GC/MS를 이용한 뇨 중 환경 에스트로겐들의 동시 프로필 (Profiling of Urinary Environmental Estrogens by Gas Chromatography/Mass Spectrometry)

  • 에스트로겐처럼 행동하여 호르몬 수용체와 결합하거나 세포의 신호전달 과정에 영향을 미침으로서 내분비계를 교란시킬 수 있는 환경 에스트로겐 19종 (phytoestrogen: 12종, mycoestrogen: 5종, synthetic estrogen: 2종)의 동시 프로필 분석을 시도하였다. Gas Chromatography/Mass Spectrometry (GC/MS)의 selected ion monitoring (SIM) 방법을 기본으로 하였으며, 액체-고체 추출, 효소 기수분해, 액체-액체 추출 그리고 trimethylsilyl (TMS)-ether 형태의 유도체화를 거치는 비교적 간편한 전처리 방법으로 47.6~99.5%의 회수율과 0.66~9.33%, 1.66~16.14%의 within-a-day 및 day-to-day 분석의 RSD 값을 얻었다. 이 방법을 적용하여 정상 성인 여성과 남성의 뇨 시료에 존재하는 대상 물질들의 농도범위를 결정함으로서 환경 에스트로겐의 영향을 평가하는데 기준이 될 수 있는 한국인 정상인 참고치를 설정하였다.

알파분광법과 중성자방사화분석법에 의한 극미량의 악티늄계원소 (Am, Pu, Th, U)분석연구 (Determination of trace actinide (Am, Pu, Th, U) using alpha spectrometry and neutron activation analysis)

  • 환경시료중의 극미량의 악티늄계 동위원소들을 분석하기는 무척 어렵다. 이들 원소들은 개별 분리하는 작업이 필요하며, 알파분광법으로 분석한 어떤 핵종들은 검출감도도 높은 편이다. 이런 극미량의 악티늄계 동위원소들을 분석하기 위해 용매추출법이 결합된 TRU-Spec 이온교환수지와 음이온 교환수지를 사용하여 악티늄계 원소들을 분리한 후 알파분광법으로 검출하였다. 그리고 U과 Th의 검출한계를 낮추기 위해 중성자방사화분석법을 적용하였다. 중성자방사화분석법을 적용하기 위한 바탕물질로 고순도 V foil을 사용하여 검출감도를 10배 향상시킬 수 있었으며, 이 분석법을 표준시료인 NIST-4354, IAEA-368 퇴적물 시료에 적용한 결과 표준값과 10% 이내에서 잘 일치하였다.

Assessment of Bandsharing Values in RAPD-PCR Analysis of Dwarf Cattle of Kerala

  • Random amplified polymorphic DNA (RAPD-PCR) analysis of 56 animals of four different genetic groups of dwarf cattle in Kerala was done as a single step analysis. Bandsharing (BS) values were calculated for animals of each group and between groups as an analytical tool to find out genetic variation among animals. The different factors affecting BS values were estimated using Harvey''s Least squares analysis. The effects of genetic group, Guanine-cytosine (GC) content of primer and gel on BS values were found significant. Bandsharing values of Kasargode-Highrange dwarf animals were significantly different from Vechur, Vatakara and their combinations. The Vechur, Vatakara and Vechur-Vatakara combinations were found to be more uniform (high BS value) compared with other combinations. The bandsharing value was lowest with primers of GC content 90% and highest with 80% GC content. The effect of gel on BS value points to the need of adjustments of gel factor for calculation of BS values.

농산물 중 메펜트리플루코나졸 및 트리티코나졸 살균제의 동시 분석법 개발 및 검증 (Development and Validation of a Simultaneous Analytical Method for the Detection of Mefentrifluconazole and Triticonazole Fungicide in Agricultural Crops)

  • BACKGROUND: Mefentrifluconazole and triticonazole are the triazole fungicides. The maximum residue levels for agricultural products need to be set up. Therefore, development of the official analytical method for determination of mefentrifluconazole and triticonazole residues from agricultural crops was necessary due to safety management, and then a simultaneous analytical method was developed for the determination of mefentrifluconazole and triticonazole in agricultural crops. METHODS AND RESULTS: Samples were extracted using acetonitrile and purified using dispersive solid phase extraction, and then detected with liquid chromatograph-tandem mass spectrometry (LC-MS/MS). Matrix-matched calibration curves (0.0025-0.25 ㎍/mL) were linear into a sample extract with r2>0.99. For validation, the recovery test was carried out at three fortification levels (LOQ, 10 LOQ and 50 LOQ) from agricultural samples. The results for mefentrifluconazole and triticonazole ranged between 92.3 to 115.3% and 91.4 to 108.5%, respectively and RSD (relative standard deviation) values were also below 6.0%. Furthermore, inter-laboratory was conducted to validate the method. CONCLUSION: All values were corresponded with the criteria ranges requested by both the CODEX (CAC/GL 40-1993, 2003) and MFDS guidelines (2016). Therefore, the proposed method can be used as an official analytical method for determination of mefentrifluconazole and triticonazole (triazole fungicides) in the Republic of Korea.

