limits of nutrient loading for this potentially beneficial outcome.

Feeding ecology of three tonguefishes, genus Cynoglossus (Cynoglossidae) in the Seto Inland Sea, Japan

  • Feeding ecology was compared among the three tonguefishes Cynoglossus abbreviatus, C. joyneri and C. robustus, collected from Seto Inland Sea, Japan, from June 2000 to May 2001. They are benthivores, consuming mainly gammarid amphipods, shrimps, crabs, gastropods, bivalves and ophiuroids. C. abbreviatus consumed greater proportions of ophiuroids whereas C. joyneri and C. robustus ate more amphipods and shrimps. While C. abbreviatus consumed mostly ophiuroids in all size classes, the diets of C. joyneri and C. robustus showed ontogenetic changes in feeding habits; smaller individuals of C. joyneri and C. robustus consumed gammarid amphipods, whereas larger C. joyneri ate shrimps and gastropods, and larger C. robustus fed mainly on gastropods, crabs, bivalves and polychaetes. Cluster analysis based on diet similarities emphasized that the three Cynoglossus species could be categorized on a size-related basis into three feeding groups: smaller C. joyneri and C. rubustus (<25 cm TL) could be classified as group A, and the larger of them (>25 cm TL) as group B, whereas C. abbreviatus was categorized as group C. This means that some degree of resource partitioning can occur among the three Cynoglossus species. The seasonal changes in the diets were also significant for the three Cynoglossus species.

Mitochondrial Genetic Variation of Pen Shell, Atrina pectinata in Korea and Japan

  • In the northwestern Pacific region, the pen shell (Atrina pectinata) is a widely distributed bivalve and economically important in fisheries. Recently, stock of this species has been greatly reduced due to overexploitation and marine pollution, which arouses interest in conservation. Studies on genetic and taxonomic entities of pen shells have not been tried in Korea, which makes difficult to take measures for effective conservation of this marine resource. In this study, we investigated mitochondrial genetic polymorphism of pen shells collected from 4 locations in Korea and Japan using cytochrome c oxidase I (COI) gene sequences. A total of 39 haplotypes were identified among 86 individuals of pen shell. Although only 5 haplotypes were shared, no significant genetic differentiation was observed between Korean and Japanese populations. These results suggest that pen shell populations of these regions share an ancestral population which might have experienced expansion during the Pleistocene, but gene flow must have been highly restricted after expansion.

  • Composting by negative aeration is a reasonable proposition to control odor generated during composting process. Cattle manure and rice hulls mixtures were composted in a bin composting system by negative aeration. Continuous(CA) and intermittent(IA) aeration methods were applied to analyze the composting characteristics. The composting temperature and the ammonia emission during composting were investigated according to the aeration methods. The main problem for the negative aeration was the generation of condensate in the suction line of blower. The quantity of condensate was significant for continuous aeration. The aeration method should be modified to escape from the cooling effect of continuous aeration at the initial stage of composting. It took a longer time to finish a composting for intermittent aeration on account of lower aeration. It was concluded that the composting by negative aeration could be accomplished by either continuous or intermittent aeration method if the flow rate would be controlled more efficiently and the water vapor in suction line of blower could be removed effectively. Ammonia emission increased up to maximum value of 675ppm for continuous aeration while 300ppm for intermittent aeration. However, the cumulative value of ammonia emission was larger for intermittent aeration than for continuous aeration.

Antimutagenic and Antileukemic Activities Aloe vera L.

  • We investigated that the extract of Aloe vera L. and its fractions exert antimutagenic activity against Salmonella typhimurium TA98 and TA100, and antileukemic effect against K562 human leukemia cell line. The aqueous ethanolic extract of A. vera L. was revealed to have antimutagenic effect on the AF-2 (2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide) in Salmonella mutation assay. Among the three fractions (fractions A, B and C) separated by silica gel chromatography, fraction C $(50\;{\mu}g/plate)$ exhibited the greatest antimutagenic effect on the AF-2 with inhibition rate of 84 and 90% in Salmonella typhimurium TA98 and TA100, respectively. The fraction C $(500\;{\mu}g/ml)$ inhibited the growth of K562 human leukemia cell line by 93% in MTT assay. However, the components of A. vera L. did not exhibit cytotoxic effect against MDBK bovine normal kidney in MTT assay.

