In situ Analysis of Methanogenic Bacteria in the Anaerobic Mesophilic and Thermophilic Sludge Digestion (중온 및 고온 혐기성 소화에서 메탄생성균 군집 분석에 관한 연구)

  • Anaerobic digestion has many advantages over the more conventional aerobic treatment processes such as low levels of excess sludge production, low space (area) requirements, and the production of valuable biogas. The purpose of this study was to evaluate the effect of organic loading rate of anaerobic digestion on thermophilic($55^{\circ}C$) and mesophilic($35^{\circ}C$) conditions. Fluorescent in situ hybridization (FISH) method was also used to study the microbial community in the reactors. The stabilizing time in mesophilic anaerobic reactors was shorter as approximately 20 days than 40 days in the thermophilic anaerobic reactors. The amount of methane production rate in anaerobic reactors was independent of the concentrations of supplied substrates and the amount of methanogens. When the microbial diversity in the mesophilic and thermophilic reactors, which had been treated with acetate-based artificial wastewater, were compared, it was found that methanogenesis was carried out by microbial consortia consisting of bacteria and archaea such as methanogens. To investigate the activity of bacterial and archaeal populations in all anaerobic reactors, the amount of acetate was measured. Archaea were predominant in all reactors. Interestingly, Methanothrix-like methanogens appeared in mesophilic anaerobic reactors with high feed substrate concentrations, whereas it was not observed in thermophilic anaerobic reactors.

  • Physico-chemical properties of the activated sludges(Suyoung and Changlim treatment plant), such as SVI(sludge volume index), absorbance, specific surface area, and specific resistance using Buchener funnel test were investigated with changing anaerobic storage time. This experimental condition was found that it was possible to estimate a linear relationship between their parameters such as specific surface area specific resistance, and sludge volume index(SVI). The specific surface area and the specific resistance to filtration of the activated sludges of Suyoung and Changlim treatment plant were found as 123.6~136.6$m^2$/gDS and 41.5~44.9$m^2$/gDS(dry solid), and 1.09$\times$10$^{14}$ ~5.48$\times$10$_{14}$ m/kg and 1.05$\times$10$^{14}$ ~2.48$\times$10$^{14}$ m/kg, respectively. The results gave a good linear relationship between the specific surface area and the specific resistance, r=2.25$\times$10$^{12}$ s-8.10$\times$10$^{13}$ ($R^2$=0.8885) at Suyoung treatment plant and r=1.26$\times$10$^{13}$ s-4.75$\times$10$^{14}$ ($R^2$=0.8756) at Changlim treatment plant.

  • Anaerobic digester coupled with hollow fiber membrane unit. Treatment of starch waste with anaerobic digester-membrane system was studied. $0.17\;m^2$ area of hollow fiber membrane unit of known pore size was immersed into laboratory-scale anaerobic digestion system. The gas production was about $0.74\;m^3/kg$ COD treated. The COD removal efficient was about 80-95% depending on the hydraulic retention time. Crossflow ultrafiltration as Post treatment to anaerobic filter. The study conducted with different membrane pore size indicated that membrane with 1,000,000 molecular weight cut-off size gave a higher COD removal efficiency in the range of 83-87% while giving a study flux of $120-130\;L/m^2\;{\cdot}\;h$. A study was conducted to see the long term clogging effect of membrane also.

