Population Viability Analysis of a Gold-spotted Pond Frog (Rana chosenica) Population: Implications for Effective Conservation and Re-introduction (금개구리 (Rana chosenica) 개체군의 생존분석: 개체군의 효과적인 보존과 야생복귀를 위한 제안)

  • Population viability analysis of a Gold-spotted pond frog (Rana chosenica) population at Cheongwon-gun, Chungbuk, in South Korea was conducted and we proposed several suggestions for effective conservation and re-introduction of the species. Simulating a developed model over 1,000 times predicted that the population will exist over 30 years with a relatively low growth rate of 0.113, but with a high probability of extinction as 81.1%. Population growth and extinction probability were the most greatly depended on the rate of successful metamorphosis. In the case of outbreak of amphibian diseases such as Chytridiomycosis and Ranavirus, the population will be easily extinct within 4 years with 100% probability. In a habitat of which carrying capacity is 200, to successfully re-introduce an extinct population, it is initially needed to put 100 individuals of which 83% is males and its age structure is normal-distributed. If we additionally conducts artificial supplementation of 10% individuals every 2 years from 4 years to 10 years after initial reintroduction, the population will become a stable with 0.297 growth rate and 0.290 extinction rate. Our results are the first case of amphibian population viability analysis in Korea and could be used to develop effective conservation and re-introduction plans for endangered Gold-spotted pond frog.

Ultrastructure of the Follicular Oocyte Surface in Rana dybowskii

  • Rana ovarian follicles consist of oocyte, vitelline envelope, granulosa cells, and theca/epithelial layer. Using scanning electron microscopy, the surface structure of each follicular component was investigated. Changes in oocyte surface during oocyte maturation were also examined. Theca/epithelial layer was almost transparent and some blood vessels and granulosa cells were observed underneath in intact follicle. The number of granulosa cells was estimated to be 6700-7200 per oocyte. The granulosa cells partially overlapped each other and their microvilli penetrated the vitelline membrane via holes present in the vitelline envelope and seemed to be linked to oocyte microvilli. After removal of the vitelline envelope by microforcep, oocyte microvilli were observed on the surface of the devitellined oocyte. The oocyte microvilli formed partial clusters on the surface of white spot area which appears iust before germinal vesicle breakdown (GVBD), whereas they were evenly distributed in other areas. The microvilli became shorter and less dense with oocyte maturation. The lengths of oocyte microvilli in the immature and mature oocyte were 1.5 $\mu$m and 0.6 $\mu$m, respectively. The present study suggests a fundamental structural change occurring on the oocyte surface during maturation.

Follow-up Monitoring & Adaptive Management after Ecological Restoration for the Stream - Focused the Hakui Stream in Anyang City - (생태하천 복원 후 모니터링과 적응관리 - 안양시 학의천을 중심으로 -)

  • Recent years, nationwide projects for ecological restoration are implemented with emerging issues on the stream ecosystem. In order to enhance effectiveness of the ecosystem restoration and reduce negative impact, the appraisal of effectiveness through the follow-up monitoring and the adaptive management process are executed in consecutive phase. In this study, planning phase, monitoring and adaptive management in Hakui stream which is part of An Yang stream restoration project is introduced as representative ongoing case of effective adaptive management. The aim of this study is to verify the adaptive management process and suggest direction of effective restoration. Restoration project of Hakui stream resulted in increasing number and diversity of species (vegetation, fish, bird, invertbrates, amphibian and reptilia) according to monitoring from 2004 to 2013, and enhancing natural river landscape by evaluation of river naturalness among 2001(before restoration), 2007 (after), 2015 (recent). However, excessive vegetation expansion or sediment deposition on channel over time caused unexpected results such as terrestrialization or degradation of habitats. Adaptive management action such as removing disturbance species (Humulus japonicus)(2007), coppicing willow (2007), release of march snail (2007), creation of wetland (2014) were implemented based on monitoring results. And then appraisal of management action was discussed.

NMR Spectroscopic Assessment of the Structure and Dynamic Properties of an Amphibian Antimicrobial Peptide (Gaegurin 4) Bound to SDS Micelles

  • The structure and dynamics of a 37-residue antimicrobial peptide gaegurin 4 (GGN4) isolated from the skin of the native Korean frog, Rana rugosa, was determined in SDS micelles by NMR spectroscopy. The solution structure of the peptide in SDS micelles was determined from 352 NOE-derived distance constraints and 22 backbone torsion angle constraints. Dynamic properties for the amide backbone were characterized by $^1H-^{15}N $heteronuclear NOE experiments. The structural study revealed two amphipathic helices spanning residues 2-10 and 16-32 and that the helices were connected by a flexible loop. An intraresidue disulfide bridge was formed between residues Cys31 and Cys37 near the C-terminus. The loop region (11-15) connecting the two helices are were slightly more flexible than these helices themselves. From the fact that since there is no contact NOEs between two helices, it is implied that the GGN4 peptide shows an independent motion of both helices which has an angle of about $ 60^{\circ}-120^{\circ}$ from each other.

  • A new trichostome ciliate, Balantidium honghuensis n. sp., was isolated from the recta of Rana nigromaculata and R. limnocharis during parasite surveys in Honghu Lake, Hubei Province, central China in summmer of 2010 and 2011. Its detailed morphometric characters based on LM and SEM studies were described herein. The organism is oval in shape and thickly ciliated. The vestibulum is "V" shaped and occupies about 1/3 to 2/5 of the body length. The vestibular and nearby regions possess strong peripheral fibers which form a marked axial fiber about the cytopharynx. More than 10 contractile vacuoles are distributed along the periphery of the latter body. Comparisions were made between this new species and B. sinensis Nie, 1935. They were discriminated from each other in terms of general body forms, body size, and vestibulum shapes. Besides, special attention was paid to its high-speed daughter swarmers which we believed to be the infective stage of B. honghuensis. Possible infection routes of anura amphibian balantidia were discussed.