Developmental characteristics of Zophobas atratus (Coleoptera: Tenebrionidae) larvae in different instars

  • The giant mealworm beetle, Zophobas atratus (Coleoptera: Tenebrionidae), is reared for commercial purposes as a live feed for animals. In 2011, it was officially introduced in Korea, and since then it has been considered commercially important. This beetle is a good animal feed resource because of its high protein content with balanced nutrients. However, its life stage characteristics have not been clearly described, especially of the larval stage that can be used as commercial products. To this end, in our study, we determined the number of Z. atratus larval instars, and described their characteristics at each stage, providing basic information about this beetle. Z. atratus larvae required eight to nine d of incubation period before hatching. The first instar period comprised three to four d. There were relatively large variations in each instar period, except for the first instar. Before the adults emerged, most of the individuals passed through15 to 18 instars. The highest pupation rate, 25.71%, was observed in both 16th and 17th instars. Body length gradually increased with each successive instar and it reached its maximum at the 18th instar. The color of larvae was white at the first instar, and gradually turned brown after the second instar.

  • Type 1 diabetes mellitus (T1DM) is an autoimmune disease characterized by extreme insulin deficiency due to an overall reduction in the mass of functional pancreatic ${\beta}$-cells. Several animal models have been used to study T1DM. Amongst these, the mini-pig seems to be the most ideal model for diabetes research, owing to similarities with humans in anatomy and physiology. The purpose of this study was to analyze differentially expressed pancreatic proteins in a streptozotocin (STZ)-induced mini-pig T1DM model. Pancreas proteins from mini-pigs treated with STZ were separated by two-dimensional gel electrophoresis, and 11 protein spots were found to be altered significantly when compared with control mini-pigs. The data in this study utilizing proteomic analysis provide a valuable resource for the further understanding of the T1DM pathomechanism.

  • The development of large-scale environmental friendly livestock complex in the reclaimed tideland is one of different alternatives to increase the competitiveness of internal livestock industry against an international opening markets as DDA and FTA in agricultural field. Recently, it is possible to introduce an environmental friendly livestock complex in the reclaimed tideland by an amendment of the acts for agricultural land. However more studies that are on the basis of nitrogen and phosphorus mass balance need to preserve the agricultural environments as the quality of agricultural water and soil in rural area. In this study, the reference for feasibility study is Whaong reclaimed tideland which located at Whaseong city, Gyeonggi Province, and a basic concept of environmental friendly livestock complex is the production of forage crops with the supply of liquid fertilizer and the production of bioenergy such as biogas by the recycling of pig slurry as a resource. The mass balance of nitrogen based on between forage crops such as maize, barley and liquid fertilizer supplied at the reclaimed tideland, and also it was estimated an economical efficiency as anaerobic digestion plant for treating pig slurry of $100m^3/day$ introduce in an environmental friendly livestock complex.

  • Background: Metabolic modeling has been an essential tool in metabolic reconstruction, which has dramatically advanced in the last decades as a part of systems biology. At present, the protocol for metabolic reconstruction has been systematically established, and it provides the basis for the analysis of complex systems, which has been limited in the past. Therefore, metabolic reconstruction can be adapted to analyze agricultural systems whose metabolic data has been accumulated recently. Purpose: The aim of this review is to suggest the suitability of metabolic modeling for understanding agricultural metabolic data and to encourage the potential use of this modeling in the field of agriculture. Review: We reviewed the procedure of metabolic reconstruction using computational modeling with applicable strategies and software tools. Additionally, we presented the initial attempts of metabolic reconstruction in the field of agriculture and proposed further applications.