  • This study thus looks into two treatment processess : i) Anaerobic digester coupled with hollow fibre membrane unit. Treatment of starch waste with anaerobic digester-membrane system was studied. $0.17\m^2$ area of hollow fibre membrane unit of known pore size was immersed into laboratory-scale anaerobic digestion system. The pore size of membrane was varied from 0.03 to $\0.15mu$m. The hydraulic retention time of anaerobic digester was varied from 1.5 to 10 days. The effect of hydraulic retention time on treatment efficiency was significant while effect of membrane size was not significant. The gas production was about 0.74㎥/kg COD treated. The COD removal efficient was about 80-95% depending on the hydraulic retention time. ii ) Crossflow ultrafiltration as post treatment to anaerobic filter. The effluent from anaerobic filter, which had a total COD in the range of 4,500-5,200 mg/L was treated by crossflow ultrafiltration units. The study conducted with different membrane pore size indicated that membrace with 1,000,000 molecular weight cut-off size gave a higher COD removal efficiency in the range of 83-87% while giving a study flux of $120-130 L/\m^2$.h.   • Landfill leachate is one of highly contaminated and heterogeneous wastewater. The leachate from initial landfill can be treated by anaerobic process because it contains biodegradable matters, particularly, volatile fatty acids (VFAs). However, the anaerobic treatment of leachate is generally required longer hydraulic retention time (HRT) than aerobic process and another treatment process to satisfy effluent concentration. Therefore the modification of conventional anaerobic treatment is needed. Two phase anaerobic membrane process (TPAMP) is an integrated membrane process to be able to separate anaerobic metabolism into two phase which are acidogenesis and methanogenesis for improvement of anaerobic treatment efficiency. In this study, the efficiency of TPAMP and conventional anaerobic treatment were compared in terms of HRT, effluent SCOD, VFAs Membrane used in TPAMP was the UF of capillary type with the surface area of $0.048m^2$. The average effluent SCOD of conventional anaerobic treatment was 1352 mg/L and the removal was 96 % at HRT 60 days, while in TPAMP, 927 mg/L and 98% at HRT 30 days.

Correlation between Uni-standing Balance Ability, Jumping Ability, and Anaerobic Lower Extremity Muscle Power of High School Taekwondo Players

  • Objective: This study analyzed the correlation between uni-standing balance ability, jumping ability, and anaerobic lower extremity power of high school Taekwondo players. Design: Cross-sectional study Methods: Two high schools recruited 36 taekwondo players, but 5 were dropped due to personal circumstances and preparations for the tournament, so the number was counted as 31. In order to test uni-standing balance ability, the Romberg test was performed using Biorescue to measure the sway area. In order to test jumping ability, standing long jump and standing highjump were measured. For anaerobic lower extremity muscle power test, the peak power and average power were measured through 30-second pedaling using a Wingate system bicycle. Results: Participants did not show any correlation with other measures in the uni-standing balance ability test (p>0.05). Looking at the correlation in the standing long jump, standing high jump (r=0.892, p<0.05), anaerobic peak power (r=0.698, p<0.05), anaerobic average leg power (r=0.662, p<0.05) showed a positive correlation. Looking at the correlations in the standing high jump, anaerobic peak power (r=0.663, p<0.05), anaerobic average power (r=0.697, p<0.05) showed a positive correlation. Looking at the correlation in the anaerobic peak power, average power (r=0.785, p<0.05) showed a positive correlation. Conclusions: This study shows that balance ability and anaerobic leg strength are independent of each other. The results of this study can be used as basic data for the training plan of high school taekwondo players in the future.

Comparison of Liquid Composting Efficiency using Liquid Pig Manure in Different Condition (가축분뇨 슬러리 액비 부숙 조건별 특성비교)

  • This study was conducted to evaluate the liquid composting efficiency using pig manure in different condition such as simple storage type reactor, continuous aeration reactor, anaerobic reactor and anaerobic agitation reactor. Continuous aeration reactor was the most efficiency method to BOD and malodors removal than other methods. While nitrogen loss in continuous aeration reactor was 47% of initial concentration, which was the higher amount than any other digestion methods. The digestion efficiency between anaerobic reactor and anaerobic agitation reactor were similar, but E. coli and malodor removal efficiency were a little higher in the anaerobic agitation reactor. Simple storage type reactor which was conventional digestion method in rural area gave lower efficiency than aerobic and anaerobic digestion methods in view of BOD, E. coli and malodor removal. The liquid composting efficiency which were evaluated by various indicators like pH, BOD, E, coli, malodor and nitrogen loss was high in the order of anaerobic agitation reactor>continuous aeration reactor>anaerobic reactor>simple storage type reactor.

