Wastewater Treatment by Microorganism (미생물에 의한 발효처리)

  • The process of biological treatment of organic wastewater is principally associated with those of self-purification in the natural water environment. The treatment system has e intensive function of stabilizing wastewater more effectively than in natural water, which is like natural water concentrated in a small space. Biological treatment of wastewater involves activated sludge and various modified process, trickling filter, rotating disk, oxidation ditch, etc. for aerobic decomposition and anaerobic processes such as anaerobic decomposition and methane fermentation. The basic characteristic of these processes is the use of mixed culture for the conversion of pollutants. This review forcuses on the various kinds of microorganisms related to each treatment processes. Kinetic analysis of the activated sludge process is discussed in order to understand the basis of control and maintenance of the biological treatment process.

Characterization of an Oxygen-Dependent Inducible Promoter Systems, the nar Promoter of Escherichia coli, and Gram negative host strains

  • The nar promoter of Escherichia coli was known to induce maximally under anaerobic or microaerobic conditions in the presence of nitrate. In this study, the nar promoter was tested to see whether the expression level of a reporter gene which fused lacZ gene at nar promoter's downstream, in the some gram negative host strains(Agrobacterium, Pseudomonas and Rhizobium). A nar promoter system(Combination of nar promoter and gram negative strain) was grown under aerobic conditions to absorbance at 600 nm of nearly 2.0 and then, the nar promoter was induced by lowering DO to 1-2% with alternating microaerobic and aerobic condition in the fermentor cultures, using different gram negative hosts. For a wild type nar promoter (pNW61), it was possible to maintain production of ${\beta}-galactosidase$ activity per cell(specific ${\beta}-galactosidase$ activity) at 14,000, 9600, 45 Miller units in the presence of 1% nitrate. and for a nitrate - independent nar promoter (pNW618) at 12,000, 10,400 and 58 Miller units in the absence of nitrate ion, respectively.

Characteristics of Denitrification from Municipal Wastewater Treatment using a Combined Fixed Film Reactor (CFFR) Process (복합생물막 반응기를 이용한 하수처리시 탈질화 특성)

  • A new biological nutrient removal system combining $A^2/O$ process with fixed film was developed in this work and the characteristics of denitrification were especially investigated in the combined fixed film reactor(CFFR). Media was added in the anaerobic, anoxic and aerobic reactors, respectively. Tests were made to establish the effluent level of $NO_x-N$, COD, DO and nitrite effects on $NO_x-N$ removal in the CFFR by decreasing hydraulic retention time (HRT) from 10.0 to 3.5 hours and by increasing internal recycle ratio form 0% to 200%. The influent was synthesized to levels similar to the average influent of municipal wastewater treatment plants in Korea. SARAN media with a porosity of 96.3% was packed 40% / 130% / 25% based on its reactor volume, respectively. It was found that COD rarely limited dentrification in the anoxic reactor because of high $C/NO_x/-N$ ratio in the anoxic reactor, while DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent inhibited denitrification in the anoxic reactor. It was proved that the critical points of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent were 0.15mg/L and 10%, respectively. As the internal recycle ratio increased, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent increased. Especially, at the condition of internal recycle ratio, 200%, DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent exceeded the critical points of 0.15mg/L and 10%, respectively. Then, denitrification efficiency considerably decreased. Consequently, it was represented that the control of DO concentration in the anoxic reactor and $NO_2-N/NO_x/-N$ from the aerobic effluent can assure effective denitrification.

Characterizations of Denitrifying Polyphosphate-accumulating Bacterium Paracoccus sp. Strain YKP-9

  • A denitrifying polyphosphate-accumulating bacterium (YKP-9) was isolated from activated sludge of a 5-stage biological nutrient removal process with step feed system. This organism was a Gram-negative, coccus-shaped, facultative aerobic chemoorganotroph. It had a respiratory type of metabolism with oxygen, nitrate, and nitrite as terminal electron acceptors. The 16S rRNA gene sequence of strain YKP-9 was most similar to the 16S rRNA gene sequence of Paracoccus sp. OL18 (AY312056) (similarity level, 97%). Denitrifying polyphosphate accumulation by strain YKP-9 was examined under anaerobic-anoxic and anaerobic-oxic batch conditions. It was able to use external carbon sources for polyhydroxyalkanoates(PHA) synthesis and to release phosphate under anaerobic condition. It accumulated polyphosphate and grew a little on energy provided by external carbon sources under anoxic condition, but did neither accumulate polyphosphate nor grow in the absence of external carbon sources under anoxic condition. Cells with intracellular PHA cannot accumulate polyphosphate in the absence of external carbon sources under anoxic condition. Under oxic condition, it grew but could not accumulate polyphosphate with external carbon sources. Based on the results from this study, strain YKP-9 is a new-type denitrifying polyphosphate-accumulating bacterium that accumulates polyphosphate only under anoxic condition, with nitrate and nitrite as the electron acceptors in the presence of external carbon sources.

