Biogas Production from Anaerobic Co-digestion Using the Swine Manure and Organic Byproduct (돈분과 유기성 부산물을 혼합한 혐기소화에서 바이오가스 생산)

  • Animal manure is produced annually 43.7 million tonnes in Korea. Among them, about 85.6 % are used as compost or liquid fertilizer to the agricultural land. The animal manure can be effectively utilized by mixing with organic byproducts that result in generation of biogas from anaerobic co-digestion process. This study aimed to optimize the content of total solid materials (TS) and determine the effect of organic byproduct on the co-digestion process. Prior to the byproduct treatments, determination of proper content of TS was conducted by controlling at 5 or 10 %. For the byproduct treatments, swine manure without adding the byproduct was used for control treatment, and swine manure mixed with either corn silage or kitchen waste was used for other treatments. Volume of biomethane ($CH_4$) generated from digested materials was quantified before and after byproduct treatments. In result, a 1.4-fold higher biomethane, about 0.556 L/$L{\cdot}d$, was produced when the content of TS was controlled at 10 %, compared at 5 %, about 0.389 L/$L{\cdot}d$. When the swine manure was mixed with the corn silage or kitchen waste, a two-fold higher biomethane was produced, about 1.theand 1.0heL/$L{\cdot}d$, respectively, compared to the control treatment. Biogas production from organic dry matter (odm) was a3, 362eand 2h6 L/kg odm${\cdot}$d for control, corn silage, and kitchen waste treatment, respectively. The lower biogas production in the treatment of kitchen waste than that of corn silage is associated with its relatively high odm contents. The methane concentration during the whole process ranged from 40 at the beginning to 70 % at the end of process for both the control and kitchen waste treatments, and ranged from 52 to 70 % for the corn silage treatment. Hydrogen sulfide ($H_2S$) concentration ranged between 350 and 500 ppm. All the integrated results indicate that addition of organic byproduct into animal manure can double the generation of biogas from anaerobic fermentation process.

Oxygen Sensitivity of Carbon Monoxide-Dependent Hydrogen Production Activity in Citrobacter sp.

  • A newly isolated Citrobacter sp. Y19 catalyzes the CO-dependent $H_2$ production (biological water-gas shift reaction) by the actions of CO dehydrogenase (CODH) and hydrogenase. Y 19 requires $O_2$ for fast growth, but its $H_2$ production activity is significantly inhibited by $O_2$. In the present study, the effect of $O_2$ on the activities of CODH ard hydrogenase was investigated quantitatively in both whole cells and broken cells, based on CO-dependent or methyl viologen (MV)-dependent $H_2$ production in addition to CO-dependent MV reduction. In crude cell extracts, CODH activity was mostly found in the soluble fraction. Inactivation of CODH and hydrogenase activities by $O_2$ followed the first-order decay kinetics, and the dependence of the rate constants on $O_2$ partial pressure could be expressed by the Michaelis-Menten equation. In whole cells, the maximum deactivation rate constants ($k_{d,max}$ of hydrogenase and CODH were quite similar: $0.07{\pm}0.03 min^{-1}\;and\;0.10{\pm}0.04 min^{-1}$, respectively. However, the first-order rate constant ($k_{d,max}/K_s$) of CODH ($0.25\;min^{-1}\;atm^{-1}$) at low $O_2$ partial pressures was about 3-fold higher than that of the hydrogenase, since the half-saturation constant ($K_s$) of CODH was about half of that of hydrogenase. In broken cells, both enzymes became significantly more sensitive to $O_2$ compared to the unbroken cells, while $k_{d,max}/K_s$ increased 37-fold for hydrogenase and 6.7-fold for CODH. When whole cells were incubated under anaerobic conditions after being exposed to air for 1 h, hydrogenase activity was recovered more than 90% in 2 h suggesting that the deactivation of hydrogenase by $O_2$ was reversible. On the contrary, CODH activity was not recovered once deactivated by $O_2$ and the only way to recover the activity was to synthesize new CODH. This study indicates that $O_2$ sensitivity of $H_2$ production activity of Citrobacter sp. Y19 is an important drawback as in other $H_2-producing$ bactria.