  • The purpose of this paper is to study on the thermal death time curve and F-values, and morphological and physiological characteristics observed for the identification. The three strains of thermal resistancing anaerobic bacteria isolated from unheated various cans and swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the manual of Bergeg's for the identification of the anareobic bacteria have been shown that the three strains of anaerobic bacteria are pertained to Cl. sporogenes B-41 Cl. butyricum B-72 & Cl.. botulinum Type E B-163. The optimum temperature, pH and thermal resistance, thermal death point of the anaerobic have been measured.

Treating Swine Wastewater by Anaerobic Bioreactors (혐기성 생물반응기에 의한 축산폐수의 처리)

  • Three different types of lab-scale anaerobic bioreactors, AF and two-stage ASBF-PR and ASBF-SP, were evaluated in treating swine wastewater by operating at $1{\sim}2$ days of hydraulic retention time with increasing organic loading rate upto 6.3 $kg-COD/m^3{\cdot}d$ at $35^{\circ}C$. Seeding the anaerobic bioreactors with waste anaerobic digester sludge from a municipal wastewater treatment plant was effective and a 40-day acclimation period was required for steady-state operation. Three anaerobic bioreactors were effective in treating swine wastewater with COD removal efficiency of $66.4{\sim}84.9$% and biogas production rate of $0.333{\sim}0.796m^3/kg-COD_{removed}{\cdot}d$. Increases of organic loading rate by increasing influent COD concentration and/or decreasing hydraulic retention time caused decreases in COD removal efficiency and increases in biogas production rate. At relatively high organic loading rate employed in this study, the treatment efficiency of AF and ASBF-PR were similar but superior than that of ASBF-SP, indicating that porosity and pore size of the media packed in the bioreactors are more important factors contributing the performance of to bioreactors than specific surface area of the media. TKN in swine wastewater must be removed prior to the anaerobic processes when anaerobic process is considered as a major treatment process since influent TKN concentration of $1,540{\sim}1,870mg/L$ to the bioreactors adversely affect the activity of methanogenic bacteria, resulting in decreases of treatment efficiency and biogas production rate by 50%.

Anaerobic Degradation of cis-1,2-Dichloroethylene by Cultures Enriched from a Landfill Leachate Sediment

  • The production of microbiologically enriched cultures that degrade cis- 1,2-dichloroethylene(DCE) under anaerobic conditions was investigated. Among 80 environmental samples, 19 displayed significant degradation of $10{\mu}M$ cis-DCE during 1 month of anaerobic incubation, and one sediment sample collected at a landfill area (Nanji-do, Seoul, Korea) showed the greatest degradation ($94\%$). When this sediment culture was subcultured repeatedly, the ability to degrade cis-DCE gradually decreased. However, under Fe(III)-reducing conditions, cis-DCE degradation by the subculture was found to be maintained effectively. In the Fe(III)-reducing subculture, vinyl chloride (VC) was also degraded at the same extent as cis-DCE No accumulation of VC during the cis-DCE degradation was observed. Thus, Fe(III)-reducing microbes might be involved in the anaerobic degradation of the chlorinated ethenes. However, the subcultures established with Fe(III) could function even in the absence of Fe(III), showing that the degradation of cis-DCE and VC was not directly coupled with the Fe(III) reduction. Consequently, the two series of enrichment cultures could not be obtained that degrade both cis-DCE and VC in the presence or absence of Fe(III). Considering the lack of VC accumulation, both cultures reported herein may involve interesting mechanism(s) for the microbial remediation of environments contaminated with chlorinated ethenes. A number of fermentative reducers (microbes) which are known to reduce Fe(III) during their anaerobic growth are potential candidates involved in cir-DCE degradation in the presence and absence of Fe(III).