Characteristics of Municipal Sewage Sludge Affecting the Biodegradation of a Plastic Material Under Aerobic Condition (호기적 조건에서 플라스틱 생분해에 영향을 미치는 도시 하수 오니의 성질)

  • The characteristics of activated sludge affecting the biodegradation of plastic materials under aerobic condition were studied using cellophane film as a model system. The activated sludges of site 3, which treat a mixture of domestic sewage and supernatant of septic tank, obtained from December 1993 to April 1994 showed similar biodegradation activities. Biodegradations for 28 days reached around 80%. Viable cell number of inoculums maintained at a level of 10$^{6}$~10$^{7}$ /ml. In this range, viable cell number showed no relationship with biodegradation activities. The activa- ted sludges of site 2, which treat a mixture of domestic sewage and anaerobic digest of nightsoil, obtained four times from April 1993 to April 1994 showed very different biodegradation activities ranged from 20% to 80% for 28 days. Inoculum size affects biodegradation significantly. One percent inoculum showed the best biodegradation among the inoculum sizes of 0.1, 1.0 and 10%. Ten percent inoculum revealed inhibitory effects on the biodegradation activity which can be greatly reduced by centrifugation and filtration. Filtration was better than centrifugation in reducing inhibitory effects.

Evaluation of gaseous concentrations, bacterial diversity and microbial quantity in different layers of deep litter system

  • Objective: An experiment was conducted to investigate the environment of the deep litter system and provided theoretical basis for production. Methods: The bedding samples were obtained from a pig breeding farm and series measurements associated with gases concentrations and the bacterial diversity as well as the quantity of Escherichia coli, Lactobacilli, Methanogens were performed in this paper. Results: The concentrations of $CO_2$, $CH_4$, and $NH_3$ in the deep litter system increased with the increasing of depth while the $N_2O$ concentrations increased fiercely from the 0 cm to the -10 cm depth but then decreased beneath the -10 cm depth. Meanwhile, the Shannon index, the dominance index as well as the evenness index at the -20 cm layer was significantly different from the other layers (p<0.05). On the other hand, the quantity of Escherichia coli reached the highest value at the surface beddings and there was a significant drop at the -20 cm layer with the increasing depth. The Lactobacilli numbers increased with the depth from 0 cm to -15 cm and then decreased significantly under the -20 cm depth. The expression of Methanogens reached its largest value at the depth of -35 cm. Conclusion: The upper layers (0 cm to -5 cm) of this system were aerobic, the middle layers (-10 cm to -20 cm) were micro-aerobic, while that the bottom layers (below -20 cm depth) were anaerobic. In addition, from a standpoint of increasing the nitrification pathway and inhibiting the denitrification pathway, it should be advised that the deep litter system should be kept aerobic.

Removal of Nutrients Using an Upflow Septic Tank(UST) - Aerobic Filter(AF) System (부패조와 호기성 여과공정을 이용한 영양염류 제거)

  • The objective of this study was to investigate a small sewage treatment system. This system was developed to improve a nitrogen and phosphorus removal efficiency and generate less solid using upflow septic tank(UST) - aerobic filter(AF) system. The UST equipped with an aerobic filter, the filter was fed with both raw sewage and recycled effluent from the UST to induce the denitrification and solid reduction simultaneously. Overall removal efficiencies of COD and total nitrogen(TN) were above 96% and 73% at recycle ratio of 200%, respectively. Critical coagulant dose without the biochemical activity was found to be 40 mg/L. Removal efficiency of total phosphorus(TP) in influent was above 90% by chemical and biological reactions. Although the phosphorus concentration was low under the high alkalinity in raw sewage, the pH value was unchanged by the coagulant dose.