In vitro evaluation of nano zinc oxide (nZnO) on mitigation of gaseous emissions

  • Background: Enteric methane ($CH_4$) accounts for about 70% of total $CH_4$ emissions from the ruminant animals. Researchers are exploring ways to mitigate enteric $CH_4$ emissions from ruminants. Recently, nano zinc oxide (nZnO) has shown potential in reducing $CH_4$ and hydrogen sulfide ($H_2S$) production from the liquid manure under anaerobic storage conditions. Four different levels of nZnO and two types of feed were mixed with rumen fluid to investigate the efficacy of nZnO in mitigating gaseous production. Methods: All experiments with four replicates were conducted in batches in 250 mL glass bottles paired with the ANKOM$^{RF}$ wireless gas production monitoring system. Gas production was monitored continuously for 72 h at a constant temperature of $39{\pm}1^{\circ}C$ in a water bath. Headspace gas samples were collected using gas-tight syringes from the Tedlar bags connected to the glass bottles and analyzed for greenhouse gases ($CH_4$ and carbon dioxide-$CO_2$) and $H_2S$ concentrations. $CH_4$ and $CO_2$ gas concentrations were analyzed using an SRI-8610 Gas Chromatograph and $H_2S$ concentrations were measured using a Jerome 631X meter. At the same time, substrate (i.e. mixed rumen fluid+ NP treatment+ feed composite) samples were collected from the glass bottles at the beginning and at the end of an experiment for bacterial counts, and volatile fatty acids (VFAs) analysis. Results: Compared to the control treatment the $H_2S$ and GHGs concentration reduction after 72 h of the tested nZnO levels varied between 4.89 to 53.65%. Additionally, 0.47 to 22.21% microbial population reduction was observed from the applied nZnO treatments. Application of nZnO at a rate of $1000{\mu}g\;g^{-1}$ have exhibited the highest amount of concentration reductions for all three gases and microbial population. Conclusion: Results suggest that both 500 and $1000{\mu}g\;g^{-1}$ nZnO application levels have the potential to reduce GHG and $H_2S$ concentrations.

Biological conversion of CO2 to CH4 in anaerobic fixed bed reactor under continuous operation (혐기성 고정층 생물반응기의 연속운전을 통한 이산화탄소의 메탄전환)

  • This study was carried out to examine different mole ratio of $H_2/CO_2$ and EBCT using the continuous system in the lab scale throughout biological methods with accumulated hydrogenotrophic methanogen that can convert $CO_2$ to $CH_4$. The experimental-based results with various gas mixtures of mole ratio of 4:1($H_2/CO_2$) and 5:1($H_2/CO_2$), $H_2$ was converted more than 99% conversion rate. In case of $CO_2$, 4:1($H_2/CO_2$) and 5:1($H_2/CO_2$) were $74.45{\pm}0.33%$, $95.8{\pm}10.7%$, respectively, in addition, the study was confirmed that the amount of $H_2$ was more needed than stoichiometric equations, where approach methods are empirical versus theoretical frameworks, for converting total $CO_2$. As such, we have noticed that $H_2$ was used for energy source of hydrogenotrophic methanogen for maintaining life. Regarding the results of the ratio of treatment by retention time, limitation of treatment capacity showed that $H_2$(99.9%) and $CO_2$(96.23%) at EBCT 3.3 hrs indicated stable conversion ratio, as well as appeared that methane production rate and $CO_2$ fixation rate were investigated $1.15{\pm}0.02m^3{\cdot}m^{-3}{\cdot}day^{-1}$ and $2.01{\pm}0.04kg{\cdot}m^{-3}{\cdot}day^{-1}$, respectively.

ISOLATION AND IDENTIFICATION OF LACTOBACILLUS SALIVARIUS INHIBITING THE FORMATION OF ARTIFICIAL PLAQUE AND THE PRODUCTION OF VOLATILE SULFUR COMPOUNDS (치태 형성과 휘발성 유황화합물 생성을 억제하는 Lactobacillus salivarius의 분리 및 동정)

  • There are normal inhabitants doing medically useful functions in the body. There are many kinds of bacteria performing specific functions in the oral cavity. Two strains of lactic acid bacteria were isolated from inhabitants of caries-free children's oral cavity, which inhibited the formation of artificial plaque by Streptococcus mutans and the production of volatile sulfur compounds by anaerobic bacteria. The isolates were identified by the test using API 50 CHL medium kit and 16S rDNA partial sequencing. 1. Two isolates were Gram-positive bacilli and produced hydrogen peroxide. 2. When Streptococcus mutans was cultured in the media, the mean weight of formed artificial plaque on the orthodontic wires was $124.4{\pm}30.4\;mg$, whereas being reduced to $5.2{\pm}2.0mg$ and $10.6{\pm}6.6mg$ in the media cultured with Streptococcus mutans and each isolate, respectively (p<0.05) 3. The number of viable cells of Streptococcus mutans was $3.4{\times}10^9$ per ml in the cultured solution, whereas those of Streptococcus mutans in the combined culture with each of isolates were $4.6{\times}10^8\;and\;2.4{\times}10^8$ per ml. 4. The optical density was 1.286 in the supernatant of Fusobacterium nucleatum after vortexing for 30minutes, whereas in the supernatant of combined Fusobacterium nucleatum and each isolate, they were reduced to 0.628 and 0.497, which the percentages of coaggregation between them were 29.4% and 57.8%, respectively 5. The optical density of Fusobacterium nucleatum precipitate was 1.794 in the culture media containing cysteine and $FeSO_4$, being reduced to 1.144 and 0.915 in the coaggregated precipitates of Fusobacterium nucleatum and each isolate. The optical density of Porphyromonas gingivalis precipitate was 1.932 in the culture media, being reduced to 1.170 and 1.266 in the coaggregated precipitates of Porphyromonas gingivalis and each isolate. 6. When two isolates were tested with API 50 CHL medium kit, those were identified Lactobaciallius salivarius and Lactobaciallius delbrueckii subsp. lactis. 7. The similarity values of 16S rDNA sequence between each of isolates and Lactobaciallius salivarius subsp. salicinius were 99.60% and 99.73%, respectively, meaning that isolates were Lactobaciallius salivarius subsp. salicinius. These results indicated that two strains isolated from caries-free children's saliva, which inhibited the formation of artificial plaque and the production of volatile sulfur compounds, were identified as Lactobaciallius salivarius subsp. salicinius.

