Effects of Fucoidan on Neuronal Cell Proliferation: Association with NO Production through the iNOS Pathway

  • Fucoidan, that is high-molecular-weight sulfated polysaccharides extracted from brown seaweeds has been shown to elicit various biological activities. Here, we investigated the effects of fucoidan on cell proliferation and nitric oxide (NO) production in neuronal blastoma cell (SH-SY5Y). In the present study, we demonstrated that fucoidan treatment resulted in increase of cell proliferation and NO production. When cells were treated with amyloid-${\beta}$ (A${\beta}$) in the absence or presence of fucoidan, fucoidan recovered the cell viability decreased by A${\beta}$ peptides. To further determine whether nitric oxide synthase (NOS) is involved in proliferative effect of fucoidan, cells were treated with NOS inhibitors in the absence or presence of fucoidan. Selective constitutive nitric oxide synthase (cNOS) inhibitor, diphenylene iodonium chloride (DPI), caused a decrease of cell viability, whereas cell viability was increased by specific inducible nitric oxide synthase (iNOS) inhibitor, S-methylisothiourea (SMT), in the fucoidan-untreated cells. Treatment with fucoidan inhibited the cell viability decreased in DPI-exposed cells. In contrast, fucoidan had no effect on cell growth in SMT-treated cells, indicating that cNOS may not play a role in the proliferation of fucoidan-treated cells. The present data suggest that fucoidan has proliferative and neuroprotective effects and these effects may be associated with iNOS.

양파(Allium cepa L.) 음료의 콜린성 활성 증가 및 뇌신경세포 보호로 인한 Amyloid β Peptide 유도에 대한 인지장애 개선 효과 (Onion Beverages Improve Amyloid β Peptide-Induced Cognitive Defects via Up-Regulation of Cholinergic Activity and Neuroprotection)

  • 본 연구는 양파의 불쾌치를 저감화시킨 무취음료와 양파 과피 추출물을 첨가한 생리활성 성분 강화음료의 $H_2O_2$로 유도된 산화적 스트레스에 대한 뇌신경세포 보호 효과와 $A{\beta}$로 유도된 인지기능 장애 동물모델에서의 개선 효과를 검증하고자 수행되었다. 뇌신경세포 보호 효과에서는 상대적으로 강화음료에서 무취음료 대비 우수한 산화적 스트레스 억제효과 및 생존율을 나타내었다. $A{\beta}$로 유도된 인지기능 장애 동물모델에 있어 Y-maze, passive avoidance 및 Morris water maze test에서 강화음료가 상대적으로 우수한 학습 및 기억력 개선 효과를 나타내는 것을 확인할 수 있었다. 마우스의 뇌 조직에서 강화음료 그룹은 AChE 활성을 저해하고, 신경전달물질인 ACh의 함량을 증가시킴으로써 $A{\beta}$로 유도된 cholinergic system 장애에 있어 개선 효과를 나타내었다. 또한, 마우스 뇌에서 SOD 함량의 증가, oxidized GSH/total GSH와 MDA 함량을 감소시킴으로써 $A{\beta}$와 같은 산화적 스트레스 인자에 대한 뛰어난 항산화 효과를 나타내었다. 최종적으로 무취음료와 강화음료의 주요성분들을 Q-TOF UPLC/MS system을 통하여 분석한 결과, 강화음료의 경우 무취음료보다 생리활성을 가진 2개의 steroidal saponin과 6개의 phenolic 화합물 등이 추가 검출되었다. 이러한 결과들을 종합해볼 때 강화음료는 상대적으로 protocatechuic acid와 quercetin 같은 강력한 항산화 효과를 나타내는 phenolic 화합물과 steroidal saponin 계열에 의한 우수한 인지기능 개선 효과를 기반으로 한 고부가가치 식품으로 활용될 수 있는 산업적 가능성이 있다고 판단된다.

  • ${\beta}2$-Microglobulin (${\beta}2m$) is known to be a major factor for dialysis-related amyloidosis. We have studied ${\beta}2m$ removal through an aggregation process, which was initiated by a ligand that could catch the ${\beta}2m$ monomer and promote its aggregation into fibril. As a ligand, we have prepared ${\beta}2m$ fibril fragments and used them as a seed. The seed was coupled to PEGylated-PS beads to remove the monomeric ${\beta}2m$ from solution. The ${\beta}2m$ seed-conjugated resin effectively adsorbed the ${\beta}2m$ monomers with a capacity of 3.6 mg/ml via promoting their aggregation into fibrils on the resin at pH 7.4.

  • $A{\beta}_{40}$ peptides form oligomers that later aggregate into a plaque, which is deemed to be a leading cause of Alzheimer's Disease. Its non-crystalline morphology has limited an understanding of comprehensive structural study. In this research, computational biomolecular simulations were performed in the following order: solvent and ion addition in a box, energy minimization of protein, equilibration, and periodic boundary condition disaggregation of a monomer from fibril. The result founded the two-fold model is 25% more stable in the simulation environment, and the steric zippers held on most tightly until 220 ps of simulation. The study supports the previous findings that two-fold aggregate $A{\beta}_{40}$ is more stable at 310 K and discusses further how much contribution steric-zipper and hydrogen bonding are making.