Evaluation of the Reducing Efficiency of Vertical and Horizontal Wetland Using Intermittent Flow System (간헐식 흐름방식을 활용한 수직·수평 습지의 정화효율 평가)

  • Nitrogen and phosphorus are key factors in causing eutrophication of water body. In this study, ceramics media was selected to increase the removal efficiency of nitrogen and phosphorus. We designed vertical, horizontal flow constructed wetlands to create aerobic and anaerobic flow conditions by using the media, then proceeded to performance evaluations after acrylic reactors were produced. In the case of vertical and horizontal flow constructed wetlands, we measured oxygen concentrations to evaluate aerobic and anaerobic conditions. we got the result of 2.7 mg/L in the aerobic condition, N.D in the anaerobic condition respectively, which suited our purpose. The result of the combined vertical and horizontal flow condition showed that the removal efficiency of SS was 94%, 91%, 61% at 140 min, 80 min, 60 min of running times, respectively, and the removal efficiency of T-P was 84%, 71%, 63% during each running time. In case of T-N, the removal efficiency was 63%, 49%, 42% during each running time. We found that the reactor exerted better removal efficiency when in the short time compared to 12 - 24 hr residence time of existing wetlands. In this study, we conducted experiments to explore functional effects after applying combined vertical and horizontal flow methods in the field. Further study will be carried out to identify its mechanism and administrative perspective.

Biohydrogen production from engineered microalgae Chlamydomonas reinhardtii

  • The green microalgae Chlamydomonas reinhardtti is well-known specie in the terms of $H_2$ production by photo fermentation and has been studying for a long time. Although the $H_2$ production yield is promising; there are some bottlenecks to enhance the yield and efficiency to focus on a well-designed, sustainable production and also scaling up for further studies. D1 protein of photosystem II (PSII) plays an important role in photosystem damage repair and related to $H_2$ production. Because Chlamydomonas is the model algae and the genetic basis is well-studied; metabolic engineering tools are intended to use for enhanced production. Mutations are focused on D1 protein which aims long-lasting hydrogen production by blocking the PSII repair system thus $O_2$ sensitive hydrogenases catalysis hydrogen production for a longer period of time under anaerobic and sulfur deprived conditions. Chlamydomonas CC124 as control strain and D1 mutant strains(D240, D239-40 and D240-41)are cultured photomixotrophically at $80{\mu}mol\;photons\;m^{-2}s^{-1}$, by two sides. Cells are grown in TAP medium as aerobic stage for culture growth; in logarithmic phase cells are transferred from aerobic to an anaerobic and sulfur deprived TAP- S medium and 12 mg/L initial chlorophyll content for $H_2$ production which is monitored by the water columns and later detected by Gas Chromatography. Total produced hydrogen was $82{\pm}10$, $180{\pm}20$, $196{\pm}20$, $290{\pm}30mL$ for CC124, D240, D239-40, D240-41, respectively. $H_2$ production rates for mutant strains was $1.3{\pm}0.5mL/L.h$ meanwhile CC124 showed 2-3 fold lower rate as $0.57{\pm}0.2mL/L.h$. Hydrogen production period was $5{\pm}2days$ for CC124 and mutants showed a longer production time for $9{\pm}2days$. It is seen from the results that $H_2$ productions for mutant strains have a significant effect in terms of productivity, yield and production time.

The Optimum Condition for the Co-digestion of Food waste and Sewage Sludge (하수처리장에서의 음식물 쓰레기와 농축 슬러지의 혐기성 병합 처리 조건 선정)

  • The objective of this study was to evaluate the possibility of co-digestion of food waste and sewage sludge mixture using anaerobic system. The Biochemical methane Potentials of cabbage and food waste were $297ml\;CH_4/g$ VS and $306.7ml\;CH_4/g$ VS, respectively. The biodegradability of food waste was 60%. The concentrations of acetate, propionate, and isobutyrate produced during the aerobic acidogenesis of food waste for 36 hours were 7,000~7,200 ppm, 260~280 ppm, 380~400 ppm, and 40~50 ppm, respectively, of which acetate was over 85%. The concentrations of acetate, propionate, and isobutyrate produced during the anaerobic acidogenesis for 36 hours were 1,400~1,600 ppm, 30~40 ppm, 220~250 ppm, and 260~300 ppm, respectively, of which acetate was over 70%. The biodegradabilities of aerobic and anaerobic acidogenesis were 30% and 25%, respectively. Methanogensis could be activated under 1 % of NaCl and 1,000 ppm of volatile fatty acids at the range of pH 6.8~7.2. The maximum mixture ratio of food waste and sewage sludge in the present study was 2:8 by the result of VS removal rate and Methane production.
