Plant Regeneration from Unpollinated Ovary Culture in Allium tuberosum Rottl (부추의 미수분 자방배양에 의한 식물체 재분화)

  • This study was carried out to determine the optimal conditions for the production of plants derived from the unpollinated ovary culture of Chinese chive (Allium tuberosum Rottl.). The Chinese chive collected from Korea showed much higher frequency of plantlet formation than those from Japan in the culture of unpollinated ovaries. Among the collections, 'Youngiljaerae' showed the highest frequency of plantlet formation. The MS basal medium was superior to B/sub 5/ in plantlet formation. The ovaries inoculated on the 2,4-D-free medium were directly induced plantlets without callus formation. Floral parts inoculated as a unit played important roles in callus formation and plant production. The frequency of callus and plantlet formation was higher in the culture of ovary with anthers than that of ovary alone.

Physical and Sensory Characteristics of Cookies with Added Leek(Allium tuberosum Rottler) Powder (부추 분말을 첨가한 쿠키의 물리적, 관능적 특성 연구)

  • In this study, we assessed the physicochemical and sensory characteristics of cookies containing various concentrations (0, 3, 5, 7 and 9%) of leek powder. The L values of cookies containing leek powder were significantly lower than those of the controls. The spread ratio decreased proportionally with the quantity of leek powder added to the cookie recipe. The loss rate and leavening rate of samples to which 3% and 5% leek powder was added evidenced a higher value than the samples to which 7% and 9% leek powder was added. The L value of brightness was significantly reduced when leek powder was added to the cookie recipe. The a and b values were also significantly reduced with the addition of leek powder. Hardness evidenced significantly high levels in the samples to which 3% and 5% leek powder was added. The results of our sensory evaluation evidenced significantly better appearance, flavor, color, texture and overall acceptability as compared to the 3% and 5% added samples. The observed quality characteristics of the 3% leek powder added samples and 5% leek powder added samples indicated the possibility that leek cookies might be developed.

A Comparative Study of the Changes in Volatile Flavor Compounds from Dried Leeks (Allium tuberosum R.) following ${\gamma}$-Irradiation

  • This study was performed to examine the effects of ${\gamma}$-irradiation on the volatile flavor compounds of dried leeks (Alliums tuberosum R.). Volatile compounds of dried leeks were extracted using simultaneous steam distillation and extraction (SDE), and analyzed by gas chromatography/mass spectrometry (GC/MS). Forty-one, 51, 45, and 42 compounds were tentatively identified in control, 1, 3, and 10 kGy irradiated samples, respectively. The constituents of flavor compounds in irradiated dried leeks were similar to non-irradiated samples. However, the intensities of the peaks were clearly different between them. Sulfur-containing compounds were detected as dominant compounds in all samples and their amounts decreased after ${\gamma}$-irradiation. ${\gamma}$-Irradiation reduced the total concentration of volatile compounds from leeks by 23.19, 15.09, and 30.23% at 1, 3, and 10 kGy doses, respectively.

  • Volatile compounds of $\gamma-irradiation$ leeks (Aillum tuberosum R.) were isolated by simultaneous steam distillation and extraction method (SDE) and analyzed by gas chromatography/mass spectrometry (GC/MS) along with the non-irradiated sample. Leeks were irradiated at dose of 20 kGy and non-irradiated samples were considered as control sample. 56 and 54 compounds were identified in non-irradiated and irradiated samples, respectively. These components included the functional groups; alcohols, aldehydes, esters, ketones, N- and S-containing compounds. S-containing compounds such as dimethyl trisulfide, methyl-(Z)-l-proenyl trisulfide, dimethyl disulfide, 3-ethyl-l,2-dithi-5-ene and methyl allyl disulfide were the major compounds. Methyl-(Z)-l-propenyl trisulfide, diallyl trisulfide, (E)-2-hexanal and 3-ethyl-l,2-dithi-5-ene were clearly decreased by the $\gamma-irradiation$, but dimethyl disulfide, diallyl sulfide, allyl methyl sulfide and pyridine were increased by the $\gamma-irradiation$.