An Analytical Model for Predicting the Effective Thermal Conductivity of Woven Wire Wick Structure

  • Woven wire wick is a very effective structure because of its easiness to insert inside of pipe for a miniature heat pipe. The present study was conducted to predict the porosity and the effective thermal conductivity of liquid-saturated woven wire wick. The porosity and the effective thermal conductivity of the evaporator region indicate different values from those of the condenser region due to the existence of non-flow region. The minimum value of the effective thermal conductivity indicates on condition of the $\theta$=$45^{Wcirc}$ and the values of the effective thermal conductivity increases symmetrically centering around the minimum value. The values of the effective thermal conductivity in the evaporator region at the angle of $45^{Wcirc}$ indicate about 60~80% higher than those in the condenser region for various combinations of copper, and stainless with water and ethanol.

Detection of a concentrated damage in a parabolic arch by measured static displacements

  • The present paper deals with the identification of a concentrated damage in an elastic parabolic arch through the minimization of an objective function which measures the differences between numerical and experimental values of static displacements. The damage consists in a notch that reduces the height of the cross section at a given abscissa and therefore causes a variation in the flexural stiffness of the structure. The analytical values of static displacements due to applied loads are calculated by means of the principle of virtual work for both the undamaged and damaged arch. First, pseudo-experimental data are used to study the inverse problem and investigate whether a unique solution can occur or not. Various damage intensities are considered to assess the reliability of the identification procedure. Then, the identification procedure is applied to an experimental case, where displacements are measured on a prototype arch. The identified values of damage parameters, i.e., location and intensity, are compared to those obtained by means of a dynamic identification technique performed on the same structure.

New coefficients to find natural period of elevated tanks considering fluid-structure-soil interaction effects

  • The main purpose of the current study is to develop the new coefficients for consideration of soil-structure interaction effects to find the elevated tank natural period. Most of the recommended relations to find the natural period just assumed the fixed base condition of elevated tank systems and the soil effects on the natural period are neglected. Two different analytical systems considering soil-structure- fluid interaction effects are recommended in the current study. Achieved results of natural impulsive and convective period, concluded from mentioned models are compared with the results of a numerical model. Two different sets of new coefficients for impulsive and convective periods are developed. The values of the developed coefficients directly depend to soil stiffness values. Additional results show that the soil stiffness not only has significant effects on natural period but also it is effective on liquid sloshing wave height. Both frequency content and soil stiffness have significant effects on the values of liquid wave height.

J-적분의 유한요소해석에 관한 연구 (A Study on the Finite Element Analysis of J-Integral)

  • One of the important subjects in fracture mechanics study is to analyze the stress intensity factor. In this paper, the stress intensity factor in Mode I ($K^{I}$) is determined by J-integral using the finite element method. In this investigation, the values of $K^{I}$ are computed for distorted and undistorted elements of 8-noded isoparametric finite elements. The numerical results obtained are summarized as follows. (1) Through a relatively coarse mesh, the $K^{I}$ values obtained by this method are fairly good accuracy. (2) The $K^{I}$ values for the distorted elements appear to be better than those obtained using the undistorted mesh. (3) Within the limits of these analyses, the solutions obtained through the integral paths in the medium region of elements approach to the analytical solution most closely.

  • The evaluation of habitability to building vibration is conducted by the values of natural frequency, amplitude displacement, damping ratio. These values can be obtained from test or analytical results. Data acquisition through test may be possible in existing building, however, to estimate the serviceability of the building, it is necessary to evaluate those values at the stage of design. The natural frequency is important and basic factor for the evaluation of the serviceability. Calculation method of the effective stiffness in RC slab is proposed. To prove the efficiency of the proposed method, sample results of the analysis and the test are compared. These results proved that the effective width proposed to calculate the effective stiffness is proper to evaluate the natural frequency.

Numerical method to determine the elastic curve of simply supported beams of variable cross-section

  • In this paper a new numerical method to determine the elastic curve of the simply supported beams of variable cross-section is demonstrated. In general case it needs to solve linear or small nonlinear second order differential equations with prescribed boundary conditions. For numerical solution the initial values of the slope and the deflection of the end cross-section of the beam is necessary. For obtaining the initial values a lively procedure is developed: it is a special application of the shooting method because boundary value problems can be transformed into initial value problems. As a result of these transformations the initial values of the differential equations are obtained with high accuracy. Procedure is applied for calculating of elastic curve of a simply supported beam of variable cross-section. Results of these numerical procedures, analytical solution of the linearized version and finite element method are compared. It is proved that the suggested procedure yields technically accurate results.