Evaluation of Odors and Odorous Compounds from Liquid Animal Manure Treated with Different Methods and Their Application to Soils (액상 가축분뇨의 처리 및 토양환원에 따른 악취 및 악취물질의 평가)

  • To comply with stricter regulations provoked by increasing odor nuisance, it is imperative to practice effective odor control for sustainable livestock production. This study was conducted to assess odor and odorous compounds emitted from liquid animal manure with different treatment methods such as Fresh Manure(without treatment, FM), Anaerobic Digestion(AD) and Thermophilic Aerobic Digestion(TAD) and their application to soil. Air samples were collected at the headspace of liquid manure, upland and paddy soil, and analyzed for odor intensity and offensiveness using an olfactometry; odor concentration index using odor analyser; nitrogen-containing compound such as ammonia(NH3) using fluorescence method; and sulfur containing compounds such as hydrogen sulfide(H2S), methyl mercaptan(MeSH), dimethyl sulfide(DMS) and dimethyl disulfide(DMDS) using gas chromatography-pulsed flame photometric detector, respectively. Odor intensity, offensiveness and concentration index from TAD liquid manure was statistically lower than those from FM and AD(p<0.01). Mean concentrations of H2S, MeSH, DMS, DMDS and NH3 were 65.93ppb, 18.55ppb, 5.26ppb, 0.33ppb and 10.57ppm for liquid manure with AD; and 5.15ppb, 0.97ppb, 0.80ppb, 0.56ppb and 1.34ppm for liquid manure with TAD, respectively. More than 60% of malodorous compounds related to nitrogen and sulfur were removed by heterotrophic microorganisms during TAD treatment. When liquid manure was applied onto upland and paddy soil, NH3 removal efficiencies ranged from 51 to 94% and 22 to 91% for AD and TAD liquid manure, respectively. The above results show that liquid manure with TAD is superior to AD and FM with respect to the odor reduction and odor problem caused by land applied liquid manure is directly related to the degree of odor generated by the manure treatment method.

Assessment of Abnormality in Skeletal Muscle Metabolism in Patients with Chronic Lung Desease by $^{31}P$ Magnetic Resonance Spectroscopy ($^{31}P$ 자기 공명분석법을 이용한 만성 폐질환 환자에서의 골격근대사 이상에 관한 연구)

  • The functional derangement of skeletal muscles which may be attributed to chronic hypoxia has been accepted as a possible mechanism of exercise impairment in patients with chronic obstructive pulmonary disease (COPD). The metabolic changes in skeletal muscle in patients with COPD are characterized by impaired oxidative phosphorylation, early activation of anaerobic glycolysis and excessive lactate and hydrogen ion production with exercise. But the cause of exercise limitation in patients with chronic lung disease without hypoxia has not been known. In order to evaluate the change in the skeletal muscle metabolism as a possible cause of the exercise limitation in chronic lung disease patients without hypoxia, we compared the muscular metabolic data of seven male patients which had been derived from noninvasive $^{31}P$ magnetic resonance spectroscopy(MRS) with those of five age-matched normal male control persons. $^{31}P$ MRS was studied during the sustained isometric contraction of the dominant forearm flexor muscles up to the exhaustion state and the recovery period. Maximal voluntary contraction(MVC) force of the muscle was measured before the isometric exercise, and the 30% of MVC force was constantly loaded to each patient during the isometric exercise. There were no differences of intracellular pH (pHi) and inorganic phosphate/phosphocreatine(Pi/PCr) at baseline, exhaustion state and recovery period between two groups. But pHi during the exercise was lower in patients group than the control group (p < 0.05). Pi/PCr during the exercise did not show significant difference between two groups. These results suggest that the exercise limitation in chronic lung disease patients without hypoxia also could be attributed to the abnormalities in the skeletal muscle metabolism.