Insecticidal activity of native plant extracts against Culix pipiens pallens and Musca domestica (자생식물 추출물의 모기 및 집파리에 대한 살충활성)

  • Methanol extracts of 9 kinds of native medicinal plants(Taraxacum platycarpum, leaf; Pinus densiflora, leaf; Artemisia prinseps, leaf; Allium tuberosum, leaf; Cassia obtussifolia, whole; Sophora angestifolia, root; Stemonae sessilifolia, root; Lonicera japonica stem, leaf, flower; and Clivia miniata) were investigated for insecticidal effect. Methanol extracts of Pinus densiflora leaves and Sophora angestifolia radix showed relatively good insecticidal activity against Culex pipiens pallens larvae. Strong larvicidal activity against the Musca domestica larvae was produced from methanol extracts of Taraxacum platycarpum leaves and Allium tuberosom leaves. while Stemonae radix showed moderate activity. All materials tested revealed little or weak insecticidal activity against M. domestica adults.

  • In counting chromosome number and karyotype study, it is necessary to let chromosomes on metaphase by pretreatment before fixation. For this purpose, colchicine, or 8-oxyquinoline are generally used. The author found out that chromosomes could be scattered by illuminating cyanoferrate complex solution in which root-tips were sunk. As materials, 8 sorts of plant such as Allium fisturosum, allium tuberosum Rottler, Triticum vulgare were used. Their root-tips were sunk on the bottom of beaker in potassium ferricyanide solution $3{\times}10-4M$ and illuminated through the solution by sterilizing lamp for 1~2 hours in dark room, keeping 10 cm distance from light source to the surface of solution and 2cm depth of solution. Then again, they were illuminated to the light which was somewhat weaker intensity than the former (distance, 16cm; depth, 3cm) for 1.5~2 hours after immersed in 1/100N-HCl and washed in water for each 5minutes. By such methods chromosomes could be scattered. About the mechanism of scattering, it is supposed that CN and Fe(CN)x ions $(x {\leq}5)$ which were gradually produced in the process of photodissociation acted together on the scattering of chromosomes.

  • Between February 2002 and June 2003, A clinical study on Noctural Enurosis Patients had been conducted by the Department of Internal medicine at Dongshin University Oriental hospital. In this study, we treated nine patients with ear acupuncture using seed of Allium tuberosum ROTTLER and Herbal medicine. And we got the efficacious results. Moreover, the patients who were treated for more than 20days show more efficient result.

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

Growth Retardation of Escherichia coli and Staphylococcus aureus by Leek Extract (부추 추출무에 의한 Escherichia coli 및 Staphylococcus aureus의 생육 저해효과)

  • The growth retardation of Escherichia coli and Staphylococcus aureus by heat or acid treated leek (Allium tuberosum) extract was observed. Antimicrobial activity of the leek was the most effective when fresh leek extract was used, but it was stable after heat treatment at 68$^{\circ}C$ for 30 min or 98$^{\circ}C$ for 20 min. It was also relatively stable after incubated at pH 2.0 for 3 hrs. The antimicrobial activity in leek was not detected after dialysis with molecular weight cutoff of 12,000. Therefore it seems to be small molecule with molecular weight lower than 12,000.

Leaf Blight of Chinese Chive Caused by Stemphylium vesicarium in Korea

  • A leaf spot disease occurred on Allium tuberosum Roth. (Chinese chive) in Korea. All the isolates of Stemphylium sp. from the lesions of the diseased plant parts were identified as S. vesicarium (Waller.) Simmons, based on the morphological characteristics of conidia and conidiophores. Pathogenicity of the fungus was proved by artificial inoculation on Chinese chive plants. This is the first record of leaf blight on Chinese chive caused by S. vesicarium in Korea.