  • Agriculture, rural landscapes are accompanied by a variety of environmental issues. Therefore, it is necessary to study on biodiversity and ecosystem services. Horticulture complex is low groundwater recharge function, it can be evaluated as a facility that biodiversity is impaired. The ecosystem services in agricultural landscapes were obtained 19 kinds of functions. Experts survey Groundwater recharge function (4.13) teeth chapter higher, Water storage (4.05), Amphibian & Reptile habitat (3.96), Aquatic insect habitat (3.92), Flood control (3.87), Water purification (3.86), Avian habitat (3.76 ), Creating landscape (3.74), Vegetation diversity (3.71), Experience, Education (3.69), Biological control (3.48), Fishery habitat (3.42), Climate regulation (3.30), Mammal habitat (3.30), Air quality regulation (3.25 ), Mainenance of genetic diversity (3.25), were analyzed in order Rest area (3.14). Improving capabilities in the Detention Pond, Wetland, Green space, Corridor, Non-Chemical, Program development, Green spaces, Rainwater storage facilities, Water cycle system, Surface water storage facilities, Infiltration trench, Water purification facilities, Permeable pavement. Environmentally friendly, and to contribute to sustainable agricultural development through ecological planning.

Determining widths of riparian ecosystem zone for water quality and ecosystem conservation - A case study for the Jinwee stream (수질개선과 생태서식환경을 고려한 수변생태구역 너비 결정 방법 - 진위천 적용을 중심으로 -)

  • Riparian management has become important as stream water quality as well as riparian ecosystem gain more public attentions. The objective of this study was to determine riparian widths based on the functions of nutrient removal and wildlife habitat protection and to apply for the Jinwee stream area as a preliminary case. Nitrogen and phosphorus filtration efficiencies were considered in water quality aspect, while the habitat radii of amphibian and reptiles were used for wildlife conservation purpose. In addition, observation of endangered species and human impact on wildlife disturbance in riparian area were also taken into account in determining riparian widths. The stream confluence zone was emphasized by doubling the riparian widths as the focal point for wildlife habitat conservation. As the results, three different levels of riparian widths were proposed depending on the major riparian functions and applied to the Jinwee stream section as the case study. The proposed method can be used to determine riparian width in other stream areas based on different functional focus, ie, water quality or riparian conservation purposes.

The effect of Novel gravitational fields on early Amphibian Morphogenesis (중력장의 변화가 양서류 초기 발생에 미치는 영향)

  • 본 연구에서는 horizontal clinostat과 원심분리 방법의 이용으로 각각 저중력 상태($\mu$G)와 고중력 상태(3G)를 유도하여, 중력장의 변화가 양서류 수정란의 초기 형태 형성에 미치는 영향을 북방산개구리(Rano dybows비름를 대상으로 조사하였다. 중력장의 변화는 첫번째 수평 분열인 제3분열면의 위치에 영향을 미쳤다. 즉 저중력 상태에서는 분열면이 적도면 근처에서, 또 고중력 상태에서는 분열면이 동물극 근처에서 형성되었다. 이와 같은 변화는 포배기로 이어져 할강의 위치와 동물극 정단 세포층 수에 영향을 미쳤다. 즉 저중력에 노출된 포배는 정상배(1G)에 비해 할강이 중앙에 위치했으며 동물극 정단의 세포층 수가 증가하는 반면 고중력에 노출된 포배는 할강이 보다 동물극 폭에 위치했으며 동물극 정단의 세포층 수도 감소하였다. 그러나 낭배기에 도달하면 동물극 정단 세포층 수의 차이는 정상 상태로 조절되었다. 한편 중력장의 변화는 낭배기에 이르는 발생 속도와 원구상순부의 위치를 변경시켰다. 낭배기 이후의 발생 단계에서 중력에 의한 변화로는 tail fin circulation(stage 22)시기의 머리크기의 변화를 들 수 있다. 이상의 결과로 무미 양서류의 초기 형태 형성은 중력장에 의해 변형될 수 있으며 이러한 변화는 발생이 진행됨에 따라 조절되는 것으로 보인다.

Developmental Lesions in Amphibian Embryos Induced by Ultraviolet Irradiation of the Fertile Egg (자외선에 의한 양서류 수정난의 발생 결함)

  • Ultraviolet irradiation of the vegetal hemisphere of the fertilized frog egg prior to first cleavage resulted in alterations in neural morphogenesis. The sensitivity to UV irradiation dropped drastically at the point of 2/3 time lapse between fertilization and the appearance of the first cleavage furrow. Scannining electron micrographs revealed a decrease in size of the cells on the surface of the irradiated embryos. However, ectoderm exchange frafts indicated that the ectoderm of the irradiated embryo retains its capaciiy to form a completely normal neural morphology and epidermal surface. These facts were interpreted in terms of a decrease in the capacity for invagination during gastrulation, and subsequent primary embryonic induction.

  • The comparative karyological analysis of the Korean treefrogs, Hyla japonica and Hyla suweonensis, were performed by C-banding method. Heteromorphic sex chromosomes, female heterogamety, has been identified in the 3rd chromosomes of H. suweonensis H. suweonensis seem to have sex chromosomes which are Zz/ZW type. The Z chromosomes contain large amount of constitutive heterochromatin, but little heterochromatin is located in the W chromosomes. This is in contrast to all previously known amphibian and most other vertebrate's W or Y chromosomes, except Gastrotheca oui'ern and G. walken (Schmid et al., 1988).