  • We investigated whether the mushroom extracts can protect neuronal death and ameliorate memory deficits in Alzheimer"s disease induced by $\beta$-amyloid peptide[A$\beta$(25-35)]. Cellular model of Alzheimer"s disease was produced by using SK-N-SH human neuronal cells treated with $A\beta$. Treatment with 40uM $A\beta$ for 48hours caused a 46% loss of cell viability. First, we examined the effects of 22 mushroom extracts on neuronal death using MTT assay. We found that 3 mushroom extracts increased viability of the cells from 46% to 87%. (omitted)

ALDH2 효소 활성과 8주간 에탄올 노출에 따른 해마조직의 아밀로이드 베타 발현 (Amyloid-β Levels in Mice Hippocampus According to the ALDH2 Enzyme Activity followed Ethanol Exposure for 8-Weeks)

  • 알츠하이머 질환은 인지능력과 행동능력 두 가지에 모두 영향을 미치는 진행적 노인성 치매증의 일종으로 정확한 발병기전은 아직 알려져 있지 않으나 환경적 요인 및 유전적 요인이 모두 중요한 위험인자로 알려져 있다. 본 연구에서는 8주간 에탄올에 노출된 Aldh2 knockout mouse 뇌조직을 분리하여 알츠하이머 질환의 지표로 잘 알려진 아밀로이드 베타와 NF-kB 발현을 평가하고 이러한 변화가 ALDH2 효소의 활성에 따라 어떻게 달라지는 지 비교하였다. 그 결과, 8주간 에탄올을 경구 투여한 마우스에서 ALDH2 효소의 활성에 따른 NF-kB 농도의 차이는 확인할 수 없었으나, ALDH2 효소 활성은 알코올 노출에 의한 해마조직의 아밀로이드 베타 축적에서 중요한 영향을 미치는 것으로 나타났다(p<0.05). 이러한 결과는 ALDH2 효소가 결핍된 사람이 결핍되지 않은 사람에 비해 알코올에 의한 알츠하이머 질환의 발생에 보다 민감할 수 있음을 시사한다.

Computational Study of Human Calcitonin (hCT) Oligomer

  • We have performed long time REMD simulation on 15-19 residues of human calcitonin hormone (DFNKF) which is known to form highly ordered amyloid fibril. The simulation started from randomly oriented multiple DFNKF strand. Using all-atom level simulations with the generalized Born solvation (GB) model (param99MOD3), we observed spontaneous formation of ${\beta}$-sheet for tetramer. Interestingly, the current simulation gives anti-parallel sheet as a major conformation, consistent with experiments. The major interaction stabilizing the anti-parallel sheet seems to be the inter-strand hydrogen bond.

Structure of CT16 in the C-terminal of Amyloid Precursor Protein Studied by NMR Spectroscopy

  • C-terminal fragments of APP (APP-CTs), that contain complete Abeta sequence, are found in neuritic plaques, neurofibrillary tangles and the cytosol of lymphoblastoid cells obtained from AD patients. CT16, Lys649-Asp664 (KKQYTSIHHGVVEVD) has been known as the most toxic part in the C-terminal fragment of amyloid precursor protein (APP). The solution structure of CT16 was investigated using NMR spectroscopy in various membrane-mimicking environments. According to Circular Dichroim (CD) spectra, CT16 has a random structure in aqueous solution, while conformational change was induced by addition of TFE and SDS micelle. Tertiary structure as determined by NMR spectroscopy shows that CT16 has a ${\beta}$-turn conformation in trifluoroethanol-containing aqueous solution.

원지와 석창포 혼합추출액의 pCT105로 유도된 신경세포암 세포주에 대한 항치매 효과 (The Effects of anti-Alzheimer in pCT105-induced Neuroblastoma cell lines by Radix Polygalae and Rhizoma Acori Graminei mixture extract)

  • Numerous lines of evidence indicate that some of the neurotoxicity associated with Alzheimer's disease (AD) is due to proteolytic fragments of the amyloid precursor protein (APP). Most research has focused on the amyloid 6 (M). However, the possible role of other cleaved products of APP is less clear. Lately It has been reported that a recombinant carboxy-terminal 105 amino acid fragment (CT105) of APP induced strong nonselective inward currents in Xenopus oocyte. In a brain with Alzheimer's disease (AD), to investigate the roles of carboxyl-terminal fragment (CT105) of amyloid precursor protein (APP) in apoptosis processes possibly linked to neurodegeneration associated with AD, we examined the effects of the CT of APP with 105 amino acid residues (CT105) on the alteration of apoptosis triggers in neubroblastoma cells. We have investigated whether Radix Polygalae and Rhizoma Acori Graminei mixture extract (RP+RAG) inhibits CT105-induced apoptosis of neuroblastoma cells. We found that RP+RAG inhibits CT105-induced apoptosis in SK-N-SH cells. Treatment of the cells with RP+RAG inhibited CT105-induced DNA fragmentation and Tunel assay of nuclear chromatin and inhibited the caspase-3 expression in SK-N-SH cells. As the result of this study, In RP+RAG group, the apoptosis in the nervous system is inhibited, the repair against the degerneration of neuroblastoma cells by CT105 expression is promoted. These results indicate that RP+RAG possess strong inhibitory effect of apoptosis in the nervous system and repair effect against the degeneration of neuroblastoma cells by CT105 expression
